Asarum heterotropoides (AH) is a notable medicinal plant in traditional Chinese medicine (TCM), but distinguishing and assessing its quality from various origins is challenging due to similar morphology and chemical profiles, using traditional chemical analysis methods. In this study, we developed an effective approach to identify AH from different origins by combining high-performance liquid chromatography (HPLC) fingerprint with chemometrics. The contents of three characteristic constituents (Asarinin, sesamin and methyleugenol) were selected for quantitive analysis, it found that the conten

Our previous study demonstrated that our novel herbal remedy, a mixture of Asarum sieboldii, Platycodon grandiflorum, and Cinnamomum Cassia extracts, exhibits a therapeutic effect in 1-chloro-2,4-dinitrobenzene (DNCB)-induced mice by inhibiting the Th-2 inflammatory response upon oral administration. It also ameliorated imbalances in lipid metabolism related to the skin barrier function in keratinocytes, indicating its potential as a topical agent. This study aims to further investigate the therapeutic effects and metabolic mechanisms of its topical application. The anti-atopic effect was eval

Pain is the cardinal symptom of many debilitating diseases and results in heavy health and economic burdens worldwide. Asarum (Asarum sieboldii Miq.) is a commonly used analgesic in Chinese medicine. However, the analgesic components and mechanisms of asarum in acute and chronic pain mice model remain unknown. In this study, we first generated asarum water extract and confirmed strong analgesic properties in mice in both the acute thermal and mechanical pain models, as well as in the complete Freund's adjuvant (CFA) induced chronic inflammatory pain model. Second, we identified higenamine as a

Aristoloxazines (AXs), aristolochic acids (AAs), and aristolactams (ALs) are potent genotoxins found in Asarum and Aristolochia plants, many of which are commonly used as herbal medicines. Emerging evidence indicates that these compounds contaminate arable soil during the cultivation of Aristolochiaceae herbs. Currently, no method exists for their simultaneous detection. In this study, we developed a high-performance liquid chromatography (HPLC)-based method for their determination. This method employs an iron powder-packed microreactor to convert non-fluorescent AXs and AAs into naturally flu

INTRODUCTION: The similarity in appearance of poisonous and medicinal plants, such as Asarum heterotropoides (AH) and Cynanchum paniculatum (CP), poses safety risks due to frequent confusion. Since AH contains toxic ingredients, the traditional methods of olfactory and gustatory identification cannot be used to distinguish AH from CP.
METHODS: To differentiate them systematically, we proposed a novel strategy based on dual electronic sensors (DES) and dual fingerprint spectra (DFS). The DES included two intelligent sensors, namely the E-nose and E-tongue, which differentiated AH and CP based

Aristolochic acids (AAs) widely exist in such plants as Aristolochia and Asarum. The renal toxicity of AAs as well as its carcinogenicity to urinary system have been widely known. In 2003 and 2004, China prohibited the use of Aristolochiae Radix, Aristolochiae Manshuriensis Caulis and Aristolochiae Fangchi Radix, and required administering other AAs-containing medicines in accordance with the regulations for prescription drugs. In this paper, we retrieved literatures on the content determination of AAs in recent 10 years in China. It suggested that the AAs content is lower in Asarum herb, espe

ETHNOPHARMACOLOGICAL RELEVANCE: Asarum heterotropoides F. Schmidt, a classic Chinese herb, traditionally treats liver deficiency with its pungent, warm nature and liver-qi smoothing properties. Asarinin (ASA), an abundant and characteristic lignan in Asarum, has the potential to protect the liver and has shown measurable systemic exposure in the body, making it a key candidate for verifying this traditional use.
AIM OF THE STUDY: We aim to delineate the molecular mechanisms underlying the hepatoprotective activity of ASA.
MATERIALS AND METHODS: Liver injury and fibrosis was generated in C57B

Bisaspochalasin F (1), a new cytochalasan homodimer featuring a fused tetrahydrofuran ring, was isolated from the endophytic fungus Aspergillus flavipes KIB-636, derived from Asarum heterotropoides Fr. Schmidt. Its structure, including the absolute configuration, was unambiguously established by single-crystal X-ray diffraction analysis and comprehensive spectroscopic data. A biomimetic semisynthesis of 1 was successfully achieved from the natural precursor aspochalasin D via LiOH-mediated dimerization. Notably, this dimerization significantly potentiated the inhibitory effect on Cav3.2 calciu

Aristolochic acid analogues (AAAs) are well-known toxins. We performed the first comprehensive screening on AAAs in Asari Radix et Rhizoma (underground part of Asarum heterotropoides Schmidt), the only Aristolochiaceae plant widely used in clinical practice. LC-HRMS revealed 70 trace AAAs using polygonal mass defect filtering and precursor ion list strategies, 38 of which were newly discovered in A. heterotropoides. UHPLC-QTrap-MS/MS was then utilized for quantitative/semiquantitative analysis of 26 abundant compounds. Seventeen AAAs were detected from 91 batches of A. heterotropoides and 20 A

Asari Radix, the roots of Asarum heterotropoides F. Maekawa var. manshuricum F. Maekawa or A. sieboldii F. Maekawa, has traditionally been used for the treatment of various infectious diseases. Since its MeOH extract inhibited the growth of Listeria monocytogenes in a preliminary test, the aim of this study was to isolate and identify the anti-listerial compounds from the plant. Activity-guided fractionation led to the isolation of seven compounds 1-7 from the MeOH extract, and their chemical structures were identified by comparison of the spectroscopic data with those in the literature. Tomato Fusarium wilt, caused by Fusarium oxysporum f. sp. lycopersici (FOL), is a devastating disease that leads to yield losses of up to 67% in severely affected regions. Current control strategies, such as chemical treatments and resistant cultivars, face challenges including environmental toxicity and pathogen resistance. In this study, we isolated an endophytic fungus, Penicillium sp. JL76001 (JL76001), from the medicinal plant Asarum forbesii, which exhibited strong antagonism against FOL. Plate confrontation assays revealed a 40.48% inhibition of FOL growth, attributed to the secondary m

Spinal cord injury (SCI) is a highly disabling trauma, and the inflammatory response plays a critical role in disease progression. The detrimental inflammatory microenvironment exacerbates neuronal apoptosis and suppresses axonal regeneration. Methyl eugenol (ME), the primary active ingredient in the traditional Chinese medicine Asarum, possesses multiple pharmacological activities, including anti-inflammatory and antioxidant. In this study, we developed glutathione-chitosan modified ME liposomes (GCME-LIPO) for the intranasal delivery in SCI treatment. The GCME-LIPO was achieved by encapsulat

Aristolochic acid (AA), mainly derived from herbal Aristolochia and Asarum plants, was listed as a human carcinogen class I in 2002. Aristolochic acid nephropathy (AAN) is a rapidly progressive tubulointerstitial nephritis and urothelial cancer caused by AA. However, the targeting molecular mechanisms of AAs-induced nephrotoxicity are largely unclear. This study aims to dissect targeting molecular mechanisms of AA-induced nephrotoxicity. Activity-based protein profiling (ABPP) in combination with cellular thermal shift assay (CETSA) was performed to identify the AAs binding target proteins. Ou
