Expression of basigin in reproductive tissues of estrogen receptor-{alpha} or -{beta} null mice.

Basigin plays important roles in both male and female reproduction because basigin (Bsg) null male and female mice are infertile. The aim of the present study was to determine whether basigin expression in reproductive organs requires estrogen receptor-alpha (ESR1, ERalpha) or -beta (ESR2, ERbeta). Expression of basigin protein in the testis, ovary, and male and female reproductive tracts was studied in adult wild-type (WT), Esr1-null (alphaERKO), and Esr2-null (betaERKO) mice by immunohistochemistry and immunoblotting. Basigin mRNA levels in ovary and uterus were examined by quantitative RT-P

Basigin Associates with Integrin in Order to Regulate Perineurial Glia and Drosophila Nervous System Morphology.

The Drosophila nervous system is ensheathed by a layer of outer glial cells, the perineurial glia, and a specialized extracellular matrix, the neural lamella. The function of perineurial glial cells and how they interact with the extracellular matrix are just beginning to be elucidated. Integrin-based focal adhesion complexes link the glial membrane to the extracellular matrix, but little is known about integrin's regulators in the glia. The transmembrane Ig domain protein Basigin/CD147/EMMPRIN is highly expressed in the perineurial glia surrounding the Drosophila larval nervous system. Here w

Evaluating Basigin as a Potential Biomarker of Blood-Brain Barrier Dysfunction in Cerebral Amyloid Angiopathy.

AIMS: Blood-brain barrier (BBB) dysfunction may be involved in the pathophysiology of neurodegenerative disorders, including sporadic cerebral amyloid angiopathy (CAA). Because basigin (BSG) may induce activity of matrix metalloproteinases and thereby BBB breakdown, we investigated BSG expression in CAA brain tissue with immunohistochemistry and as cerebrospinal fluid biomarker for BBB dysfunction in patients with CAA. METHODS: Using immunohistochemistry, we quantified BSG expression within the cortical microvasculature of the temporal lobe of 50 CAA patients (16 with intracerebral haemorrhag

The reticulocyte restriction: invasion ligand RBP1a of Plasmodium vivax targets human TfR1, prohibitin-2, and basigin.

INTRODUCTION: Plasmodium vivax is the most widespread cause of malaria outside Africa. Developing effective controls is challenging because P. vivax exclusively invades reticulocytes, immature erythrocytes that are scarce and short-lived. This limits opportunities to culture the parasite and investigate the receptor-ligand interactions crucial for host cell invasion. METHODS: The erythroid cell lines JK-1 and BEL-A were evaluated in vitro as reticulocyte surrogates to assess their susceptibility to P. vivax invasion. Comparative membrane proteomics of these cell lines, reticulocytes, and matu

Basigin links altered skeletal stem cell lineage dynamics with glucocorticoid-induced bone loss and impaired angiogenesis.

Glucocorticoid (GC) induced osteoporosis (GIOP) and osteonecrosis remain a significant health issue with few approved therapies. Here, we investigate the cellular and molecular processes by which GCs affect osteogenesis and angiogenesis. We find that GC treatment reduces bone mass through decreased bone formation by skeletal stem cells (SSCs). Concomitantly, endothelial cells increase in number but display distorted phenotypical features. Transplantation studies of SSCs combined with molecular analysis by single cell RNA-sequencing and functional testing of primary human cells tie GC-induced s

Erythrocyte invasion-neutralising antibodies prevent Plasmodium falciparum RH5 from binding to basigin-containing membrane protein complexes.

To evaluate the tolerance and the outcome of radical surgery in the management of elderly patients with uterine cervical cancer in Korea, seventy-seven patients with stage Ib or IIa squamous cell carcinoma of the uterine cervix, diagnosed at Seoul National University Hospital from January 1996 to June 2003, were retrospectively reviewed. Sixty-six patients undergoing radical surgery were divided into two groups according to their age: 41-50 years (group A; n = 43) and 65 years or older (group B; n = 23). Statistical analysis was conducted by using Mann-Whitney U test and Fisher's exact t test.

Urinary Basigin/CD147 is a useful marker of acute T cell-mediated rejection in kidney transplant recipients.

BACKGROUND: Acute T cell-mediated rejection (ATCMR) is a severe negative outcome of kidney transplantation; however, it currently has no reliable marker in Japan. METHODS: This cross-sectional study was conducted at the Japanese Red Cross Aichi Medical Center Nagoya Daini Hospital from 2016 to 2018 to determine whether plasma or urinary Basigin/CD147 is an effective marker of ATCMR. Plasma and urinary samples were obtained when episode graft biopsies were performed. RESULTS: Forty-six kidney transplant recipients received graft biopsies. Three of them missed plasma and urinary samples and th

Direct Tumor Suppression by Anti-Basigin Antibody and Enhancement of Macrophage-Mediated Cancer Cell Phagocytosis under SIRPα Blockade.

The transmembrane protein CD47 expressed on tumor cells suppresses Fcγ receptor-mediated antibody-dependent cellular phagocytosis (ADCP) through its interaction with signal regulatory protein α (SIRPα), a transmembrane protein expressed on phagocytic cells such as macrophages. Although blockade of the CD47-SIRPα interaction enhances the efficacy of tumor-targeting antibodies such as rituximab, its antitumor activity remains limited in cancers lacking defined tumor-associated antigens. In this study, we identified the membrane protein Basigin and SIRPα as broadly applicable therapeutic targets

Majority of the erythrocyte binding proteins of the Pvfam "a" family of Plasmodium vivax interact with Basigin to assist parasite entry into the host cell.

Molecular mechanisms of red cell invasion by the Plasmodium vivax parasite remain obscure since information on receptor-ligand interaction is scarce. Several proteins of the P. vivax Pvfam "a" family are known to bind with host erythrocytes. Some of them share their erythrocyte receptors with each other and vice versa, but the identification of these receptors is awaited with the exception of PvTRAg38. Here, we demonstrate by using solid-phase binding assay and surface plasmon resonance that majority (7 out of 10) of these erythrocyte binding proteins (PvTRAg, PvTRAg33.5, PvTRAg35.2, PvTRAg34,

Multimodal Single-Cell Network Analysis Identifies Basigin/Cluster of Differentiation 147 as a Tumor-Associated Signaling Hub in Hepatocellular Carcinoma.

BACKGROUND AND AIMS: Hepatocellular carcinoma (HCC) ranks among the leading causes of cancer mortality globally, driven by late diagnosis and inadequate sensitivity of biomarkers like α-fetoprotein for early or indeterminate lesions. Single-cell RNA sequencing (scRNA-seq) enables precise mapping of tumor heterogeneity and early malignant transitions. This study aims to apply scRNA-seq analysis to detect clinically important molecular patterns that define the early stages of malignant transformation in HCC and facilitate the diagnosis of small or ambiguous lesions. METHODS: Here, we integrated

Identification and analyses of natural compounds as potential inhibitors of TRAF6-Basigin interactions in melanoma using structure-based virtual screening and molecular dynamics simulations.

The interaction of the proteins, tumor necrosis factor receptor-associated factor6 (TRAF6) and Basigin (CD147), is known to be associated with the over-expression of matrix metalloproteinases (MMPs) in melanoma cells. MMPs are known to be responsible for melanoma metastasis. Hence, the TRAF6-Basigin complex can act as a potential therapeutic target. In previous studies, amino acid residues Lys340, Lys 384, Glu417 and Glu511 of TRAF6 were identified as the most vital residues on the basis of their contributions to interaction energy, relative solvent accessibility and electrostatic interactions

High-Mannose-Type Glycan of Basigin in Endothelial Cells Is Essential for the Opening of the Blood-Brain Barrier Induced by Hypoxia, Cyclophilin A, or Tumor Necrosis Factor-α.

Pathologic opening of the blood-brain barrier accelerates the progression of various neural diseases. Basigin, as an essential molecule for the opening of the blood-brain barrier, is a highly glycosylated transmembrane molecule specified in barrier-forming endothelial cells. This study analyzed the involvement of basigin in the regulation of the blood-brain barrier focusing on its glycosylation forms. First, basigin was found to be expressed as cell surface molecules with complex-type glycan as well as those with high-mannose-type glycan in barrier-forming endothelial cells. Monolayers of endo

Tissue expression of lactate transporters (MCT1 and MCT4) and prognosis of malignant pleural mesothelioma (brief report).

BACKGROUND: Malignant pleural mesothelioma (MPM) is an aggressive neoplasm of the pleura, mainly related to asbestos exposure. As in other solid tumors, malignant cells exhibit high glucose uptake and glycolytic rates with increased lactic acid efflux into the interstitial space. Lactate transport into and out of cells, crucial to maintaining intracellular pH homeostasis and glycolysis, is carried out by monocarboxylate transporters (MCTs) and the chaperone basigin (CD147). We set out to examine the clinical significance of basigin, MCT1 and MCT4 in the context of MPM and to evaluate their exp

Assessing the functional impact of PfRh5 genetic diversity on ex vivo erythrocyte invasion inhibition.

The PfRh5-Basigin ligand-receptor interaction is an essential step in the merozoite invasion process and represents an attractive vaccine target. To reveal genotype-phenotype associations between naturally occurring allelic variants of PfRh5 and invasion inhibition, we performed ex vivo invasion inhibition assays with monoclonal antibodies targeting basigin coupled with PfRh5 next-generation amplicon sequencing. We found dose-dependent inhibition of invasion across all isolates tested, and no statistically significant difference in invasion inhibition for any single nucleotide polymorphisms. T

Development and application of a high-throughput screening assay for identification of small molecule inhibitors of the P. falciparum reticulocyte binding-like homologue 5 protein.

The P. falciparum parasite, responsible for the disease in humans known as malaria, must invade erythrocytes to provide an environment for self-replication and survival. For invasion to occur, the parasite must engage several ligands on the host erythrocyte surface to enable adhesion, tight junction formation and entry. Critical interactions include binding of erythrocyte binding-like ligands and reticulocyte binding-like homologues (Rhs) to the surface of the host erythrocyte. The reticulocyte binding-like homologue 5 (Rh5) is the only member of this family that is essential for invasion and

CD147 Is Essential for the Development of Psoriasis via the Induction of Th17 Cell Differentiation.

Th17 cells play an important role in psoriasis. The differentiation of naïve CD4+ T cells into Th17 cells depends on glycolysis as the energy source. CD147/basigin, an integral transmembrane protein belonging to the immunoglobulin superfamily, regulates glycolysis in association with monocarboxylate transporters (MCTs)-1 and -4 in cancer cells and T cells. We examined whether CD147/basigin is involved in the pathogenesis of psoriasis in humans and psoriasis-model mice. The serum level of CD147 was increased in patients with psoriasis, and the expression of CD147 and MCT-1 was elevated in their

A role for plasma membrane Ca2+ ATPases in regulation of cellular Ca2+ homeostasis by sphingosine kinase-1.

Sphingosine-1-phosphate (S1P) is a ubiquitous lipid mediator, acting via specific G-protein-coupled receptors (GPCR) and intracellularly. Previous work has shown that deletion of S1P lyase caused a chronic elevation of cytosolic [Ca2+]i and enhanced Ca2+ storage in mouse embryonic fibroblasts. Here, we studied the role of sphingosine kinase (SphK)-1 in Ca2+ signaling, using two independently generated EA.hy926 cell lines with stable knockdown of SphK1 (SphK1-KD1/2). Resting [Ca2+]i and thapsigargin-induced [Ca2+]i increases were reduced in both SphK1-KD1 and -KD2 cells. Agonist-induced [Ca2+]i

The ELAV/Hu protein Found in neurons regulates cytoskeletal and ECM adhesion inputs for space-filling dendrite growth.

Dendritic arbor morphology influences how neurons receive and integrate extracellular signals. We show that the ELAV/Hu family RNA-binding protein Found in neurons (Fne) is required for space-filling dendrite growth to generate highly branched arbors of Drosophila larval class IV dendritic arborization neurons. Dendrites of fne mutant neurons are shorter and more dynamic than in wild-type, leading to decreased arbor coverage. These defects result from both a decrease in stable microtubules and loss of dendrite-substrate interactions within the arbor. Identification of transcripts encoding cyto
