Role of hippocampal NO in the acquisition and consolidation of inhibitory avoidance learning.

Nitric oxide (NO), an unconventional neurotransmitter in the brain, has been postulated as a retrograde intercellular messenger necessary for the induction, but not the maintenance phase, of activity-dependent forms of synaptic plasticity in the hippocampus. Here we report on the effects of an inhibitory avoidance learning task on hippocampal NO synthase (NOS) activity and on the effects of intrahippocampal infusion of a NOS inhibitor in the acquisition and consolidation of this task in rats. NOS activity increases by 45% in the hippocampus immediately after training (0 min) but not at 60 min

Effects of learned flavor avoidance on grooming behavior in rats.

In Experiment 1, rats were conditioned to avoid saccharin in tapwater by pairing it with LiCl in carboxymethylcellulose (CMC) applied to the fur. Conditioned flavor avoidance (CFA) of saccharin was then assessed in drinking and grooming tests. In Experiment 2, rats were given saccharin CMC on their fur and NaCl in water (or vice-versa) as conditioned stimuli in a CFA paradigm. Two-choice tests (saccharin vs. NaCl) followed in drinking and grooming contexts. In Experiment 3, rats were given saccharin CMC on one flank and vehicle (CMC only) on the other. After grooming, animals were injected wit

Better safe than sorry: Avoidance of interoceptive threat following observational fear learning.

Learning through observation is often adaptive, allowing individuals to acquire important threat-related information without exposing themselves to danger. However, it can become maladaptive when it leads to excessive fear or avoidance in the absence of actual threat. This study examined whether observational learning of an interoceptive threat promotes avoidance behavior and how conditioned responding changes when avoidance is prevented. Forty healthy participants completed a three-phase observational fear conditioning paradigm. In the observational learning phase, participants watched a demo

Diminished reward circuit response underlies pain avoidance learning deficits in problem drinkers.

Individuals engaging in problem drinking show impaired proactive pain avoidance. As successful pain avoidance is intrinsically rewarding, this impairment suggests reward deficiency, as hypothesized for those with alcohol and substance misuse. Nevertheless, how reward circuit dysfunctions impact avoidance learning and contribute to drinking behavior remains poorly understood. Here, we combined functional imaging and a probabilistic learning go/nogo task to examine the neural processes underlying proactive pain avoidance learning in 103 adult drinkers. We hypothesized that greater drinking sever

Chronic Social Defeat Stress Gives Rise to Social Avoidance Through Fear Learning.

Chronic social defeat stress (CSDS), a widely used rodent model of stress, reliably leads to decreased social interaction in stress susceptible animals. Here, we investigate a role for fear learning in this response using 129Sv/Ev mice, a strain that is more vulnerable to CSDS than the commonly used C57BL/6 strain. We first demonstrate that defeated 129Sv/Ev mice avoid a CD-1 mouse, but not a conspecific, indicating that motivation to socialize is intact in this strain. CD-1 avoidance is characterized by approach behavior that results in running in the opposite direction, activity that is cons

Enhanced avoidance behavior in social anxiety: evidence from a probabilistic learning task.

BACKGROUND AND OBJECTIVES: Social phobia is characterized by avoidance of feared social situations. Although avoidance is a central feature of social anxiety, few studies have examined avoidance learning. METHODS: We used a probabilistic instrumental learning paradigm where participants had to learn by trial and error which response led to the disappearance of a neutral or angry face. 20 high socially anxious and 20 non-socially anxious individuals with an average level of social anxiety learned to avoid an angry or a neutral face by choosing one of two cues. Each of the cues led to the disap

The Neural Basis of Avoidance Learning and Its Relation to Safety Processing.

Persistent avoidance plays a major role in maintaining anxiety disorders. The few studies that have explored the neural basis of avoidance have predominantly examined established avoidance behaviors rather than the learning processes that establish avoidance responses. We aimed to characterize the temporal dynamics and neural mechanisms of avoidance learning using a novel paradigm combined with 7 tesla fMRI in 82 healthy participants (37 male and 45 female). Participants were first conditioned to two threat stimuli and a compound safety stimulus. Next, they were trained to avoid one threat sti

The role of learning in risk-avoidance strategies during spider-ant interactions.

Cognitive abilities used by arthropods, particularly predators, when interacting in a natural context have been poorly studied. Two neotropical sympatric predators, the golden silk spider Nephila clavipes and the ectatommine ant Ectatomma tuberculatum, were observed in field conditions where their interactions occurred regularly due to the exploitation of the same patches of vegetation. Repeated presentations of E. tuberculatum workers ensnared in their web triggered a progressive decrease in the capture response of N. clavipes. All the spiders that stopped trying to catch the ant on the secon

Learning to avoid spiders: fear predicts performance, not competence.

We used an immersive virtual environment to examine avoidance learning in spider-fearful participants. In 3 experiments, participants were asked to repeatedly lift one of 3 virtual boxes, under which either a toy car or a spider appeared and then approached the participant. Participants were not told that the probability of encountering a spider differed across boxes. When the difference was large (Exps. 1 and 2), spider-fearfuls learned to avoid spiders by lifting the few-spiders-box more often and the many-spiders-box less often than non-fearful controls did. However, they hardly managed to

Learned helplessness and social avoidance in the Wistar-Kyoto rat.

The Wistar-Kyoto (WKY) rat is an established depression model characterized by elevated anxiety- and depression-like behavior across a variety of tests. Here we further characterized specific behavioral and functional domains relevant to depression that are altered in WKY rats. Moreover, since early-life experience potently shapes emotional behavior, we also determined whether aspects of WKYs' phenotype were modifiable by early-life factors using neonatal handling or maternal separation. We first compared WKYs' behavior to that of Sprague-Dawley (SD), Wistar, and Spontaneously Hypertensive (SH

Asymmetric learning to avoid heterospecific males in Mesocricetus hamsters.

If a female mates with a male of a closely related species, her fitness is likely to decline. Consequently, females may develop behavioral mechanisms to avoid mating with heterospecific males. In some species, one such mechanism is for adult females to learn to discriminate against heterospecific males after exposure to such males. We have previously shown that adult, female Syrian hamsters (Mesocricetus auratus) learn to discriminate against male Turkish hamsters (Mesocricetus brandti) after exposure to a single heterospecific male during 8 days across a wire-mesh barrier. Here we repeated th

Food allergy and eosinophilic esophagitis: learning what to avoid.

Food allergies have increased in prevalence significantly in the past decade and so, apparently, has eosinophilic esophagitis. Although the cause of eosinophilic esophagitis is unknown, allergic responses including food allergies have been implicated. This article reviews both conditions, focusing on how to detect and manage them.

Active avoidance learning differentially activates ERK phosphorylation in the primary auditory and visual cortices of Roman high- and low-avoidance rats.

Understanding the mechanisms underlying conditioned avoidance is a critical step toward the development of novel treatments of anxiety. In this context, the two-way active avoidance (2WAA) task is a validated paradigm to investigate uncontrolled avoidance, a hallmark of anxiety disorders. The outbred Roman high- (RHA) and low-avoidance (RLA) rat lines are selected for respectively rapid vs. poor acquisition of active avoidant behavior, and emotional reactivity appears to be the most prominent behavioral difference between the two lines, with RLA rats being more fearful/anxious than their RHA c

The impact of social support cues on pain-related fear and avoidance learning.

Recently, social support cues have been suggested to be "unique" safety signals possessing fear inhibitory properties. Yet, previous research only included autonomic fear measures (i.e., skin conductance) in passive fear conditioning procedures, therefore the impact of social support cues on operant pain-related avoidance learning and self-reported fear and pain-expectancy is unknown. In contrast, previous research showed that social threat cues facilitate pain-related fear learning, but their impact on operant pain-avoidance learning has never been investigated. Pain-free participants (N = 81

Critical roles for anterior insula and dorsal striatum in punishment-based avoidance learning.

The division of human learning systems into reward and punishment opponent modules is still a debated issue. While the implication of ventral prefrontostriatal circuits in reward-based learning is well established, the neural underpinnings of punishment-based learning remain unclear. To elucidate the causal implication of brain regions that were related to punishment learning in a previous functional neuroimaging study, we tested the effects of brain damage on behavioral performance, using the same task contrasting monetary gains and losses. Cortical and subcortical candidate regions, the ante

Hyperalgesia, low-anxiety, and impairment of avoidance learning in neonatal caffeine-treated rats.

RATIONALE: The nonselective adenosine receptor antagonist caffeine is used clinically to treat apnea in preterm infants. The brain developmental stage of preterm infants is usually at a period of rapid brain growth, referred as brain growth spurt, which occurs during early postnatal life in rats and is highly sensitive to central nervous system (CNS) acting drugs. OBJECTIVES: The aim of this work was to study whether caffeine treatment during brain growth spurt produces long-term effects on the adenosine receptor-regulated behaviors including nociception, anxiety, learning, and memory. Corticosteroid receptor antagonists are amnestic for passive avoidance learning in day-old chicks.

Glucocorticoids can modulate behavioural processes and neural plasticity. They are released during learning situations and can trigger neural actions through binding to brain receptors. We hypothesized that a glucocorticoid action could play a critical role in the mechanisms involved in long-term memory formation. In order to test this hypothesis, chicks were trained on a passive avoidance learning task and given bilateral intracerebral injections of selective mineralocorticoid (RU-28318) or glucocorticoid (RU-38486) receptor antagonists. The results showed that both antagonists alter informat
