Semen extracellular vesicles mediate vertical transmission of subgroup J avian leukosis virus.

Subgroup J avian leukosis virus (ALV-J) is a highly oncogenic retrovirus that has been devastating the global poultry industry since the late 1990s. The major infection model of ALV-J is vertical transmission, which is responsible for the congenital infection of progeny from generation to generation. Increasing evidence has suggested that extracellular vesicles (EVs) derived from virus-infected cells or biological fluids have been thought to be vehicles of transmission for viruses. However, the role of EVs in infection and transmission of ALV-J remains obscure. In the present study, semen extr

Anti-CD81 antibody blocks vertical transmission of avian leukosis virus subgroup J.

Control of ALV-J in breed of chicken is still a serious issue that need more attention to be paid. Vertical transmission of ALV-J often give rise to more adverse pathogenicity. However, the way to elimination of ALV-J underlying vertical transmission remains not-well understood. In addition, effective vaccines or drugs have not been developed to prevent and control the transmission of ALV-J so far. CD81, a member of the tetraspanins superfamily, plays important roles in regulating membrane proteins, facilitating cells adhesion or fusion, and also participates in viral infection. The purpose of

Differential response to avian leukosis virus infection exhibited by two chicken lines.

Infection of susceptible chickens with avian leukosis virus (ALV) results in the development of bursal lymphomas. These neoplasms develop within the bursa of Fabricius following a latent period of several months. The response exhibited by two previously uncharacterized chicken lines to ALV infection has been examined. The two lines, Hyline SC and FP, responded differently to ALV infection. During a 24-week period following intravenous ALV infection, 27 of 50 SC chickens developed bursal lymphomas. No lymphomas developed in the 36 FP chickens tested. A majority of the SC chickens that developed

Avian sarcoma/leukosis virus (RCAS)-mediated over-expression of IFITM3 protects chicks from highly pathogenic avian influenza virus subtype H5N1.

Broad-spectrum antiviral activities of interferon-induced transmembrane proteins (IFITMs) are primarily attributed to in vitro inhibition of viral entry. Here, we used an avian sarcoma-leukosis virus (RCAS)-based gene transfer system and successfully generated chicks that constitutively express chicken IFITM3 (chIFITM3). The chIFITM3-overexpressing chicks showed significant protection and disease tolerance against highly pathogenic avian influenza virus (HPAIV) H5N1 (Clade 2.2.1.2). The chicks, overexpressing chIFITM3, also showed delayed onset of clinical symptoms, reduced viral shedding, and

Isolation and identification of avian leukosis virus subgroup J from Luohé Ma chicken.

Henan Province is rich in livestock and poultry resources, home to well-known local breeds, including Gushi, Lushi, and Luohé Ma chickens. These breeds are of substantial agricultural and economic importance and play a pivotal role in preserving biodiversity as well as fostering genetic innovation. Avian leukosis (AL) is a major viral disease affecting the poultry industry, with the J subgroup of avian leukosis virus (ALV-J) emerging as a significant cause of economic losses. In this study, we performed reverse transcription -polymerase chain reaction (RT-PCR) identification on chickens suspec

A novel multi-variant epitope ensemble vaccine against avian leukosis virus subgroup J.

The hypervariable antigenicity and immunosuppressive features of avian leukosis virus subgroup J (ALV-J) has led to great challenges to develop effective vaccines. Epitope vaccine will be a perspective trend. Previously, we identified a variant antigenic neutralizing epitope in hypervariable region 1 (hr1) of ALV-J, N-LRDFIA/E/TKWKS/GDDL/HLIRPYVNQS-C. BLAST analysis showed that the mutation of A, E, T and H in this epitope cover 79% of all ALV-J strains. Base on this data, we designed a multi-variant epitope ensemble vaccine comprising the four mutation variants linked with glycine and serine.

Body weight suppression in broilers naturally infected with avian leukosis virus subgroup J.

Broiler progeny were hatched from avian leukosis virus (ALV)-negative and ALV subgroup J (ALV-J)-positive breeders. ALV-J infection in progeny was identified by p27 antigen capture enzyme-linked immunosorbent assay and reverse transcriptase-polymerase chain reaction specific for ALV-J performed on serum or plasma samples obtained at hatch. Hatch weights did not differ between ALV-negative broilers and ALV-J-positive broilers. Body weights of ALV-J-positive broilers were 64.4% of those of ALV-negative broilers at 1 wk of age. At 8 wk of age, weights of broilers with congenital ALV-J infection w

Resistance to avian leukosis virus lymphomagenesis occurs subsequent to proviral c-myc integration.

Most chicken strains are highly susceptible to avian leukosis virus (ALV) induction of bursal lymphoma, involving proviral integration within the c-myc proto-oncogene, while certain strains are genetically resistant to lymphomagenesis. A nested PCR assay was developed to analyse the appearance of proviral c-myc integrations after ALV infection of lymphoma-susceptible birds, and to determine whether these integrations arise in lymphoma-resistant birds. Proviral c-myc integrations are detected in bursa and other tissues from 6 day-old lymphoma-susceptible birds infected as embryos. The abundance

Development of a quadruplex RT-qPCR for the detection of avian leukosis virus, chicken infectious anemia virus, avian reovirus, and fowl adenovirus.

Avian leukosis virus (ALV), chicken infectious anemia virus (CIAV), avian reovirus (ARV), and fowl adenovirus (FAdV) are important viral pathogens that can transmitted horizontally and vertically, and induce immunosuppression to the poultry flocks. They exhibit diverse pathogenic characteristics in clinical settings, and pose continuous threat to the health of poultry flocks. Here, the specific primers and probes were designed for the env gene of ALV, the VP1 gene of CIAV, the M1 gene of ARV, and the ORF1 gene of FAdV. The RNA standards for ALV, and ARV, and the plasmid DNA standards for CIAV,

Preparation of a detailed restriction map of the avian leukosis virus MAV-2(O).

Unintegrated MAV-2(O) DNA was isolated from infected chicken embryo fibroblasts and inserted into the lambda bacteriophage vector lambda gtWES lambda B. Three x 10(6) bacteriophage plaques were screened, yielding a total of seven clones, six of which contained DNA representing the complete MAV-2(O) genome. Viral DNA was isolated from four of the clones and was used to transfect chicken embryo fibroblasts. All four clones produced virus as monitored by reverse transcriptase assay. When the four cloned viruses were inoculated into 10-day-old embryos, all hatched chickens developed osteopetrosis.

Independent recombination between avian leukosis virus terminal sequences and host DNA in virus-induced proliferative disease.

A cDNA transcript of Rous sarcoma virus, which contained the long terminal repeat (LTR) and some additional 3'-terminal sequences, was inserted into the plasmid pBR322. This recombinant plasmid, p53, was then used as a hybridization probe to detect viral terminal sequences in DNA from a number of tissues of birds with a variety of avian leukosis virus (ALV)-induced proliferative diseases. Using restriction endonuclease digestion and blot hybridization analysis, we detected, in addition to standard ALV genomes, viral terminal sequences linked to host DNA and not to viral genes. In DNA from burs

MARCH2 inhibits avian leukosis virus replication by targeting gp85 for ubiquitination and degradation.

The replication of avian leukosis virus (ALV), a simple retrovirus that induces neoplastic diseases in chickens, is regulated by host factors. Investigating the antiviral activity of these host factors and the underlying mechanisms could provide a theoretical foundation for the prevention and control of ALV infections. In this study, RNA sequencing revealed that ALV-A infection upregulates the expression of membrane-associated RING-CH 2 (MARCH2). Increasing MARCH2 levels inhibited ALV-A replication by 3.47- to 16.98-fold in viral titer, while conversely, knockout of MARCH2 promoted it by 5.04-

Efficacy of an autophagy-targeted DNA vaccine against avian leukosis virus subgroup J.

Infection with the avian leukosis virus subgroup J (ALV-J) can lead to neoplastic disease in chickens, inflicting significant economic losses to the poultry industry. Recent reports have identified inhibitory effects of ALV-J on autophagy, a process involving in innate and adaptive immunity. Inspired by this connection between autophagy and immunity, we developed a novel DNA vaccine against ALV-J which includes co-administration of rapamycin to stimulate autophagy. To measure the efficacy of the developed prototype vaccine, five experimental groups of seven-day-old chickens was immunized three

Avian endogenous retrovirus EAV-HP shares regions of identity with avian leukosis virus subgroup J and the avian retrotransposon ART-CH.

The existence of novel endogenous retrovirus elements in the chicken genome, designated EAV-HP, with close sequence identity to the env gene of avian leukosis virus (ALV) subgroup J has been reported (L. M. Smith, A. A. Toye, K. Howes, N. Bumstead, L. N. Payne, and K. Venugopal, J. Gen. Virol. 80:261-268, 1999). To resolve the genome structure of these retroviral elements, we have determined the complete sequence of two proviral clones of EAV-HP from a line N chicken genomic DNA yeast artificial chromosome library and from a meat-type chicken line 21 lambda library. The EAV-HP sequences from t

Differences in pathogenicity among strains of the same or different avian leukosis virus subgroups.

An efficient induction of wasting disease in chickens by avian leukosis virus (ALV), particularly ALV subgroup C, requires >102 infectious units virus inoculated in mid embryogenesis. The most conspicuous symptoms of the disease were induced by ALV subgroup C; however, significant differences in the occurrence of wasting disease were found among individual members of this subgroup. Almost comparable pathogenicity was exhibited by ALV subgroup D, whereas viruses of subgroups B and A proved to be moderately and almost non-pathogenic, respectively. Using antibodies to cellular antigens, tissue al

Replication of an osteopetrosis-inducing avian leukosis virus in fibroblasts, osteoblasts, and osteopetrotic bone.

Avian leukosis virus (ALV)-induced osteopetrosis is caused by the abnormal growth and differentiation of osteoblasts. To evaluate the role of infection in osteopetrosis induction, the replication of an osteopetrosis-inducing virus (Br21) has been compared in osteopetrotic bone, calvarial-derived osteoblasts, and chick embryo fibroblasts. Much higher levels of infection occurred in diseased bone than in the cultures. Severe cases of osteopetrosis contained 10 times more viral DNA, 30 times more mature capsid protein, 5 to 10 times more Gag precursor protein, and 2 to 3 times more Env protein th

Avian leukosis virus-induced osteopetrosis is associated with the persistent synthesis of viral DNA.

DNAs from 19 cases of avian leukosis virus-induced osteopetrosis have been analyzed for viral sequences. Among these were instances of rapid, intermediate, and slow onset osteopetrosis. The DNAs from osteopetrotic bone contained no evidence for osteopetrosis being caused by proviral insertions into or viral transductions of a host protooncogene. Instead, DNAs from osteopetrotic bone displayed evidence for osteopetrosis being associated with the persistent synthesis of viral DNA. Each of the 19 DNAs contained unintegrated as well as integrated viral DNA. Rapid onset osteopetrosis contained abou
