Apolipoprotein E4 and meningeal lymphatics in Alzheimer disease: a conceptual framework.

The potential existence and roles of the meningeal lymphatic system in normal and pathological brain function have been a long-standing enigma. Recent evidence suggests that meningeal lymphatic vessels are present in both the mouse and human brain; in mice, they seem to play a role in clearing toxic amyloid-beta peptides, which have been connected with Alzheimer disease (AD). Here, we review the evidence linking the meningeal lymphatic system with human AD. Novel findings suggest that the recently described meningeal lymphatic vessels could be linked to, and possibly drain, the efferent parava

Leisure activities, apolipoprotein E e4 status, and the risk of dementia.

BACKGROUND/PURPOSE: Leisure activities have been associated with a decreased risk of dementia. However, to date, no study has explored how apolipoprotein E (ApoE) e4 status or vascular risk factors modified the association between leisure activities and dementia risks. METHODS: This case-control study recruited patients (age ≥ 60 years) with Alzheimer's disease (AD; n = 292) and vascular dementia (VaD; n = 144) and healthy controls (n = 506) from three teaching hospitals in Taiwan between 2007 and 2010. Information on patient's leisure activities were obtained through a questionnaire. Conditi

Apolipoprotein E e4 allele affects risk of hyperhomocysteinemia in the elderly.

BACKGROUND: Apolipoprotein E (APOE) plays a central role in VLDL metabolism. Both APOE e4 allele (APOE4) and C-reactive protein (CRP) are associated with greater risk of dementia and vascular disease, but APOE4 carriers have lower blood concentrations of CRP than do noncarriers, possibly through a mechanism favoring the clearance of the CRP VLDL-bound fraction. Homocysteine, another risk factor for vascular disease and dementia, also binds to VLDL in blood. However, the association between APOE4 and hyperhomocysteinemia has never been thoroughly investigated. OBJECTIVE: We investigated in an

Apolipoprotein E4 promotes the early deposition of Abeta42 and then Abeta40 in the elderly.

The apolipoprotein Eepsilon4 allele (ApoEepsilon4) is associated with a selective increase in deposition of the 40-amino acid form of the beta-amyloid peptide (Abeta40) in endstage Alzheimer's disease. To determine how apoE genotype affects the early events in beta-amyloid pathogenesis, we analyzed the medial temporal lobes of 244 elderly persons who were not clinically demented using antibodies selective for the C termini of Abeta40 and Abeta42. We found that: (1) the number of both Abeta42- and Abeta40-positive senile plaques increase with age; (2) Abeta42 appears at younger ages, and in mor

Aerobic exercise improves hippocampal blood flow for hypertensive Apolipoprotein E4 carriers.

Cerebrovascular dysfunction likely contributes causally to Alzheimer's disease (AD). The strongest genetic risk factor for late-onset AD, Apolipoprotein E4 (APOE4), may act synergistically with vascular risk to cause dementia. Therefore, interventions that improve vascular health, such as exercise, may be particularly beneficial for APOE4 carriers. We assigned cognitively normal adults (65-87 years) to an aerobic exercise intervention or education only. Arterial spin labeling MRI measured hippocampal blood flow (HBF) before and after the 52-week intervention. We selected participants with hype

Coronary artery disease and plasma apolipoprotein E4 in mild cognitive impairment.

BACKGROUND: Atherosclerosis and apolipoprotein E4 (APOE4) are known risks for Dementia. We sought to evaluate the relationship between coronary atherosclerosis and APOE4 with mild cognitive impairment (MCI). METHODS: In a case-control study, subjects with age more than 60 years and recent coronary angiography were evaluated by mini-mental state examination and neuropsychiatry unit cognitive assessment tool (NUCOG) to find the patients with MCI (n = 40) and the controls with normal cognition (n = 40). Coronary angiography records were re-assessed to find the severity of coronary artery disease

Membrane-Coated DNA Nanoflowers Attenuate Cognitive Decline in Apolipoprotein E4 Mice.

APOE4-driven mitochondrial dysfunction is one of the important primary drivers of cognitive decline in neurodegenerative diseases. However, achieving mitochondrial-targeted drug delivery in the nervous system requires overcoming multiple barriers, necessitating the development of a safer and more efficient bionanoparticle drug delivery system. DNA nanoflowers (DNFs), generated via rolling circle amplification (RCA), offer programmable platforms for creating targeted drug delivery vehicles. Here, we designed a multifunctional DNF surface conjugated with ferrocene groups, loaded with resveratrol

Pathogenesis of Alzheimer's disease: current status on apolipoprotein E4 gene research.

Alzheimer's disease (AD) affects millions of people throughout the world. The financial and emotional costs that are inflicted by this devastating disease are enormous. Genes responsible for the early-onset familial AD have been cloned and found to be membrane associated transport proteins. Evidence suggests that apolipoprotein E4 may be the risk factor for the late-onset form of the disease. Beta-amyloid and potassium channel dysfunction have also been implicated in the development of AD. Hyperphosphorylation of "tau" has been indicated for the generation of neurofibrillary tangles in the bra

Progressive loss of synaptic integrity in human apolipoprotein E4 targeted replacement mice and attenuation by apolipoprotein E2.

Inheritance of the APOE4 allele is a well established genetic risk factor linked to the development of late onset Alzheimer's disease. As the major lipid transport protein in the central nervous system, apolipoprotein (apo) E plays an important role in the assembly and maintenance of synaptic connections. Our previous work showed that 7 month old human apoE4 targeted replacement (TR) mice displayed significant synaptic deficits in the principal neurons of the lateral amygdala, a region that is critical for memory formation and also one of the primary regions affected in Alzheimer's disease, co

Apolipoprotein E4 and immune dysfunction in Alzheimer's disease: mechanisms and therapeutic implications.

Emerging evidence suggests that apolipoprotein E4 (APOE4) functions as an organizer of brain-periphery immune crosstalk, coupling maladaptive microglial states with peripheral immune remodeling to establish a self-reinforcing neuroimmune loop that may accelerate the progression of Alzheimer's disease. In the central nervous system, APOE4 pushes microglia away from homeostatic surveillance toward metabolically stressed, inflammatory, and synaptotoxic phenotypes, thereby weakening pathological protein clearance and promoting neurodegeneration. In the periphery, APOE4 primes innate and adaptive i

Role of apolipoprotein E4 in alzheimer's disease pathogenesis and emerging therapeutic strategies.

Alzheimer’s disease, the leading cause of dementia worldwide, is profoundly influenced by the apolipoprotein E4 allele, the primary genetic risk factor for sporadic Alzheimer’s disease. This allele exacerbates amyloid-beta pathology, tau phosphorylation, mitochondrial dysfunction, neuroinflammation, synaptic impairment, vascular dysfunction, and gut-brain axis alterations, contributing to disease progression. Recent advances elucidate apolipoprotein E4’s molecular mechanisms, including its role in lipid metabolism and receptor interactions, which drive these pathological processes. This review

Apolipoprotein E4 Reduction with Antisense Oligonucleotides Decreases Neurodegeneration in a Tauopathy Model.

OBJECTIVE: Apolipoprotein E (ApoE) genotype is the strongest genetic risk factor for late-onset Alzheimer's disease, with the ε4 allele increasing risk in a dose-dependent fashion. In addition to ApoE4 playing a crucial role in amyloid-β deposition, recent evidence suggests that it also plays an important role in tau pathology and tau-mediated neurodegeneration. It is not known, however, whether therapeutic reduction of ApoE4 would exert protective effects on tau-mediated neurodegeneration. METHODS: Herein, we used antisense oligonucleotides (ASOs) against human APOE to reduce ApoE4 levels in

Apolipoprotein E4 effects on topological brain network organization in mild cognitive impairment.

The Apolipoprotein E isoform E4 (ApoE4) is consistently associated with an elevated risk of developing late-onset Alzheimer's Disease (AD); however, less is known about the potential genetic modulation of the brain networks organization during prodromal stages like Mild Cognitive Impairment (MCI). To investigate this issue during this critical stage, we used a dataset with a cross-sectional sample of 253 MCI patients divided into ApoE4-positive (‛Carriers') and ApoE4-negative ('non-Carriers'). We estimated the cortical thickness (CT) from high-resolution T1-weighted structural magnetic images

In Silico Identification of Novel Apolipoprotein E4 Inhibitor for Alzheimer's Disease Therapy.

INTRODUCTION: Apolipoprotein E4 (ApoE) is a major genetic factor for developing Alzheimer's disease (AD). It plays a vital role in brain to maintain a constant supply of neuronal lipids for rapid and dynamic membrane synthesis. Aggregation of beta amyloid plaques (Aβ) and neurofibrillary tangles in brain are responsible for onset of AD. The current study is designed to predict a drug against over activity of apoE4. 22 natural compounds (marine, microorganism and plant derivative) were used in current study. METHODS: These compounds were used as inhibitors to target apoE4 protein activity. Mor

Effect of low-density lipoprotein apheresis on plasma levels of apolipoprotein e4.

Apolipoprotein E4 (apoE4) is a positively charged proinflammatory apolipoprotein bound to high-density lipoprotein (HDL) cholesterol and remnant lipoproteins. ApoE4 is associated with an increased risk of cardiovascular and cerebrovascular disease. Low-density lipoprotein (LDL) apheresis, a therapy for patients with familial hypercholesterolemia, removes apolipoprotein B and other positively charged plasma proteins but negatively charged proteins such as HDL cholesterol are generally spared. Despite their negative charge, LDL apheresis still removes 10% to 15% of HDL cholesterol, in particular

Free-water imaging of the nucleus basalis of Meynert in apolipoprotein E4 carriers.

INTRODUCTION: The apolipoprotein E (APOE) ε4 allele is the strongest genetic risk factor for late-onset Alzheimer's disease (AD) and cardiovascular disease. This study aimed to investigate the interactive effects of APOE ε4 genotype and cardiovascular risk on the microstructure of the nucleus basalis of Meynert (NBM), a key cholinergic region affected early in AD, using advanced diffusion magnetic resonance imaging. METHODS: This cross-sectional study included 167 cognitively unimpaired older adults from the Arizona APOE Cohort. Participants were stratified by genotype: APOE ε4 non-carriers (

The relationship between apolipoprotein E4 and lipid metabolism is impaired in Alzheimer's disease.

BACKGROUND: Human apolipoprotein (Apo) E4 (ApoE4) is an important determinant of lipid metabolism and cell-to-cell cholesterol transport. It is also a major genetic risk factor for both vascular disease and familial and sporadic late-onset Alzheimer's disease (AD). Since vascular pathology could dramatically reduce neuronal reserve capacity, a biological chain of events between ApoE4, hypercholesterolemia and AD has been postulated. The aim of the present study is to explore the relationship between lipid metabolism and ApoE isoforms in a large series of elderly subjects in relation to the pre

Apolipoprotein E4 and synaptic dysfunction in Alzheimer's disease: Mechanisms and therapeutic implications.

Alzheimer's disease (AD) is characterized by progressive cognitive decline, with synaptic dysfunction as the strongest correlate of clinical symptoms. The apolipoprotein E ε4 (ApoE4) allele is the most potent genetic risk factor for late-onset AD. Beyond its roles in amyloid-β aggregation and tau hyperphosphorylation, ApoE4 disrupts synaptic integrity by perturbing lipid metabolism, neuroimmune regulation, mitochondrial dynamics, and activity-dependent plasticity. These ApoE4-driven mechanisms impair presynaptic vesicle trafficking, destabilize postsynaptic receptor and scaffolding networks (i
