Pain relief represents a critical unresolved medical need. Consequently, the search for new analgesic agents is intensively studied. Annona crassiflora, a native species of the Brazilian Savanna, represents a potential source for painful treatment. This study aimed to investigate the antinociceptive potential of A. crassiflora fruit peel, focusing on its major alkaloid, stephalagine, in animal models of pain evoked by the activation of transient receptor potential ankyrin 1 (TRPA1) and vanilloid 1 (TRPV1) channels. Male C57BL/6/J mice were submitted to formalin-, cinnamaldehyde-, and capsaicin

Background: Pancreatic cancer cells adapt to nutrient stress by activating metabolic pathways that facilitate survival and proliferation within the tumor microenvironment, a phenomenon known as austerity. Targeting this resilience presents a strategy for developing chemotherapeutic agents with anti-austere properties. Methods: Phytochemical investigation of Nelumbo nucifera petals led to the isolation of six alkaloids: three benzylisoquinolines (1-3), two aporphines (4, 5), and one proaporphine (6). Preferential cytotoxicity under nutrient deprivation was evaluated. The most active compound wa

Aporphine alkaloids have shown affinity for serotonin receptors (5-HTRs), and there has been a recent upsurge of interest in aporphines as 5-HT2CR ligands. 1,2,9,10-Tetraoxygenated aporphine alkaloids in particular have demonstrated good affinity for 5-HTRs. In continued efforts to understand the impacts of structural modification of the 1,2,9,10-tetraoxygenated aporphine template on affinity, selectivity, and activity at 5-HT2R subtypes, we used (+)-boldine (8) as a semisynthetic feedstock in the preparation of C-2-alkoxylated (+)-predicentrine analogues. Compound 10n, which contains a benzyl

INTRODUCTION: The Annonaceae family is known as a promising abundant source of secondary metabolites, especially annonaceous acetogenins, terpenoids and isoquinoline-derived alkaloids. Although widely investigated from the phytochemical viewpoint, this family still presents some largely unexplored genera, e.g. the Bocageopsis.
OBJECTIVE: To investigate the alkaloid content of Bocageopsis pleiosperma Maas using direct infusion electrospray ionisation ion trap tandem mass spectrometry (ESI-IT-MS(n)) analysis.
METHODOLOGY: Dichloromethane extracts of aerial parts were subjected to acid-base par

Aporphines are a class of isoquinoline alkaloids that are endowed with a range of biological activities. The 5-HT7R is an emerging biological target for prostate cancer therapeutics. In this manuscript, we report the synthesis and evaluation of aporphine enantiomers as 5-HT7R ligands, as well as their activity in inhibiting the proliferation of prostate cancer cells (specifically, PC3). The (S)-enantiomers displayed higher affinity at the 5-HT7R than the racemates and the (R)-enantiomer counterparts. The (S)-enantiomers were found to be antagonists at the 5-HT7R. Racemates as well as their res

A proposed dopamine (DA) receptor labeling agent, [3H](-)N-chloroethylnorapomorphine (3H-NCA) underwent relatively little chemical change at 25 degrees C and pH 6.4 up to an hour of incubation. At low (nM) concentrations it bound rapidly and avidly to a membrane preparation of calf caudate nucleus, but the binding did not saturate over two hours of incubation or at ligand concentrations between 0.2 nM and 10 microM. While similarly bound [3H]-(-)apomorphine was rapidly displaced by DA and other agents that interact with DA receptors, 3H-NCA could not be displaced by unlabeled DA, apomorphine a

While alkylated and acylated aporphine derivatives are well-represented in the literature, O-arylated analogues remain scarce. We report here a practical and regioselective methodology for the synthesis of 9-O-arylboldine derivatives. Our approach utilizes a copper-catalyzed Chan-Lam coupling reaction, enabling the efficient preparation of 30 novel 9-O-arylboldine derivatives. This operationally simple procedure proceeds at room temperature under ambient air conditions, employing readily available phenylboronic acids as coupling partners. Extensive one- and two-dimensional NMR studies unequivo

Benzylisoquinoline alkaloids (BIAs) are a structurally diverse group of plant specialized metabolites found mainly in members of the order Ranunculales, including opium poppy (Papaver somniferum), for which BIA biosynthetic pathways leading to the critical drugs morphine, noscapine, and sanguinarine have been elucidated. Sacred lotus (Nelumbo nucifera), in the order Proteales, accumulates medicinal BIAs in the proaporphine, aporphine, and bisbenzylisoquinoline structural subgroups with a prevalence of R enantiomers, opposed to the dominant S configuration occurring in the Ranunculales. The D2 high receptor, functional high-affinity state of the dopamine D2 receptor, has been shown to be upregulated in the context of neurological disorders. We had synthesized a specific D2 receptor agonist, the aporphine derivative [3H]-MCL-524. Previous, preliminary data indicated that [18F]-MCL-524 specifically binds to the activated form of the D2 receptor, D2 high. Here, we studied the binding characteristics of [3H]-MCL-524 in vitro, with the aim of unambiguously demonstrating its specificity for D2 high. In saturation binding studies using Chinese Hamster Ovary (CHO) cell membranes expr

One new p-quinonoid aporphine alkaloid, obtusipetadione (1), and eleven known compounds (2-12) were isolated from the acetone extract of the twigs of Dasymaschalon obtusipetalum. Their structures were elucidated by spectroscopic methods. The cytotoxic and antimalarial activities of the isolated compounds were evaluated. Compound 1 showed significant in vitro antiplasmodial activity against the P. falciparum strains TM4 and K1 (multidrug resistant strain) with IC50 values of 2.46 ± 0.12 and 1.38 ± 0.99 μg/mL, respectively with no cytotoxicity. Compound 9 had more modest antiplasmodial activity,

Morphinan alkaloids have attracted constant attention since the isolation of morphine by Sertürner in 1805. However, a group of 45 compounds possessing a complete ent-morphinan backbone can also be found in the literature. These compounds are related to the morphinandienone subgroup and display a substitution pattern which is different from the morphinans. In particular, these alkaloids could be substituted at position C-2 and C-8 either by a hydroxy function or a methoxy moiety. Four groups of ent-morphinan alkaloids can be proposed, the salutaridine, pallidine, cephasugine and erromangine se

Brazil is a country with the greatest biodiversity in the world, but much of it is still largely unexplored. Trigynaea axilliflora (Annonaceae) is a Brazilian plant species that has not been previously studied. This research aimed to characterize the chemical constituents and evaluate the biological activities of the extract, fractions and isolated compounds. Xylopine, a major compound isolated from this extract, is an aporphine alkaloid and a chemotaxonomic marker of this family, whose structure was confirmed by 1H/13C NMR and HRESIMS. HPLC-UV-ESI-qTOF dereplication indicated that xylopine an

BACKGROUND: Croton draco is an arboreal species and its latex as well as some other parts of the plant, are traditionally used in the treatment of a wide range of ailments and diseases. Alkaloids, such as magnoflorine, prevent early atherosclerosis progression while taspine, an abundant constituent of latex, has been described as a wound-healer and antitumor-agent. Despite the great interest for these and other secondary metabolites, no omics resources existed for the species and the biosynthetic pathways of these alkaloids remain largely unknown.
RESULTS: To gain insights into the pathways i
