Induction of macrophage procoagulant activity by Bacteroides fragilis.

Fibrin deposition in the peritoneal cavity during acute peritonitis appears to predispose the host to abscess formation by providing an environment for bacterial proliferation protected from host defenses. The purpose of the present study was to determine whether the potent abscess-inducing anaerobe Bacteroides fragilis could promote fibrin deposition by inducing mononuclear cells to express procoagulant activity (PCA). B. fragilis stimulated PCA in a dose-dependent fashion, achieving a maximum at 10(7) CFU/ml. Heat-killed B. fragilis induced comparable levels of PCA, while a nonspecific phago

Bacterial Pyomyositis in the Neck Due to Bacteroides Fragilis.

Bacterial pyomyositis is characterized by the formation of abscesses in skeletal muscle groups, frequently in the lower extremities. It is most prevalent in tropical climates and associated with Staphylococcus aureus. The presentation of pyomyositis in the neck is rare. We present a unique case of pyomyositis caused by a gut bacterium, Bacteroides fragilis. Additionally, the case highlights the difficulty in identifying the cause of spread of the bacteria from the gut to the affected musculature. A 77-year-old diabetic Caucasian male presented with progressive bilateral neck swelling and limit

Bacteroides fragilis - an overlooked cause of abortion in the mare?

A near late-term pregnant mare developed purulent vaginal discharge, fever and peripheral edema. Preterm foaling was medically induced, and the mare delivered a weak foal that was subsequently euthanized. Necropsy revealed an extensive, severe allantochoroiditis affecting the entire corpus segment. Necrosuppurative choroiditis with multiple colonies of gram-negative bacilli and suppurative bronchioalveolitis was found at histology. Anaerobic culture of the chorion revealed a mixed culture clearly dominated by Bacteroides fragilis. B. fragilis has not previously been associated with abortion in

Clinical significance of and outcomes for Bacteroides fragilis bacteremia.

BACKGROUND AND PURPOSE: Bacteroides fragilis is a virulent anaerobic pathogen, resulting in considerable mortality. This study was conducted to investigate the clinical characteristics, significance of polymicrobial bacteremia, and treatment outcomes of B. fragilis bacteremia. METHODS: This retrospective analysis enrolled 199 adult patients with B. fragilis bacteremia, who were admitted to hospital between January 2004 and May 2007. Chi-squared and Fisher's exact tests were used for comparison. A p value of <0.05 was considered statistically significant. RESULTS: 142 patients with B. fragili

The clinical significance of clindamycin-resistant Bacteroides fragilis.

The Bacteroides fragilis group of bacteria are the most numerous microorganisms in the colon and the most common anaerobic isolates from human infections. Although clindamycin hydrochloride is still considered the antibiotic of choice for treatment of infections with these bacteria, resistant strains are becoming more common. To determine the clinical significance of clindamycin-resistant bacteroides, we reviewed the charts of 14 patients with resistant isolates. Clindamycin resistance was a major factor in the clinical course of at least four patients. Three of these did not recover until the

Colonic mucus attenuates Bacteroides fragilis virulence in vitro.

OBJECTIVES: Enterotoxigenic Bacteroides fragilis (ETBF) is a subspecies of B. fragilis that is considered to contribute to colorectal carcinogenesis through the activity of the B. fragilis toxin (BFT). However, colonic carriage of ETBF is also evident in a subset of asymptomatic individuals. In vitro studies using the HT29 cell line have linked BFT to the disruption of cell-cell adhesion and inflammation. This cell line, however, fails to consider the role of the mucus barrier in protecting the colonic epithelium in healthy individuals. This study aimed to investigate the role of a cell-associ

Bacteroides fragilis vertebral osteomyelitis: A rare clinical entity.

Acute pyogenic vertebral osteomyelitis is mainly attributed to haematogenous spread of aerobic bacteria, while anaerobic osteomyelitis results from contiguous spread of polymicrobial infections through breaks in the gut mucosal barrier and involves the vertebral bodies in only about 2%-5%. Herein, we report two cases of vertebral osteomyelitis due to Bacteroides fragilis. It is noteworthy that cases of vertebral osteomyelitis due to Bacteroides fragilis have been attributed to the extension of intra-abdominal or pelvic floor infections. However, in the two cases described, there was no history

Bacteroides fragilis-stimulated interleukin-10 contains expanding disease.

Commensal symbionts may become pathogens upon escaping their habitat. In the gut, Bacteroides fragilis protects against colitis through induction of interleukin 10 (IL-10) by CD4(+) T cells. When intestinal integrity is disrupted, B. fragilis and colonic contents escape into the peritoneum, causing abscesses and bacteremia. Whether the virulence mechanisms employed by B. fragilis during infections differ from those employed for symbiosis during commensalism is unknown. We demonstrate T cell-independent IL-10 production in response to B. fragilis during its pathogenic interactions with the host

Beta-lactamase-mediated imipenem resistance in Bacteroides fragilis.

Imipenem has excellent antimicrobial activity owing in part to beta-lactamase stability. We found that only 2 of over 350 Bacteroides fragilis group clinical isolates were resistant to imipenem, with an MIC of more than 16 micrograms/ml. These two isolates from the Tufts Anaerobe Laboratory (TAL) were resistant to all other beta-lactam agents tested. The organisms were able to inactivate imipenem in broth cultures and contained similar beta-lactamases that were able to hydrolyze carbapenems, cephamycins, cephalosporins, and penicillins. The molecular sizes of the beta-lactamases in TAL2480 and

[Detection of Bacteroides fragilis enterotoxin from extraintestinal clinical isolates of B. fragilis group organisms].

A total of 300 extraintestinal clinical isolates of Bacteroides fragilis group organisms were assayed for enterotoxin using HT29/C1 cells. Of the 179 strains of B. fragilis, 36 (20.0%) were positive for cell culture assay, a positivity which was neutralized by anti-B. fragilis enterotoxin serum; all 36 strains were confirmed to be enterotoxigenic B. fragilis (ETBF). Among B. fragilis isolated from blood, 18 (31%) of 59 strains were ETBF while 18 (15.0%) of 120 strains isolated from non-blood specimens were ETBF (p < 0.05). None of the 121 strains of the B. fragilis group other than B. fragilis

Bacteroides fragilis toxin promotes gall bladder cancer in mice.

Enterotoxigenic Bacteroides fragilis (ETBF), which secretes B. fragilis toxin (BFT) and has been associated with colorectal cancer, can be enriched in patients with gall bladder cancer (GBC). Whether and how ETBF contributes to GBC is unclear. Here we confirm, through analysis of patient samples, that ETBF is enriched in GBC tumours, while experiments in mice show that ETBF colonizes the gall bladder. In vitro and in patient-derived organoids, ETBF promoted GBC proliferation. ETBF also promoted tumour growth in a BFT-dependent manner in a mouse GBC allograft model. Mechanistically, the tumorig

Bacteroides fragilis in saliva: investigating links with ulcerative colitis.

BACKGROUND: Ulcerative colitis (UC) is a long-term bowel inflammation of unknown cause. Recent research points to gut microbiota, especially Enterotoxigenic Bacteroides fragilis (ETBF), in UC's development. This study examined the presence of Bacteroides fragilis (B. fragilis) and ETBF in the saliva of UC patients and Healthy Controls (HCs) in Iran. METHODS: A total of 40 UC patients and 40 healthy controls were included in the study. Saliva samples were collected and analyzed for the presence of B. fragilis and ETBF using real-time polymerase chain reaction (PCR). RESULTS: B. fragilis was m

Characterization of a Bacteriophage GEC_vB_Bfr_UZM3 Active against Bacteroides fragilis.

Bacteroides fragilis is a commensal gut bacterium that is associated with a number of blood and tissue infections. It has not yet been recognized as one of the drug-resistant human pathogens, but cases of the refractory infections, caused by strains that are not susceptible to the common antibiotic regimes established for B. fragilis, have been more frequently reported. Bacteriophages (phages) were found to be a successful antibacterial alternative to antibiotic therapy in many cases of multidrug-resistant (MDR) bacterial infections. We have characterized the bacteriophage GEC_vB_Bfr_UZM3 (UZM

Spondylodiscitis due to Bacteroides fragilis: two cases and review.

Non-iatrogenic spondylodiscitis caused by anaerobic bacteria remains exceptional. We describe 2 cases of spondylodiscitis with epidural abscess due to Bacteroides fragilis, 1 after colonoscopy with biopsy and 1 in a cirrhotic patient. The clinical and imaging findings were not discriminant relative to other pyogenic spondylodiscitis. One should consider B. fragilis when treating a spondylodiscitis with epidural abscess, especially in patients with a possibly digestive portal of entry.

Newer beta-lactam agents and the Bacteroides fragilis group.

A continuing nationwide susceptibility survey of the Bacteroides fragilis group, begun in 1981, is being conducted at the New England Medical Center. Review of susceptibility testing in years 1986 through 1988 is reported here. Totals of 557 strains in 1986, 506 in 1987, and 534 in 1988 were obtained from seven centers in the United States. The most active beta-lactam drugs were beta-lactamase-inhibitor combinations ticarcillin-clavulanic acid, ampicillin-sulbactam, cefoperazone-sulbactam and the carbapenem imipenem. These drugs had virtually no resistance detected. Their rank order of activit

To resist and persist: Important factors in the pathogenesis of Bacteroides fragilis.

Bacteroides fragilis is a most frequent anaerobic pathogen isolated from human infections, particularly found in the abdominal cavity. Different factors contribute to the pathogenesis and persistence of B. fragilis at infection sites. The knowledge of the virulence factors can provide applicable information for finding alternative options for the antibiotic therapy and treatment of B. fragilis caused infections. Herein, a comprehensive review of the important B. fragilis virulence factors was prepared. In addition to B. fragilis toxin (BFT) and its potential role in the diarrhea and cancer dev

The antibiotic sensitivity of the Bacteroides fragilis group in the United Kingdom.

Eight hundred and sixty-two cultures of penicillin-resistant anaerobic Gram-negative bacilli collected from 14 centres in the United Kingdom were identified. Six hundred and fifty-one were members of the Bacteroides fragilis group, and their sensitivity to 14 antibiotics was determined by a microtitre method. Among the beta-lactams, moderate resistance to ampicillin (MIC greater than or equal to 8 mg/l) was common, (B. ovatus 100%, B. thetaiotaomicron 95%, B. fragilis 76%, B. distasonis 50%, B. vulgatus 25%) but high-level resistance (MIC greater than 128 mg/l) was uncommon (B. fragilis 4%, B.

Infection with Bacteroides fragilis: pathogenesis and immunoprophylaxis in an animal model.

Organisms of the genus Bacterioides represent the major group of obligate anaerobes involved in human infections. Bacteroides usually cause either bacteremia or localized abscesses. Of the numerous species of Bacteroides, Bacteroides fragilis is the single most frequent clinical isolate. B. fragilis and Bacteroides melaninogenicus have chemically incomplete lipopolysaccharides as compared with the lipopolysaccharides (endotoxins) of aerobic bacteria, and the lipopolysaccharides of Bacteroides lack of biologic potency characteristic of endotoxin. This inactivity may account for the very infrequ
