Evaluating the Phytohormone Proficiencies of Multifarious Bacillus rugosus for Growth Promotion in Arachis hypogaea (L.).

The application of Plant Growth-Promoting Rhizobacteria (PGPR) in agriculture is increasingly emphasized as a sustainable alternative to hazardous agrochemicals. This study aimed to isolate and characterize PGPR strains from the rhizospheric soil of Arachis hypogaea L., hypothesizing that the rhizosphere of a healthy plant harbors beneficial microbes with significant plant growth-promoting (PGP) attributes. The isolate AB1 demonstrated promising PGP traits, including phosphate solubilization (56.44 µg mL⁻¹), zinc solubilization (6.1 µg mL⁻¹), ammonia production (3.8 µM µg mL⁻¹), and the synthe

Autotetraploid Induction of Three A-Genome Wild Peanut Species, Arachis cardenasii, A. correntina, and A. diogoi.

A-genome Arachis species (AA; 2n = 2x = 20) are commonly used as secondary germplasm sources in cultivated peanut breeding, Arachis hypogaea L. (AABB; 2n = 4x = 40), for the introgression of various biotic and abiotic stress resistance genes. Genome doubling is critical to overcoming the hybridization barrier of infertility that arises from ploidy-level differences between wild germplasm and cultivated peanuts. To develop improved genome doubling methods, four trials of various concentrations of the mitotic inhibitor treatments colchicine, oryzalin, and trifluralin were tested on the seedlings

Net Blotch and Necrotic Warts Caused by Streptomyces scabies on Pods of Peanut (Arachis hypogaea).

Peanut (Arachis hypogaea L.) grown in rotation with potato (Solanum tuberosum L.) in the Northern Cape Province and at one locality in the Free State Province, South Africa, have developed unusual pod symptoms since the 1994 to 1995 season. Symptoms ranged from a net blotch with scattered lesions to dark brown, necrotic, wartlike lesions on the cv. Sellie. Streptomyces scabies (2) was consistently isolated from both types of lesions. Pathogenicity was confirmed in greenhouse tests. Inoculum was prepared by growing colonies on yeast malt extract agar for 21 days. Ten milliliters of sterile, dis

Soil Bacterial Communities Across Seven Rhizoma Peanut Cultivars (Arachis glabrata Benth.) Respond to Seasonal Variation.

Soil microorganisms play key roles in soil nutrient transformations and have a notable effect on plant growth and health. Different plant genotypes can shape soil microbial patterns via the secretion of root exudates and volatiles, but it is uncertain how a difference in soil microorganisms induced by crop cultivars will respond to short-term seasonal variations. A field experiment was conducted to assess the changes in soil bacterial communities of seven rhizoma peanut (Arachis glabrata Benth, RP) cultivars across two growing seasons, April (Spring season) and October (Fall season). Soils' ba

INVESTIGATION OF AMOEBICIDAL POTENTIAL OF ARACHIS HYPOGAEA L. PERICARP ON CYSTS OF ACANTHAMOEBA ASTRONYXIS T7 GENOTYPE.

Acanthamoeba is an opportunistic pathogen cauAing keratitis and fatal encephalitis. Early diagnosis, followed by intense treatment using a drugs mixture is a necessity for effective therapy. Many natural compounds have proved lethal effects, yet the search for original natural amebicidal agents is still of current concern. This study investigated the acanthamoebicidal effect of A. hypogaea L. pericarp; total ethanol extract and its successive fractions, n-hexane, dichloro-methane, ethyl acetate, and methanol as well as resveratrol. Acanthamoebae were isolated and cultivated on E. coli seeded n

Identification and characterization of phages parasitic on bradyrhizobia nodulating groundnut (Arachis hypogaea L.) in South Africa.

In this study, three lytic phages (namely, PRSA-1, PRSA-2 and PRSA-26) were isolated and characterized for their morphology, host range, profile and restriction endonuclease banding pattern of genome size. The susceptible rhizobial isolates were identified by nifH and glnII sequence analysis. The results showed that all phages had polyhedral head with non-contractile tail which confirmed their relationship with the Siphoviridae family. All the three phages produced highly distinct plaques on their host bradyrhizobial lawn, and were highly sensitive to chloroform. The phage genome sizes ranged

Identification photosynthesis-related genes in peanut (Arachis hypogaea L.) by hyperspectral phenomics, GWAS and RNA-seq.

Peanut (Arachis hypogaea L.) contributes importantly to the global food supply, and its yield potential is closely linked to leaf photosynthetic efficiency. However, conventional measurement of leaf net-photosynthetic-rate (Leaf Pn) is time- and labor-intensive, limiting its application to large breeding populations. We established a phenomics-driven framework integrating hyperspectral reflectance and genome-wide association studies for high-throughput evaluation and genetic dissection of Leaf Pn. Machine-learning models were trained on preprocessed spectral data from 1,200 leaf samples collec

Characterization of a pathogen induced thaumatin-like protein gene AdTLP from Arachis diogoi, a wild peanut.

Peanut (Arachis hypogaea L) is one of the widely cultivated and leading oilseed crops of the world and its yields are greatly affected by various biotic and abiotic stresses. Arachis diogoi, a wild relative of peanut, is an important source of genes for resistance against various stresses that affect peanut. In our previous study a thaumatin-like protein gene was found to be upregulated in a differential expression reverse transcription PCR (DDRT-PCR) study using the conidial spray of the late leaf spot pathogen, Phaeoisariopsis personata. In the present study, the corresponding full length cD

Identification of HsfB Family in Peanut (Arachis hypogea) and Role of AhHsfB1-5A in High-Temperature Stress.

Global warming-triggered heat stress severely restricts plant growth and crop productivity. Peanut (Arachis hypogaea L.), a vital oilseed and cash crop that is susceptible to high temperatures throughout its growth cycle, exhibits inhibited peg and pod development, growth retardation, and premature leaf senescence under heat stress, which ultimately causes substantial yield losses. Heat shock factors (Hsfs) serve as core regulatory modulators of plant abiotic stress tolerance, among which the HsfB subfamily exerts a critical function in thermotolerance modulation. Nevertheless, the biological

Enhanced Recovery and Bioactivity of Flavonoids From Peanut Shells (Arachis hypogaea): Antioxidant and Acetylcholinesterase Inhibitory Properties.

Peanut shells, a by-product of the peanut processing industry, are rich in flavonoid compounds with a range of bioactivities. This study aimed to develop an efficient extraction and fractionation process to enhance the recovery and in vitro antioxidant and acetylcholinesterase (AChE) inhibition activities of these compounds from peanut shells cultivated in Vietnam. Peanut shell samples were subjected to maceration with ethanol, followed by liquid-liquid partitioning (LLP) and column chromatography (CC) using a hexane and ethyl acetate solvent system at varying ratios. This process yielded a cr

Transcriptomic analysis of AhAHL23-mediated root development and space-induced mutations in peanut (Arachis hypogaea L.).

Peanut seedling development encompasses four distinct tissues (leaf, stem, hypocotyl, root) with largely uncharacterized transcriptional regulatory networks, so we conducted RNA-seq on the space mutant line ZHM112. Transcriptome analysis showed that differentially expressed genes (DEGs) in peanut seedlings were enriched in basic biosynthesis and physiological metabolism pathways, with photosynthetic metabolism prominent in leaves and hormone metabolism in roots. The weighted gene co-expression network analysis (WGCNA) identified eight modules related to the four tissues. By integrating differe

Histopathology of Thecaphora frezzii Colonization: A Detailed Analysis of Its Journey Through Peanut (Arachis hypogaea L.) Tissues.

Over the past few decades, peanut smut, caused by Thecaphora frezzii, has evolved from an emerging disease to a major global threat to peanut production. However, critical knowledge gaps persist regarding the anatomical pathways and host responses involved in infection, colonization, and sporulation. This study examines the pathosystem and histopathology of the biotrophic phase of T. frezzii in the susceptible cv. Granoleico. Anatomical analyses were conducted using light microscopy, confocal laser scanning, and scanning electron microscopy. Our findings reveal that T. frezzii enters the host

Arachis hypogaea PGPR isolated from Argentine soil modifies its lipids components in response to temperature and salinity.

The aim of this work was to clarify the mechanism related to plant growth promoting of a bacterial strain (L115) isolated from Arachis hypogaea rhizospheres and the effects of high growth temperature and salinity on phospholipids and fatty acids composition. L115 was isolated from peanut rhizospheres and identified according to the sequence analysis of the 16S rRNA gene. Phenotypic, metabolic and plant growth promoting rhizobacteria (PGPR) characteristics of L115 were tested. Inoculation test in plant growth chamber was performed. In addition, L115 was exposed to a 37 °C and 300 mM NaCl and ph

Multi-environment evaluations reveal that peanut (Arachis hypogaea L.) quality traits are equally determined by genotype and environment.

BACKGROUND: Peanut (Arachis hypogaea L.) is a widely planted cash and oil crop, and regional and meteorological factors, as the most critical environmental factors, play crucial roles in peanut kernel quality traits. To explore the relationship between peanut quality traits and environmental factors, this study dissected the quality traits and the genotype-by-environment interaction factors using seven varieties across multiple environments. METHODS: Analysis of variance (ANOVA) was used to partition the variance of kernel quality traits, correlation analysis was used to examine the associati

Slaked lime improves growth, antioxidant capacity and reduces Cd accumulation of peanut (Arachis hypogaea L.) under Cd stress.

Slaked lime has been used to remediate contaminated agricultural soils as an in situ chemical immobilization amendment for a long time. However, the effects of slaked lime on peanut and soil cadmium (Cd) levels remain poorly understood with respect to remediating Cd-contaminated soil. In this study, six rates of slaked lime (e.g., 0, 300, 600, 900, 1200 and 1500 kg ha-1) were applied to evaluate the effects of slaked lime treatments on soil pH and the growth, Cd accumulation and physiology characteristics of peanut, which were in Cd-contaminated soil, and 0 kg ha-1 was taken as the control. Expression of δ-endotoxin Cry1EC from an inducible promoter confers insect protection in peanut (Arachis hypogaea L.) plants.

BACKGROUND: Spodoptera litura (F.) is a polyphagous foliage insect and a major pest on peanut (Arachis hypogaea L.). Constitutive expression of δ-endotoxin Cry1EC gives protection against S. litura, as reported earlier. In this study, insect bites and salicylic acid induced high-level expression of Cry1EC was achieved in peanut. In order to achieve this, the expression of pathogenesis responsive promoter PR-1a was enhanced by placing it downstream of the CaMV35S promoter in the pCAMBIA 1300 backbone. The resultant promoter CaMV35S(r)PR-1a expressed a high level of insecticidal δ-endotoxin Cry1
