RNA interference (RNAi) has strong antiviral activity in a range of animal phyla, but the extent to which RNAi controls virus infection in chordates, and specifically mammals remains incompletely understood. Here we analyze the antiviral activity of RNAi against a number of positive-sense RNA viruses using Argonaute-2 deficient human cells. In line with absence of virus-derived siRNAs, Sindbis virus, yellow fever virus, and encephalomyocarditis virus replicated with similar kinetics in wildtype cells and Argonaute-2 deficient cells. PURPOSE: Argonaute proteins are key players in small RNA-guided gene silencing processes. Ago2 is the member of the Argonaute subfamily with slicer endonuclease activity and is critical for microRNA homeostasis and indispensable for biological development. However, the impact of Ago2 dysregulation in the retina remains to be fully explored. In this study, we studied the role of Ago2 in mouse retina.
METHODS: We explored the function of Ago2 in the mouse retina through an adeno-associated virus-mediated Ago2 disruption mouse model. An ERG was carried out to determine the retinal function. Spec

Abasic substitutions within DNA or RNA are tools for evaluating the impact of absent nucleobases. Because of the importance of abasic sites in genetic damage, most research has involved DNA. Little information is available on the impact of abasic substitutions within RNA or on RNA interference (RNAi). Here, we examine the effect of abasic substitutions on RNAi and allele-selective gene silencing. Huntington's disease (HD) and Machado Joseph Disease (MJD) are severe neurological disorders that currently have no cure. HD and MJD are caused by an expansion of CAG repeats within one mRNA allele en

Prokaryotic Argonaute proteins (pAgos) are nucleic acid-guided endonucleases with diverse functions. Mucilaginibacter paludis Argonaute (MbpAgo) is unusual in using guide DNA (gDNA) to cleave target RNA (tgRNA), but the structural basis for this activity has been unclear. Here we present cryo-electron microscopy structures of MbpAgo in apo, binary, and ternary states at up to 2.55 Å resolution. The apo structure reveals a conserved bilobal scaffold with unique insertions in the PIWI and MID domains that stabilize the catalytic conformation. Upon gDNA binding, MbpAgo forms a dimer stabilized by

Short prokaryotic Argonaute associated with TIR-APAZ (SPARTA) is a bacterial defense system that oligomerizes to activate the TIR domain, depleting NAD+ upon guide-mediated target DNA recognition. Previous studies have shown a marked activity divergence between thermophilic Crenotalea thermophila SPARTA (CrtSPARTA) and Maribacter polysiphoniae SPARTA (MapSPARTA); however, the underlying mechanism remains unclear. Here, through biochemical and structural analysis, we found that compared with the relatively rigid CrtSPARTA, MapSPARTA exhibits much more flexibility in its TIR domain, which flips

Injury response in the peripheral nervous system (PNS) is characterized by rapid alterations in the genetic program of Schwann cells. However, the epigenetic mechanisms modulating these changes remain elusive. Here we show that sciatic nerve injury in mice induces a cohort of 22 miRNAs, which coordinate Schwann cell differentiation and dedifferentiation through a combinatorial modulation of their positive and negative gene regulators. These miRNAs and their targeted mRNAs form functional complexes with the Argonaute-2 protein to mediate post-transcriptional gene silencing. MiR-138 and miR-709

Small RNAs, including small interfering RNAs (siRNAs), microRNAs (miRNAs) and Piwi-associated interfering RNAs (piRNAs), are powerful gene expression regulators. This RNA-mediated regulation results in sequence-specific inhibition of gene expression by translational repression and/or mRNA degradation. siRNAs and miRNAs are generated by RNase III enzymes and subsequently loaded into Argonaute protein, a key component of the RNA induced silencing complex (RISC), to form the core of the RNA silencing machinery. RNA silencing acts as an ancient cell defense system against molecular parasites, such

Bioactive molecules can pass between microbiota and host to influence host cellular functions. However, general principles of interspecies communication have not been discovered. We show here in C. elegans that nitric oxide derived from resident bacteria promotes widespread S-nitrosylation of the host proteome. We further show that microbiota-dependent S-nitrosylation of C. elegans Argonaute protein (ALG-1)-at a site conserved and S-nitrosylated in mammalian Argonaute 2 (AGO2)-alters its function in controlling gene expression via microRNAs. By selectively eliminating nitric oxide generation b

The Piwi subfamily is one of two Argonaute family proteins, which are characterized by the presence of Piwi and Piwi‑Argonaute‑Zwille domains, and are well known for their role in RNA silencing. Hiwi, a human member of the Piwi subfamily, is restricted to the germ line, where it binds Piwi‑interacting RNAs and functions in stem cell self‑renewal and gametogenesis. Previous reports have indicated that abnormal Hiwi expression may be associated with a poor prognosis of numerous types of human cancer, including hepatocellular carcinoma (HCC). However, little is currently known about the oncogenic

Non-coding RNAs serve as key regulators of gene expression at both the transcriptional and post-transcriptional levels. Among them, piRNAs, a class of small non-coding RNAs, have emerged as significant mediators in cancer biology. PIWI proteins, members of the Argonaute family highly expressed in germline cells, interact with piRNAs to form piRNA/PIWI complexes, which play a key role in the development, recurrence, and metastasis of several malignancies, including gliomas. This review describes the biological functions of PIWI proteins and piRNAs, emphasizing their roles in cancer-related proc

Thermus thermophilus Argonaute (TtAgo) has shown exceptional promise in biosensing and molecular diagnostics owing to its programmable and DNA-guided specific cleavage activity. However, the potential of regulating TtAgo activity through precise structural modification of guide DNA (gDNA) remains to be fully utilized, thereby limiting the flexible expansion of TtAgo-based biosensing strategies. In this work, TtAgo achieves optimal cleavage activity strictly with a specific linear single-stranded gDNA (linear gDNA) length range (about 15-21 nt), whereas an excessively long linear gDNA fails to

Machado-Joseph disease (MJD) is a polyglutamine neurodegenerative disorder characterized by the formation of neuronal intranuclear Ataxin-3 inclusions. The nucleocytoplasmic transport is known to be profoundly dysregulated from early stages of neurodegeneration, further aggravating its progressive nature. To understand the contribution of these proteins in MJD, we screened the effect of 34 transport proteins on Ataxin-3 aggregation. Importin-8 (Ipo8 gene) was selected for further neuropathology and behavior analysis to evaluate its modulatory effect in vitro and in vivo. Our results showed tha

BACKGROUND: PIWI proteins belong to the Argonaute family. The human PIWI subfamily genes encode four PIWI (also known as PIWI-like) proteins: PIWIL1, PIWIL2, PIWIL3, and PIWIL4. The dysregulated expression of PIWI proteins appears to be associated with tumorigenesis.
OBJECTIVE: To explore the expression of PIWI proteins in primary and metastatic tumors from patients with stage III epithelial ovarian cancer (EOC), evaluate the diagnostic value of PIWI in various tissues, and analyze the characteristics of each PIWI protein associated with metastasis.
METHODS: A total of 20 patients with stage

Many neurodegenerative diseases are characterized by the presence of intracellular protein aggregates, resulting in alterations in autophagy. However, the consequences of impaired autophagy for neuronal function remain poorly understood. In this study, we used cell culture and mouse models of huntingtin protein aggregation as well as post-mortem material from patients with Huntington's disease to demonstrate that Argonaute-2 (AGO2) accumulates in the presence of neuronal protein aggregates and that this is due to impaired autophagy. Accumulation of AGO2, a key factor of the RNA-induced silenci

This study introduces an innovative electrochemiluminescence (ECL) biosensor for the highly sensitive and specific detection of Argonaute 2 (Ago2) activity. Ago2, a key enzyme in the RNA interference (RNAi) pathway, plays a crucial role in gene regulation, and its dysregulation is associated with diseases such as cancer and viral infections. The biosensor integrates hybridization chain reaction (HCR) amplification and the CRISPR/Cas12a system, leveraging a multi-stage signal amplification strategy. The detection mechanism begins with Ago2-mediated cleavage of a designed hairpin RNA (HP-RNA), r

Extracellular particles, including extracellular vesicles (EVs) and non-vesicular extracellular particles (NVEPs), enable intercellular communication by transferring regulatory miRNAs and other biomolecules. While EVs have been studied for drug delivery, NVEPs remain relatively unexplored. Exomeres, a recently discovered class of NVEPs enriched in RNAi proteins, preferentially carry miRNAs and deliver them to cells more effectively than EVs, underscoring their potential as vehicles for therapeutic RNAs. One current limitation to studying and applying exomeres for therapeutic RNA delivery is th

Argonaute (Ago) proteins play pivotal roles in nucleic acid targeting across eukaryotes and prokaryotes. While eukaryotic Ago (eAgo) proteins utilize RNA guides to cleave complementary RNA, prokaryotic Ago (pAgo) proteins often employ DNA guides for DNA targeting. This study focuses on the short pAgo system from Pleomorphomonas koreensis (Pko-SPARDA), which consists of a DUF4365-APAZ protein (PkoRE) and an Ago protein (PkoAgo). Upon RNA-guided target DNA recognition, Pko-SPARDA exhibits nonspecific cleavage activity toward ssDNA, dsDNA, and RNA, demonstrating potential for nucleic acid detecti
