Reward network mechanism in anhedonia and depression.

BACKGROUND: Depression is one of the most burdensome mental disorders. Anhedonia, a core symptom of major depressive disorder (MDD), is characterized by abnormal resting-state reward network (RN). However, it is unclear whether anhedonia symptom and depressive episode share similar resting-state RN mechanism, as well as whether the RN mechanism is a state or trait-like marker of depression. This study aims to clarify the two points by recruitingboth current and remitted depression. METHODS: Using functional Magnetic Resonance Imaging (fMRI) scans, this study observed the resting-state RN func

The effect of vortioxetine on anhedonia in patients with schizophrenia.

OBJECTIVE: Anhedonia is a common symptom of depression, but is also a negative symptom of schizophrenia. The purpose of this study was to examine the effects of vortioxetine on anhedonia in patients with schizophrenia. METHODS: A total of 120 patients with schizophrenia in remission who met inclusion criteria were randomized 1:1 by the envelope method into intervention and control groups. All participants in both groups were divided into three subgroups based on the antipsychotic therapy they were receiving (olanzapine, risperidone, or aripiprazole). Vortioxetine was administered to those in

State Anhedonia and Suicidal Ideation in Adolescents.

Background: Recent work suggests that state anhedonia and its social aspect of loss of interest in people was an important predictor of suicidal ideation in adults. Aim: The current study investigated the relationship between state anhedonia, trait anhedonia, suicidal ideation, and suicide attempts in adolescents. Method: State anhedonia was assessed using the anhedonia subscale from the Child Mood and Feelings Questionnaire, while trait social anhedonia was assessed using the Adolescent Anticipatory and Consummatory Interpersonal Pleasure Scale and the Temporal Experience of Pleasure Scale. Making Sense of Rodent Models of Anhedonia.

A markedly reduced interest or pleasure in activities previously considered pleasurable is a main symptom in mood disorder and psychosis and is often present in other psychiatric disorders and neurodegenerative diseases. This condition can be labeled as "anhedonia," although in its most rigorous connotation the term refers to the lost capacity to feel pleasure that is one aspect of the complex phenomenon of processing and responding to reward. The responses to rewarding stimuli are relatively easy to study in rodents, and the experimental conditions that consistently and persistently impair th

Levetiracetam-associated loss of libido and anhedonia.

The relationship between the older antiepileptic drugs (AEDs) and sexual dysfunction has long been known and it is likely to be related to sexual hormonal changes. Instead, rare reports on sexual disorders related to new AEDs suggest the possibility of complex and poorly understood mechanisms, mainly involving central nervous system neurotransmitters such as glutamate, serotonin, and dopamine. Herein, we describe two young men with epilepsy who experienced severe loss of libido and anhedonia after levetiracetam intake.

Early stimulus processing in dysthymia and anhedonia.

Previous event-related brain potential (ERP) research has found that dysthymic subjects differ from control subjects during later stages of information processing. An important issue that emerges from this literature is whether differences found in these ERP components, typically associated with cognitive processing, can be attributed to earlier differences in basic perceptual processing. This study was undertaken to determine whether early processing deficits are apparent in dysthymic persons. Responses of dysthymics (n = 23) were compared with those of anhedonic (n = 15) and normal control (

Circuit Mechanisms of Reward, Anhedonia, and Depression.

Pleasure and motivation are important factors for goal-directed behavior and well-being in both animals and humans. Intact hedonic capacity requires an undisturbed interplay between a number of different brain regions and transmitter systems. Concordantly, dysfunction of networks encoding for reward have been shown in depression and other psychiatric disorders. The development of technological possibilities to investigate connectivity on a functional level in humans and to directly influence networks in animals using optogenetics among other techniques has provided new important insights in th

Anhedonia, depressed mood, and smoking cessation outcome.

OBJECTIVE: Although the relation between lifetime depression and smoking cessation outcome has been well studied, the proposition that different symptomatic expressions of depression exert disparate predictive effects on risk of smoking cessation failure has largely gone uninvestigated. This study analyzed the individual contributions of depression's 2 hallmark affective symptoms, anhedonia (i.e., diminished interest in normally enjoyable activities) and depressed mood (i.e., elevated sadness), to the prediction of smoking cessation outcome. METHOD: Participants were adult daily smokers (N =

Hindlimb unloading elicits anhedonia and sympathovagal imbalance.

The hindlimb-unloaded (HU) rat model elicits cardiovascular deconditioning and simulates the physiological adaptations to microgravity or prolonged bed rest in humans. Although psychological deficits have been documented following bed rest and spaceflight in humans, few studies have explored the psychological effects of cardiovascular deconditioning in animal models. Given the bidirectional link established between cardiac autonomic imbalance and psychological depression in both humans and in animal models, we hypothesized that hindlimb unloading would elicit an alteration in sympathovagal ton

Physical anhedonia, perceptual aberration, and psychosis proneness.

Two groups of hypothetically psychosis-prone subjects were chosen from among college students who scored deviantly high on scales of Physical Anhedonia (n = 50) or Perceptual Aberration (n = 65). Scores on these two scales had a small negative correlation, indicating that the scales identify different sets of deviant subjects. These experimental subjects and a control group (n = 66) were interviewed using a modification of the Schedule for Affective Disorders and Schizophrenia--Lifetime Version. A second interview covered social and academic adjustment. Psychotic and psychotic-like symptoms (a

A parallel-arm, randomized trial of Behavioral Activation Therapy for anhedonia versus mindfulness-based cognitive therapy for adults with anhedonia.

BACKGROUND: Anhedonia, deficits in motivation and pleasure, is a transdiagnostic symptom of psychopathology and negative prognostic marker. METHODS: In this randomized, parallel-arm clinical trial, a novel intervention, Behavioral Activation Treatment for Anhedonia (BATA), was compared to an individually administered Mindfulness-Based Cognitive Therapy (MBCT) in a transdiagnostic cohort of adults with clinically significant anhedonia (ClinicalTrials.gov Identifiers NCT02874534 and NCT04036136). Participants received 8-15 individual psychotherapy sessions, once weekly, with either BATA (n = 61

Schizotypy, affective temperaments and anhedonia in bipolar depression.

OBJECTIVES: "Schizotypy" is a term describing personality traits reflected in emotional, perceptual and cognitive styles. Affective temperaments are trait-like features which were observed to be stable in time and predispose to mood disorders. The purpose of this study was to examine relationship between schizotypal features, affective temperaments and anhedonia in patients with bipolar depression. METHODS: 54 patients with bipolar depression were included in the study. Participant were administered the following psychometric tools: Dimensional Anhedonia Rating Scale (DARS), Snaith-Hamilton P

Understanding the neural code of stress to control anhedonia.

Anhedonia, the diminished drive to seek, value, and learn about rewards, is a core feature of major depressive disorder1-3. The neural underpinnings of anhedonia and how this emotional state drives behaviour remain unclear. Here we investigated the neural code of anhedonia by taking advantage of the fact that when mice are exposed to traumatic social stress, susceptible animals become socially withdrawn and anhedonic, whereas others remain resilient. By performing high-density electrophysiology to record neural activity patterns in the basolateral amygdala (BLA) and ventral CA1 (vCA1), we iden

Anhedonia in combat veterans with penetrating head injury.

Anhedonia is a common symptom following traumatic brain injury. The neural basis of anhedonia is poorly understood, but believed to involve disturbed reward processing, rather than the loss of sense of pleasure. This analysis was undertaken to determine if injury to specific regions of prefrontal cortex (PFC) result in anhedonia. A CT-based lesion analysis was undertaken in 192 participants of the Vietnam Head Injury Study, most with penetrating head injury. Participants were divided into left and right ventrolateral prefrontal, bilateral ventromedial prefrontal, and other injury locations. An

Alcohol protracted withdrawal syndrome: the role of anhedonia.

AIMS: The aim of the present study is to characterize the relevance of withdrawal symptoms during the first 12 months of abstinence and their relation to anhedonia and craving. METHODS: 102 detoxified subjects meeting clinical criteria for Alcohol Dependence in Remission were recruited at various time since the detoxification and subdivided into four groups according to the length of abstinence (group 1: 15-30 days; group 2: 30-90 days; group 3: 90-180 days; group 4: 180-360). Withdrawal symptomatology was assessed through the Clinical Institute Withdrawal Assessment for Alcohol (CIWA-Ar). Th

Stress, depression, and anhedonia: caveats concerning animal models.

Numerous animal models of depression have been advanced, each having multiple attributes and some limitations. This review provides caveats concerning etiologically valid animal models of depression, focusing on characteristics of the depressive subtype being examined (e.g. typical vs atypical major depression, dysthymia, melancholia), and factors that contribute to the interindividual behavioral variability frequently evident in stressor-related behavioral paradigms. These include the stressor type (processive vs systemic stressors), and characteristics of the stressor (controllability, predi

Defining anhedonia subgroups using the dimensional anhedonia rating scale in active depression: Findings from the Texas resilience against depression study.

BACKGROUND: Anhedonia is a core symptom of major depressive disorder (MDD). We aimed to validate the Dimensional Anhedonia Rating Scale (DARS), explore data-driven subgroups of anhedonia among individuals with depression, and determine associations with anxiety, suicidal ideation, neuroticism, and childhood trauma. METHODS: Participants with current major depressive episode (n = 474) enrolled in the D2K arm of the Texas Resilience Against Depression (T-RAD) study were included in these analyses. Structure and reliability of the DARS were assessed using a factor analysis with a PROMAX rotation

The association between anhedonia and obesity: A systematic review.

BACKGROUND: Anhedonia, conceptualized here as a multidimensional transdiagnostic construct encompassing deficits in both hedonic experience (''liking'') and motivational reward processing (''wanting''), is frequently reported in individuals with obesity; however, the nature of this relationship remains incompletely characterized. This systematic review synthesizes clinical evidence on the association between anhedonia and obesity, with an emphasis on behavioral, metabolic, and genetic mechanisms. METHODS: Following PRISMA guidelines, we systematically searched PubMed, Ovid (MEDLINE and APA Ps

Anhedonia in Parkinson's disease: a systematic review of the literature.

Anhedonia, defined as lowered ability to experience physical or social pleasure, is a key symptom of several psychiatric illnesses. In this systematic review, we aimed to evaluate the role of anhedonia in Parkinson's Disease and its relationships with other clinical characteristics, dopamine dysfunction, and antiparkinsonian therapy. The database was selected using PubMed Services. Relevant journals were hand-searched, and the bibliographies of all the important articles were scrutinized to find additional publications. Fifteen studies assessed the topic of anhedonia in Parkinson's disease fro
