Barbiturate enhancement of spike frequency adaptation in Aplysia giant neurons.

Pentobarbital and phenobarbital inhibit repetitive firing of action potentials in neurons of Aplysia by enhancing the normal process of spike frequency adaptation. Voltage clamp studies reveal a slowly developing outward current which is unique to neurons demonstrating spike frequency adaptation. Enhancement of this current by barbiturates was demonstrated and is the probable mechanism by which these drugs enhance adaptation. In experiments in which the pH of the applied drug solutions was altered, barbiturates were more effective in increasing both the slow outward current and adaptation at l

Cycle-to-cycle variability of neuromuscular activity in Aplysia feeding behavior.

The objective of this study was to assess the impact of two sequential long, static, anterior lumbar flexions on the development of a neuromuscular disorder and to compare it with previously obtained data from a series of short static flexion periods of the same cumulative time (Sbriccoli P, Solomonow M, Zhou BH, Baratta RV, Lu Y, Zhu MP, and Burger EL, Muscle Nerve 29: 300-308, 2004). Static flexions with loads of 20, 40, and 60 N were applied to the lumbar spine over two 30-min periods with a 10-min rest in between. The reflex EMG activity from the multifidus muscles and supraspinous ligamen

Synaptic block of a calcium-activated potassium conductance in Aplysia neurones.

In the preceding paper (Kehoe, 1985) it was shown that the firing of any one of three neurones (I, II, III) presynaptic to the medial cells of the pleural ganglion of Aplysia californica causes a diminution of the cholinergically controlled K conductance in those cells. Firing of the same three presynaptic neurones was shown here to cause a similar diminution in a depolarization-induced K-dependent conductance in the same post-synaptic cells. The depolarization-induced K conductance was found to disappear when Ca ions were removed from the sea water bathing the ganglion or when the cell was in

Synaptic block of a transmitter-induced potassium conductance in Aplysia neurones.

A voltage-clamp study was made of a slow excitatory post-synaptic potential (slow e.p.s.p.) that can be elicited in the medial cells of the left pleural ganglion of Aplysia californica by the firing of at least three different presynaptic neurones (labelled I, II and III). Each of these three neurones elicits other permeability changes in addition to the slow e.p.s.p., and all elements of these synaptic responses were shown to be mediated monosynaptically. The slow e.p.s.p., associated with an increase in membrane resistance, was shown to be due to a decrease in K permeability. When the slow e

Active transport of potassium by the giant neuron of the aplysia abdominal ganglion.

We measured the internal potassium activity, a(i) (K), and membrane potential, E(m), simultaneously in 111 R2 giant neurons of Aplysia californica. a(i) (K) was 165.3 +/- 3.4 mM, E(m) was -47.8 +/- 0.9 mv, and E(K) calculated using the Nernst equation was -76.9 +/- 0.05 mv. Such values were maintained for as long as 6 hr of continuous recording in untreated cells, a(i) (K) fell exponentially after the following treatments: cooling to 0.5 degrees -4 degrees C, ouabain, zero external potassium, 2,4-dinitrophenol, and cyanide. The effects of cooling and zero potassium were reversible. Potassium p

Aplysia temptin - the 'glue' in the water-borne attractin pheromone complex.

Temptin, a component of the complex of water-borne protein pheromones that stimulate attraction and mating behavior in the marine mollusk Aplysia, has sequence homology to the epidermal growth factor (EGF)-like domains of higher organisms that mediate protein-cell surface contact during fertilization and blood coagulation. In this work, recombinant temptin for structural and functional studies was produced in Escherichia coli using a cold shock promoter and purified by RP-HPLC. CD spectra confirmed a predominantly beta-sheet structure. Two disulfide bonds were determined via limited proteolysi

Calcitonin induces a decreased Na+ conductance in identified neurons of Aplysia.

The ionic mechanism of the effect of extracellularly ejected calcitonin (CT) on the membrane of identified neurons R9 and R10 of Aplysia was investigated with voltage-clamp, micropressure ejection, and ion substitution techniques. Micropressure-ejected CT caused a marked hyperpolarization in the unclamped neuron. Heat-inactivated CT was without effect. Clamping the same neuron at its resting potential level (-60 mV) and re-ejecting CT with the same dose produced a slow outward current (Io(CT), 30-40 sec in duration, 4-6 nA in amplitude) associated with a decrease in input membrane conductance.

Comparative study of histamine immunoreactivity in nervous systems of Aplysia and Pleurobranchaea.

The distribution of histamine in the nervous system of the marine molluscs Aplysia californica and Pleurobranchaea californica was studied by using a newly available immunohistochemical localization technique and specific antiserum against histamine-protein conjugate. We examined several sets of complete histological sections through the major ganglia of both animals, as well as all nerve roots of the buccal and cerebral ganglia and the corresponding target tissues. The results indicate that histamine is present in several neurons and/or nerve fibers of all major ganglia. An especially dense h

Effect of acetylcholine on ventrocaudal sensory neurons in the pleural ganglia of Aplysia.

1. The effects of acetylcholine (ACh) on the soma of cultured ventrocaudal sensory neurons from the pleural ganglia of Aplysia kurodai were characterized. 2. Whole-cell recording was used for current and voltage clamping. ACh and other drugs were microapplied to the membranes of the cultured neurons. 3. Microapplication of ACh induced an outward current mediated by a conductance increase. No desensitization to repeated applications of ACh was detected. The threshold was 10(-7) M and the maximum response was at 10(-5) M. 4. The reversal potential in normal seawater is -80 mV, close to the K+ eq

Effects of barbiturates on inhibitory and excitatory responses to applied neurotransmitters in Aplysia.

The effects of barbiturates on responses to iontophoretically applied acetylcholine and gamma-aminobutyric acid were studied in Aplysia californica by using a voltage clamp. Dose-response data were obtained for barbiturate concentrations ranging from 1 microM to 3 mM. Both phenobarbital and pentobarbital depress chloride-dependent inhibitory responses to both acetylcholine and gamma-aminobutyric acid. This depression occurs at approximately the same concentrations which attenuate excitatory responses. Potassium-dependent inhibitory responses were minimally affected. In on case was enhancement

Charybdotoxin selectively blocks small Ca-activated K channels in Aplysia neurons.

The action of charybdotoxin (ChTX), a peptide component isolated from the venom of the scorpion Leiurus quinquestriatus, was investigated on membrane currents of identified neurons from the marine mollusk, Aplysia californica. Macroscopic current recordings showed that the external application of ChTX blocks the Ca-activated K current in a dose- and voltage-dependent manner. The apparent dissociation constant is 30 nM at V = -30 mV and increases e-fold for a +50- to +70-mV change in membrane potential, which indicates that the toxin molecule is sensitive to approximately 35% of the transmembra

Analysis of decreased conductance serotonergic response in Aplysia ink motor neurons.

Micropressure ejection of serotonin (5-hydroxytryptamine, 5-HT) produced excitatory responses in the L14 ink motor neurons of Aplysia that depended on the site of application. Ejection of 5-HT onto the cell body produced a slow response that showed variability in voltage sensitivity between preparations. In contrast, ejection of 5-HT onto the neuropil underneath the cell body produced a response whose amplitude was consistently a linear function of the holding potential, reversing near the predicted potassium equilibrium potential. Subsequent analyses focused on this second response. The neuro

Aplysia Neurons as a Model of Alzheimer's Disease: Shared Genes and Differential Expression.

Although Alzheimer's disease (AD) is the most common form of dementia in the United States, development of therapeutics has proven difficult. Invertebrate alternatives to current mammalian AD models have been successfully employed to study the etiology of the molecular hallmarks of AD. The marine snail Aplysia californica offers a unique and underutilized system in which to study the physiological, behavioral, and molecular impacts of AD. Mapping of the Aplysia proteome to humans and cross-referencing with two databases of genes of interest in AD research identified 898 potential orthologs of

Isolation and characterization of haemoporin, an abundant haemolymph protein from Aplysia californica.

In the present study, we show the isolation and characterization of the protein haemoporin, which constitutes the second most abundant protein fraction in the haemolymph of the marine gastropod Aplysia californica. Although Aplysia is commonly used to investigate the molecular basis of learning, not much is known about the proteins in its haemolymph, which is in contact with the neurons owing to the open circulatory system of molluscs. In the native state, haemoporin is a macromolecular complex forming a cylinder with a central solvent-filled pore. The native complex most probably is a homopen

Control of pedal and parapodial movements in Aplysia. II. Cerebral ganglion neurons.

1. Intracellular stimulation of individual neurons in the two symmetrical A neuron clusters of the cerebral ganglion evoked contractions of both the foot and parapodia. Electrical stimulation of pedal and parapodial nerves caused antidromic action potentials in A neurons. Units recorded in the nerves followed the driven somatic spike 1:1. This suggests that the A neurons are presumptive pedal and parapodial motor neurons.2. Individual A neurons evoked both bilteral and unilateral contractions of the parapodia or split foot. Contractions in the parapodia were independent of those in the foot. A

Cholinergic depolarization recruits a persistent Ca2+ current in Aplysia bag cell neurons.

Many behaviors and types of information storage are mediated by lengthy changes in neuronal activity. In bag cell neurons of the hermaphroditic sea snail Aplysia californica, a transient cholinergic synaptic input triggers an ∼30-min afterdischarge. This causes these neuroendocrine cells to release egg laying hormone and elicit reproductive behavior. When acetylcholine is pressure-ejected onto a current-clamped bag cell neuron, the evoked depolarization is far longer than the current evoked by acetylcholine under voltage clamp, suggesting recruitment of another conductance. Our earlier studies

Sign stimulus activates a peptidergic neural system controlling reproductive behavior in Aplysia.

1. In the marine mollusk Aplysia, egg laying is a complex behavior that lasts for up to several hours. We used behavioral and electrophysiological methods to determine how egg laying occurs in groups of animals and how it is related to other aspects of reproductive behavior. 2. Prolonged contact with an existing egg mass by the lips and tentacles of an animal is a sign stimulus for release of egg-laying behavior and two other fixed action patterns in the same individual, mating as a female during egg laying and mating as a male after egg laying. 3. Prolonged contact with the egg mass initiated
