Antipyretic activity of copper salicylate.

Thermal responses to copper II salicylate (200 mg/kg iv) were investigated in febrile rabbits (treated with E. coli lipopolysaccharide) at 3 ambient temperatures (Ta) of 5, 20, 28 degrees C. The compound produced antipyresis, which unlike that induced by sodium salicylate (200 mg/kg iv), increased with the drop of Ta. This antipyretic activity was accompanied by enhanced heat elimination which became most evident in thermoneutral conditions. The metabolic rate was intensified after treatment with both compounds. Possible mechanisms responsible for copper salicylate antipyresis are discussed.

Effect of antipyretics on performance of influenza surveillance in Ghana.

BACKGROUND: The World Health Organization's case definition for influenza-like illness (ILI) includes a measured temperature of ≥38°C. We conducted this study to assess the effect of antipyretics on performance of ILI surveillance in Ghana. METHODS: A cross-sectional study was conducted in two districts of Ghana from September 2013 to May 2014. We collected epidemiological data and respiratory specimens from an expanded ILI case definition, which included patients presenting to health facilities with measured temperature ≥38°C or reported fever (but afebrile at the time of evaluation), and co

Electron attachment to antipyretics: possible implications of their metabolic pathways.

The empty-level structures and formation of negative ion states via resonance attachment of low-energy (0-15 eV) electrons into vacant molecular orbitals in a series of non-steroidal anti-inflammatory drugs (NSAIDs), namely aspirin, paracetamol, phenacetin, and ibuprofen, were investigated in vacuo by electron transmission and dissociative electron attachment (DEA) spectroscopies, with the aim to model the behavior of these antipyretic agents under reductive conditions in vivo. The experimental findings are interpreted with the support of density functional theory calculations. The negative an

Eugenol: antipyretic activity in rabbits.

The analgesic agent eugenol reduced fever when given intravenously in small doses to rabbits made febrile by interleukin 1. To test this observation further, the drug was given intragastrically and its antipyretic effect was compared with that of a molar equivalent dose of acetaminophen. Eugenol was more effective in reducing fever than acetaminophen. Tests with intracerebroventricular injections indicated that eugenol could act centrally to reduce fever in doses much smaller than those which were effective peripherally. Other tests with the presumed fever mediators sodium archidonate and pros

Gastrointestinal effects of antipyretic analgesics.

Aspirin and paracetamol (acetaminophen) are the most commonly used minor analgesics, but their effects on the gastrointestinal tract differ widely. Aspirin is significantly associated with major upper gastrointestinal hemorrhage, whereas acetaminophen is not. Short-term use of aspirin produces erythema, erosions, and occasionally ulcers; acetaminophen use does not. Chronic gastric ulcer is linked to aspirin intake in patients with rheumatic disease, and epidemiologically in all heavy aspirin users; paradoxically, in only one epidemiologic study was a significant association found between aceta

History of antipyretic analgesic therapy.

The use of naturally occurring plant materials for the relief of pain dates back to 3,000 B.C., although rapid advances in antipyretic analgesic therapy have been made more recently. Salicylic acid was synthesized in 1860, and the pyrazolone group, first represented by antipyrine, in 1883. Phenacetin was developed in 1886. Acetaminophen has been in use since the 1890s.

Renal effects of antipyretic analgesics.

Most antipyretic analgesics can cause acute nephrotoxic effects, including acute tubular necrosis, acute interstitial nephritis, glomerular toxicity, and functional changes, such as "salicyl edema," following large doses of sodium salicylate. Most functional changes are related to acute suppression of prostaglandin synthesis, "the acute prostaglandin-effect," and have been primarily noted with the use of indomethacin. The association between prolonged and excessive consumption of compound analgesics and the development of renal disease and renal failure, characterized by renal papillary necros

Pharmacokinetics of common analgesics, anti-inflammatories and antipyretics in children.

Non-steroidal anti-inflammatory drugs (NSAIDs) are a major component of paediatric therapeutics. This paper summarises the clinical pharmacology and pharmacokinetics of some commonly used NSAIDs as well as paracetamol (acetaminophen), which has very weak anti-inflammatory activity. Available information is reviewed in an attempt to evaluate the basis for paediatric dosing recommendations and to underline known or likely differences in drug disposition that result from diseases and changes in developmental physiology. Clinically important general considerations are stressed, including areas in

Spinal cord effects of antipyretic analgesics.

Tissue damage results in the release of inflammatory mediators, including prostaglandins, which sensitive fine nerve endings in the periphery to mechanical and thermal changes. Sensitisation of these nerve endings, or nociceptors, contributes to the phenomenon of hyperalgesia, which routinely accompanies tissue damage. It has been shown that the acidic antipyretic analgesics reduce or down-regulate the enhanced nociceptor sensitivity in damaged tissue, an effect probably attributable to inhibition of prostaglandin synthesis. Recent studies suggest that these drugs may have an additional mechan

Antipyretic effect of centrally administered CRF.

CRF injected into the third cerebral ventricle (0.5-2.5 micrograms) caused dose-related reductions in fever induced in rabbits by IV administration of leukocytic pyrogen. Control injections of CRF when the same animals were afebrile did not alter normal body temperature. Intravenous injections of 5 and 20 micrograms CRF, doses known to release ACTH and corticosteroids into the bloodstream in other species, did not reduce fever. CRF injected into the cerebral ventricles may be antipyretic per se, or it may reduce fever by virtue of central release of the antipyretic peptides ACTH and alpha-MSH.

Review of comparative antipyretic activity in children.

Fever is one of the most common medical complaints referred to physicians for diagnosis and therapy. In addition, consumers frequently medicate themselves for fever associated with common, self-limited illnesses. The pathogenesis of fever suggests that pharmacologic therapy, which lowers the hypothalamic set-point, is an essential element in treatment. Not all fevers need to be treated; however, when indicated, therapy with antipyretics is necessary. The major antipyretic agents, acetaminophen, aspirin, and pyrazolone derivatives, are equally effective in reducing fever. However, after compari

Use of antipyretic analgesics in the pediatric patient.

Fever and pain are the most common issues in pediatric patient management. Acetaminophen, aspirin, and dipyrone are the most commonly used drugs and are equivalent in their efficacy. Dipyrone, used in many parts of the world, but not in the United States, is an effective agent; however, it has been implicated in producing agranulocytosis and anaphylactic shock. The salicylates have anti-inflammatory effects making them appropriate for the treatment of patients with juvenile rheumatoid arthritis, but they are gastric irritants, may impair clotting, and, because of saturable kinetics, may lead t

Hypothermia induced by therapeutic doses of antipyretics: misdiagnosis of cold sepsis.

Fever is a natural and almost universal mammalian response to infection. There exists a fear amongst general public and healthcare providers regarding fever being harmful leading to its overzealous management with antipyretics. Although the National Institute for Health and Care Excellence (NICE) guidelines suggest the use of single antipyretic agent for management of fever, combination therapy with paracetamol, and ibuprofen is common in paediatric practice in the United Kingdom. These antipyretics at times can cause significant adverse events even when administered at regular therapeutic dos

Antipyretic measures for treating fever in malaria.

BACKGROUND: Fever is common in malaria, and drugs and sponging are widely used for symptomatic relief. Some researchers have suggested that fever reduction may prolong malaria illness. Care in the use of ibuprofen as an antipyretic in children.

Ibuprofen is being widely used as an antipyretic in children. Recent studies indicate that it is as efficacious and with no significant difference in side-effects when compared to paracetamol. We describe three cases that illustrate that renal complications can occur when ibuprofen is prescribed in the presence of intravascular volume depletion and/or pre-existing renal problems. We discuss the mode of action of ibuprofen and recommend that its use as an antiypretic in children should be avoided in actual or potential intravascular volume contraction and in cases with pre-existing renal proble

Vasopressin functions as an endogenous antipyretic in the newborn.

These experiments sought to determine the role of arginine vasopressin (AVP) in the inability of newborns to produce a fever. Our data demonstrate that the AVP analog, [1-(beta-mercapto-beta,beta-cyclopentamethylene propionic acid)-2-(O-methyl) tyrosine]arginine vasopressin (M-AVP), administered centrally to adult rats, prevented the antipyretic action of centrally injected AVP. Behaviorally thermoregulating 3-day-old rat pups failed to respond to endoxin with a fever, similar to neonates of other species, but when central AVP antipyretic receptors were blocked by pretreatment with M-AVP, the
