Hyper-activation of Aurora kinase a-polo-like kinase 1-FOXM1 axis promotes chronic myeloid leukemia resistance to tyrosine kinase inhibitors.

BACKGROUND: Chronic myeloid leukemia (CML) is a myeloproliferative disease caused by the constitutive tyrosine kinase (TK) activity of the BCR-ABL1 fusion protein. Accordingly, TK inhibitors have drastically changed the disease prognosis. However, persistence of the transformed hematopoiesis even in patients who achieved a complete response to TK inhibitors and the disease relapse upon therapy discontinuation represent a major obstacle to CML cure. METHODS: Thiostrepton, Danusertib and Volasertib were used to investigate the effects of FOXM1, AKA and Plk1 inhibition in K562-S and K562-R cells

High-throughput discovery and characterisation of pentafluorobenzene sulfonamide modifiers of Aurora A kinase.

Covalent modification can enable understanding and modulation of protein function, and the identification of new therapeutic opportunities. A "direct to biology" workflow was developed that harnesses sulfonylation as a connective reaction for the synthesis of diverse sets of reactive fragments. The workflow expanded the diversity of accessible reactive fragment sets, and facilitated the discovery of pentafluorobenzene sulfonamides that modify Aurora A kinase, NEK7 kinase, and UbcH5B. Characterisation of several of the Aurora A-modifying reactive fragments revealed both their modification rates

Aurora Kinase A: The Prominent Oncogenic Link in Helicobacter pylori-Driven Gastric Carcinogenesis.

Chronic Helicobacter pylori (H. pylori) infection leads to gastric carcinoma (GC), while aurora kinase A (AURKA) is known to be upregulated in several cancers. However, the direct association between AURKA and H. pylori remains largely unexplored. The significance of AURKA in H. pylori infection was investigated using an RNAi-mediated silencing method. The expression of downstream signaling genes and apoptotic markers was analyzed through qRT-PCR and western blot. Cancerous properties were evaluated through scratch wound assay, cell counting through trypan blue, and genomic instability assay.

Targeting Aurora A Kinase (AAK) in Platinum-Resistant High Grade Serous Ovarian Cancer.

Aurora A kinase (AAK) involved in G2-M transition is functionally involved in centrosome maturation and maintaining an active spindle assembly checkpoint. We tested the hypothesis that in platinum-taxane resistant high grade serous ovarian cancer (HGSOC) inhibition of AAK involved in G2-M transition would enhance the anti-tumor activity of cisplatin (CP) or paclitaxel (PT). Using HGSOC cell lines from platinum-taxane refractory patients that do not harbor BRCA1/2 mutations, we tested the anti-tumor activity of CP, or PT alone or in combination with the AAK inhibitor alisertib (AL). Treatment w

Aurora Kinases in Ocular Malignancies: Functional Significance and Potential Implications.

BACKGROUND: Ocular malignancies are rare compared to tumors affecting other organs. The treatment options for these cancers remain limited, mostly restricted to nonspecific chemotherapeutic drugs, radiation therapy, and surgery. Because of their infrequency, these tumors have received minimal attention in the broader field of cancer biology. In recent years, efforts have focused on elucidating the role of molecular pathways driving these ocular tumors with the goal of developing targeted therapies. SUMMARY: Aurora kinases (AURKs), specifically aurora kinase A (AURKA) and aurora kinase B (AURK

The activity of Aurora kinase B is required for dengue virus release.

Flaviviruses, such as Dengue (DENV), Zika, Yellow Fever, Japanese Encephalitis and West Nile are important pathogens with high morbidity and mortality. The last estimation indicates that ∼390 millions of people are infected by DENV per year. The DENV replicative cycle occurs mainly in the cytoplasm of the infected cells and different cytoplasmic, nuclear and mitochondrial proteins participate in viral replication. In this paper we analyzed the participation of Aurora kinase B (AurKB) in the DENV replicative cycle using the specific AurKB inhibitor ZM 447439. The kinase inhibition does not alte

An update on the pharmacokinetics and pharmacodynamics of alisertib, a selective Aurora kinase A inhibitor.

Human Aurora kinases, including Aurora kinase A (AURKA), B (AURKB), and C (AURKC), play an essential role in mitotic events such as monitoring of the mitotic checkpoint, creation of bipolar mitotic spindle and alignment of centrosomes on it, also regulating centrosome separation, bio-orientation of chromosomes and cytokinesis. AURKA and AURKB are key regulators of mitosis and centrosome via polymerizing microfilaments and controlling chromatid segregation. In particular, AURKA plays critical roles in the regulation of mitotic entry, centrosome function, bipolar spindle assembly, and chromosome

Arabidopsis TPXL Proteins Mediate a Selective Aurora Kinase-Microtubule Association During Cell Division.

Microtubule organization during cell division requires coordinated regulation by microtubule-associated proteins and protein kinase signaling pathways. In animals, TPX2 (Targeting Protein for Xklp2) regulates spindle assembly by binding to and activating Aurora A kinase. In Arabidopsis, individual TPX2-like (TPXL) proteins have been implicated in Aurora-associated functions, but whether the TPXL family members share common microtubule-associated properties and how TPXL-Aurora interactions differ across the family remain incompletely understood. Here, we investigated the structural and cellular

Regulation of Aurora-A kinase gene expression via GABP recruitment of TRAP220/MED1.

The TRAP/Mediator coactivator complex serves as a functional interface between DNA-bound transactivators and the RNA polymerase II-associated basal transcription apparatus. TRAP220/MED1 is a variably associated subunit of the complex that plays a specialized role in selectively targeting TRAP/Mediator to specific genes. Ablation of the Trap220/Med1 gene in mice impairs embryonic cell growth, yet the underlying mechanism is unknown. In this report, we identified distinct cell growth regulatory genes whose expression is affected by the loss of TRAP220/MED1 by RNA interference. Among the down-reg

Effect of Aurora kinase B on polyploidy and decidualization in mouse uterus.

RESEARCH QUESTION: Decidualization is critical to the establishment of mouse normal pregnancy. The fibroblast-like stromal cells in the process form polyploid multinucleated cells. Aurora kinase B (Aurora B) has previously been shown to regulate polyploidy in various cells. However, whether Aurora B regulates the formation of decidual cell polyploidization and its regulatory mechanisms remain poorly understood. DESIGN: Establish decidualization model of mouse primary endometrial stromal cells in vitro. Construct pseudopregnancy mouse models and delayed-activation mouse models. Detect Aurora B

Analysis of Aurora-A and hMPS1 mitotic kinases in mantle cell lymphoma.

Aurora-A and hMPS1 are kinases involved in spindle checkpoint and centrosome duplication regulation and whose alterations have been associated with cell transformation and chromosome instability in different tumor models. In this study, we have examined the possible alterations of these genes in 58 mantle cell lymphomas (MCLs) and 4 MCL-related cell lines. Aurora-A was also examined in 46 diffuse large B-cell lymphomas (DLBCLs). Aurora-A and hMPS1 mRNA expression levels were related to tumor proliferative activity. Interestingly, a MCL case with the highest number or chromosomal imbalances als

AT9283 exhibits antiproliferative effect on tyrosine kinase inhibitor‑sensitive and ‑resistant chronic myeloid leukemia cells by inhibition of Aurora A and Aurora B.

Imatinib is the gold standard in the conventional treatment of chronic myeloid leukemia (CML). However, some patients become resistant to imatinib therapy. To overcome this resistance, second‑generation (dasatinib, nilotinib, and bosutinib) and third‑generation (ponatinib) tyrosine kinase inhibitors (TKIs) have been developed and have been shown to be effective against refractory CML. Although these TKIs provide many benefits for patients with CML, advanced patients show resistance even to these TKIs. Therefore, novel therapeutic strategies are urgently needed for the treatment of TKI‑resistan

Integrated computational and experimental identification of Aurora A kinase inhibitors for breast cancer therapy.

The present study aimed to identify potential Aurora A kinase (AURKA) inhibitors using an integrated computational and experimental approach. A total of 281 curated AURKA inhibitors from the ChEMBL database, with IC50 ≤ 1 nM, were analysed using physicochemical descriptors and clustering techniques, with K-means showing the best performance (0.504). The compounds were grouped into three clusters and used to generate a pharmacophore model. Hydrogen-bond acceptors, donors, hydrophobicity, and aromatic rings were identified as key pharmacophoric features. Among the generated models, ADDRR and AAA

Deletion of Aurora kinase A prevents the development of polycystic kidney disease in mice.

Aurora Kinase A (AURKA) promotes cell proliferation and is overexpressed in different types of polycystic kidney disease (PKD). To understand AURKA's role in regulating renal cyst development we conditionally deleted the gene in mouse models of Autosomal Dominant PKD (ADPKD) and Joubert Syndrome, caused by Polycystin 1 (Pkd1) and Inositol polyphosphate-5-phosphatase E (Inpp5e) mutations respectively. We show that while Aurka is dispensable for collecting duct development and homeostasis, its deletion prevents cyst formation in both disease models. Cross-comparison of transcriptional changes im

Effects of AZD1152, a selective Aurora B kinase inhibitor, on Burkitt's and Hodgkin's lymphomas.

We studied the effects of AZD1152, an Aurora B kinase inhibitor, on Burkitt's lymphoma (BL) and Hodgkin's lymphoma (HL) in human tissues and cell cultures and in a murine xenograft model of lymphoma. Aurora kinase A and B levels were assessed by RT-PCR and immunohistochemistry. They were aberrantly expressed in BL and HL cell lines, and in lymph nodes from patients with BL and HL. Next, activation of the Aurora B promoter was detected by reporter gene assays. The promoter activity of Aurora B kinase was high in BL cell lines and the Aurora B promoter contained a positive regulatory region betw
