Amantadine in chronic granulomatous disease.

Chronic granulomatous disease (CGD) is a rare genetically determined immunodeficiency. Neutrophils from CGD patients show a defective killing of phagocytosed fungi and bacteria, due not only to an impairment in oxidative burst, but also to absence of normal pH value within phagocytic vacuole following phagocytosis. Because a weak base such as amantadine could potentially reverse these pH abnormalities, the authors used this drug to treat 2 CGD patients. They observed modifications of both phagosomal pH and killing activity on their neutrophils compared to those of healthy controls. Since the d…

Acute psychosis after amantadine overdose.

Amantadine is an antiviral agent that is also used in the treatment of parkinsonism and neuroleptic-induced extrapyramidal symptoms. Toxic effects of amantadine relate primarily to the central nervous system and range from mild symptoms to disorientation and hallucinations. Anti-cholinergic agents may exacerbate these effects. We report a case of unsuspected amantadine overdose in a previously healthy 35-year-old woman who presented with acute psychosis manifested by delirium and visual hallucinations. Concomitant use of diphenhydramine contributed to the clinical presentation. Amantadine toxi…

[Treating treatment resistant catatonia with amantadine].

Catatonia is one of the most severe psychiatric syndromes, and clinical symptoms and etiology are very heterogeneous. When accompanied by au … to both lorazepam and ECT. We performed an experimental treatment with the NMDA-receptor antagonist amantadine. This treatment-strategy seemed promising, but remission was short lived. Recovery occurred with high-dose clozapine.

Amantadine-induced livedo reticularis--Case report.

Livedo reticularis is a spastic-anatomical condition of the small vessels which translates morphologically by a reticular pattern, interspersing cyanosis, pallor and erythema. The same can be congenital or acquired. Among the acquired, we highlight the physiological livedo reticularis and the idiopathic livedo by vasospasm; the latter configures the most common cause. The drug-induced type is less common. The drugs amantadine and norepinephrine are often implicated. Cyanosis is usually reversible if the causative factor is removed, however, with chronicity, the vessels may become permanently d…

Creutzfeldt-Jakob disease treated with amantadine.

A case of Creutzfeldt-Jakob disease treated with amantadine showed considerable initial improvement, followed by a period of almost five years during which the patient remained in a relatively stable condition until he died accidentally. The diagnosis was confirmed histologically.

Toxicokinetics and postmortem redistribution of amantadine in rats.

OBJECTIVES: Amantadine (AMD) is an antiviral and antiparkinsonian drug with a narrow therapeutic window and a recognized risk of severe intoxication. Interpretation of postmortem drug concentrations is complicated by postmortem redistribution (PMR), yet systematic toxicokinetic and multi-tissue PMR data for amantadine remain limited. METHODS: An integrated investigation of amantadine toxicokinetics and postmortem redistribution was conducted in male rats. For toxicokinetic assessment, a single oral dose of 450 mg/kg (LD50) was administered, and concentrations were quantified in blood and nine…

Amantadine withdrawal in a patient with spinocerebellar ataxia.

We report a case of a man with spinocerebellar ataxia (SCA) on high-dose amantadine who was admitted for acute on chronic dysphagia secondary to progression of SCA. Four days after oral medications were held due to patient's dysphagia, he developed fever, tachycardia and mild rigidity in extremities and became obtunded. Despite antibiotics treatment, the vitals and mental status changes persisted for 8 days. When amantadine was resumed, the patient's vital signs and encephalopathy improved within 2 days. This is among the first reports of amantadine withdrawal syndrome (AWS) in a patient witho…

Novel use of amantadine: to treat hiccups.

Persistent hiccups may have a considerable impact on general health through disturbance of diet, sleep, and mood. They can cause exhaustion, malnutrition, dehydration, wound dehiscence, and even death in extreme cases. We report a complex clinical case of intractable hiccups in a patient with cancer of the pancreas and Parkinson's disease and some of the problems encountered when attempting symptom control. We also discuss a potential therapeutic response to a novel agent, amantadine, unlicensed in the treatment of hiccups.

[An experimental study of the psychostimulant action of amantadine].

According to previous experimental and clinical data, we have tested the hypothetic psycho-stimulating activity of amantadine on experimental sensitized models. We obtained a strong action on the spontaneous sleep of rats at a dose of 10 mg/kg and a less striking action in diminishing the effects of barbitol on the righting reflex of mice and in reducing the narcosis of rats induced by mebubarbital. This action does not seem to be due to a release of catecholamine because the lesions produced by electrocoagulation in the ascending aminergic pathways or pretreatment by alpha-methylparatyrosine …

Corneal endothelial dysfunction associated with amantadine toxicity.

PURPOSE: To describe bilateral corneal endothelial dysfunction in a patient with Parkinson disease who was treated with long-term amantadine. METHODS: We employed an A-B-A-B single-subject research design that included clinical history, clinical findings, photographic images, and specular microscopic findings of ocular changes. RESULTS: A 52-year-old woman with Parkinson disease who had taken amantadine for 6 years had bilateral corneal edema for 2 months at baseline. After cessation of amantadine, the edema resolved, and the endothelial cell densities were <or=600/mm. Corneal edema recurred…
