Axially-directed extrusive loads of 0.01-0.1 N were applied to the lathyritic mandibular incisors of 12 rabbits. Tooth position was monitored continuously with a variable capacitance displacement transducer. Compared with pair-fed controls, lathyritic teeth showed markedly reduced resistance to extrusive loading, indicating that the periodontal collagen has a role in supporting the tooth. The pattern of tooth mobility, however, was little affected. The results may have implications for the eruptive mechanism. Monepantel (MOP) is a new anthelmintic drug intended for the treatment and control of gastrointestinal roundworms (nematodes) infection and associated disease in sheep. The aim of our study was to find out metabolic pathways of MOP in sheep in vivo and in its parasite Haemonchus contortus ex vivo. MOP biotransformation in two H. contortus strains with different sensitivity to anthelmintics was also compared. Ultra high-performance liquid chromatography with tandem mass spectrometry (UHPLC-MS/MS) technique is used for the identification of MOP metabolites in ovine urine, faeces, and nematodes.

BACKGROUND: Haemonchus contortus, a gastrointestinal nematode parasite of sheep, is mainly controlled by anthelmintics; the occurrence of anthelmintic resistance leads to treatment failures and increases economic burden. Because molecular mechanisms involved in drug resistance can be elucidated by genomic studies, an extreme quantitative trait locus (X-QTL) mapping approach was used to identify co-segregation of the resistance phenotype with genetic markers to detect the genome-wide variants associated with monepantel resistance in H. contortus.

Collagen fibres in the periodontal ligament may have two functions: to resist displacing forces and to cause the tooth to erupt. Their function was examined in the continuously-erupting incisor of the rat using various concentrations and types of lathyrogens.
