Dolastatins are a class of naturally occurring antimitotic peptides that have inspired the development of some of the most active and widely used anticancer agents. Here, we report on the development of synthetic methodologies for the preparation of parallel libraries of small peptides inspired by dolastatin 15 and its analogs cemadotin and tasidotin. The approaches rely on the use of either one or multiple Ugi-multicomponent reactions to generate amide N-substituted dolastatin-like skeletons, which allow the exploration of tertiary amide chemical spaces that have not been assessed previously.

OBJECTIVE: Cancer continues to pose a significant worldwide health concern, underscoring the need for the identification of innovative, safer, and more effective therapeutic agents. Phytochemicals obtained from medicinal plants are progressively acknowledged for their anticancer efficacy. This work investigates the antimitotic and antiproliferative properties of the ethanolic extract of Benincasa hispida seeds (EeBHS), known as winter melon, from the Cucurbitaceae family.
METHODS: Antimitotic activity was assessed using the Allium cepa root tip meristem test and the seed germination assay. Th

Background: Antimitotic agents are very successful antitumor therapies, but lack tumor selectivity, causing toxicity. Antitumor alkylphospholipids (APLs) selectively accumulate in tumor cells but display low potencies. Hypothesis: Incorporating APL moieties onto antimitotic N-trimethoxyphenyl naphthalene sulfonamides (TMNS) might afford SMDCs with the potency of antimitotics and the tumor selectivity of APLs. Methods: 24 new TMNSs with spacers of different lengths (4 to 9 atoms) and nature (alkanes or ethers) on the sulfonamide nitrogen and capped with phosphorus-containing groups such as diet

Diazepam (DZP) is a widely prescribed drug for central nervous system disorders. However, it also exhibits a significant inhibitory effect on tubulin and the mitotic kinesin Eg5. These novel mitotic mechanisms explain its cytotoxicity and provide valuable insights for designing safer, more targeted antimitotic agents. Studies in mammalian cancer and noncancerous cell lines, including HeLa, MCF-7, A549, and L929, demonstrated that DZP inhibits cell proliferation in a concentration-dependent manner, with IC50 values ranging from 42 μM to 76 μM. Fluorescence spectroscopy confirmed direct binding

Pharmacological blockade of fibroblastic proliferation after glaucoma filtration surgery by using antimitotic agents in different formulations (liposomes, nanospheres) is of great clinical interest. However, only limited comparative data are available on the effect of encapsulated drugs on intraocular pressure (IOP) after filtering surgery. Therefore we have studied the effect on IOP of liposomes, nanospheres and a solution of mitoxantrone (MTO), a well-known antimitotic, in rabbits before and after sclerectomy. MTO in solution form, as well as in a liposome formulation, improved the outcome o

Polyploid giant cancer cells (PGCC) are common in tumors and have been associated with resistance to cancer therapy, tumor relapse, malignancy, immunosuppression, metastasis, cancer stem cell production, and modulation of the tumor microenvironment. However, the molecular mechanisms that cause these cells to form are not yet known. In this study, we discover that Aurora kinases are synergistic determinants of a switch from the proliferative cell cycle to polyploid growth and multinucleation in lung cancer cell lines. When Aurora kinases were inhibited together, lung cancer cells uniformly grew

MYCN amplification occurs in approximately 20-30% of neuroblastoma patients and correlates with poor prognosis. The TH-MYCN transgenic mouse model mimics the development of human high-risk neuroblastoma and provides strong evidence for the oncogenic function of MYCN. In this study, we identified mitotic dysregulation as a hallmark of tumor initiation in the pre-cancerous ganglia from TH-MYCN mice that persists through tumor progression. Single-cell quantitative-PCR of coeliac ganglia from 10-day-old TH-MYCN mice revealed overexpression of mitotic genes in a subpopulation of premalignant neurob

Isoniazid is a safe and very effective antituberculosis drug. Antimitotic agents routinely cause alopecia. Drug-induced alopecia is usually reversible upon withdrawal of the drug. Isoniazid, thiacetazone and ethionamide are the antituberculosis drugs which have been associated with alopecia. Isoniazid-induced alopecia was observed in one case and confirmed by the finding that hair growth resumed when drug removed from the regimen.

The role of microtubules in the thyrotropin- or adenosine 3',5' cyclic monophosphate (cyclic AMP)-stimulated accumulation of cytoplasmic colloid droplets and secretion of iodine from the mouse thyroid gland has been investigated by means of different classes of agents that affect the stability of microtubules. The onset of inhibition of secretion by colchicine, the uptake of colchicine-(3)H by thyroid lobes, and the binding of colchicine-(3)H to thyroidal soluble protein are shown to have similar time courses Colloid droplet accumulation is also inhibited and does not readily resume upon remov

Reported therapies for primary pterygium have included surgical excision with or without adjunct radiation, antimitotic agents, and conjunctival transplants. The effectiveness of these strategies may be limited by recurrences, side effects of the treatment, or expensive and time-consuming procedures. We report the results of using thermal cautery to treat primary pterygium in three patients. In all cases the pterygia were successfully eliminated without recurrence.

Stroke increases neurogenesis. The authors investigated whether neural stem cells or progenitor cells in the adult subventricular zone (SVZ) of rats contribute to stroke-induced increase in neurogenesis. After induction of stroke in rats, the numbers of cells immunoreactive to doublecortin, a marker for immature neurons, increased in the ipsilateral SVZ and striatum. Infusion of an antimitotic agent (cytosine-beta-D-arabiofuranoside, Ara-C) onto the ipsilateral cortex eliminated more than 98% of actively proliferating cells in the SVZ and doublecortin-positive cells in the ipsilateral striatum

Colchicine-site-binding antimicrotubule agents remain promising antimitotic compounds, particularly when structural modification enables retention of tubulin-directed activity while modulating multidrug resistance (MDR) phenotypes. Building on the N-benzyl colchicine scaffold, we synthesised and evaluated a focused series of N-substituted analogues in which the native N-acetyl group was replaced by adamantylmethyl, ferrocenylmethyl or ruthenocenylmethyl, or isomeric o-, m-, and p-ferrocenylbenzyl substituents. The derivatives displayed low-nanomolar antiproliferative activity in drug-sensitive

Methotrexate is an antimitotic and immunosuppressive agent used for the treatment of cancer, psoriasis, and rheumatologic disorders and for the termination of ectopic pregnancies. Physicians advising patients on the use of methotrexate need to be aware of its possible side effects, including photosensitivity. We present a patient who received methotrexate for the termination of an ectopic pregnancy and experienced a severe reactivation of her sunburn. The literature was reviewed on the types of photosensitivity and their relationship to methotrexate.

Leiodermatolide is an antimitotic macrolide isolated from the marine sponge Leiodermatium sp. whose potentially novel tubulin-targeting mechanism of action makes it an exciting lead for anticancer drug discovery. In pursuit of a sustainable supply, we report a highly stereocontrolled total synthesis (3.2% yield) based on a convergent sequence of palladium-mediated fragment assembly and macrolactonization. Boron-mediated aldol reactions were used to configure the three key fragments 2, 5, and 6 by employing the appropriate enantiomer of the lactate-derived ketone 7.

Pyoderma gangrenosum (PG) is a neutrophilic dermatosis of unknown origin. Systemic agents occasionally administered provide either incomplete long-term control of the disease or have been associated with serious adverse side effects after chronic administration. We present two patients with PG successfully treated with low-dose colchicine. Antimitotic, anti-inflammatory and immunomodulating properties of colchicine might account for its beneficial effects in PG patients. Colchicine is effective and well tolerated in low doses by most patients. In addition, it is inexpensive and safer for long-

Despite advances in the current treatment strategies for breast cancer management, chemotherapeutic resistance and recurrence are now recognised as a substantial global health problem. Microtubule-targeting agents are key clinical candidates in breast cancer therapy that abrogate tubulin dynamics, lead to arrest in mitosis, and enhance apoptosis of cancer cells. However, traditional agents like taxanes and vinca alkaloids suffer from the serious issue of toxicity, suboptimal pharmacokinetics, and multidrug resistance. Novel scaffolds and molecular hybrids have been reported to exhibit potent a
