Thiamine-producing lactic acid bacteria and their potential use in the prevention of neurodegenerative diseases.

Thiamine or vitamin B1, an essential micronutrient mainly involved in energy production, has a beneficial impact on the nervous system, and its deficiency can be associated with the development and progression of neurodegenerative diseases. The aim of this work was to select thiamine-producing lactic acid bacteria (LAB) and study their physiological effects using neuron cell cultures. In this study, 23 LAB able to produce thiamine were identified by growth in thiamine-free synthetic medium. Intra- and extracellular thiamine concentrations were determined using a microbiological method and resu

Thiamine deficiency induces endoplasmic reticulum stress in neurons.

Thiamine (vitamin B1) deficiency (TD) causes region selective neuronal loss in the brain; it has been used to model neurodegeneration that accompanies mild impairment of oxidative metabolism. The mechanisms for TD-induced neurodegeneration remain incompletely elucidated. Inhibition of protein glycosylation, perturbation of calcium homeostasis and reduction of disulfide bonds provoke the accumulation of unfolded proteins in the endoplasmic reticulum (ER), and cause ER stress. Recently, ER stress has been implicated in a number of neurodegenerative models. We demonstrated here that TD up-regulat

Sea cucumber extract enhances glutathione-related antioxidant defense and reduces apoptosis in Eimeria papillata-infected mice.

Eimeria spp. are globally significant parasites infecting many vertebrate hosts and causing coccidiosis. The widespread use of anticoccidial drugs has led to increased parasite resistance, highlighting the need for alternative therapies. This study aimed to evaluate the anticoccidial, antioxidant, and anti-apoptotic effects of Holothuria polii extract (HpE) compared with the reference drug amprolium in mice experimentally infected with Eimeria papillata. Seven groups of mice were studied: a negative control; a group treated with HpE only (200 mg/kg); a positive control (infected untreated); an

Cerebro-cortical necrosis (C.C.N.) in calf. An experimental reproduction of the disease.

In four ruminant calves a state of thiamine deficiency was brought about with a thiamine antagonist, amprolium. Twice daily the calves were given Amprolmix® containing 25 % amprolium through a stomach tube. The dosage was 350–500 mg/kg body weight daily during the entire experimental period. After 31 days the first calf showed fully developed symptoms; two other calves fell ill after another four and eight days respectively. The fourth calf was killed before the disease became clinically manifest. In all calves cerebrocortical necroses were found on histological examination. The clinical pictu

Biosynthesized Nanosilver from Ginger Extract Exhibits Antioxidant and Hepatic Responses during Eimeria papillata Infection.

Although several anticoccidial medications have long been used to prevent coccidiosis, their adverse effects necessitate the use of alternative control methods. In this study, Eimeria papillate was used to infect the mouse jejunum, and the response of the liver to induced coccidiosis on treatment with nanosilver synthesized from Zingiber officinale (NS) and the reference anticoccidial drug amprolium was compared. Mice were infected with 1000 sporulated oocysts to induce coccidiosis. NS was able to inhibit the sporulation of E. papillate by approximately 73%, and also, the NS treatment improved

Thiamine and selected thiamine antivitamins - biological activity and methods of synthesis.

Thiamine plays a very important coenzymatic and non-coenzymatic role in the regulation of basic metabolism. Thiamine diphosphate is a coenzyme of many enzymes, most of which occur in prokaryotes. Pyruvate dehydrogenase and 2-oxoglutarate dehydrogenase complexes as well as transketolase are the examples of thiamine-dependent enzymes present in eukaryotes, including human. Therefore, thiamine is considered as drug or diet supplement which can support the treatment of many pathologies including neurodegenerative and vascular system diseases. On the other hand, thiamine antivitamins, which can int

Pyrimidinyl nicotinic acid and cerebrocortical necrosis.

Pyrimidinyl nicotinic acid (PNA) exhibits great structural similarities to amprolium, which is a known thiamine antagonist. Experimental cerebrocortical necrosis (CCN) has been induced by oral administration of amprolium. PNA has been demonstrated in ruminai fluid from animals suffering from CCN. The aim of this investigation was to study whether PNA can act as thiamine antagonist and whether it can give rise to CCN. For this purpose PNA was synthesized and was daily given intravenously to a calf. The dose corresponded to roughly 10 times the content of thiamine in the blood. After three weeks

Anticoccidial and jejunum-protective effects of Krameria lappacea roots extract on experimental Eimeria papillata infection.

Coccidiosis is a protozoan parasitic disease caused by Eimeria species and affects wild and domestic animals. Coccidiostats are currently available to control this disease, although drug resistance has been confirmed for all of them. As a result, there is an urgent need to identify eco-friendly agents to control and treat this disease. This study aimed to investigate the ameliorative role of the Krameria lappacea roots extract (KLRE) on the outcome of coccidiosis induced by Eimeria papillata. Male C57BL/6 mice were divided into seven groups (5 mice/group), as follows: Group 1: noninfected-nont

Biochemical and pathological observations on sheep showing various clinical manifestations of experimental cerebrocortical necrosis.

For observing chronological changes of biochemical and pathological aspects in experimental cerebrocortical necrosis, 14 sheep were given amprolium (600 mg/kg/day), and killed at various clinical stages. At the onset of abnormal electroencephalograms, total thiamine concentrations in the cerebrum, liver, heart and blood were reduced significantly. After anorexia was noted, total thiamine concentrations in them did not decrease further as symptoms developed severely. Autofluorescence and necrotic lesions in the cerebral cortex were observed in animals killed one day after the onset of abnormal

Tissue responses of Clarias gariepinus (Catfish) to experimental Eimeria subepithelialis infection.

Coccidial infection is a common feature in most African fishes. As a result of the chronic nature of the disease in fish, mortality is gradual (Kim and Paperna, 1993a) and is often overlooked in most farms, with losses becoming evident only at the end of the production cycle. This study evaluates the tissue responses of Clarias gariepinus (Catfish) in an experimental infection with Eimeria subepithelialis using 200 laboratory-bred Juveniles. Four groups (A, B, C and D) of 50 Juveniles each were preconditioned for 2 weeks. Groups A and B were infected per os with 200 E. subepithelialis sporocys

Thiamine deficiency--induced partial necrosis and mitochondrial uncoupling in neuroblastoma cells are rapidly reversed by addition of thiamine.

Culture of neuroblastoma cells in a medium of low-thiamine concentration (6 nM) and in the presence of the transport inhibitor amprolium leads to the appearance of overt signs of necrosis; i.e., the chromatin condenses in dark patches, the oxygen consumption decreases, mitochondria are uncoupled, and their cristae are disorganized. Glutamate formed from glutamine is no longer oxidized and accumulates, suggesting that the thiamine diphosphate-dependent alpha-ketoglutarate dehydrogenase activity is impaired. When thiamine (10 microM) is added to the cells, the O2 consumption increases, respirato

Biosynthesized selenium nanoparticles to rescue coccidiosis-mediated oxidative stress, apoptosis and inflammation in the jejunum of mice.

One of the most crucial approaches for treating human diseases, particularly parasite infections, is nanomedicine. One of the most significant protozoan diseases that impact farm and domestic animals is coccidiosis. While, amprolium is one of the traditional anticoccidial medication, the advent of drug-resistant strains of Eimeria necessitates the development of novel treatments. The goal of the current investigation was to determine whether biosynthesized selenium nanoparticles (Bio-SeNPs) using Azadirachta indica leaves extract might treat mice with Eimeria papillata infection in the jejunal

[Cerebrocortical necrosis in calves, sheep and goats].

Cerebrocortical necrosis was induced through the oral application of amprolium to calves (0.250 g/kg to 1.00 g/kg) and weaned lambs and goats (1.25 g/kg) in the course of 25 to 35 days. In order to shed light on the etiology of the disease an experiment was carried out with a ration rich in carbohydrates for calves and sheep. Clinically, the disease was manifested with nervous symptoms. Biochemically, a drop of the amount of B1 in the blood serum, liver, and brain was established, along with a rise of pyruvic acid in the blood serum. Morphologically, most characteristic were the laminar necros
