Despite convincing evidence that T cells are critical for both cellular and humoral immunity against haemoprotozoan parasites, the difficulty of performing meaningful experiments in cattle that would define the role of T cells in immunity to Babesia spp. has impeded research in this area. However, experiments performed ex vivo with immune T cells can reflect in-vivo events, and provide valuable insight into the nature of immunogenic proteins and the responding lymphocytes. The progress made towards identification of the immunogenic proteins and epitopes that stimulate anamnestic CD4+ type-1 (i

Uruguay is situated in a marginal area for the development of Boophilus microplus (30 degrees 35 degrees South Lat.) with important areas of enzootic instability for Babesia bovis and B. bigemina. The livestock products represent 70% of our exports, for which reason it is fundamental to evaluate the losses in the production that these haemoparasites cause as basic information to take future decisions. In the period 1988-1990, several works were carried out by our laboratory to know the incidence of babesiosis in the reduction of liveweight gains. The results are shown and discussed in the work

Bovine babesiosis caused by Babesia species is an economically significant disease of cattle. Severe clinical babesiosis in cattle is caused by Babesia bovis, B. bigemina, and the recently discovered Babesia sp. Mymensingh. Mongolia is an agricultural country with a large cattle inventory. Although previous studies have detected active infections of B. bovis and B. bigemina in Mongolian cattle, only a few provinces were surveyed. Additionally, the endemicity of Babesia sp. Mymensingh in Mongolia remains unknown. We screened blood DNA samples from 725 cattle reared in 16 of the 21 Mongolian pro

Babesia bigemina and Babesia bovis, are the two major causes of bovine babesiosis, a global neglected disease in need of improved methods of control. Here, we describe a shared method for the stable transfection of these two parasites using electroporation and blasticidin/blasticidin deaminase as a selectable marker. Stably transfected B. bigemina and B. bovis were obtained using a common transfection plasmid targeting the enhanced green fluorescent protein-BSD (egfp-bsd) fusion gene into the elongation factor-1α (ef-1α) locus of B. bigemina and B. bovis under the control of the B. bigemina ef

Two groups of cattle, one previously exposed to Babesia bigemina and one not, were challenged with Babesia bovis. The group previously infected with Babesia bigemina was only mildly affected upon challenge with B. bovis, whereas four of five of the other group were severely affected. Immunoblotting studies performed in both homologous and heterologous systems showed that there were polypeptides of similar molecular weight in both species, but species-specific polypeptides were demonstrated only in B. bovis by the homologous B. bovis reaction. B. bovis antisera reacted avidly with B. bigemina-i

The tick-transmitted hemoprotozoan Babesia bovis is a major causative agent of bovine babesiosis, an often fatal disease of cattle. The disease is widespread in the northeastern region of Argentina, where an increasing part of the livestock is composed of water buffalos. Although clinical cases of buffalo babesiosis have not been reported so far, the pathogen-transmitting tick vector has been occasionally observed by us to be feeding on water buffalos. We therefore set out to examine whether buffalos may constitute a reservoir of the parasite. Competitive enzyme-linked immunosorbent assay (cEL

We established a new highly sensitive method, PCR-ELISA, for the dectection of Babesia bovis in cattle for farms in Thailand. The detection of around 2.4 x 10(-8)% parasitemia (equivalent to 1 infected erythrocyte per 2 ml) was achieved by PCR amplification followed by the ELISA detection of a biotin tagged gene. When comparing the sensitivity of PCR-ELISA with the microscopic method, our PCR-ELISA method is more sensitive than thin blood smears by at least 1,000 times. The established PCR-ELISA also showed high specificity to B. bovis with no cross reaction to other endemic parasites except f

Babesia bovis and Plasmodium falciparum are both vector-borne parasites primarily infecting the erythrocytes of their respective hosts. They have obvious differences, yet the diseases caused by these parasites share many common features. Both have generated a considerable body of research but, perhaps because of the classical distinction between veterinary and medical parasitology, many of the similarities between the two have been neglected. As this review shows however, many of the pathophysiological changes in B. bovis infections are poorly described for P. falciparum - and vice versa. Exam

Susceptible Hereford cattle of different ages were inoculated with 2 X 10(8) Babesia bovis organisms. Experiment I consisted of cows aged 6 to 7 years, steers aged 17 to 18 months and calves aged 5 to 6 months, while experiment 2 consisted of cows aged 6.5 to 7.5 years, steers aged 23 to 24 months and yearlings aged 11 to 12 months. Daily measurements of temperature, parasitaemia and packed cell volume were made in order to determine susceptibility of the different ages. Twenty-four of the 36 animals in experiment I, which included all 12 cows, required treatment. One cow died as a result of a

Babesia bovis was cultured in a suspension of bovine erythrocytes incubated at 37 degrees C in Medium 199 with 50% bovine serum. The cells in culture were kept in suspension by slow stirring in spinner flasks and the medium was replaced at 24-hour intervals. Persistent multiplication of the parasite in a short series of subcultures suggests the feasibility of this approach for continuous culture.

Babesiosis is a global zoonotic disease acquired by the bite of a Babesia-infected Ixodes tick or through blood transfusion with clinical relevance affecting humans and animals. In this study, we evaluated a series of small molecule compounds that have previously been shown to target specific apicomplexan enzymes in Plasmodium, Toxoplasma and Cryptosporidium. The compounds, bumped kinase inhibitors (BKIs), have strong therapeutic potential targeting apicomplexa-specific calcium dependent protein kinases (CDPKs). We investigated if BKIs also show inhibitory activities against piroplasms such as

Although Babesia bovis and Babesia bigemina infections cause economic losses in the cattle industry in northern Thailand, there is inadequate information on Babesia isolates present in the area. Therefore, to determine the prevalence and genetic relationship between Babesia isolates, we screened 200 blood samples of cattle from Chiang Rai, Chiang Mai, and Lumpang provinces of northern Thailand. A nested polymerase chain reaction using primers targeting B. bovis spherical body protein 2 (BboSBP2) and B. bigemina rhoptry-associated protein 1a (BbiRAP-1a) genes revealed a prevalence of 12 and 21

The indirect fluorescent antibody test was used to measure the antibody response of cattle for 8 weeks after infection with either Babesia bigemina or Babesia bovis, or a combination of both species. Serological cross-reactions were observed between the 2 species, but these were most marked when B. bigemina antigen was used. In animals infected with both Babesia spp., the B. bigemina reaction appeared to suppress the B. bovis reaction.

Molecular assays have been widely used for the detection and quantification of bovine babesiosis due to their high sensitivity and specificity. However, variations in the sensitivity of pathogen detection may occur depending on the selected target gene. Thus, this study aimed to compare the detection sensitivity (DS) of Babesia bovis and B. bigemina infection levels in artificially and naturally infected cattle using quantitative PCR (qPCR) and six target genes. For B. bovis, the merozoite surface antigen genes 2b and 2c (msa-2b and msa-2c), and the mitochondrial cytochrome b gene (cybmt) were

Serological responses of field cattle (260) on a farm in KwaZulu-Natal, South Africa were determined before and after vaccination with the commercial Babesia bovis live-blood vaccine, using the indirect fluorescent antibody test (IFAT). All the cattle tested negative for B. bovis antibodies before vaccination while 83% of them had significant antibody titres (>or=1/80) to Babesia bigemina, indicating a high degree of natural exposure to the latter parasite. By Day 60 post-vaccination only 53% of the cattle had seroconverted to B. bovis. This raised the question as to why only half of the vacci

Babesia bovis is the blood parasite causing the highest morbidity and mortality of cattle in Cuba. Thus a control program based on immunization with a vaccine using an attenuated strain of the parasite was developed. For this purpose, a virulent B. bovis strain was isolated and then attenuated by the traditional method in which biological characteristics, proteolytic activity and vector transmissibility were studied. The protection conferred by the vaccine was measured by vaccinating cattle which then was challenged with a heterologous strain. In order to assess postvaccination accident risks,

This paper addresses the identification, cloning, expression, purification and functional characterization of thioredoxin reductase from Babesia bovis, the etiological agent of babesiosis. The work deals with in vitro steady state kinetic studies and other complementary analyses of the thioredoxin reductase found in the pathogenic protist. Thioredoxin reductase from B. bovis was characterized as a homodimeric flavoprotein that catalyzes the NADPH-dependent reduction of Trx with a high catalytic efficiency. Moreover, the enzyme exhibited a disulfide reductase activity using DTNB as substrate, b
