Endoscopic manometry of the sphincter of Oddi in dogs. Pharmacological effects of anticholinergic and selective antimuscarinic agents.

A dog model was used for direct measurement of the sphincter of Oddi using endoscopy. The effects of anticholinergic agents and a selective antimuscarinic agent on the canine sphincter of Oddi motility were studied. Manometric tracing at the canine sphincter showed rhythmical phasic contractions which were not affected by respiration. The mean values of the basal motility were as follows: the peak pressure was 61.5 +/- 2.2 (m +/- SE) cmH2O, trough pressure was 2.18 +/- 1.2 cmH2O, and the frequency was 17.4 +/- 0.2 waves/min. Intravenous administration of atropine sulfate, butropium bromide, bu

Behavioral effects of phencyclidine and ketamine alone and in combination with other drugs.

The behavioral effects of phencyclidine (PCP) and ketamine administered alone and in combination with naloxone, atropine, methyl atropine, chlorpromazine and d-amphetamine were studied in squirrel monkeys trained to press a response lever under a fixed-ratio 30 schedule maintained by the termination of a stimulus associated with electric shock presentation. Under non-drug conditions, a period of high-rate responding in the presence of the stimulus associated with shock presentation was followed by a period of no responding during a 40-s timeout scheduled between fixed-ratio components. Pineal hyperactivity in spontaneously hypertensive rats: muscarinic regulation of indole metabolism.

1. Choline acetyltransferase activity and [3H]quinuclidinyl benzylate-binding sites were detected in the pineal gland of normotensive Wistar-Kyoto rats and of spontaneously hypertensive rats. 2. In vitro, muscarinic activation by pilocarpine increased the pineal metabolic production of hydroxyindole derivatives up to 5-hydroxytryptamine and produced a less marked stimulation of melatonin biosynthesis. 3. Electrical field stimulation of pineal gland slices caused similar metabolic effects. 4. Muscarinic blockade with atropine inhibited the effects on hydroxyindole metabolism. 5. [3H]Quinuclidin

Bladder electromyograms and function in monkeys after atropine.

Voiding induced in conscious monkeys by infusion of sterile saline into the bladder via a chronically implanted bladder catheter was stable over many months. Artefact-free recordings of electrical activity were obtained from the bladder neck and dome of these animals during bladder filling and voiding. Characteristic voiding electromyograms were recorded from both sites but were seen first in the record from the neck. Small doses of methyl-atropine abolished both voiding and the electromyograms; evoked responses to sacral ventral root stimulation were similarly prevented. Therefore the bladder

Duration and effect of single-dose atropine: paralysis of accommodation in penalization treatment of functional amblyopia.

BACKGROUND AND PURPOSE: Atropine dilates the pupil and paralyzes the ciliary muscle accommodation, blurring vision, and therefore is an effective penalization of the sound eye in the treatment of functional amblyopia of the other eye. The degree of blur induced is a function of the amount of the patient's uncorrected hyperopia and the distance from the eye of the viewed material or object. Another factor determining effectiveness of atropine penalization is the duration of the effect of the atropine. It is the purpose of this study to investigate these factors. METHODS: Six normal children un

Ipratropium bromide: a review of its pharmacological properties and therapeutic efficacy in asthma and chronic bronchitis.

Ipratropium bromide is an anticholinergc bronchodilator administered by inhalation. Although producing bronchodilation in most patients with obstructive airways disease, it is somewhat less effective than beta 2-adrenoceptor agonist drugs such as salbutamol or fenoterol in patients with asthma, but is at least as effective as these agents in bronchitis. As with the beta 2-adrenoceptor agonists, the onset of maximum effect with ipratropium (about 1.5 to 2 hours) is slower than with isoprenaline (although significant bronchodilation usually occurs within seconds or minutes of ipratropium inhalat

A heat-stressed rat model to determine relative anticholinergic and anticholinesterase drug potency.

We have reported that atropine, the prototype of muscarinic anticholinergic drugs, elicits a dose-dependent increase in core temperature of heat-stressed rats. In the present study, we have quantified the effects of other anticholinergic drugs on increments in core temperature and have derived the following potencies relative to atropine: imipramine 0.004, amitriptyline 0.02, chlorpromazine 0.1, atropine 1, L-hyoscyamine 2, atropine methyl nitrate 4, and scopolamine 16. Additionally, we quantified the efficacy of carbamates to reduce the elevated heating rate of atropinized rats as a measure o

A protein kinase C-dependent mechanism(s) is involved in atropine-resistant accumulation of diacylglycerol in rat parotid gland.

We investigated the atropine-resistant accumulation of sn-1,2-diacylglycerol (DAG) on stimulation of 1 microM carbachol (CCh) in parotid acinar cells. CCh-induced DAG accumulation was biphasic, peaking at 5 and 20 min. Atropine inhibited two peaks in a dose-dependent manner, but the first peak was inhibited much more than the second one. Atropine (10 microM) completely inhibited both DAG accumulation and the breakdown of phosphatidylinositol 4-monophosphate and 4,5-bisphosphate (PIP2) 5 min after stimulation with CCh. In contrast, the breakdown of PIP2 at 20 min was completely inhibited by 10

The disordering effect of hyoscyamine drugs on phospholipid membranes.

The effect of hyoscyamine drugs on the fluidity of dipalmitoylphosphatidylcholine liposomes has been studied by differential scanning calorimetry (DSC), electron spin resonance spectroscopy (ESR), fluorescence polarization and freeze-fracture electron microscopic techniques. DSC results indicate that anisodamine, anisodine, atropine and scopolamine all increase the fluidity of dipalmitoylphosphatidylcholine liposomes but with different degrees of efficiency. The increasing of fluidity of dipalmitoylphosphatidylcholine liposomes by hyoscyamine drugs is in a dose-dependent way. Increase of the f

Unexpected potentializing effect of a tacrine derivative (9-amino-7-methoxy-1,2,3,4 tetrahydroacridine) upon the non-epileptic myoclonus in baboons Papio papio.

1. The influence of 7-methoxytacrine (7-MEOTA) on the non epileptic myoclonus of the Papio papio baboon was studied in 5 animals. 2. This type of myoclonus is thought to depend on a cholinergic system dysfunction since it can be induced by atropine and blocked by physostigmine. 3. 7-MEOTA, a tacrine derivative, is believed to display a conspicuous anticholinesterase activity but, surprisingly, it here potentiated the non epileptic myoclonus occuring either spontaneously or induced by atropine. 4. In baboons not spontaneously presenting the non epileptic myoclonus, 7-MEOTA induced the myoclonus

Anticholinergic behavioral effect of phencyclidine.

Phencyclidine (PCP) impaired spatial alternation performance in rats. This effects was mimicked by antimuscarinic anticholinergics (scopolamine, atropine) and PCP derivatives (ketamine, cyclohexamine) but not by a variety of other agents. Muscarinic cholinergic agonists antagonized PCP. Impairment of spatial alteration performance by PCP appears to be mediated, at least in part, by and anticholinergic action; this is the first instance of a behavioral effect of PCP that can be largely attributed to a specific mechanism.

Autonomic nervous system activity in Macaca fascicularis.

This study was performed to determine the contributions of the sympathetic and parasympathetic nervous systems to cardiovascular control. Hexamethonium was administered to block the autonomic ganglia, propranolol to block beta adrenergic receptors of the sympathetic nervous system, and methylatropine to block the parasympathetic nervous system. The results of this study indicate high sympathetic tone and low parasympathetic tone in resting Macaca fascicularis. These findings are different from those in man but are similar to other nonhuman primates.

Radial arm maze performance of mice: acquisition and atropine effects.

CD-1 mice were successfully trained in a six-arm radial maze in which half of the arms were baited, a procedure which had been used to differentiate between reference and working memory. Stable performance was achieved following eight daily training sessions, as measured by decreasing running time and number of errors. This finding strengthens the foraging hypothesis as a basis for the performance of rodents in the radial maze. Acute subcutaneous administration of the cholinergic antagonist atropine sulfate (1-6 mg/kg) to trained mice produced dose-related increases in running time and working

Cholinergic activation produces psychosomatic digitalis toxicity.

The purpose of this study was first to develop an animal model for stress-induced digitalis toxicity in such a way that physical and psychological factors might be kept separate. The other purpose was to begin an analysis of the mechanism of this psychosomatic effect. To do this, free-ranging guinea pigs were subjected to repeated sessions of signaled shock (Pavlovian fear conditioning) or to signal alone. On a probe day in which signals were delivered as usual but shock was not administered, guinea pigs were infused i.v. with ouabain after an i.p. injection of either saline, atropine sulfate

Uniformly sized molecularly imprinted polymer for atropine and its application to the determination of atropine and scopolamine in pharmaceutical preparations containing Scopolia extract.

A uniformly sized molecularly imprinted polymer (MIP) for atropine has been prepared. The MIP was prepared using 2-(trifluoromethyl) acrylic acid and ethylene glycol dimethacrylate as a functional monomer and cross-linker, respectively, by a multi-step swelling and thermal polymerization method. The selectivity factor, which is defined as the ratio of the retention factors (k) on the molecularly imprinted and non-imprinted polymers, k(imprinted)/k(non-imprinted), was 2.2 for atropine on the MIP. The obtained MIP was applied for the determination of tropane alkaloids (atropine and scopolamine)

Cost-effectiveness analysis of myopia management: A systematic review.

The rising prevalence of myopia is a major global public health concern. Economic evaluation of myopia interventions is critical for maximizing the benefits of treatment and the healthcare system. This systematic review aimed to evaluate the cost-effectiveness of interventions for treating myopia. Five databases were searched - Embase, Emcare, PubMed, Web of Science, and ProQuest - from inception to July 2022 and a total of 2,099 articles were identified. After careful assessments, 6 studies met the eligibility criteria. The primary outcomes of this systematic review were costs, quality-adjust

Central versus peripheral anticholinergic activity as assessed by two in vivo procedures in mice.

The activity of tertiary and quaternary anticholinergic drugs was compared in two different test procedures designed to measure cholinolytic activity in mice. The four drugs utilized were atropine sulfate, atropine methylnitrate, scopolamine hydrobromide, and scopolamine methylnitrate. The results led to the conclusion that one of these test procedures, the induction of mydriasis (increase in pupil size), primarily measures peripheral anticholinergic activity whereas the other procedure, inhibition of physostigmine lethality, primarily measures anticholinergic activity in the CNS. These two te

Twice-daily administration improves the effectiveness of 0.01% atropine for controlling myopia in slowing axial elongation and refractive progression.

PURPOSE: It remains unclear whether increasing the frequency of administration of 0.01% atropine improves its effectiveness in slowing axial elongation or refractive progression. This study compared twice-daily 0.01% atropine administration and traditional once-nightly administration. METHODS: A hybrid design was adopted incorporating both retrospective and prospective elements, with participants observed for 1 year. The control group (received saline eye drops) and once-daily 0.01% atropine group (Atropine-1 group) were derived from two previous trials, while the twice-daily 0.01% atropine g

Atropine effects on delayed discrimination performance of rats.

The effects of atropine sulfate (ATS) and atropine methyl nitrate (ATM) on the conditional discrimination behavior of rats were investigated in eight-hour experimental sessions. Responding of rats was reinforced on either a lighted or a darkened lever depending on whether lights over both levers had been on during the preceding sample portion of the trial. Zero-delay and four-second-delay trials were randomly interspersed. Quality of performance was analyzed using the A' sensitivity measure of signal detection theory. Both drugs reduced both sensitivity and the percentage of trials on which re

Effects of 8-(2-fluoroethyl)-3 alpha-hydroxy-1 alpha H, 5 alpha H-tropaniumbromide benzilate (Ba598Br) on allergy- and drug-induced asthmas.

The effects of Ba598Br, a new atropine derivative possessing a quarternary ammonium salt structure, on the canine airway, including its antiasthmatic effects, were investigated. The in vivo airway resistance was determined using the modified Konzett-Rössler method. Inhalation of 0.01% of Ba598Br had an inhibitory effect against acetyl-choline (ACh, 10 micrograms/kg i.v.)-evoked bronchoconstriction. The effect of Ba598Br was more powerful and longer lasting than that of the same dose of atropine. Pretreatment with Ba598Br (0.3%) and atropine (0.3%) by inhalation produced a remarkable inhibitory
