Chromosome-level genome assemblies of the malaria vectors Anopheles coluzzii and Anopheles arabiensis.

BACKGROUND: Anopheles coluzzii and Anopheles arabiensis belong to the Anopheles gambiae complex and are among the major malaria vectors in sub-Saharan Africa. However, chromosome-level reference genome assemblies are still lacking for these medically important mosquito species. FINDINGS: In this study, we produced de novo chromosome-level genome assemblies for A. coluzzii and A. arabiensis using the long-read Oxford Nanopore sequencing technology and the Hi-C scaffolding approach. We obtained 273.4 and 256.8 Mb of the total assemblies for A. coluzzii and A. arabiensis, respectively. Each asse

Temperature impacts the environmental suitability for malaria transmission by Anopheles gambiae and Anopheles stephensi.

Extrinsic environmental factors influence the spatiotemporal dynamics of many organisms, including insects that transmit the pathogens responsible for vector-borne diseases (VBDs). Temperature is an especially important constraint on the fitness of a wide variety of ectothermic insects. A mechanistic understanding of how temperature impacts traits of ectotherms, and thus the distribution of ectotherms and vector-borne infections, is key to predicting the consequences of climate change on transmission of VBDs like malaria. However, the response of transmission to temperature and other drivers i

Detection of Anopheles rivulorum-like, a member of the Anopheles funestus group, in South Africa.

BACKGROUND: The Anopheles gambiae sensu lato (s.l.) and Anopheles funestus s.l. species complexes contain the most important malaria vectors in Africa. Within the An. funestus group of at least 11 African species, the vector status of all but the nominal species An. funestus appears poorly investigated, although evidence exists that Anopheles rivulorum and Anopheles vaneedeni may play minor roles. A new species, An. rivulorum-like, was described from Burkina Faso in 2000 and subsequently also found in Cameroon and Zambia. This is the first paper reporting the presence of this species in South

Spatiotemporal distribution of insecticide resistance in Anopheles culicifacies and Anopheles subpictus in Sri Lanka.

The malaria situation in Sri Lanka worsened during the 1990s with the emergence and spread of resistance to the drugs and insecticides used for control. Chloroquine resistance has increased rapidly over this period, but adverse changes in malaria transmission are more closely associated with insecticide use rather than drug resistance. Insecticide susceptibility tests were routinely carried out in key anopheline vectors across the country for more than a decade. These sentinel data were combined with data collected by other research programmes and used to map the spatial and temporal trends of

Residual malaria transmission and the role of Anopheles arabiensis and Anopheles melas in central Senegal.

Understanding the behavior and ecology of local malaria vectors is essential for the effectiveness of the commonly used vector-targeted malaria control tools in areas of low malaria transmission. This study was conducted to determine species composition, biting behavior and infectivity of the major Anopheles vectors of Plasmodium falciparum in low transmission settings in central Senegal. Adult mosquitoes were collected using human landing catches during 2 consecutive nights and Pyrethrum Spray Catches in 30-40 randomly selected rooms, from July 2017 to December 2018 in 3 villages. Anopheline

Impact of a spatial repellent product on Anopheles and non-Anopheles mosquitoes in Sumba, Indonesia.

BACKGROUND: The East Nusa Tenggara province, Indonesia, contributed to 5% of malaria cases nationally in 2020, with other mosquito-borne diseases, such as dengue and filariasis also being endemic. Monitoring of spatial and temporal vector species compositions and bionomic traits is an efficient method for generating evidence towards intervention strategy optimization and meeting disease elimination goals. METHODS: The impact of a spatial repellent (SR) on human biting mosquitoes was evaluated as part of a parent cluster-randomized, double-blinded, placebo-controlled trial, in Sumba, East Nusa

Environmental factors associated with the malaria vectors Anopheles gambiae and Anopheles funestus in Kenya.

BACKGROUND: The Anopheles gambiae and Anopheles funestus mosquito species complexes are the primary vectors of Plasmodium falciparum malaria in sub-Saharan Africa. To better understand the environmental factors influencing these species, the abundance, distribution and transmission data from a south-eastern Kenyan study were retrospectively analysed, and the climate, vegetation and elevation data in key locations compared. METHODS: Thirty villages in Malindi, Kilifi and Kwale Districts with data on An. gambiae sensu strict, Anopheles arabiensis and An. funestus entomological inoculation rates

Comparative mitochondrial genome and phylogenetic analysis of malaria mosquitoes Anopheles hyrcanus and Anopheles messeae.

Anopheles mosquitoes serve as a significant vector for the transmission of malaria and arboviruses holding great medical importance, but their phylogenetic relationships have not yet been clearly elucidated. This study was the first to sequence and analyze the mitochondrial genomic features of An. hyrcanus and An. messeae, and to incorporate them into a phylogenetic analysis with 102 other species within the genus Anopheles. Both genomes contain 37 genes, including 22 tRNAs, 13 protein-coding genes and 2 rRNAs, as well as one control region. The genome characteristics, codon usage, and tRNA se

Natural Plasmodium infections in Anopheles darlingi and Anopheles benarrochi (Diptera: Culicidae) from eastern Peru.

Malaria, both Plasmodium falciparum (Welch) and Plasmodium vivax (Grassi & Feletti), has reemerged as a significant public health disease issue in Peru, especially in forested areas in the eastern part of the country. The spread of Anopheles darlingi Root, the principal South American malaria vector, into new areas of Peru is thought to be a factor in this resurgence. However, epidemiological evidence suggests that in malaria endemic areas of eastern Peru where An. darlingi does not occur, other species are involved in malaria transmission. The objective of this study was to analyze Anopheles

Delayed mating in the malaria vector Anopheles funestus compared to Anopheles arabiensis (Diptera: Culicidae).

Mating is a vital behavior for mosquito reproduction, yet it remains poorly understood under captive conditions. We examined the copulation dynamics of 2 key malaria vectors, Anopheles funestus sensu stricto Giles and Anopheles arabiensis Patton, under laboratory settings in Tanzania. We conducted observations in 24-h cycles and monitored copulation events and insemination of females, initially using flashlights for nighttime visibility, followed by red lights in subsequent experiments. We observed how variations in mosquito age and artificial lighting influenced mating success for these 2 mos

Spontaneous mutation rate estimates for the principal malaria vectors Anopheles coluzzii and Anopheles stephensi.

Using high-depth whole genome sequencing of F0 mating pairs and multiple individual F1 offspring, we estimated the nuclear mutation rate per generation in the malaria vectors Anopheles coluzzii and Anopheles stephensi by detecting de novo genetic mutations. A purpose-built computer program was employed to filter actual mutations from a deep background of superficially similar artifacts resulting from read misalignment. Performance of filtering parameters was determined using software-simulated mutations, and the resulting estimate of false negative rate was used to correct final mutation rate

Isolation of Japanese encephalitis virus from Anopheles annularis and Anopheles vagus in Lombok, Indonesia.

Three strains of Japanese encephalitis (JE) virus were recorded from mosquitoes collected in Lombok, Indonesia, during March 1979, from pools of Anopheles vagus, An. annularis and Culex tritaeniorhynchus respectively. This is believed to be the first report of isolation of JE virus from An. vagus. The frequencies of JE viral infection in zoophilic Anopheles species were higher than in Cx tritaeniorhynchus, the principle vector of JE virus in Asia. The low frequency of infection in Cx tritaeniorhynchus and the relatively infrequent raising of pigs may account for the low prevalence of JE neutra

Cryopreservation of Anopheles gambiae Larvae.

Mosquitoes remain a significant global health concern due to their role in transmitting deadly diseases including malaria, dengue, and yellow fever. Recent advances in genetic modification technologies have enabled researchers to better study disease transmission and is an emerging approach for vector control. However, a major bottleneck is the need for continuous rearing of each new strain, an intensive process requiring substantial manual effort and space with inherent risks of cross-contamination, genetic drift, and catastrophic loss. Cryopreservation offers a potential solution, where stra

Larvicidal effects of selected medicinal plant extracts against Anopheles arabiensis, Anopheles stephensi, and Aedes aegypti.

BACKGROUND: The emergence of resistance to synthetic (chemical) insecticides along with their harmful effects on human health, non-target organisms and the environment necessitates the development of new complementary bioinsecticides that are effective, environmentally friendly, biodegradable and target-specific. This study was undertaken to evaluate larvicidal activities of 80% methanol and n-hexane extracts of four plants that are traditionally used by communities against mosquitoes. METHODS: The dried plant parts of Ocimum lamiifolium, Amaranthus hybridus, Premna schimperi, and Lepidium sa

Evaluation of sustainable susceptibility to Plasmodium vivax infection among colonized Anopheles darlingi and Anopheles deaneorum.

BACKGROUND: The colonization of mosquitoes susceptible to Plasmodium vivax via direct membrane feeding assay (DMFA) has the potential to significantly advance our knowledge of P. vivax biology, vector-parasite interaction and transmission-blocking vaccine research. Anopheles darlingi and Anopheles deaneorum are important vectors of malaria in the Western Brazilian Amazon. Since 2018, well-established colonies of these species have been maintained in order to mass produce mosquitoes destined for P. vivax infection. Plasmodium susceptibility was confirmed when the colonies were established, but

Stable high-density and maternally inherited Wolbachia infections in Anopheles moucheti and Anopheles demeilloni mosquitoes.

Wolbachia, a widespread bacterium that can reduce pathogen transmission in mosquitoes, has recently been reported to be present in Anopheles (An.) species. In wild populations of the An. gambiae complex, the primary vectors of Plasmodium malaria in Sub-Saharan Africa, Wolbachia DNA sequences at low density and infection frequencies have been detected. As the majority of studies have used highly sensitive nested PCR as the only method of detection, more robust evidence is required to determine whether Wolbachia strains are established as endosymbionts in Anopheles species. Here, we describe hig

Rapid discrimination between Anopheles gambiae s.s. and Anopheles arabiensis by High-Resolution Melt (HRM) analysis.

There is a need for more cost-effective options to more accurately discriminate among members of the Anopheles gambiae complex, particularly An. gambiae and Anopheles arabiensis. These species are morphologically indistinguishable in the adult stage, have overlapping distributions, but are behaviorally and ecologically different, yet both are efficient vectors of malaria in equatorial Africa. The method described here, High-Resolution Melt (HRM) analysis, takes advantage of minute differences in DNA melting characteristics, depending on the number of incongruent single nucleotide polymorphisms
