This study was performed to investigate the role of cell-mediated immunity in primary glomerular diseases (PGD), by estimating natural killer (NK) activity from peripheral blood mononuclear cells (PBMC) assayed against K562 cells, and antibody-dependent cell-mediated cytotoxicity (ADCC) activity from PBMC assayed against chicken red blood cells. The proportion of PBMC was examined by flow cytometry using the monoclonal antibodies OKT and Leu series. NK activity in patients with nephrotics of minimal change nephrotic syndrome (MCNS), membranous glomerulonephritis (MGN) and membranoproliferative

Three anti K562 monoclonal antibodies (MAb) 4.3, 4.6 and 4.8 reacting predominantly with cells of myeloid lineage, were tested for antibody dependent cellular cytotoxicity (ADCC) and complement dependent cytotoxicity (CDC). MAb 4.6 (IgG3k) effectively mediated ADCC against K562 cells and fresh leukemic targets with effectors from healthy donors. However, for ADCC on chronic myeloid leukemia (CML) targets, effectors from CML patients in remission needed modulation with IL-2. All MAb showed significant CDC against peripheral blood (PB) and bone marrow (BM) cells obtained from CML patients in chr

Natural killer (NK) cells are critical innate effector cells against human immunodeficiency virus type 1 (HIV-1) transmission and pathogenesis. Among the diverse NK cell functions, antibody-dependent cellular cytotoxicity (ADCC) is the most critical for HIV-1 control. Nevertheless, multiple reports have demonstrated dysfunctional NK cell activity in people living with HIV-1 (PLWH), including impaired ADCC. As a putative mechanism, CD16 signaling activation in PLWH is perturbed via ADAM17-mediated CD16 downregulation, and ADAM17 blockade can reverse CD16 downregulation in PLWH NK cells. However

Double-negative T (DNT) cells (TCRαβ+CD4-CD8-NK1.1-/CD56-) exhibit strong tumor-killing capabilities. Our single-cell transcriptome analysis has revealed high Fcer1g expression in DNT cells, but its role in tumor immunity remains unclear. In this study, we demonstrated that IgG1 stimulation significantly upregulated IgG Fc receptors and cytotoxic molecules in DNT cells, enhancing their cytotoxicity against MC38 tumor cells in vitro. FcεRIγ-deficient DNT cells failed to respond effectively to IgG1 stimulation. Inhibiting the downstream spleen tyrosine kinase (Syk) of FcεRIγ reduced cytotoxicity

Engineering antibodies to elicit antibody-dependent cell-mediated cytotoxicity (ADCC) can be used to eliminate target cells. Here, we described how single domain VHH arms on a bispecific antibody (BsAb) format can be engineered to modulate cell binding and ADCC activity. The BsAbs, comprising two anti-epidermal growth factor receptor (EGFR) nanobodies, 7D12 and EGA1, were engineered onto a human IgG1 Fc domain in monovalent, bivalent, and tandem formats. While 7D12 had a stronger ADCC activity than EGA1, the tandem 7D12-EGA1 mediated a significantly stronger ADCC activity without significant c

Zika virus (ZIKV) has become a global public health issue due to its teratogenicity and ability to cause Guillain-Barré syndrome in adults. Although anti-ZIKV envelope protein neutralizing antibodies correlate with protection, the non-neutralizing function of ZIKV antibodies including antibody-dependent cell-mediated cytotoxicity (ADCC) is incompletely understood. To study the role of ADCC antibodies during ZIKV infections, we generated a stably transfected, dual-reporter target cell line with inducible expression of a chimeric ZIKV prM-E protein on the cell surface as the target cell for the

We demonstrate that the FATAL cytolysis assay can be adapted into a rapid and fluorometric antibody-dependent cellular cytotoxicity assay (RFADCC). The RFADCC relies on double-staining target cells with a membrane dye (PKH-26) and a viability dye (CFSE) prior to the addition of antibody and effector cells. We used the RFADCC to assess dose-dependent and envelope-specific anti-human immunodeficiency virus (HIV) ADCC responses mediated by monoclonal antibody-2G12 and human sera. Using the assay, we also detected early anti-simian immunodeficiency virus (SIV) ADCC responses in rhesus macaques inf

Antibody Dependent Cell Cytotoxicity (ADCC) appears to be involved in Autoimmune Thyroid Disease (AITD). Homologous system may trigger non-specific reactions which might obscure specific ADCC. Heterologous target cells may be useful for studying ADCC, provided relevant antigen(s) are expressed. We therefore tested the capacity of porcine thyroid cells to elicit ADCC reaction in the presence of sera from various patients with AITD. Porcine thyroid cells were used in a 4-hr chromium release assay in the presence of 1/10 heat inactivated human sera and human peripheral blood lymphocytes at a 30:1

Anti-HER2 monoclonal antibody (mAb) is the standard first-line therapy for advanced HER2+ gastric cancer. However, resistance to anti-HER2 therapy remains a significant clinical challenge. In this study, we identified a novel resistance mechanism to anti-HER2 therapy in gastric cancer and proposed a strategy to enhance therapeutic efficacy by activating T cells. The association between intratumoral immune cells and clinical responses to anti-HER2 therapy in gastric cancer was investigated. Peripheral blood mononuclear cells (PBMCs) were co-cultured with HER2+ gastric cancer cell lines or organ

Previous studies in our laboratory characterized a panel of highly mutated HIV-specific conformational epitope-targeting antibodies (Abs) from a panel of HIV-infected long-term nonprogressors (LTNPs). Despite binding HIV envelope protein and having a high number of somatic amino acid mutations, these Abs had poor neutralizing activity. Because of the evidence of antigen-driven selection and the long CDR3 region (21 amino acids [aa]), we further characterized the epitope targeting of monoclonal Ab (MAb) 76-Q3-2C6 (2C6). We confirmed that 2C6 binds preferentially to trimeric envelope and recogni

A growing body of evidence underscores the role of antibody-dependent cellular cytotoxicity (ADCC) in antiviral immunity. Yet, the mechanisms underlying the ability of certain antibodies (Abs) to mediate potent ADCC activity remain poorly characterized. In particular the contribution of features within the antigen-binding Fab region remains largely unexplored. In this study, we leveraged a collection of 142 SARS-CoV-2 monoclonal Abs to systematically dissect the determinants of ADCC activity. We analyzed their epitope domain target, binding characteristics, neutralization potency, somatic hype

Natural killer (NK) activity of peripheral mononuclear cells (PMNC) from patients with adult T-cell leukemia (ATL), anti-HTLV-I antibody positive healthy carriers, and anti-HTLV-I antibody negative healthy persons (Ab-negative persons) was investigated using various target cells. PMNC from patients with ATL and healthy carriers had reduced NK activity against the NK-sensitive non-HTLV-I producing target cells, compared with the controls. In contrast, PMNC from patients with ATL, healthy carriers, and Ab-negative persons did not exhibit significant NK cell lysis against HTLV-I producing cells.

Ab-mediated signaling in tumor cells and Ab-dependent cell-mediated cytotoxicity (ADCC) are both considered as relevant effector mechanisms for Abs in tumor therapy. To address potential interactions between these two mechanisms, we generated HER-2/neu- and CD19-derived chimeric target Ags, which were expressed in experimental tumor target cells. HER-2/neu-directed Abs were documented to mediate effective ADCC with both mononuclear cells (MNCs) and polymorphonuclear granulocytes (PMNs), whereas Abs against CD19 were effective only with MNCs and not with PMNs. We generated cDNA encoding HER-2/C

T-lymphocyte subpopulations and antibody-dependent, cell-mediated cytotoxicity (ADCC) were studied in patients with alopecia areata and universalis. The proportions of T-cells with the receptors for IgG (Tg) and T-cells with receptor for IgM (Tm) were determined in 16 cases. The ADCC of unfractionated lymphocytes was examined in 5 cases. A higher proportion of Tg-cells (T-cell with low affinity for sheep red cells) and a more pronounced ADCC of the peripheral lymphocytes were found in patients with alopecia areata and universalis than in age- and sex-matched normal donors.

BACKGROUND: Seasonal influenza vaccines aim to induce strain-specific neutralizing antibodies. Non-neutralizing antibodies may be more broadly cross-reactive and still protect through mechanisms including antibody-dependent cell-mediated cytotoxicity (ADCC). Influenza vaccines may stimulate ADCC antibodies in adults, but whether they do so in children is unknown. Here we examined how vaccination affects cross-reactive ADCC antibody responses in children after receipt of inactivated trivalent vaccine (IIV3) or quadrivalent live-attenuated vaccine (LAIV4).
METHODS: Children aged 5-17 were recru

Natural killer (NK) cells are key components of the innate immune system. In murine cardiac transplant models, donor-specific antibodies (DSA), in concert with NK cells, are sufficient to inflict chronic allograft vasculopathy independently of T and B cells. In this study, we aimed to determine the effector mechanism(s) required by NK cells to trigger chronic allograft vasculopathy during antibody-mediated rejection. Specifically, we tested the relative contribution of the proinflammatory cytokine interferon gamma (IFN-γ) versus the contact-dependent cytotoxic mediators of perforin and the CD9

Chronic-active antibody mediated rejection (c-aABMR) contributes significantly to late renal allograft failure. The antibodies directed against donor-derived antigens, e.g. anti-HLA antibodies, cause inflammation at the level of the microvascular endothelium. This is characterized by signs of local activation of the complement system and accumulation of immune cells within the capillaries. Non-invasive biomarkers of c-aABMR are currently not available but could be valuable for early detection. We therefore analyzed the activation profiles of circulating T and B cells, NK cells and monocytes in

CAR-T, TIL, and TCR-based T cell immunotherapies have revolutionized cancer treatment; however, their clinical application remains limited by severe adverse effects such as CRS, ICANS, and HLH, as well as the risk of GvHD in the allogeneic setting. In contrast, NK cell-based therapies have emerged as a safer alternative with minimal adverse effects and can be developed from diverse cellular sources. Nevertheless, clinical translation of NK cell therapies is challenged by difficulties in large-scale expansion, donor variability, and high sensitivity to cryopreservation. In this study, we establ
