Aspartame: Should Individuals with Type II Diabetes be Taking it?

BACKGROUND: Individuals with Type II Diabetes (T2D) have to manage blood glucose levels to sustain health and longevity. Artificial sweeteners (including aspartame) are suggested sugar alternatives for these individuals. The safety of aspartame in particular, has long been the centre of debate. Although it is such a controversial product, many clinicians recommend its use to T2D patients, during a controlled diet and as part of an intervention strategy. Aspartame is 200 times sweeter than sugar and has a negligible effect on blood glucose levels, and it is suggested for use so that T2D can con

Nephrotoxic Effect of Aspartame as an Artificial Sweetener: a Brief Review.

Aspartame is one of the most popular artificial sweeteners over the world. Although its consumption is considered to be safe in acceptable daily intake ranges which were set by the United States Food and Drugs Administration and other regulatory agencies, there are lots of controversies regarding its safety nowadays. Some of the recent experimental and epidemiological studies showed that consumption of aspartame may causes some adverse health effects including obesity, metabolic syndrome, and alteration in gut microbiota. Moreover, studies on the nephrotoxic effect of aspartame have increased.

The effect of oral aspartame administration on the balance of magnesium in the rat.

The aim of the present work was to determine the effect of aspartame administration on the excretion of magnesium and its distribution in the various rat tissues and organs. The present results have shown that aspartame administration influences the balance of magnesium in the organism, since in some organs and tissues (heart, lungs, kidneys, adrenals, jejunum, hair and blood) it is accumulated, while other organs (liver and testes) are deprived of it. Aspartame administration also affects the excretion of magnesium from the organism, since it decreases the concentration of magnesium in both u

Aspartame prevents the karyomegaly induced by ochratoxin A in rat kidney.

Ochratoxin A (OTA) is a mycotoxin produced by Aspergillus ochraceus as well as other moulds. This mycotoxin contaminates animal feed and food. OTA is immunosuppressive, genotoxic, teratogenic, carcinogenic and is nephrotoxic in all animal species studied so far. OTA inhibits protein synthesis and induces lipid peroxidation. Since it seems impossible to avoid completely contamination of foodstuffs by toxigenic fungi, it is necessary to investigate the possible ways of limiting such toxicity. An attempt to prevent OTA-induced nephrotoxic and genotoxic effects, mainly the karyomegaly, has been ma

Aspartame. Review of safety issues. Council on Scientific Affairs.

This report examines the safety issues related to the nutritive sweetener aspartame, including possible toxic effects of aspartame's component amino acids, aspartic acid and phenylalanine, and its major decomposition products, methanol and diketopiperazine, and the potential synergistic effect of aspartame and dietary carbohydrate on brain neurochemicals. Available evidence suggests that consumption of aspartame by normal humans is safe and is not associated with serious adverse health effects. Individuals who need to control their phenylalanine intake should handle aspartame like any other so

Evaluation of an aspartame loading test for the detection of heterozygotes for classical phenylketonuria.

Classical phenylketonuria (PKU) is an inborn error of metabolism of autosomal recessive inheritance characterized by the accumulation of phenylalanine (Phe) in tissues due to Phe-4-hydroxylase deficiency. Several methods have been developed for the detection of PKU heterozygotes based on the determination of plasma Phe and tyrosine (Tyr) levels, on the analysis of the Phe/Tyr and Phe2/Tyr ratios and on the use of discriminant functions. The objective of the present study was to test the value of loading with aspartame (a sweetener consisting of Phe, aspartate and methanol) for the identificati

The biological properties of aspartame. III. Examination for endocrine-like activities.

A series of studies with aspartame were run in mice, rats and rabbits using standard procedures to characterize possible estrogenic, androgenic, progestational and glucocorticoid activities. Aspartame was administered orally at levels (ca 300 mg/kg/day) substantially in excess of expected maximal human intake when used as a sweetening agent. No significant hormone-mimetic response was observed in the endocrine target organs evaluated. In similar studies, when administered simultaneously with the steroid hormones, it did not reduce the response expected with the steroid. Thus, it was concluded

Formaldehyde derived from dietary aspartame binds to tissue components in vivo.

Adult male rats were given an oral dose of 10 mg/kg aspartame 14C-labelled in the methanol carbon. At timed intervals of up to 6 hours, the radioactivity in plasma and several organs was investigated. Most of the radioactivity found (>98% in plasma, >75% in liver) was bound to protein. Label present in liver, plasma and kidney was in the range of 1-2% of total radioactivity administered per g or mL, changing little with time. Other organs (brown and white adipose tissues, muscle, brain, cornea and retina) contained levels of label in the range of 1/12 to 1/10th of that of liver. In all, the ra

Aspartame and Human Health: A Mini-Review of Carcinogenic and Systemic Effects.

Aspartame, a widely used artificial sweetener, has been at the center of ongoing debates concerning its safety, particularly its potential role in cancer development. This review provides an overview and analysis of the current research exploring the carcinogenic effects of aspartame. It examines findings from in vitro studies, in vivo experiments, and epidemiological investigations to offer a comprehensive perspective on the controversy. The results from these studies remain inconsistent-some suggest a possible association between high aspartame intake and increased cancer risk, while others

The biological properties of aspartame. II. Actions involving the gastrointestinal system.

Aspartame (APM) was investigated in several pharmacological tests to delineate any effects on the gastrointestinal system. The compound did not affect food consumption at one hour following a single intragastric dose of 200 mg/kg in rats. There was no evidence of inhibition or stimulation of the gastric juice secretion rate, the concentration of gastric acid, acid output or proteolytic activity following an intragastric dose of 250 mg/kg in five-hour pylorus-ligated rats. Likewise, APM at the same dosage did not significantly affect gastric ulceration induced by nineteen hours of pylorus-ligat

Aspartame and Phe-Containing Degradation Products in Soft Drinks across Europe.

Phenylketonuria and tyrosinemia type 1 are treated with dietary phenylalanine (Phe) restriction. Aspartame is a Phe-containing synthetic sweetener used in many products, including many 'regular' soft drinks. Its amount is (often) not declared; therefore, patients are advised not to consume aspartame-containing foods. This study aimed to determine the variation in aspartame concentrations and its Phe-containing degradation products in aspartame-containing soft drinks. For this, an LC-MS/MS method was developed for the analysis of aspartame, Phe, aspartylphenylalanine, and diketopiperazine in so

Aspartame promotes intestinal tumor progression via Akkermansia muciniphila -mediated 5-hydroxytryptophan production.

Aspartame, one of the most widely consumed artificial sweeteners globally, has been associated with increased cancer risk in epidemiological studies, yet the underlying mechanisms remain poorly understood. This study investigated the effects of subchronic aspartame exposure on intestinal tumorigenesis using the Apcmin/+ spontaneous intestinal tumor mouse model and elucidated the mechanistic pathways involved. Mice were exposed to physiologically relevant doses of aspartame (0.07 and 0.2 mg/mL) in drinking water for 3 months, followed by comprehensive analysis of tumor development, gut microbio

Aspartame and neurobehavioural symptoms: a bibliometric synthesis across four decades (1985-2025).

This bibliometric synthesis maps the global research landscape on aspartame (ASP) and neurobehavioral symptoms over a 40-year period (1985-2025). Working from 30 included records, we quantified temporal trends, authorship, journal distribution, study designs, and recurring thematic emphases. Interest in the topic has revived, with most records (approximately 60%, 18/30) published from 2015 onwards. The literature is dispersed across many outlets, and animal/in-vivo studies (56.7%, 17/30) outnumber human/clinical studies (30.0%, 9/30). Recurring domains were behavioral, cognitive, and memory-re

Cellular Stress, Inflammation and Barrier Damage in Gut Epithelial Cells Caused by Aspartame.

Aspartame has been widely used as a sweetener in foods and beverages since the 1980s. In this study, we aimed to assess its effects on gut epithelial cell biology, inflammation and the epithelial barrier. We found that aspartame induces cytotoxic effects, disrupts the epithelial barrier and triggers proinflammatory cytokine release in gastrointestinal epithelial cells, organoids and gut-on-a-chip models at concentrations corresponding to daily consumed doses in food products. Cellular cytotoxicity was observed at doses as low as 1.25 mg/mL. RNA sequencing analysis revealed that aspartame signi

Aspartame-induced cognitive dysfunction: Unveiling role of microglia-mediated neuroinflammation and molecular remediation.

Aspartame, an artificial sweetener, is consumed by millions of people globally. There are multiple reports of aspartame and its metabolites affecting cognitive functions in animal models and humans, which include learning problems, headaches, seizures, migraines, irritable moods, anxiety, depression, and insomnia. These cognitive deficits and associated symptoms are partly attributed to dysregulated excitatory and inhibitory neurotransmitter balance due to aspartate released from aspartame, resulting in an excitotoxic effect in neurons, leading to neuronal damage. However, microglia, a central
