Interspecies scaling: predicting pharmacokinetic parameters of antiepileptic drugs in humans from animals with special emphasis on clearance.

The objective of this study was to test the interspecies-scaling approach in a series of antiepileptic drugs. Clearance, volume of distribution, and elimination half-life were scaled up from animal data obtained from literature. Four different methods were utilized to generate plots to scale up the clearance values: (i) clearance vs body weight (simple allometric equation); (ii) the product of clearance and maximum life-span potential (MLP) vs body weight (an approach recommended in literature); (iii) the two-term power equation which incorporates both body weight and brain weight suggested by

Large-scale survey, animal models, and computational modeling identify histological neurodegenerative biomarkers for traumatic optic neuropathy.

BACKGROUNDTraumatic optic neuropathy (TON) is a leading cause of blindness following closed traumatic brain injury, with no effective treatments available. Previous interventional clinical trials were complicated by its low prevalence, variability in neurodegenerative severity, and unavailability of reliable biomarkers.METHODSWe analyzed data from 1,226 patients enrolled in the prospective National Multi-Center Collaborative Clinical Research Program of China (2017-2024) to establish a clinical profile and identify noninvasive biomarkers for neurodegenerative severity. Subgroup analysis of pat

Which animal type contributes the most to the emission of antibiotic resistance genes in large-scale swine farms in China?

Modern swine farms generally contain several animal types and rely extensively on the feed additives, including antibiotics and heavy metals, to augment animal growth. Nonetheless, as an important reservoir of antibiotic resistance genes (ARGs) in the environment, the ARGs emission of each animal type from swine farms has not been characterized. The goal of this study is to determine which animal type contributes the most to the ARGs emission into the environment in typical swine farms of China. Results showed that chlortetracycline (CTC), Cu and Zn were the typical feed additives, and the con

The tale of a modern animal plague: tracing the evolutionary history and determining the time-scale for foot and mouth disease virus.

Despite significant advances made in the understanding of its epidemiology, foot and mouth disease virus (FMDV) is among the most unexpected agricultural devastating plagues. While the disease manifests itself as seven immunologically distinct strains their origin, population dynamics, migration patterns and divergence times remain unknown. Herein we have assembled a comprehensive data set of gene sequences representing the global diversity of the disease and inferred the time-scale and evolutionary history for FMDV. Serotype-specific rates of evolution and divergence times were estimated usin

Chondrogenesis in primitive tracheal neocartilage: insights from 3D-printed silicone grafts in a large-scale animal model.

OBJECTIVES: Tracheal cartilage has limited regenerative potential in adults, posing a significant challenge for tracheal repair. Here, we observed marked neocartilage growth in a porcine model after transplanting it with a three-dimensional (3D)-printed silicone tracheal graft. METHODS: Virtual silicone tracheal grafts of 2 cm in length were first generated using a 3D printer, and simulated a 3-month-old porcine trachea based on data from physical stress tests. After a segmental resection, a graft underwent end-to-end anastomosis at both the proximal and distal tracheal sections. Neotissue sa

In vivo models of infection: Large animals - Mini review on human-scale one-stage revision in a porcine osteomyelitis model.

Animal models are essential for orthopedic infectious research. However, only few models are currently able to capture the complex and multidisciplinary treatment approach for osteomyelitis. To replicate treatment situations in their entirety, large animal models are needed, and the most used species are sheep and pigs. Herein, we review a well-characterized and reproducible porcine model of human-scale one-stage revision of implant-associated osteomyelitis that can be used for robust preclinical testing of operative and post-operative interventions. The pros and cons of the model are discusse

Large-Scale Neural Recording Technologies for Investigating Circuit Dysfunction and Functional Biomarker Discovery in Animal Models of Alzheimer's Disease.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder associated with amyloid beta accumulation, tau pathology, and neuronal loss. Increasing evidence suggests that AD is associated with disruptions in large-scale circuit activity across multiple disease animal models, even before clear cognitive decline. These findings highlight the need for functional biomarkers that capture dynamic changes in neuronal network activity, rather than relying solely on molecular or anatomical measures. Recent advances in neural recording technologies now allow AD-related network dysfunction to be

Peritonitis-related bacterial infections: a large-scale case-series retrospective study in 160 domestic animals (2009-2022).

Bacterial peritonitis infections comprise a life-threatening clinical condition in domestic animals that commonly lead to sepsis and high mortality. A set of bacterial pathogens have been identified in septic peritonitis in livestock and companion animals. Nonetheless, most descriptions are restricted to case reports or limited to only one domestic species, and a restrict number of comprehensive studies involving this infection has focused on a great number of domestic animals. Here, we retrospectively investigated selected epidemiological data (with an emphasis in outcome), clinical signs, ba

High-quality, large-scale, semi-thin, & ultra-thin sections of the optic nerve in large animals: An optimized procedure.

Large animal model of optic nerve (ON) injury is an essential tool for translational medicine. Perfusion fixation with paraformaldehyde is mainly used for preparing the semi-thin (1-2 μm thick) and ultra-thin (<0.5 μm thick) sections of the ON tissues. However, this conventional fixation technique in large animals needs a large volume of fixatives, which increases the risk of toxic exposure and is environmentally unfriendly. Additionally, fixed residual ON cannot be used for other tests that require fresh tissue samples. Although conventional immersion fixation is feasible for preparing a semi

What drives small-scale farmers to vaccinate their multiple livestock species animals against common infectious diseases in Myanmar?

Livestock rearing is an important income source for small-scale farmers in Myanmar, but Foot and Mouth Disease (FMD) and Newcastle disease (ND) are major constraints to livestock production. A study was conducted to identify perceptions of farmers about FMD and ND disease risks and perceptions about vaccination practices by using the modified health belief model. The majority of livestock farmers (>70%) reported that they were aware of the risk and impact of FMD and ND and were willing to vaccinate their livestock (>60%). Focusing on three main livestock farmer groups, about 17.0% of cattle, 1

Standardized Minimum Number of Individuals (MNI) Enumeration and Veterinary Forensic Assessment in a Large-Scale Animal Starvation Scene: A Technical Note.

Mass companion animal mortality scenes pose major veterinary forensic and animal welfare challenges, particularly when remains are commingled, variably decomposed, or skeletonized. In March 2023, the remains of 1256 dogs and cats were recovered from a private residence in Yangpyeong-gun, Gyeonggi-do, Republic of Korea, after prolonged deprivation of food and water. Because whole-carcass enumeration was not feasible across the entire site, a standardized skull-based minimum number of individuals (MNI) approach was used to derive the scene-wide count by combining direct counts of intact, non-dup

Mitophagy modulation rescues single large-scale mitochondrial DNA deletion (SLSMD) disease symptoms in the C. elegans uaDf5 animal model.

S ingle large s cale m itochondrial DNA (mtDNA) d eletions (SLSMD) underlie a range of sporadic or maternally inherited primary mitochondrial diseases having significant morbidity and mortality, including Pearson syndrome, Kearns-Sayre Syndrome, or Chronic Progressive External Ophthalmoplegia. Therapeutic development has been hindered by limited existing knowledge on mtDNA quality control and a lack of SLSMD animal models. To address this challenge, we utilized the C. elegans heteroplasmic SLSMD strain, uaDf5, to objectively screen for potential therapies. As mitophagy modulation has been impl

A high level estimation of the net economic benefits to small-scale livestock producers arising from animal health product distribution initiatives.

INTRODUCTION: A fundamental challenge for charities that facilitate distribution of animal health products to small-scale livestock producers (SSPs) in low and middle income countries (LMICs) is identifying the products and market mechanisms that provide the greatest positive impact for SSPs and estimating their associated impact. This paper describes a pragmatic approach to modeling the impact of market-led product distribution initiatives based on estimating the net economic benefit of administration of animal health products. METHODS: The model estimates the economic impact of diseases at

Optimizing large-scale autologous human keratinocyte sheets for major burns-Toward an animal-free production and a more accessible clinical application.

BACKGROUND AND AIMS: Autologous keratinocyte sheets constitute an important component of the burn wound treatment toolbox available to a surgeon and can be considered a life-saving procedure for patients with severe burns over 50% of their total body surface area. Large-scale keratinocyte sheet cultivation still fundamentally relies on the use of animal components such as inactivated murine 3T3 fibroblasts as feeders, animal-derived enzymes such as trypsin, as well as media components such as fetal bovine serum (FBS). This study was therefore aimed to optimize autologous keratinocyte sheets by

Chromosome-scale genome assembly and gene annotation of the hydrothermal vent annelid Alvinella pompejana yield insight into animal evolution in extreme environments.

BACKGROUND: The Pompeii worm Alvinella pompejana, a terebellid annelid, has long been an exemplar of a metazoan that lives in an extreme environment, on the chimney wall of deep-sea hydrothermal vents, but this very environment has made it difficult to study. Comprehensive assessment of Alvinella pompejana genome content, and the factors that could explain its ability to thrive in seemingly hostile conditions has been lacking. RESULTS: We report the chromosome-level genome sequence of Alvinella pompejana and population-level sequence variants. We produced a set of gene models and analysed the

Association between animal welfare indicators and mastitis pathogens in bulk tank milk: the reality of small- and medium-scale dairy cattle farms in Italy.

INTRODUCTION: Animal welfare is increasingly recognized as a key determinant of dairy herd health, yet its relationship with the presence of mastitis pathogens at the herd level remains insufficiently explored. This study aimed to evaluate the association between animal welfare indicators and the presence of major mastitis pathogens in bulk tank milk (BTM) from dairy cattle herds in the Aosta Valley, Italy, while also characterizing microbial profiles using bacteriological culture and polymerase chain reaction (PCR). METHODS: A broad microbiological screening was performed on unique BTM sampl

Large-scale animal model study uncovers altered brain pH and lactate levels as a transdiagnostic endophenotype of neuropsychiatric disorders involving cognitive impairment.

Increased levels of lactate, an end-product of glycolysis, have been proposed as a potential surrogate marker for metabolic changes during neuronal excitation. These changes in lactate levels can result in decreased brain pH, which has been implicated in patients with various neuropsychiatric disorders. We previously demonstrated that such alterations are commonly observed in five mouse models of schizophrenia, bipolar disorder, and autism, suggesting a shared endophenotype among these disorders rather than mere artifacts due to medications or agonal state. However, there is still limited rese

Extremely Elevated Total Mercury and Methylmercury in Forage Plants in a Large-Scale Abandoned Hg Mining Site: A Potential Risk of Exposure to Grazing Animals.

Ninety-five wild forage plants (belonging to 22 species of 18 families) and their corresponding rhizosphere soil samples were collected from wastelands of a large-scale abandoned Hg mining region for total Hg (THg) and methylmercury (MeHg) analysis. The forage plant communities on the wastelands were dominated by the Asteraceae, Crassulaceae, and Polygonaceae families. The THg and MeHg concentrations in the forage plants varied widely and were in the range of 0.10 to 13 mg/kg and 0.19 to 23 μg/kg, respectively. Shoots of Aster ageratoides showed the highest average THg concentration of 12 ± 1.
