Effect of Bacillus clausii in attenuating symptoms of DSS-induced ulcerative colitis by modulating NFkB pathway and oxidative stress in mice.

Ulcerative colitis (UC) is a condition characterized by inflammation and ulcer formation in the colon and rectum due to genetic and environmental factors. It is a common condition, with a global prevalence rate exceeding 0.3%. Current treatments have limited efficacy and can cause unwanted side effects, leading to a high recurrence rate and reduced quality of life for patients. This study suggests that Bacillus clausii has a beneficial role in reducing intestinal inflammation and relieving colitis symptoms in mice. The study aimed to examine B. clausii's potential to reduce the progression and

Protective effects of Bacillus clausii against Amoxicillin-induced hepatic injury are associated with Attenuation of LPS-TLR4-NF-κB signalling.

Amoxicillin (AMX) is a widely used β-lactam antibiotic that can cause liver injury through oxidative stress and inflammation, often disrupting the gut-liver axis. This study explored the hepatoprotective effects of the probiotic Bacillus clausii (B. clausii, BCL) in male Swiss albino mice divided into four groups: control, BCL, AMX, and AMX + BCL, with treatments administered orally for 45 days.AMX treatment led to significant liver dysfunction, evidenced by elevated aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), and tumor necrosis factor-alpha (T

Bacillus clausii spores maintain gut homeostasis in murine ulcerative colitis via modulating microbiota, apoptosis, and the TXNIP/NLRP3 inflammasome cascade.

Ulcerative colitis (UC), a persistent immune-mediated disorder lacking effective treatment, is distinguished by gut microbiota dysbiosis, abnormal activation of the NLRP3 inflammasome pathway, and apoptosis. Despite growing attention to these factors, understanding their significance in UC pathogenesis remains a challenge. The present study explores the potential therapeutic impact of Bacillus clausii (Bc) spores in a murine UC model induced by drinking 4 % (w/v) dextran sulfate sodium (DSS) in C57BL/6 mice. Subsequently, the DSS-induced mice were orally administered either Bc at varying conce

Effects of mannan oligosaccharide, Bacillus clausii, and their synbiotic supplementation on growth performance, humoral immunity, intestinal morphology, and ghrelin gene expression in broiler chickens.

This study evaluated the effects of a prebiotic (mannan-oligosaccharide, MOS), a probiotic (Bacillus clausii), and their combinations on growth performance, immune response, intestinal morphology, and ghrelin gene expression in broiler chickens. Two hundred fifty day-old Hubbard chicks were randomly allotted to five treatments (control, prebiotic, probiotic, Combo-I 50:50, Combo-II 100:100), each with five replicates of 10 birds. Combo-II supplementation improved body-weight gain and feed conversion ratio during the starter (p = 0.0256 and 0.0428), grower (p = 0.0425 and 0.0483), and finisher

Biorefinery-inspired, two-step valorization strategy to manage plant-based recalcitrant organic waste, involving solvent extraction, and fermentation with Bacillus clausii-a proof of concept study.

Approximately 40-50% of municipal solid waste is organic and causing biogenic malodor and infections, due to inefficient treatment methods. Biorefinery-based bioremediation and valorization is in vogue against these conventional strategies since it combines unit operations for better efficiency and productivity. Deriving inspiration, the proposed strategy puts together a unique and compatible combination of processes. This novel two-step valorization workflow involves the extraction of small molecules using organic solvents, and fermentation of resulting denatured residues (increased biodegrad

Alkalihalobacillus clausii bacteremia after probiotic use for acute diarrhea: a case report.

Alkalihalobacillus clausii (previously known as Bacillus clausii) is a spore-forming bacterium used as a probiotic due to its resistance to gastric acidity and intestinal conditions. It is marketed for managing acute diarrhea, though evidence of its efficacy and safety is limited. We present the case of an 87-year-old man with acute viral diarrhea treated with Alkalihalobacillus clausii, who developed septic shock with blood cultures confirming Alkalihalobacillus clausii bacteremia. Intensive care and daptomycin therapy led to clinical resolution.

Biostimulated-sesame sprout extracts as potential agents against Leishmania mexicana.

Leishmania mexicana is one of the causal agents of cutaneous leishmaniasis. Current antileishmanial chemotherapeutics have demonstrated adverse side effects; thus, alternative treatments are needed. In this study, we performed in silico and in vitro analyses of the leishmanicidal potential of the most abundant phenolic compounds identified in black sesame sprouts biostimulated with Bacillus clausii. The molecular docking analysis showed strong interactions (binding free energies between -6.5 and -9.5 kcal/mol) of sesaminol 2-O-triglucoside, pinoresinol dihexoside, isoverbascoside, and apigenin

Development of a Tool for High-Efficiency, Markerless and Iterative Genome Editing in Shouchella clausii.

Shouchella clausii is a spore-forming, Gram-positive bacterium with intrinsic antibiotic resistance and promising potential in biotherapeutics, industrial biotechnology and environmental applications. Its genetic intractability, due to a rigid cell wall and lack of natural competence, has limited its development as a microbial chassis. To facilitate its genetic transformation, a hyperosmotic electroporation protocol was optimised using cell wall weakening agents, achieving efficiencies comparable to other recalcitrant bacilli. A comprehensive and reusable genetic tool was developed centred on

Comparative Genomics and Physiological Characterization of Two Aerobic Spore Formers Isolated from Human Ileal Samples.

Spore formers are ubiquitous microorganisms commonly isolated from most environments, including the gastro-intestinal tract (GIT) of insects and animals. Spores ingested as food and water contaminants safely transit the stomach and reach the intestine, where some of them germinate and temporarily colonize that niche. In the lower part of the GIT, they re-sporulate and leave the body as spores, therefore passing through their entire life cycle in the animal body. In the intestine, both un-germinated spores and germination-derived cells interact with intestinal and immune cells and have health-b
