Temporary modification of salivary protein profile and individual responses to repeated phenolic astringent stimuli.

The extent of the change in salivary protein characteristics after repeated stimulations was shown to be correlated to differences in perceived astringency. Salivary characteristics of 77 subjects were compared after masticatory (S1) and taste/masticatory (S2) stimulations. The variations (S2 minus S1) of protein concentration and saliva haze-forming capacity (HFC) were used to define 3 subject groups: low responding (LR, n = 20), medium responding (MR, n = 37), and high responding (HR, n = 20). Salivary protein concentration did not change in LR subjects; decreased a little, but significantly

Astringent Effects of Red Wine Associated with Responses of Aquaporins Found in Human Tongue and Salivary Tissues.

Aquaporin (AQP) water channels facilitate fluid transport across cell membranes, are implicated in rodent oral water sensing, and were examined in the human tongue for their modulation by mouthfeel compounds and ensuing effects on perception and saliva tribology. Immunohistochemistry demonstrated abundant AQP1 and AQP2 and moderate AQP5 in human tongue and high AQP5 in salivary gland. Trained human tasting panels evaluated astringency intensities; results correlated with wine tannin measures and inhibition of AQP1 water flux (R2 ≥ 0.9). Wine tannin extract additions of 1 g/L reduced cell swell

Study of Serial Exposures of an Astringent Green Tea Flavonoid Extract with Oral Cell-Based Models.

It is well known that repeated exposure to phenolic compounds (PCs) raises astringency perception. However, the link between this increase and the oral cavity's interactions with salivary proteins (SPs) and other oral constituents is unknown. To delve deeper into this connection, a flavonoid-rich green tea extract was tested in a series of exposures to two oral cell-based models using a tongue cell line (HSC3) and a buccal mucosa cell line (TR146). Serial exposures show cumulative PC binding to all oral models at all concentrations of the green tea extract; however, the contribution for the fi

Chemical Drivers of Sweetness, Sourness, and Astringency in Apple Juice Identified Using Support Vector Machine Regression.

The role of the main apple compositional parameters (sugars, organic acids, polyphenols, and volatile organic compounds) and their interactions on the perceived sensory attributes of Sweetness, Sourness, and Astringency was investigated in apple juice. Bivariate correlations and Support Vector Machine Regression (SVMR) analyses were used to examine the relationships between all the analyzed features and their relative importance in an interactive mode. By analyzing the contribution of the features to the SVMR models using Shapley values, we found that sweetness was strongly influenced by sucro

Impact of oral astringent stimuli on surface charge and morphology of the protein-rich pellicle at the tooth-saliva interphase.

The proteinaceous pellicle layer, which develops upon contact with saliva on the surface of teeth, is important for the formation of oral biofilms and for the protection of teeth from abrasion and chemically induced erosion. Astringent food ingredients comprising polyphenols, cationic macromolecules, and multivalent metal salts are known to interact with the pellicle. However, astringent-induced changes in the physicochemical properties of the tooth-saliva interphase are not yet completely understood. Here we provide comprehensive insights into interfacial charging, ultrastructure, thickness,

Effect of Different Enological Tannins on Oxygen Consumption, Phenolic Compounds, Color and Astringency Evolution of Aglianico Wine.

BACKGROUND: In the wine industry, in addition to condensed tannins of grape origin, other commercial tannins are commonly used. However, the influence of oxygen uptake related to different tannin additions during the post fermentative phase in wine has not been completely investigated. In this study, we evaluated the influence of four different commercial tannins (namely, condensed tannins, gallotannins, ellagitannins and tea tannins) during four saturation cycles. METHOD: Wine samples were added with four different tannin classes (30 g/hL) as to have 5 different experimental samples: control

Dual Mechanisms of pH-Dependent Tea Astringency: Binding and Metabolism of Polyphenols Revealed by In Vivo and In Vitro Oral Processing.

pH critically influences the astringency of tea, yet the underlying mechanisms remain poorly understood. This study aimed to elucidate how pH modulates tea astringency through dual mechanisms (polyphenol-saliva binding and polyphenol oral metabolism) with comparable in vivo and in vitro oral processing (OP) models. After optimizing OP parameters (10 mL, 10 s), the effect of pH (3.0-6.0) on green tea infusion and representative polyphenols (epigallocatechin-3-O-gallate (EGCG), epigallocatechin (EGC), and gallic acid (GA)) was investigated by sensory evaluation, turbidimetry, and paper spray mas

Identification of potential flavonoids strongly correlated with bitterness and astringency in quinoa seedling vegetables via metabolomics and electronic tongue.

Quinoa (Chenopodium quinoa Willd.) seedlings are nutrient-dense leafy vegetables, but their inherent bitterness and astringency limit consumer acceptance. The chemical basis underlying these sensory attributes remains largely unclear. In this study, four quinoa seedling varieties differing in seed coat colour were evaluated using electronic tongue analysis in combination with comprehensive metabolomics. Among the detected flavor attributes, bitterness and astringency were the most prominent in the four identified quinoa seedling varieties. Meanwhile, 2060 metabolites were identified across the

Fatty Acid-Induced Modulation of Saliva Protein-Polyphenol Interactions: Molecular Signatures Underlying the Camembert Effect of Astringency.

The first contact of polyphenols with the human body occurs in the mouth, where they are known to interact with proline-rich proteins (PRPs), leading to physicochemical and biochemical changes in the salivary pellicles and epithelia of the oral cavity. These interactions are important at the sensory level for the development of astringency. Organoleptic tests have also shown that astringency can be attenuated when fatty foods are consumed, a phenomenon known as the Camembert effect. Employing isothermal titration calorimetry, fluorescence, and CD spectroscopy, we investigated the binding proce

L-Proline-Mediated Modulation of Astringency in Black Chokeberry Puree: Molecular Interactions, Process Optimization, and Quality Preservation.

Aronia melanocarpa puree is rich in anthocyanins and proanthocyanidins, but its pronounced tannin-derived astringency limits product acceptance. This study developed a non-removal astringency-modulation strategy using food-grade L-proline and evaluated its molecular basis, processing window, and quality effects. Ultraviolet-visible (UV-Vis) and Fourier-transform infrared (FT-IR) spectroscopic analyses suggested that L-proline altered the local microenvironment of procyanidin B2 (PC-B2) through hydrogen bonding, hydrophobic association, and molecular packing rearrangement, without evident disru

Impact of food additives (sodium carbonate and ammonium bicarbonate) on interactions between astringent compounds and an oral cell model.

Many foods contain additives like sodium carbonate and ammonium bicarbonate to ensure safety, preserve quality, or extend shelf life. Recent studies suggest these additives may influence interactions between astringent compounds and oral cells. Using a tongue epithelium cell model, we investigated how these salts affect interactions between astringent compounds (phenolic and non-phenolic) and oral constituents. The salts reduced the interaction of green tea flavanol-rich extract (GTE) with salivary proteins and altered interactions with tannic acid and alum. The effects varied depending on the

Comprehensive Phenolic Profiling and Antioxidant Evaluation of Calyx, Rind, and Edible Tissues From Non-Astringent Persimmons (Diospyros kaki).

Persimmon (Diospyros kaki L.) processing generates substantial quantitiesof underutilized by-products despite their richness in bioactive compounds. This study evaluated the phenolic content, antioxidant potential, and phytochemical composition of four tissues (calyx, rind, pulp, inner pulp) from two non-astringent cultivars, namely, Fuyu and Jiro. Phenolic estimation (TPC, TFC, and TTC) and seven antioxidant assays (DPPH, ABTS+, ·OH-RSA, FRAP, RPA, TAC, and FICA) revealed strong tissue-specific variation. Calyx tissues displayed the highest phenolic concentrations and antioxidant activities,

Prefrontal hemodynamic responses to astringency aftertaste across alcoholic and alcohol-free matrices: A functional near-infrared spectroscopy study.

Astringency is an important mouthfeel attribute in many beverages, yet its neurophysiological correlates remain poorly understood. It also remains unclear how beverage matrix shapes frontal hemodynamic responses to astringency over time. This study used functional near-infrared spectroscopy (fNIRS) to characterize prefrontal hemodynamic responses to proanthocyanidin (PAC)-induced astringency in three matrices: water, ethanol, and red wine. To capture temporal aspects of the perceptual experience, responses were assessed separately during the induction and aftertaste phases. Sensory evaluation

Integrated Transcriptomic and Proteomic Analysis Identifies Novel Regulatory Genes Associated with Plant Growth Regulator-Induced Astringency in Grape Berries.

Astringency influences the sensory characteristics and flavor quality of table grapes. We tested the astringency sensory attributes of berries and investigated the concentration of flavan-3-ols/proanthocyanidins (PAs) in skins after the application of the plant growth regulators CPPU and GA3 to the flowers and young berries of the "Summer Black" grape. Our results showed that CPPU and GA3 applications increase sensory astringency perception scores and flavan-3-ol/proanthocyanidin concentrations. Using integrated transcriptomic and proteomic analysis, differentially expressed transcripts and pr

Astringency, bitterness and color changes in dry red wines before and during oak barrel aging: An updated phenolic perspective review.

To understand effects of using oak barrels on the astringency, bitterness and color of dry red wines, phenolic reactions in wines before and after barrel aging are reviewed in this paper, which has been divided into three sections. The first section includes an introduction to chemical reactivities of grape-derived phenolic compounds, a summary of the phenolic reactions that occur in dry red wines before barrel aging, and a discussion of the effects of these reactions on wine astringency, bitterness and color. The second section introduces barrel types that determine the oak barrel constituent

A study on the dynamic differences and component correlations of astringency in green tea, black tea and oolong tea based on TI/TDS and LC-MS.

The dynamic patterns and chemical drivers of astringency across tea varieties remain underexplored. This study integrated time-intensity (TI), temporal dominance of sensations (TDS), electronic tongue, and LC-MS analyses to evaluate astringency in six green, black, and oolong teas. TI results indicated that green tea exhibited higher peak intensity, larger area under the curve (AUC), longer duration, and faster onset compared to oolong and black teas. TDS analysis revealed dominant sensations: "harsh" and "coarse grain" for green tea, "smooth" and "dry" for black tea, and alternating "rough" a

Green Synthesized WO3-PEDOT:PSS Composites into Benzoid-Rich Conformation for Nonenzymatic Bioelectronic Tongues for Tannic Acid-Based Wine Astringency Detection.

This study introduces electrochemical sensor materials based on a conductive polymer PEDOT:PSS and a green-synthesized WO3 electrode (i.e., polymer composites of PEDOT:PSS@WO3-n, where n = 1-3 with three wt. ratios of PEDOT:PSS/WO3 = 1/1, 1/2, and 1/3, respectively) to directly measure tannic acid in beverages. Besides the conductive polymer PEDOT:PSS alone, the electro-exploding wire technique is used to prepare WO3 nanoparticles and their modified polymer composites of PEDOT:PSS@WO3-n (n = 1-3), thereby imparting a benzoid-rich conformation and improving its electrochemical sensitivity to ta

Green tea polyphenol epigallocatechin gallate activates TRPA1 in an intestinal enteroendocrine cell line, STC-1.

A characteristic astringent taste is elicited by polyphenols. Among the polyphenols, catechins and their polymers are the most abundant polyphenols in wine and tea. A typical green tea polyphenol is epigallocatechin gallate (EGCG). Currently, the mechanism underlying the sensation of astringent taste is not well understood. We observed by calcium imaging that the mouse intestinal endocrine cell line STC-1 responds to the astringent compound, EGCG. Among major catechins of green tea, EGCG was most effective at eliciting a response in this cell line. This cellular response was not observed in HE
