Attract or repel Amblyomma sculptum ticks: Screening of semiochemicals.

Amblyomma sculptum is a tick of medical-veterinary importance. Areas with high infestations need to be monitored, and parasitized hosts treated accordingly. Carbon dioxide (CO2) traps and acaricides are commonly deployed as control measures, although with some disadvantages such as high costs, challenging logistics and vertebrate intoxication. Semiochemicals have the potential to improve tick attraction to traps and monitoring devices and alleviate the burden of A. sculptum infestations. Four concentrations (10, 5, 2.5 and 1.25 %) of 13 semiochemical candidates (CO2 only at 5 % as the standard

Rickettsia rickettsii Co-feeding Transmission among Amblyomma aureolatum Ticks.

Amblyomma aureolatum ticks are vectors of Rickettsia rickettsii, the etiologic agent of Rocky Mountain spotted fever in Brazil. Maintenance of R. rickettsii in nature depends on horizontal transmission along tick generations. Although such transmission is known to occur when uninfected and infected ticks feed simultaneously on susceptible animals (co-feeding systemic transmission), we investigated co-feeding nonsystemic transmission, which was based on R. rickettsii-infected and -uninfected A. aureolatum ticks feeding simultaneously on guinea pigs immune to R. rickettsii. Our acquisition and t

Equine piroplasmosis associated with Amblyomma cajennense Ticks, Texas, USA.

We report an outbreak of equine piroplasmosis in southern Texas, USA, in 2009. Infection prevalence reached 100% in some areas (292 infected horses). Amblyomma cajennense was the predominant tick and experimentally transmitted Theileria equi to an uninfected horse. We suggest that transmission by this tick species played a role in this outbreak.

Altitudinal Assessment of Amblyomma aureolatum and Amblyomma ovale (Acari: Ixodidae), Vectors of Spotted Fever Group Rickettsiosis in the State of São Paulo, Brazil.

Amblyomma aureolatum (Pallas) and Amblyomma ovale Koch are common ectoparasites of domestic dogs in São Paulo state, southeastern Brazil, where they are vectors of distinct spotted fever group rickettsioses, one caused by Rickettsia rickettsii (transmitted by A. aureolatum), and the other caused by Rickettsia sp. strain Atlantic rainforest (transmitted by A. ovale). For the present study, we performed an altitudinal assessment of all 1992-2012 records of A. aureolatum and A. ovale retrieved from a tick collection. The municipalities with A. ovale records presented significantly (P < 0.05) lowe

Direct injection of Amblyomma americanum ticks with Cytauxzoon felis.

Cytauxzoon felis is a tick-borne hemoprotozoan parasite that causes life-threatening disease in domestic cats in the United States. Currently, the platforms for C. felis research are limited to natural or experimental infection of domestic cats. This study aims to develop an alternative model by infecting Amblyomma americanum ticks with C. felis via direct injection. Amblyomma americanum adults were injected with C. felis-infected feline erythrocytes through two routes: directly into the digestive tract through the anal pore (IA injection), or percutaneously into the tick hemocoel (IH injectio

Vectorial competence of Amblyomma tonelliae to transmit Rickettsia rickettsii.

The aim of this work was to test the vectorial competence of Amblyomma tonelliae (Ixodida: Ixodidae) to transmit Rickettsia rickettsii (Rickettsiales: Rickettsiaceae), the agent of Rocky Mountain spotted fever (RMSF). All parasitic stages of A. tonelliae were exposed to R. rickettsii by allowing each stage to feed on hosts inoculated with this pathogen. Thereafter, ticks were fed on uninfected hosts. All stages of A. tonelliae were able to acquire the R. rickettsii infection and maintain it by transstadial and transovarial transmission. When infected ticks fed on uninfected hosts, the hosts de

Experimental infection of Amblyomma aureolatum ticks with Rickettsia rickettsii.

We experimentally infected Amblyomma aureolatum ticks with the bacterium Rickettsia rickettsii, the etiologic agent of Rocky Mountain spotted fever (RMSF). These ticks are a vector for RMSF in Brazil. R. rickettsii was efficiently conserved by both transstadial maintenance and vertical (transovarial) transmission to 100% of the ticks through 4 laboratory generations. However, lower reproductive performance and survival of infected females was attributed to R. rickettsii infection. Therefore, because of the high susceptibility of A. aureolatum ticks to R. rickettsii infection, the deleterious e

Amblyomma imitator ticks as vectors of Rickettsia rickettsii, Mexico.

Real-time PCR of Amblyomma imitator tick egg masses obtained in Nuevo Leon State, Mexico, identified a Rickettsia species. Sequence analyses of 17-kD common antigen and outer membrane protein A and B gene fragments showed to it to be R. rickettsii, which suggested a potential new vector for this bacterium.

The bacterial community of the lone star tick (Amblyomma americanum).

BACKGROUND: The lone star tick (Amblyomma americanum), an important vector of a wide range of human and animal pathogens, is very common throughout the East and Midwest of the USA. Ticks are known to carry non-pathogenic bacteria that may play a role in their vector competence for pathogens. Several previous studies using the high throughput sequencing (HTS) technologies reported the commensal bacteria in a tick midgut as abundant and diverse. In contrast, in our preliminary survey of the field collected adult lone star ticks, we found the number of culturable/viable bacteria very low. METHOD

Occurrence of Amblyomma dissimile on wild crocodylians in southern Mexico.

Ticks are common ectoparasites of amphibians and reptiles but very few reports of such parasites on crocodylians exist worldwide. Herein, we report the first detailed observations of Amblyomma dissimile Koch, 1844 on the 3 crocodylian species present in Mexico, with the first report of tick parasitism on Crocodylus acutus and the second on Caiman crocodilus chiapasius. This is also the first report of A. dissimile in the state of Quintana Roo. Proportions of infested individuals found in this study ranged from 0.51 to 1.96%, suggesting that tick parasitism in crocodylians is likely opportunist

Progress towards the eradication of Amblyomma variegatum from the Caribbean.

Progress in the Caribbean Amblyomma Program (CAP) is reviewed since its inception in 1995 when regional eradication activities for Amblyomma variegatum ticks were initiated using Bayticol pour-on. Technical achievements in the various islands were slow initially, and showed a wide diversity in attaining eradication targets. St. Kitts, considered as a model program. eliminated the tropical Bont tick (TBT) from most of the island in less than 3 years. However, the elimination of very low numbers of residual adult TBT that persisted in three 'hot-spots' took another 3 years. A similar problem was

The molecular basis of the Amblyomma americanum tick attachment phase.

Towards discovery of molecular signaling cascades that trigger and/or facilitate the tick attachment and formation of its feeding lesion, suppressive subtractive hybridization, high throughput sequencing and validation of differential expression by cDNA dot blot hybridization were performed on Amblyomma americanum ticks that had attained appetence and were exposed to feeding stimuli. This approach allowed for identification of 40 genes that are up regulated before ticks begin to penetrate the host skin. Based on BLAST and secondary structure homology searches as well as motif scan analyses, pr

Influence of microhabitat use and behavior of Amblyomma sculptum and Amblyomma dubitatum nymphs (Acari: Ixodidae) on human risk for tick exposure, with notes on Rickettsia infection.

Brazilian spotted fever (BSF) is a potentially lethal human disease caused by Rickettsia rickettsii transmitted by ticks, including Amblyomma sculptum. However, in Southeast Brazil, where most BSF cases occur, capybaras are key hosts for both A. sculptum and Amblyomma dubitatum. We therefore compared the risk of human exposure to these ticks at a non-endemic anthropogenic site in Southeast Brazil where both tick species are maintained by capybaras and occur at high abundance. Cloth dragging, human baits and CO2 traps were used to assess tick abundance and risk for human exposure. The two tick

Efficacy of oral afoxolaner against Amblyomma maculatum infestations in dogs.

Amblyomma maculatum, the Gulf Coast tick, is a species of significant veterinary and public health importance, especially because it is a vector of important diseases, such as American canine hepatozoonosis and tidewater spotted fever. Amblyomma maculatum infests a wide range of vertebrates including livestock, dogs, cats, and humans. Two experimental studies were conducted to evaluate the efficacy of afoxolaner formulated in an oral tablet (NexGard®) against induced infestations of A. maculatum in dogs. These Good Clinical Practice (GCP) studies used a randomized, negative controlled and mask

Amblyomma sculptum Salivary Protease Inhibitors as Potential Anti-Tick Vaccines.

Amblyomma sculptum is the main tick associated with human bites in Brazil and the main vector of Rickettsia rickettsii, the causative agent of the most severe form of Brazilian spotted fever. Molecules produced in the salivary glands are directly related to feeding success and vector competence. In the present study, we identified sequences of A. sculptum salivary proteins that may be involved in hematophagy and selected three proteins that underwent functional characterization and evaluation as vaccine antigens. Among the three proteins selected, one contained a Kunitz_bovine pancreatic tryps

Red Meat Allergies after Lone Star Tick (Amblyomma americanum) Bites.

Red meat allergies have followed tick bites on every continent except Antarctica. The sensitizing antigen is galactose-α-1,3-galactose (α-gal), an oligosaccharide constituent of nonprimate blood and meat, acquired by ticks during animal bloodfeeding. Because red meat allergy after tick bites is a worldwide phenomenon, the objectives of this review were to describe the global epidemiology of red meat allergy after tick bites and its immunological mechanisms; to identify the human risk factors for red meat allergy after tick bites; to identify the most common tick vectors of red meat allergy wor

Life cycle of Amblyomma integrum (Acari: Ixodidae) under laboratory conditions.

Amblyomma integrum is a hard tick infesting mainly buffalo and cattle and has been identified as an agent of human otoacariasis in Sri Lanka. Data on the life cycle pattern of A. integrum were collected by experimental infestation on New Zealand white rabbits under laboratory conditions. Wild-caught females laid 55-7389 eggs for 2-35 days after spending a latent period of 10-25 days. Egg incubation period was 31-105 days and the newly emerged larvae started feeding after 4-11 days. Larvae dropped off after feeding and they moulted into nymphs after 10-16 days. Nymphs actively fed on rabbits fo

Population genetics of Amblyomma americanum (Acari: Ixodidae) collected from Arkansas.

Lone star ticks, Amblyomma americanum L. (Acari: Ixodidae),infest multiple hosts such as birds, and mammals of various sizes (rodents to white-tailed deer) and can harbor human pathogens such as Borrelia lonestari and Ehrlichiosis chaffeensis. The population structure of 251 A. americanum ticks, collected from canines and two white-tailed deer in six Arkansas ecoregions, was examined using DNA sequences of a 247-bp region of the mitochondrial DNA ribosomal RNA 16S gene. Of the 247 nucleotide characters, 26 were variable. Thirty-three haplotypes were identified of which 25 haplotypes occurred o

Amblyomma cajennense ticks induce immediate hypersensitivity in horses and donkeys.

Since host immune reaction to ticks interferes with tick-borne pathogen transmission, it is important to recognize naturally occurring tick-host immune relationships to better understand the epidemiology of such infectious diseases. Amblyomma cajennense is an important tick-borne disease vector in the Neotropical region and horses maintain it in domestic environments. In the present work intradermal testing of A. cajennense tick exposed horses and donkeys using crude tick antigens was used to evaluate the type of hypersensitivity induced by infestations. Animals sensitized by A. cajennense inf
