Antimutagenic effects of stobadine: review of results.

The paper summarizes the results of our previously published studies testifying the hypothesis of the antimutagenic effect of stobadine (STB) in vivo and in vitro. The micronucleus test was used in in vivo experiments with ICR mice. Oral pretreatment with STB significantly decreased the mutagenic effect of cyclophosphamide (CP) in a concentration-dependent way. The protective effect of STB was confirmed in fetuses of CP-treated mice. STB pretreatment exerted also a radioprotective effect in Co60-irradiated mice. The ineffectiveness of STB posttreatment is indicative of its effect operative in

Antimutagenic action of the live yeast can be transmitted to the offspring of Drosophila melanogaster. A genetic study using the wing spot assay.

The present study evaluates whether the protective effect of live yeast (LY) against direct and indirect mutagenic agents, persists in the offspring from individuals fed with LY. The wing-spot test in Drosophila was used; four different mates were performed: a) neither females nor males were fed with LY-enriched food (NLYxNLY); b) only females were fed (LYxNLY); c) males were fed (NLYxLY) or d) both progenitors were fed (LYxLY). Results confirm that LY strongly stimulates fecundity in females but not in males and provides strength to the egg for survive. A greater reduction in mutation rate wa

Assessment of antimutagenic effects of stobadine dihydrochloride on MNNG-induced mutations in Chinese hamster cells V79.

Mutagenicity of N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) and antimutagenic effect of antioxidant stobadine (STB) were investigated by so called HGPRT/V79 system. Cells were treated by STB before, during and after MNNG-treatment. Our results showed that the highest antimutagenic effect of STB was observed if the drug was given as a pretreatment before exposure of cells to MNNG. This effect was not concentration-dependent within the framework of 1.5-9 mmol. All other combinations of MNNG- and STB-treatment led to the weaker but statistically significant decrease of 6-TGr mutations. Inhibition

Preliminary assessment of mutagenic and anti-mutagenic potential of some aminoalkanolic derivatives of xanthone by use of the Vibrio harveyi assay.

The Vibrio harveyi assay was used to evaluate mutagenic and anti-mutagenic effects of four new aminoalkanolic derivatives of xanthone with anticonvulsant activity, to select the potentially safe compounds for further in vivo studies in animal models. The study showed that at a concentration of 40 ng/ml the test compounds were not mutagenic. Additionally, two of the investigated compounds, namely the (R,S)-N-methyl-1-amino-2-propanol derivative of 6-methoxyxanthone (compound III) and the (R)-N-methyl-2-amino-1-butanol derivative of 7-chloroxanthone (compound IV) were strong inhibitors of the mu

Radioprotection by antioxidants.

The role of reactive oxygen species in ionizing radiation injury and the potential of antioxidants to reduce these deleterious effects have been studied in animal models for more than 50 years. This review focuses on the radioprotective efficacy and the toxicity in mice of phosphorothioates such as WR-2721 and WR-151327, other thiols, and examples of radioprotective antioxidants from other classes of agents. Naturally occurring antioxidants, such as vitamin E and selenium, are less effective radioprotectors than synthetic thiols but may provide a longer window of protection against lethality a

Anticarcinogenic effects of selenium.

Selenium (Se) exerts its anticarcinogenic effects by multiple mechanisms. In the physiological dosage range, Se appears to function as an antimutagenic agent, preventing the malignant transformation of normal cells and the activation of oncogenes. These protective effects of Se seem to be primarily associated with its presence in the glutathione peroxidases, which are known to protect DNA and other cellular components from damage by oxygen radicals. Selenoenzymes are also known to play roles in carcinogen metabolism, in the control of cell division, oxygen metabolism, detoxification processes,

N-acetylcysteine (NAC) and glutathione (GSH): antioxidant and chemopreventive properties, with special reference to lung cancer.

Lung cancer arises as a focal transformation of chronically injured epithelium with cigarette smoke as one of its well-recognized causes. Apart from oxidants (free radicals), cigarette smoke contains such a multitude of (pre)carcinogens that it is astonishing that not every heavy smoker becomes a victim of malignancy. This points to the interindividual variability in susceptibility to carcinogens; several lines of evidence suggest that metabolic factors are involved in such variability. Metabolism of carcinogens as well as the subsequent (multi)steps of carcinogenesis are affected by host fact
