Anti-proliferative effects of progesterone antagonists in the primate endometrium: a potential role for the androgen receptor.

In women and non-human primates, treatment with anti-progestins suppresses oestrogen-dependent mitotic activity in the endometrial glands. This anti-proliferative effect is paradoxical, because anti-progestins do not bind to the oestrogen receptor. Although this phenomenon has been termed a 'non-competitive anti-oestrogenic effect', it does not occur in all species or in other regions of the primate reproductive tract, so is best referred to as an 'endometrial anti-proliferative effect'. The abundance of androgen receptors is greatly increased by anti-progestin treatment, especially in the gla

Design, synthesis and activity evaluation of benzene sulfonamide derivatives as novel androgen receptor antagonist.

The androgen receptor (AR) is a critical therapeutic target for the endocrine treatment of prostate cancer (PCa). Current AR antagonists, which primarily target the hormone binding pocket (HBP) within the ligand-binding domain (LBD), are often limited by the emergence of resistance mutations, calling for novel strategies of AR inhibition. In this study, a series of substituted benzene sulfonamide derivatives were designed and synthesized based on IMB-A6, a lead compound previously identified by our group to target the activation function 2 (AF2) region of AR-LBD. These compounds were evaluated

Inflammation impacts androgen receptor signaling in basal prostate stem cells through interleukin 1 receptor antagonist.

Chronic prostate inflammation in patients with benign prostate hyperplasia (BPH) correlates with the severity of symptoms. How inflammation contributes to prostate enlargement and/or BPH symptoms and the underlying mechanisms remain unclear. In this study, we utilize a unique transgenic mouse model that mimics chronic non-bacterial prostatitis in men and investigate the impact of inflammation on androgen receptor (AR) in basal prostate stem cells (bPSC) and their differentiation in vivo. We find that inflammation significantly enhances AR levels and activity in bPSC. More importantly, we ident

Inflammation Impacts Androgen Receptor Signaling in Basal Prostate Stem Cells Through Interleukin 1 Receptor Antagonist.

The majority of patients with benign prostate hyperplasia (BPH) exhibit chronic prostate inflammation and the extent of inflammation correlates with the severity of symptoms. How inflammation contributes to prostate enlargement and/or BPH symptoms and the underlying mechanisms are not clearly understood. We established a unique mouse model Prostate Ovalbumin Expressing Transgenic 3 (POET3) that mimics chronic non-bacterial prostatitis in men to study the role of inflammation in prostate hyperplasia. After the injection of ovalbumin peptide-specific T cells, POET3 prostates exhibited an influx

Androgen receptor antagonist flutamide modulates estrogen receptor alpha expression in distinct regions of the hypospadiac rat penis.

INTRODUCTION: Intrauterine exposure to endocrine disrupting chemicals (EDCs), particularly anti-androgens, has been implicated in hypospadias by disrupting fetal masculinization of the genital tubercle (GT). Other pathways, including estrogen signaling, may also contribute but remain poorly characterized, especially in rats - a key model in chemical toxicity testing. Estrogen signaling has also been linked to hypospadias in mice, raising questions about androgen-estrogen interactions in guiding GT differentiation. METHODS: We induced hypospadias in male rat offspring via intrauterine exposure

Efficient extraction of fluorinated non-steroidal androgen receptor antagonists on a novel trifluoromethyl functionalized magnetic microporous organic network.

Given the widespread clinical use, environmental persistence, and potential ecological and health risks of non-steroidal androgen receptor antagonists (NSAAs), developing a highly selective and sensitive method to detect their residues in aquatic environments is of significant importance and poses considerable challenges. Based on the hydrophobicity and fluorine-containing characteristics of NSAAs, this study designed and prepared a novel trifluoromethyl functionalized magnetic microporous organic network (MMON-CF3) for the efficient and selective magnetic solid-phase extraction (MSPE) of thre

Falls and fractures in men with prostate cancer taking second-generation androgen receptor antagonists: A retrospective chart review.

IntroductionSecond-generation androgen receptor antagonists, including enzalutamide, apalutamide, and darolutamide, are commonly used to treat metastatic and non-metastatic prostate cancer. Although these medications are typically well-tolerated, falls were reported in 4.2-15.6% of patients and fractures in 4.2-11.7% of patients enrolled in the phase III clinical trials evaluating these drugs in prostate cancer. However, post-marketing studies have reported a lower incidence than reported in clinical trials. The objective of this study is to determine the prevalence of falls and fractures in p

Occurrence of androgens in sewage treatment plants influents is associated with antagonist activities on other steroid receptors.

The occurrence of endocrine disrupting chemicals such as estrogens in raw urban sewage is well documented. By contrast, the presence of other steroidal activities in wastewater has been poorly studied, although they can cause undesirable biological responses in the environment. In this work, extracts of raw wastewater were tested for agonist and antagonist activities on estrogen, androgen, progesterone, mineralocorticoid and glucocorticoid receptors. We detected strong estrogenic activities that correlated well with the concentration of natural estrogens (estrone, estriol and 17β-estradiol) me

Discovery of N-(1,2,4-Thiadiazol-5-yl)benzo[b]oxepine-4-carboxamide Derivatives as Novel Antiresistance Androgen Receptor Antagonists.

The ligand-binding pocket of the androgen receptor (AR) is the targeting site of all clinically used AR antagonists. However, various drug-resistant mutations emerged in the pocket. We previously reported a new targeting site at the dimer interface of AR (dimer interface pocket) and identified a novel antagonist M17-B15 that failed in oral administration. In this study, the head part of M17-B15 was substituted with divergent structures. Potent antagonist Z10 with benzo[b]oxepine was first identified. Subsequent structural optimization on the 2-oxopropyl moiety of Z10 generated the more powerfu

Comparative fall risk of patients treated with novel androgen receptor antagonists in prostate cancer: a systematic review and meta-analysis.

OBJECTIVE: Androgen receptor pathway inhibitors (ARPIs) are cornerstone treatments for advanced prostate cancer; however, their potential to increase fall risk remains a significant clinical concern. This meta-analysis aims to provide a rigorous, drug-specific evaluation of the association between novel ARPIs and the risk of falls. METHODS: We systematically searched PubMed, Web of Science, Cochrane Library, and ClinicalTrials.gov for Phase 2 or 3 randomized controlled trials (RCTs) comparing ARPIs (enzalutamide, apalutamide, and darolutamide) with control groups (placebo or non-steroidal ant

Rapid Development of Spinal Epidural Lipomatosis after Treatment of Metastatic Castration-Resistant Prostate Cancer with Second-Generation Androgen Receptor Antagonists.

BACKGROUND: Previous studies have described the association of spinal epidural lipomatosis with several conditions including chronic steroid therapy, Cushing's syndrome, obesity, Paget disease, and hypothyroidism. We present a report of rapid development of spinal epidural lipomatosis after treatment with second-generation anti-androgen therapy, a new strategy for treatment of metastatic castration-resistant prostate cancer that has been increasingly employed in the past few years. A comprehensive discussion of the underlying molecular networks involving androgen receptor blockage and adipocyt

Discovery of Novel Bifunctional Agents as Potent Androgen Receptor Antagonists and Degraders for the Treatment of Enzalutamide-Resistant Prostate Cancer.

Bifunctional agents that simultaneously antagonize and degrade various AR proteins more effectively block the AR signaling pathway, offering a promising strategy for the treatment of mCRPC patients. Herein, we report the discovery and development of a series of small-molecule AR degraders with 3,8-diazabicyclo[3.2.1]octan scaffold. The optimal compound 20i exhibited potent AR antagonistic and degrading activities, effectively overcoming multiple resistance mechanisms and showing significant antiproliferative effects against enzalutamide-resistant PCa cell lines. Moreover, compound 20i exhibite

Uncertain magnitude of benefit of early second-generation androgen receptor antagonist treatment in advanced prostate cancer.

Guidelines currently recommend the early use of second-generation androgen receptor signaling inhibitors (ARSIs) in both castrate-resistant and -sensitive metastatic prostate cancer (with or without chemotherapy) and also in biochemical non-metastatic castrate-sensitive relapse. We highlight design issues in the randomized controlled trials (RCTs) that bias the results of early ARSI use, suggesting that the magnitude of benefit might be overestimated and, therefore, uncertain. Issues include sub-optimal post-protocol care, inadequate control arms, and the inclusion of highly selected patients,

Discovery of Potential First-in-Class Dual Ligand-Directed Degrader and Antagonist of the Androgen Receptor: BMS-986365.

Androgen receptor (AR) signaling is central to the progression of prostate cancer, including castration-resistant disease. Targeted protein degradation, particularly through heterobifunctional compounds, represents a significant advancement in eliminating disease-causing proteins beyond traditional inhibition. Here, we describe the medicinal chemistry discovery of BMS-986365, a novel dual-function AR degrader and an antagonist. BMS-986365 selectively and efficiently degrades both wild-type and clinically relevant mutant ARs while also antagonizing residual AR activity. This dual mechanism resu

The fungicide procymidone alters sexual differentiation in the male rat by acting as an androgen-receptor antagonist in vivo and in vitro.

Procymidone is a dicarboximide fungicide structurally related to the well-characterized fungicide vinclozolin. Vinclozolin metabolites bind to mammalian androgen receptors (AR) and act as AR antagonists, inhibiting androgen-dependent gene expression in vivo and in vitro by inhibiting AR-binding to DNA. The current study was designed to determine if procymidone acted as an AR antagonist in vitro and to describe the dosage levels of procymidone that alter sexual differentiation in vivo. In vitro, procymidone inhibited androgen from binding the human AR (hAR) in COS (monkey kidney) cells transfec

A novel small-molecule androgen receptor antagonist selective for the T878A-mutant AR in prostate cancer therapy.

Prostate cancer (PCa) is one of the most prevalent malignancies in males. While androgen receptor (AR) antagonists play a crucial role in PCa therapy, their efficacy is often limited by resistance arising from AR mutations. Here, we identified S-94, corresponding to the S-enantiomer of compound 94, as a novel AR antagonist with marked selectivity for the T878A-mutant AR. In vitro, S-94 inhibited AR transcriptional activity (IC50 = 0.38 μM) and suppressed cell proliferation (IC50 = 10.32 μM) in LNCaP cells harboring the T878A-mutant AR, while exhibiting lower cytotoxicity toward non-cancerous c
