Basophils in Immunity: Activation Pathways, Roles in Allergy and AllergoOncology and Application of the Basophil Activation Test.

Basophils, the least abundant leukocytes, are increasingly recognised as potent immunomodulators. Upon activation, they rapidly release preformed granule-associated mediators including histamine and lipid mediators such as LTC4, while cytokine production occurs over a longer timescale, contributing to downstream immune responses. Basophil activation is driven by IgE-dependent FcεRI crosslinking or IL-3, complement and Toll-like receptor (TLR) pathways, enabling diverse effector responses, including type I hypersensitivity to allergens. Through IL-4 and IL-13 secretion, basophils bridge innate

Mast cells and basophils in cutaneous immune responses.

Mast cells and basophils share some functions in common and are generally associated with T helper 2 (Th2) immune responses, but taking basophils as surrogate cells for mast cell research or vice versa for several decades is problematic. Thus far, their in vitro functions have been well studied, but their in vivo functions remained poorly understood. New research tools for their functional analysis in vivo have revealed previously unrecognized roles for mast cells and basophils in several skin disorders. Newly developed mast cell-deficient mice provided evidence that mast cells initiate contac

Basophils and mast cells in chronic idiopathic urticaria.

Chronic idiopathic urticaria (CIU) is diagnosed in patients when urticarial eruptions recur for more than 6 weeks, and no specific cause is determined. Given that urticaria resembles the lesions induced by injection of histamine or allergen into the skin, a role for mast cells or basophils has been proposed in the generation of localized urticarial lesions. However, currently, the exact mechanisms governing regional mast cell or basophil activation are unknown. In the past decade, there has been mounting interest in viewing CIU as an autoimmune disease, given the presence of circulating autoan

Effector cells of anaphylaxis: mast cells and basophils.

Systemic anaphylaxis arises when mast cells, possibly along with other cell types, are provoked to secrete mediators that evoke a systemic response. Mast cells in perivascular, respiratory, gastrointestinal and cutaneous tissues are likely involved, regardless of whether IgE or non-IgE-dependent pathways are invoked. Alpha/beta tryptases are selectively and abundantly produced by mast cells. Tryptase levels in the circulation provide a precise indicator of mast cell involvement. Mature beta tryptase is stored in secretory granules and is released when the cells are activated to degranulate, as

Interleukin 2 regulates the activation of human basophils.

Basophils are important effector cells in allergic disorders and anti-parasitic immune response. A number of activators including interleukin 3 (IL-3) and IgE have been identified in the regulation of human basophils expressing mediators such as histamine and leukotriene C4 (LTC4) and cytokines, including IL-4 and IL-13. Human basophils express high levels of IL-2 receptors. However, the function of the IL-2 pathway in basophils remains unknown. Here, we identified that IL-2 induced the activation of human basophils in vitro to express a variety of inflammatory cytokines and chemokines includi

An inherited anomaly of human eosinophils and basophils.

Inclusions staining gray or blue-gray in Romanowsky treated films were found in eosinophils and basophils in a family as an autosomal dominant anomaly without overt clinical effects. Associated with the inclusions were distinctive crystals which were different from the crystalloids of eosinophil granules and from Charcot-Leyden crystals. The restriction of the anomaly to eosinophils and basophils indicates that these are related more closely to each other than to neutrophils and mast cells, and suggests the existence of a separate stem cell for eosinophils and basophils.

Chronic Basophilic Leukaemia in a Dog.

A 13-year-old neutered female mixed-breed dog with a clinical history of emaciation, inappetence and vomiting for 2 months was presented. Blood tests showed marked leucocytosis with increased neutrophil and basophil count, mild thrombocytosis and anaemia. Seven days after the initial visit, the dog died and was submitted for necropsy examination. Grossly, the bone marrow was red in colour and hepatomegaly and splenomegaly with discolouration were observed. A bone marrow smear showed an increased proportion of basophilic lineage cells. Histologically, the bone marrow showed high cellular densit

Basophil responsiveness in chronic urticaria.

Chronic urticaria is a common skin disease without an etiology in the majority of cases. The similarity of symptoms and pathology to allergen-induced skin reactions supports the idea that skin mast cell and blood basophil IgE receptor activation is involved; however, no exogenous allergen trigger has been identified. Recent evidence supports a role for blood basophils in disease expression. Specifically, blood basopenia is noted in active disease with the recruitment of blood basophils to skin lesional sites. In addition, blood basophils display altered IgE receptor-mediated degranulation that

Mast cell and basophil development.

Mast cells and basophils express certain remarkable similarities in mediator content, histochemical characteristics, and function. Yet a large body of evidence now indicates that the mast cell and basophil lineages are distinct. Stem cell factor, the ligand for the receptor encoded by c-kit, is a major growth factor for mast cells in both rodent and primate species, and can modulate mast cell secretory function. Moreover, abnormalities affecting the stem cell factor receptor or stem cell factor might contribute to some cases of mastocytosis or mast cell neoplasms. By contrast, basophils can de

Basophils in the regulation of allergies and inflammatory disorders.

Basophils are rare granulocytes that were long overlooked because of their scarcity and phenotypic overlap with mast cells. Recent advances in genetic models and antibody-based depletion strategies have redefined basophils as versatile regulators of inflammation, particularly in the context of type 2 immune responses such as atopic dermatitis (AD) and allergic asthma. In patients with AD and asthma, basophils infiltrate sites of inflammation, and studies using mouse models have demonstrated their crucial roles in allergic inflammation of the skin and lungs. Emerging evidence further indicates

Basophils activate oncostatin M receptor-expressing vagal sensory neurons.

BACKGROUND: Vagal sensory neurons (VSNs) relay signals from internal organs to the brainstem via the vagus nerve. Published transcriptomic data sets suggest that VSNs express Mas-related G protein-coupled receptor family member D and oncostatin M (OSM) receptor, markers also found in dorsal root ganglia-resident pruriceptor neurons. However, the cellular source of OSM and its potential interactions with VSNs remain unknown. OBJECTIVE: We sought to validate the expression of pruritogen receptors on VSNs, identify basophils as a source of OSM, and determine how OSM modulates VSN activity. METH

Are Basophils and Mast Cells Masters in HIV Infection?

The World Health Organization AIDS epidemic update estimates that more than 37 million people are living with HIV infection. Despite the unprecedented success of antiretroviral treatments, significant challenges remain in the fight against HIV. In particular, how uninfected cells capture HIV and transmit virions to target cells remains an unanswered question. Tissue mast cells and peripheral blood basophils can be exposed to virions or HIV products during infection. Several HIV proteins (i.e., envelope glycoproteins gp120 and gp41, Tat, and Nef) can interact with distinct surface receptors exp

Selective depletion of basophils ameliorates immunoglobulin E-mediated anaphylaxis.

Basophils, which are the rarest granulocytes, play crucial roles in protective immunity against parasites and development of allergic disorders. Although immunoglobulin (Ig)E-dependent responses via receptor for IgE (FcεRI) in basophils have been extensively studied, little is known about cell surface molecules that are selectively expressed on this cell subset to utilize the elimination in vivo through treatment with monoclonal antibody (mAb). Since CD200 receptor 3 (CD200R3) was exclusively expressed on basophils and mast cells (MCs) using a microarray screening, we have generated anti-CD200

Signaling pathways activated by a protease allergen in basophils.

Allergic diseases represent a significant burden in industrialized countries, but why and how the immune system responds to allergens remain largely unknown. Because many clinically significant allergens have proteolytic activity, and many helminths express proteases that are necessary for their life cycles, host mechanisms likely have evolved to detect the proteolytic activity of helminth proteases, which may be incidentally activated by protease allergens. A cysteine protease, papain, is a prototypic protease allergen that can directly activate basophils and mast cells, leading to the produc

Basophils in atopic dermatitis: Immunologic roles and clinical implications.

Atopic dermatitis (AD) or eczema is a chronic inflammatory skin disease, characterized by dysfunction of the epidermal skin barrier causing increased sensitization to environmental allergens. The resulting type 2-dominated local and systemic immune responses cause epidermal hyperplasia, dermal infiltration by T cells and eosinophilia, and elevated levels of total and allergen-specific IgE. In addition, the skin of patients with AD is often colonized with Staphylococcus aureus which correlates with diseases severity. Recent emerging studies highlight a significant role for basophils in both loc
