Modern extraction technique-pressurized liquid extraction (PLE) was optimised for extraction of lycorine and galanthamine (Amaryllidaceae alkaloids) from Narcissus jonquilla 'Pipit'. Crude extracts were purified on Oasis MCX cartridges, and the alkaloids eluted with 80-100% recoveries using methanol-10% ammonia solution (3:1, v/v). Quantitative results were obtained by both HPTLC-densitometry on silica gel plates and RP-HPLC with diode array (DAD) on XTerra C(18) stationary phase. Both methods were fully validated in terms of specificity, precision (including intra- and inter-day measurements)…

Natural products have always played a significant role in the search for new drugs. One of the most relevant alkaloid-containing plant groups is the Amaryllidaceae family, a source of exclusive structures with a wide variety of pharmacological activities. The aim of this work was to determine the alkaloid composition and biological potential of an extract from the bulbs of an endemic Peruvian Amaryllidaceae species Ismene amancaes (Ker Gawl.) Herb. The alkaloid profiling was carried out by GC-MS, which revealed the presence of 13 compounds, 2 of them unidentified. The plant extract was found t

BACKGROUND: Amaryllidaceae alkaloids (AAs) are a large group of plant-specialized metabolites displaying an array of biological and pharmacological properties. Previous investigations on AA biosynthesis have revealed that all AAs share a common precursor, norbelladine, presumably synthesized by an enzyme catalyzing a Mannich reaction involving the condensation of tyramine and 3,4-dihydroxybenzaldehyde. Similar reactions have been reported. Specifically, norcoclaurine synthase (NCS) which catalyzes the condensation of dopamine and 4-hydroxyphenylacetaldehyde as the first step in benzylisoquinol…

Targeting protein translation is a promising strategy in oncology, especially for malignancies dependent on short-lived survival factors. The clinical translation inhibitor homoharringtonine (HHT) was recently discontinued in the U.S. due to poor solubility and cardiotoxicity, creating an urgent need for new leads. We investigated structurally distinct Amaryllidaceae alkaloids, narciclasine (NCS), lycorine (LYC), and haemanthamine (HAEM), in models of chronic lymphocytic leukemia (CLL) and Richter transformation (RT). Using an optimized 10 L Soxhlet extraction of 'Carlton' daffodil bulbs, an u…

In the present work the qualitative and quantitative analysis of Amaryllidaceae-type alkaloids in the aerial parts and bulbs of Galanthus reginae-olgae Orph. subsp. vernalis Kamari is presented for the first time using GC-MS analysis. The alkaloids galanthamine, lycorine, and tazettine were identified in both extracts while crinine and neronine were found only in the bulbs. The yield of alkaloid fraction from bulbs (36.8%) is very high compared to the yield from aerial parts (9.34%). Lycorine was the major component in both fractions. The antioxidant potential was determined by three complemen

Patients with Alzheimer's disease (AD) have two types of abnormal protein buildups: amyloid plaques and neurofibrillary tangles, in addition to the early synaptic dysfunction associated with the enzymes acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE). Impairment of the glutamatergic system is also crucial for neuronal survival, as it can cause synaptic dysfunction that overstimulates glutamate receptors, especially N-methyl-d-aspartate receptors (NMDARs). Another protein affecting neuronal health is glycogen synthase kinase-3 (GSK3), a widely preserved serine/threonine protein ki

RATIONALE: The bulbs of Narcissus tazetta L. var. chinensis Roem (BNTCR) has been used as an herbal medicine for the treatment of carbuncles in China. The previous phytochemical studies indicated that the main active components of BNTCR were Amarllidaceae alkaloids (AAs). However, the comprehensive chemical and metabolic profiling of AAs in BNTCR remains underexplored. Therefore, it is necessary to establish an accurate and rapid approach for chemical characterization and metabolic profiling of AAs in BNTCR.
METHODS: The structural elucidation of natural products by ultra-high performance liq

Amaryllidaceae is a well-known family for its high alkaloidal content. These alkaloids comprise a unique group of bases that have been found to occur in this family. The Amaryllidaceae alkaloids represent a large and still expanding group of isoquinoline alkaloids, the majority of which are not known to occur in any other family of plants. This article reports on the phytochemical investigation of the alkaloidal content of the flowers of clivia nobilis cultivated in Egypt which resulted in the isolation of four alkaloids; lycorine with pyrrolo{de}phenanthridine nucleus (lycorine-type) which is…

The plant family Amaryllidaceae is of provenance in the South African region which is known to harbor about a third of the global complement of around 1000 species. It has widespread usage in the traditional medicinal practices of the indigenous peoples of the region. As a consequence and given its unique alkaloid principles, its members have provided a viable platform for phytochemical based drug discovery. The medicinal potential of the family has been realized through the commercialization of galanthamine as an Alzheimer's drug due to its potent and selective inhibitory activity against the
