Structure-Based Strategy Reveals a Novel Ligand Binding Site in Staphylococcus aureus Catabolite Control Protein A: Implications for Transcriptional Regulation and Drug Design.

Staphylococcus aureus is a major human pathogen responsible for a broad spectrum of infections and is recognized by the World Health Organization as a high-priority antimicrobial-resistant organism. The global transcription factor (TF) catabolite control protein A (CcpA), a central regulator of metabolism and virulence, represents a promising drug target. However, efforts to design inhibitors are hindered by their closed crystallographic conformation and the absence of well-defined ligand-binding cavities. Here, we present a structure-guided alignment strategy to identify potential ligand-bind

Arsenate microbial reducing behavior regulated by the temperature fields in landfills.

Attention should be paid to the As(V) reducing behavior in landfills under different temperature fields. In this study, microcosm tests were conducted using enrichment culture from a landfill. The results revealed that the reduction rate of As(V) was significantly affected by the temperature field, with the highest reduction rate observed at 50 °C, followed by 35 °C, 25 °C, and 10 °C. Different As cycling pathways were observed under various temperature fields. At room and medium temperatures, As4S4 was detected, indicating that both biomineralization and methylation processes occurred after A

A safeguarding factor monitors septal pore stability to secure compartmentalization during sporulation in Bacillus subtilis.

Bacterial endospore formation begins with a polar septum that compartmentalizes two transcriptionally distinct cells, the mother cell and forespore. In Bacillus subtilis, a complex composed of SpoIIIE, SpoIIIM and PbpG maintains compartmentalization at a septal pore during hydrolysis of septal peptidoglycan (PG), by coordinating two critical functions: chromosome translocation from the mother cell to the forespore through the septal pore and preservation of septal pore stability through SpoIIIE-PG interactions and PG synthesis. Disruption of this mechanism leads to cytoplasmic leakage, failed

Cross Resistance in S-Methoprene-Resistant Culex quinquefasciatus (Diptera: Culicidae).

The juvenile hormone analog S-methoprene is the only synthetic biopesticide that is registered with the United States Environmental Protection Agency to control arthropods of economic importance in public health, livestock, pets, urban, and stored products. The high activity, relative target specificity, and benign environmental profile of S-methoprene have been well documented. While the risk of resistance in mosquitoes to S-methoprene is generally low, there is a lack of information regarding cross resistance in S-methoprene-resistant mosquitoes to other pesticides. In this paper, a populati

Parkinson's Disease, Gut Microbiota and Plasma Metabolites: Causal Pathways via a Mendelian Randomization Study.

INTRODUCTION: Mounting evidence implicates gut microbiota in PD pathogenesis, with emerging studies suggesting this influence may be mediated by plasma metabolites. However, the causal relationships between gut microbiota and PD phenotypes (PDs), as well as whether plasma metabolites act as mediators, remain unclear. METHODS: Genetic instruments for gut microbiota, plasma metabolites, and PDs were sourced from genome-wide association studies. Bidirectional two-sample Mendelian randomization and two-step mediation analyses were performed, with False Discovery Rate (FDR) correction applied to p

Traditional fermented foods of Indonesia harbour functionally redundant but phylogenetically diverse taxa.

Fermented foods represent complex microbial ecosystems that contribute to food quality, functionality, and potential health benefits, yet many traditional fermented foods remain poorly characterized. The aim of this study was to study microbial diversity, and functional potential of underexplored traditional Indonesian fermented food. The fermented products displayed substantial variation in bacterial richness, ranging from 65 to 614 bacterial amplicon sequence variants across samples. The microbial communities were dominated by bacterial taxa affiliated with the orders Bacillales and Lactobac

Korean Natural Farming practices are dominated by a limited number of microbes and decrease fungal diversity.

Korean Natural Farming (KNF) practices claim to cultivate and transfer "indigenous microorganisms (IMOs)" to donor soils as a method of probiotic soil enhancement. We investigated whether IMO cultivation can propagate unique microbiomes and maintain microbial diversity through successive IMO stages for restoration of flood contaminated soils. Employing a balanced study design using soil samples from salt marsh, deciduous forest, and urban greenspace (plus sterilized controls), samples underwent the first two IMO cultivation steps followed by 16S rRNA and ITS metagenomic sequencing. Notably, IM

Isolation and whole genome analysis of endospore-forming bacteria from heroin.

Infections caused by endospore-forming bacteria have been associated with severe illness and death among persons who inject drugs. Analysis of the bacteria residing in heroin has thus been biased towards species that affect human health. Similarly, exploration of the bacterial diversity of seized street market heroin correlated with the skin microflora of recreational heroin users insofar as different Staphylococus spp. or typical environmental endospore formers including Bacillus cereus and other Bacilli outside the B. cereus sensu lato group as well as diverse Clostridia were identified. GerAB residues predicted to interfere with water passage based on steered molecular dynamics are key to germinosome functionality.

Some Bacillales and Clostridiales bacteria form spores in unfavorable environments. These spores are dormant but can rapidly resume metabolism in germination. This process can be initiated by a variety of low molecular weight nutrients termed germinants. Structural modeling and mutagenesis studies showed that GerAB, an inner membrane (IM) protein of the Bacillus subtilis spore germinant receptor (GR) GerA, is involved in L-alanine-initiated spore germination. A previous molecular simulation study also suggested there is a water channel in GerAB. In the current work, Steered Molecular Dynamics

Spatiotemporal diversity of bacterial endophyte microbiome of mandarin (Citrus reticulata) in the northern Persian Gulf and its HCN production and N2 fixation.

Endophytes are symbionts that live in healthy plants and potentially improve the health of plant holobionts. Here, we investigated the bacterial endophyte community of Citrus reticulata grown in the northern Persian Gulf. Bacteria were isolated seasonally from healthy trees (root, stem, bark, trunk, leaf, and crown tissues) in four regions of Hormozgan province (i.e., Ahmadi, Siyahoo, Sikhoran, Roudan), a subtropical hot region in Iran. A total of 742 strains from 17 taxa, 3 phyla, and 5 orders were found, most of which belonged to Actinobacteria (Actinobacteriales) as the dominant group, foll

Effect of phosphate-solubilizing bacteria on phosphorus dynamics and the bacterial community during composting of sugarcane industry waste.

Sugarcane processing generates a large quantity of residues, such as filter cake and ashes, which are sometimes composted prior to their amendment in soil. However, important issues still have to be addressed on this subject, such as the description of bacterial succession that occurs throughout the composting process and the possibilities of using phosphate-solubilizing bacteria (PSB) during the process to improve phosphorus (P) availability in the compost end product. Consequently, this study evaluated the bacterial diversity and P dynamics during the composting process when inoculated with

Effect of Moxibustion on Intestinal Microbiome in Acute Gastric Ulcer Rats.

In Traditional Chinese Medicine (TCM), moxibustion had been used for thousands of years. Many clinical case reports and scientific studies had proved that moxibustion had a good effect in treating acute gastric ulcer (AGU). Some studies had shown that the relative content and species of bacteria in the intestinal would be changed when gastric mucosal injury happened. However, there was little research on the effect of intestinal microbiome with AGU rats that were treating by moxibustion. This study is aimed at analyzing the effect of fecal microbiome in rats with AGU by the 16S rDNA sequencing

Impact of streptozotocin-induced type 1 and type 2 diabetes on ocular surface microbial signatures in C57BL/6J mice.

The ocular surface (OS), like other mucosal sites, hosts a diverse microbiome. However, the impact of hyperglycemia associated with diabetes on OS microbial composition remains poorly understood. In this study, we established type 1 diabetes mellitus (T1DM) and type 2 diabetes mellitus (T2DM) models in C57BL/6J mice by administering high-dose streptozotocin (STZ) for T1DM and low-dose STZ combined with a high-fat diet for T2DM. The OS microbiome was characterized and analyzed using 16S rRNA sequencing. The results showed that neither T1DM nor T2DM significantly affected microbial richness comp
