M13 phage coated surface elicits an anti-inflammatory response in BALB/c and C57BL/6 peritoneal macrophages.

Bacteriophages are one of the viral components of the human microbiome. M13 phages have recently been considered for immunotherapy because they can be detected by immune cells and stimulated immune responses. Macrophages are essential innate immune cells that respond to stimuli and direct subsequent immune responses. Therefore, it is crucial to evaluate the immunomodulatory effect of phage on macrophage function. For this purpose, peritoneal macrophages from BALB/c and C57BL/6 mice were cultured on the M13 phage, M13 phage-RGD, gelatin-coated, and un-coated wells. Then macrophages were examine

Engineered M13 phage as a novel therapeutic bionanomaterial for clinical applications: From tissue regeneration to cancer therapy.

Bacteriophages (phages) are nanostructured viruses with highly selective antibacterial properties that have gained attention beyond eliminating bacteria. Specifically, M13 phages are filamentous phages that have recently been studied in various aspects of nanomedicine due to their biological advantages and more compliant engineering capabilities over other phages. Having nanofiber-like morphology, M13 phages can reach varied target sites and self-assemble into multidimensional scaffolds in a relatively safe and stable way. In addition, genetic modification of the coat proteins enables specific

Unraveling the potential of M13 phages in biomedicine: Advancing drug nanodelivery and gene therapy.

M13 phages possessing filamentous phage genomes offer the benefits of selective display of molecular moieties and delivery of therapeutic agent payloads with a tolerable safety profile. M13 phage-displayed technology for resembling antigen portions led to the discovery of mimetic epitopes that applied to antibody-based therapy and could be useful in the design of anticancer vaccines. To date, the excremental experiences have engaged the M13 phage in the development of innovative biosensors for detecting biospecies, biomolecules, and human cells with an acceptable limit of detection. M13 phage grafted with peptide motifs as a tool to detect amyloid-β oligomers in brain tissue.

Oligomeric clusters of amyloid-β (Aβ) are one of the major biomarkers for Alzheimer's disease (AD). However, proficient methods to detect Aβ-oligomers in brain tissue are lacking. Here we show that synthetic M13 bacteriophages displaying Aβ-derived peptides on their surface preferentially interact with Aβ-oligomers. When exposed to brain tissue isolated from APP/PS1-transgenic mice, these bacteriophages detect small-sized Aβ-aggregates in hippocampus at an early age, prior to the occurrence of Aβ-plaques. Similarly, the bacteriophages reveal the presence of such small Aβ-aggregates in post-mor

Topology-Dependent Antiviral Activity of H-Form Zeolites: Discovery of a Highly Active Natural Mordenite.

The global threat of viral infections highlights the urgent need for the development of effective antiviral solid materials. Proton-form zeolites have been recently reported to inactivate viruses such as influenza and M13 bacteriophage without relying on metal ion exchange, suggesting a promising approach for low-cost and nondiscoloring antiviral applications. However, these findings have so far been confined primarily to FAU-type zeolites, leaving the relationship between antiviral activity and the physicochemical properties of zeolites─such as framework topology and chemical composition─larg

Detection of constitutive molecules on Histoplasma capsulatum yeasts through single chain variable antibody fragments displayed in M13 phages.

A nonimmune library, containing single chain variable fragments (scFv) of immunoglobulin human genes displayed on the surface of M13 filamentous phages, was used to recognize molecules exposed on Histoplasma capsulatum yeasts' surface, during their growth in synthetic medium. The scFv clones were checked in their consistency by Dot-ELISA using HRP/anti-M13 conjugate, and they were tested to recognize molecules on H. Capsulatum yeasts' surface by ELISA in plates. Three out of 80 scFv cones (C2, C6, and C52) reacted consistently with H. capsulatum molecules, and they recognized molecules from bo

Bacteriophage carriers localize in the brain of a rat model of neonatal hypoxic-ischemic encephalopathy.

BACKGROUND: Neonatal hypoxic-ischemic encephalopathy arises from a reduction of oxygen and blood supply to the infant brain and can lead to severe brain damage and life-long disability. The damage is greatest at the irreversibly injured necrotic core, whereas the penumbra is the surrounding, potentially salvageable tissue populated with a mix of alive and dying cells. To date, there exists no method for targeting drugs to the brain damage. METHODS AND MAJOR RESULTS: Bacteriophages are viruses that propagate in bacteria but are biocompatible in humans and also amenable to genetic and chemical

Functional Blood-Brain Barrier Crossing by Biomimetic M13 Phage Vectors for Targeted Neuronal Delivery.

The blood-brain barrier (BBB) is a highly selective gatekeeper. While providing essential protection against pathogens, BBB also poses a major challenge by restricting the delivery of systemically administered therapeutics. Consequently, there is a pressing need for innovative strategies that can efficiently and selectively target neuronal populations to treat diverse brain disorders. Nanomaterials enable transcellular transport across the endothelial layer; however, given the biomolecular corona paradigm, designing biomimetic hybrid nanoconstructs that preserve and leverage biological functio

Phage Display Biopanning Strategy for Discovery of Nanoparticle-Binding Peptides.

The phage display technology allows to select peptides, exposed on the surface of bacteriophage virions, which can specifically bind to virtually any materials, including proteins, lipids, carbohydrates, and other organic macromolecules, as well as metals or metal oxides. In this chapter, we describe and discuss procedures for effective selection of peptides characterized with affinity to metallic or metal oxide nanoparticles. The use of a peptide library displayed on virions of the M13 bacteriophage, the most commonly used phage display system, is presented. Specific notes indicate what are c

Selection of a Biosafety Level 1 (BSL-1) surrogate to evaluate surface disinfection efficacy in Ebola outbreaks: Comparison of four bacteriophages.

The 2014 West African Ebola virus disease outbreak was the largest to date, and conflicting, chlorine-based surface disinfection protocols to interrupt disease transmission were recommended. We identified only one study documenting surface disinfection efficacy against the Ebola virus, showing a >6.6 log reduction after 5-minute exposure to 0.5% sodium hypochlorite (NaOCl) based on small-scale tests (Cook et al. (2015)). In preparation for future extensive, large-scale disinfection efficacy experiments, we replicated the Cook et al. experiment using four potential BSL-1 surrogates selected bas

Retraction of F pili.

The disappearance of F pili on Escherichia coli cells in the presence of 10(-2) M NaCN was studied by electron microscopy and serum-blocking power. The pili which disappeared from the cell did not appear as free pili in the culture medium, suggesting that the pili had retracted into the cell. New pili were produced at a normal rate approximately 3 min after NaCN was removed. The adsorption of either F pili antibody or R17 bacteriophage to the sides of pili and temperatures below 24 C prevented retraction. The disappearance of pili was accompanied by a loss in the ability of cells to adsorb R17

Mixed Ionic and Electronic Charge Transport in Conductive Protein Fibers Revealed with DC Electrical Measurements.

Naturally conductive protein nanowires have inspired efforts to engineer electrical conductivity into synthetic fibrous proteins for the development of bioelectronic materials and devices. A comprehensive analysis of charge transport in these systems requires a combination of various measurement methods, instruments and electrode designs. Measurements under direct current (DC) typically focus on charge transport without distinguishing between charged species, requiring alternating current (AC) and electrochemical methods to probe additional phenomena. Here, ionic and electronic charge transpor

Characterization of a sequence of human T cell leukemia virus type I from a patient with chronic progressive myelopathy.

DNA from peripheral blood mononuclear cells of individuals with chronic progressive myelopathy (CPM) were extensively analyzed for the presence of human T cell leukemia virus (HTLV) type I-like sequences by using the polymerase chain reaction. The DNA samples were amplified with oligonucleotides from three separate regions of HTLV viral genomes. A portion of the amplified viral genome from a representative patient was sequenced after molecular cloning into bacteriophage M13. Sequence data indicate that HTLV type I and not a related virus is associated with CPM.

Engineered phage-based therapeutic materials inhibit Chlamydia trachomatis intracellular infection.

Developing materials that are effective against sexually transmitted pathogens such as Chlamydia trachomatis (Ct) and HIV-1 is challenging both in terms of material selection and improving bio-membrane and cellular permeability at desired mucosal sites. Here, we engineered the prokaryotic bacterial virus (M13 phage) carrying two functional peptides, integrin binding peptide (RGD) and a segment of the polymorphic membrane protein D (PmpD) from Ct, as a phage-based material that can ameliorate Ct infection. Ct is a globally prevalent human pathogen for which there are no effective vaccines or mi

Biohybrid M13 phage backpacks for active blood-brain barrier traversal and precision central nervous system leukemia treatment.

Central nervous system (CNS) leukemia remains a major clinical challenge in acute lymphoblastic leukemia, largely due to the restrictive nature of the blood-brain barrier (BBB), which severely limits therapeutic delivery and contributes to high relapse rates and poor prognosis. Here, we report ALB-phage, a bioengineered anti-leukemia backpack-phage system designed for active BBB navigation and intracranial drug delivery. M13 bacteriophages were genetically engineered to display BBB-homing ligands, forming a high-aspect-ratio biological transport platform (BTP). A leukemia-targeting therapeutic

Hierarchically Engineered Phage-Activated Click Interface for Ultra-Efficient Cell Capture.

Efficient cell adhesion under flow is vital in biological processes and artificial capture systems governed by the frequency of cell-surface encounters and the probability of successful binding. While previous efforts mainly focused on enhancing these factors at the molecular level within two-dimensional interfaces, strategies that simultaneously boost both aspects across the multiscale spatial level remain largely unexplored. In this study, we developed a multiscale hierarchically engineered cell capture interface, PACE-Chip, which integrates click chemistry at the molecular level, nanofiber-

Structural tuning of bridging linkers on M13 phages for high-affinity detection of enterovirus A71.

Enterovirus A71 (EV-A71), the principal etiological agent of hand, foot, and mouth disease, poses a serious public health concern because infection can induce severe and potentially fatal neurological complications through direct invasion and inflammation of the central nervous system. Current detection methods lack rapid and sensitive analytical capabilities, particularly in the context of wastewater surveillance. This study reports a PEG4-biotin-functionalized M13 bacteriophage engineered as a molecular receptor for the colorimetric detection of EV-A71. VP1-specific phage ligands were identi

Far-UVC (222 nm) irradiation effectively inactivates ssRNA, dsRNA, ssDNA, and dsDNA viruses as compared to germicidal UVC (254 nm).

Ultraviolet-C (UVC) irradiation is being used as an effective approach for the disinfection of pathogenic viruses present in air, surfaces, and water. Recently, far-UVC radiation (222 nm) emitted by KrCl* (krypton-chloride) excimer lamps have been recommended for disinfecting high-risk public spaces to reduce the presence and transmission of infectious viruses owing to limited human health exposure risks as compared to germicidal UVC (254 nm). In this study, the UVC inactivation performances of individual filtered KrCl* excimer lamp (222 nm) and germicidal UVC lamp (254 nm) were determined aga

Inactivation and spike protein denaturation of novel coronavirus variants by CuxO/TiO2 nano-photocatalysts.

In order to reduce infection risk of novel coronavirus (SARS-CoV-2), we developed nano-photocatalysts with nanoscale rutile TiO2 (4-8 nm) and CuxO (1-2 nm or less). Their extraordinarily small size leads to high dispersity and good optical transparency, besides large active surface area. Those photocatalysts can be applied to white and translucent latex paints. Although Cu2O clusters involved in the paint coating undergo gradual aerobic oxidation in the dark, the oxidized clusters are re-reduced under > 380 nm light. The paint coating inactivated the original and alpha variant of novel coronav

Truncated M13 phage for smart detection of E. coli under dark field.

BACKGROUND: The urgent need for affordable and rapid detection methodologies for foodborne pathogens, particularly Escherichia coli (E. coli), highlights the importance of developing efficient and widely accessible diagnostic systems. Dark field microscopy, although effective, requires specific isolation of the target bacteria which can be hindered by the high cost of producing specialized antibodies. Alternatively, M13 bacteriophage, which naturally targets E. coli, offers a cost-efficient option with well-established techniques for its display and modification. Nevertheless, its filamentous
