Anaphylaxis induced by oral methylprednisolone in a 10-year-old boy.

Corticosteroids are potent anti-inflammatory and anti-allergic agents used in the treatment of various inflammatory diseases, including allergic diseases. Allergic reactions caused by corticosteroids, however, have been reported. Among these, delayed reactions to topical steroids are more common, whereas immediate reactions to systemic steroids are rare. Herein, we report the case of a 10-year old boy with methylprednisolone-induced anaphylaxis, in which the patient had a positive oral challenge test result. Physicians should be aware of the possibility of anaphylaxis or other allergic hyperse

The effects of anti-PAF and other agents on the nasal symptoms in sensitized guinea pigs.

To define the role of platelet activating factor (PAF) in allergic rhinitis, we examined the effects of anti-PAF agents (WEB2086, SM10661) on the changes of nasal airway resistance (NAR) and nasal symptoms after topical antigen challenge in actively sensitized guinea pigs and compared these with the changes brought by anti-leukotriene (LT) agent (FPL 55712), 5-lipoxygenase inhibitor (E6080), anti-allergic agent (azelastine), and anti-histamine agent (mepyramine maleate). We noted biphasic increase in NAR after antigen challenge; the first peak, 146.3 +/- 4.3% at 10 min and the second peak, 163

Recent patents and emerging therapeutics in the treatment of allergic conjunctivitis.

Ocular allergy is an inflammatory response of the conjunctival mucosa that also affects the cornea and eyelids. Allergic conjunctivitis includes seasonal allergic conjunctivitis (SAC), perennial allergic conjunctivitis (PAC), vernal keratoconjunctivitis (VKC), atopic keratoconjunctivitis (AKC) and giant papillary conjunctivitis (GPC). In general, allergic conditions involve mast cell degranulation that leads to release of inflammatory mediators and activation of enzymatic cascades generating pro-inflammatory mediators. In chronic ocular inflammatory disorders associated with mast cell activati

Ocular surface disease: a case of vernal keratoconjunctivitis.

Vernal keratoconjunctivitis (VKC) is a recurrent, chronic allergic inflammation of the ocular surface associated with remodeling of the cornea and conjunctiva, with a potential to adversely affect the patient's vision and quality of life. It affects mainly children and young adults. The purpose of this interventional case report is to present a typical case of limbal VKC, which was unresponsive to antiallergic treatment alone. A 5-year-old boy, who was previously unsuccessful with conventional anti-allergic therapy alone, presented with clinicopathological features pathognomonic of limbal VKC.

Anti-Allergic Potentials of Actinodaphne angustifolia: Integrated In Vivo Evaluation and In Silico Analysis of Bioactive Compounds.

Actinodaphne angustifolia (Lauraceae), traditionally used for treating inflammatory and allergic disorders was investigated for its anti-allergic potential using integrated in vivo and in silico approaches. Safety evaluation in accordance with OECD guidelines confirmed the non-toxic nature of the ethanolic leaf extract. In a toluene 2,4-diisocyanate (TDI)-induced allergic model, the extract reduced allergic symptoms, including sneezing, nasal scratching, and nasal scores, also normalizing elevated white blood cell counts in blood and bronchoalveolar lavage fluid. A marked reduction in serum Ig

Antihistamines in late-phase clinical development for allergic disease.

Allergic rhinitis (AR) is a global health concern and shares a high comorbidity with asthma. Recent research suggests that different allergic diseases, such as AR, asthma, allergic conjunctivitis and chronic idiopathic urticaria (CIU), are evoked by common pathological mechanisms characterised by the release of histamine and other inflammatory mediators. Although H(1) receptor antagonists are the mainstay of therapy for allergic disease, the unacceptably high incidence of anticholinergic and CNS-related side effects of first-generation H(1) antagonists led to the search for improved second-gen

Anti-allergic properties of Cissampelos sympodialis and its isolated alkaloid warifteine.

Development of new agents capable of regulating eosinophilic inflammation can uncover novel therapeutic approaches for the treatment of allergic diseases, such as asthma. Here, we evaluated the anti-allergic properties of an extract of the Brazilian Menispermaceae Cissampelos sympodialis, focusing on its effects on allergic eosinophilia. By studying two models of allergic inflammation, an asthma model and the allergic pleurisy in actively sensitized Balb/c mice, we observed that the oral pre-treatment with C. sympodialis reduced pleural eosinophil influx triggered by allergen challenge in a do

Anti-allergic effect of Perilla frutescens and its active constituents.

The leaves of Perilla frutescens Britton (Labiatae) are one of the most popular garnishes in Japan, used as an antidote for fi sh and crab meat allergy or as a food colorant. The present study was conducted to evaluate its anti-allergic effect and to identify its active constituents using mice ear-passive cutaneous anaphylaxis (PCA)-reaction. 48 h after the cutaneous injection of anti-ovalbumin serum into the ears of mice, ovalbumin and evansblue dye were intravenously injected. Perilla was extracted with boiling water, and intraperitoneally injected 15 min before ovalbumin-treatment. Thirty m

Hypersensitivity and adverse reactions to biologics for asthma and allergic diseases.

Introduction: The development of new biologic agents has provided definite therapeutic advances but, like with any new medications, safety remains a concern.Areas covered: Using PubMed, we reviewed the literature on the adverse effects (AE) to five biologics approved for asthma and/or allergic diseases: one anti-IgE (omalizumab), three anti-IL5 (mepolizumab, reslizumab, benralizumab), and one anti-IL4 (dupilumab).Expert opinion: Biologic agents approved for asthma and allergic diseases are generally safe. Most common AE are benign and tolerated, though long-term safety is lacking for most of t

Anti-allergic effects of cnidii monnieri fructus (dried fruits of Cnidium monnieri) and its major component, osthol.

Anti-allergic effects (types I and IV) of the 70% ethanol extract (CM-ext) obtained from Cnidii Monnieri Fructus (dried fruits of Cnidium monnieri) were investigated on 48 h homologous passive cutaneous anaphylaxis (PCA), 2, 4-dinitrofluorobenzene (DNFB)-induced contact dermatitis and picryl chloride (PC)-induced contact dermatitis in experimental animals. CM-ext showed inhibitory effects on these allergic models. Osthol isolated from CM-ext also had the inhibitory effects. These results suggested that Cnidii Monnieri Fructus might be useful as an agent for allergic diseases and that its anti-

Human skin safety test of green tea cell extracts in condition of allergic contact dermatitis.

Various kinds of positive effects of green tea extracts had been studied for long time which included anti-inflammation, anti-aging, and cardiometabolic effects. Although topical steroid and non-steroidal calcineurin inhibitors may control clinical symptoms of allergic contact dermatitis, some of patients also present allergic reaction to these topical agents. Therefore, we have tried green tea extracts for managing this skin disorder with expectation of anti-inflammatory effect without potential side effects including skin irritation and toxic responses. The toxicity test of green tea extract

A case of IgG4-related dacryoadenitis that regressed without systemic steroid administration.

There are no reports on the effect of anti-allergic agents against IgG4-related disease. We herein report a case of IgG4-related dacryoadenitis that is believed to have regressed due to the administration of anti-allergic agents. A 57-year-old woman consulted us because of bilateral temporal upper eyelid swelling and induration. She had also been suffering from allergic rhinitis and allergic conjunctivitis for 20 years. We performed an incisional biopsy of the lesion. With respect to the pathology, extranodal marginal zone lymphoma of mucosa-associated lymphoid tissue type was strongly suspect

Anti-allergic properties of the bromeliaceae Nidularium procerum: inhibition of eosinophil activation and influx.

New therapeutic approaches for the treatment of allergic diseases can be aided by the development of agents capable of regulating eosinophilic leukocytes. Here, we evaluated the anti-allergic properties of a crude extract of the Brazilian bromeliaceae Nidularium procerum, focusing on its effects on allergic eosinophilia. By studying allergic pleurisy in actively sensitized C57Bl/6 mice, we observed that pretreatment with N. procerum (2 mg/kg; i.p.) reduced pleural eosinophil influx triggered by allergen challenge. N. procerum was also able to reduce lipid body numbers found within infiltrating

Emerging drugs for ocular allergy.

Ocular allergy is a common condition that usually affects the conjunctiva of the eye and is, therefore, often referred to as allergic conjunctivitis. The severity of the disease can range from mild itching and redness, as seen in seasonal allergic conjunctivitis, to the more severe, sight-threatening forms such as vernal and atopic keratoconjunctivitis. The central mechanism in the pathogenesis of these diseases is IgE-mediated mast cell degranulation and activation of T lymphocytes, eosinophils and conjunctival structural cells. The pharmacotherapy of allergic conjunctivitis consists of sever

Natural Anti-inflammatory and Anti-allergy Agents: Herbs and Botanical Ingredients.

Allergies have been known to be an abnormally vigorous immune response in which the immune system fights off an allergen or antigen, initiating mast cells to release histamine into the blood. Substances that prevent mast cells from releasing histamine are considered antiallergic agents. The drugs utilized to treat allergy are mast cell stabilizers, steroids, anti-histamine, leukotriene receptor antagonists, and decongestants. Anti-histamine drugs have side effects such as drowsiness, confusion, constipation, difficulty urinating, blurred vision, etc. The use of medicinal plants for the effecti

A Biotinylated Heptapeptide Containing a D-Amino Acid Suppresses Both IgE-Dependent and -Independent Allergic Edema in Mice.

We previously found that, among three biotinylated heptapeptides (Peptide 1, Peptide 2, and Peptide 3) derived from the amino acid sequence of Asp-hemolysin, a protein toxin produced by Aspergillus fumigatus, Peptide 2, which contains a d-amino acid, exhibits potent anti-allergic activity in vitro and in vivo. In this study, we evaluated the effects of Peptide 2 on immunoglobulin E (IgE)-dependent allergic edema induced by ovalbumin (OVA) in OVA-specific IgE transgenic mice, IgE-independent allergic edema induced by compound 48/80 (C48/80), and nonallergic inflammatory edema induced by histami

Incidence of infusion reactions to anti-neoplastic agents in early phase clinical trials: The MD Anderson Cancer Center experience.

Infusion reactions (IRs) to anti-neoplastic agents require prompt recognition and immediate treatment to avert significant complications. We conducted a retrospective review of the medical records of consecutive patients who received anti-neoplastic therapy in the outpatient treatment center of the Department of Investigational Cancer Therapeutics from January 1, 2013 to November 30, 2013. Of the 597 patients who received treatment, 9 (1.5 %) had IRs (all ≤ grade 2). The most common IRs observed on first occurrence were chills (n = 5), itching, rash, and facial flushing (n = 3 each). There wer

[Effects of anti-PAF agents on nasal response after allergen challenge in guinea pigs].

We previously reported that Platelet-activating factor (PAF) activities were detected in nasal lavage fluids from patients with allergic rhinitis and in ovalbumin (OA) sensitized guinea pigs after topical allergen challenge. In guinea pigs, a topical application of PAF produced an increase in nasal vascular permeability which was inhibited by a PAF-antagonist (CV3988). To define the role of PAF in allergic rhinitis, we examined the effects of anti-PAF agents (WEB2086) and anti-allergic agents (Azelastine) on nasal airway resistance (NAR) and nasal symptoms in actively sensitized guinea pigs.
