Nowadays, bioprocesses are developed or optimized on small scale. Also, vinegar industry is motivated to reinvestigate the established repeated batch fermentation process. As yet, there is no small-scale culture system for optimizing fermentation conditions for repeated batch bioprocesses. Thus, the aim of this study is to propose a new shaken culture system for parallel repeated batch vinegar fermentation. A new operation mode - the flushing repeated batch - was developed. Parallel repeated batch vinegar production could be established in shaken overflow vessels in a completely automated oper

While perfusion culture for Chinese hamster ovary (CHO) cells offers advantages such as continuous operation and flexibility, it suffers from product loss through cell bleeding and difficulties in reaching high productivity due to sustained rapid cell growth. Growth inhibitory strategies are widely used to enhance productivity in fed‑batch processes; however, their practical implementation and comparative effectiveness in perfusion processes remain insufficiently explored. Meanwhile, process development often relies on costly trial‑and‑error approaches. Here, we systematically compared three g

Chinese hamster ovary (CHO) cells are the industrial workhorse for manufacturing biopharmaceuticals, including monoclonal antibodies. CHO cell line development requires a more data-driven approach for the accelerated identification of hyperproductive cell lines. Traditional methods, which rely on time-consuming hierarchical screening, often fail to elucidate the underlying cellular mechanisms driving optimal bioreactor performance. Big data analytics, coupled with advancements in "omics" technologies, are revolutionizing the study of industrial cell lines. Translating this knowledge into pract

Improving the productivity of a biopharmaceutical Chinese hamster ovary (CHO) fed-batch cell culture can enable cost savings and more efficient manufacturing capacity utilization. One method for increasing CHO cell productivity is the addition of histone deacetylase (HDAC) inhibitors to the cell culture process. In this study, we examined the effect of valproic acid (VPA, 2-propylpentanoic acid), a branched-chain carboxylic acid HDAC inhibitor, on the productivity of three of our CHO cell lines that stably express monoclonal antibodies. Fed-batch shake flask VPA titrations on the three differe

Monoclonal antibodies (mAbs) represent a majority of biotherapeutics in the market today. These glycoproteins undergo posttranslational modifications, such as N-linked glycosylation, that influence the structural & functional characteristics of the antibody. Glycosylation is a heterogenous posttranslational modification that may influence therapeutic glycoprotein stability and clinical efficacy, which is why it is often considered a critical quality attribute (CQA) of the mAb product. While much is known about the glycosylation pathways of Chinese Hamster Ovary (CHO) cells and how cell culture

Current industrial processes for yeast-based conversion of glucose, xylose, and arabinose mixtures in lignocellulosic hydrolysates mostly rely on monocultures of generalist strains that consume the sugars sequentially. Co-cultivation of "sugar-specialist" strains offers potential advantages regarding volumetric productivity and genetic stability of pentose-fermenting strains during prolonged operation. However, in these cultures, the slow fermentation of pentoses strongly affects the time required for complete sugar conversion. This study explores whether the kinetics of such co-cultures can b

Ethanol concentrations above 4% (v/v) are required for economic bioethanol production due to the cost of recovery from dilute solutions. Although thermophilic bacteria have many potential advantages over Saccharomyces cerevisiae as process organisms for second generation bioethanol production, they are known to be less tolerant to ethanol, typically to concentrations less than 4% (v/v). To address this issue we have investigated the application of in situ gas-stripping of ethanol using microbubbles to increase the surface area per unit volume of gas, using fed-batch and continuous cultures of

The general approach to industrial production of monoclonal antibodies is fed-batch culture using Chinese Hamster Ovary (CHO) cells. Perfusion culture is also attracting attention as a next-generation culture method. In these culture methods, optimization of amino acid and glucose concentration in the culture medium is essential, and influences cell proliferation, viability, productivity, and monoclonal antibody quality. Further, the maintenance of optimal nutrient levels - by avoiding both depletion and accumulation - is crucial. This study aimed to develop a dynamic feeding strategy based on

For bioprocesses producing live virus, such as enterovirus Coxsackievirus A21, viral titer (infectivity basis) decay rates can exceed 30% within a day. Consequently, harvest timing is paramount. To optimize titer at harvest, a continuous viral product titer model was generated to elucidate kinetics. The model leveraged experimentally determined viable cell density, cell-specific viral productivity, and viral specific decay rates. Next, three separate online process analytical technology (PAT) harvest triggers were developed to predict maximal viral titer. Finally, the PAT harvest triggers were

Lactic acid (LA) serves as a widely used platform compound and has received significant attention as a raw material for synthesis of biodegradable polylactic acid. Currently, LA is mainly produced through microbial fermentation, but its high costs undermine its competitive advantage against other materials, necessitating the development of novel production routes. Methanol bioconversion represents an emerging low-carbon circular economy, where LA could become an outstanding representative product. This study successfully established an efficient methanol-based LA synthesis route in Pichia past

Variation in the primary sequence of monoclonal antibodies (mAbs) can negatively affect their behavior in biopharmaceutical manufacturing platforms, and efforts to identify mAbs with poor "developability" characteristics lack robust methods for assessing mAb expression from an industrially relevant platform. Recent advancements in site-specific integration-based (SSI) platforms in Chinese hamster ovary (CHO) cells can mitigate the high transcriptional variation observed with random integration and the low industrial relevance of transient expression by providing a flexible platform for mAb exp

Established animal cells, such as Vero, Madin Darby canine kidney (MDCK) or chicken embryo fibroblasts (CEFs), are still the main cell lines used for viral vaccine production, although new "designer cells" have been available for some years. These designer cell lines were specifically developed as a cell substrate for one application and are well characterized. Later screening for other possible applications widened the product range. These cells grow in suspension in chemically defined media under controlled conditions and can be used for up to 100 passages. Scale-up is easier and current pro

Chlamydomonas reinhardtii CW15, a cell-wall-deficient strain, is a widely used chassis in microalgal genetic engineering. However, its low cell density under autotrophic and mixotrophic cultivation restricts industrial scalability. This study aimed to establish a cost-effective, high-density heterotrophic cultivation process for C. reinhardtii CW15. Initially, an optimized AP medium (Tris-free TAP with K2HPO4 as the sole phosphorus source) significantly reduced medium costs while achieving a cell density of 0.446 g/L. Subsequently, a fed-batch process in a 5 L fermentor yielded a record cell d

The recombinant production of extracellular matrix proteins is a promising approach for replacing animal-derived materials in biomedical applications. K. phaffii represents a favorable expression host because it combines the ability of higher eukaryotes for secreted protein production with the ability to grow to high cell densities on simple, low-cost media. Additionally, this well-studied host allows for tight control of recombinant protein expression using the methanol-inducible AOX1 promoter. In this study, different methanol feeding strategies were evaluated to optimize the expression of a

Applying a single parameter set to describe complex mammalian kinetics often is too simplistic, as it fails to capture sensitive cell-to-environment interactions that may be exploited to optimize production performance. To resolve this time dependency, intra-experimental parameter shifts as part of design of dynamic experiments (DoDE) can be performed to study mammalian growth and production kinetics in fed-batch processes. This enables growth phase-dependent optimization, aligned with cellular requirements. Here, we provide a comprehensive, head-to-head comparison of our phase-dependent optim

The use of bioreactors coupled to membrane-based perfusion systems enables very high cell and product concentrations in vaccine and viral vector manufacturing. Many virus particles, however, are not stable and either lose their infectivity or physically degrade resulting in significant product losses if not harvested continuously. Even hollow fiber membranes with a nominal pore size of 0.2 µm can retain much smaller virions within a bioreactor. Here, we report on a systematic study to characterize structural and physicochemical membrane properties with respect to filter fouling and harvesting

This study developed a magnetic nanoparticles (MNPs) harvesting and reactivation technique for rapid cyanobacteria Microcystis aeruginosa separation. The harvesting of raw MNPs achieved high efficiency of 99.6% with the MNPs dosage of 0.58g MNPs/g dry-biomass, but gradually decreased to 59.1% when directly reused 5 times. With extra ultrasonic chloroform:methanol solvent treatment, the MNPs can be effectively reactivated for M. aeruginosa harvesting with 60% efficiency after 5 times reactivation and the separation efficiency kept above 93% with 0.20g MNPs/g dry-biomass dosage. The cyanobacteri

Microwell arrays are established platforms for high-throughput microbial screening and single-cell analysis. However, most existing systems are limited to batch culture, where nutrient depletion and metabolite accumulation restrict sustained bacterial growth and compromise downstream characterization. Here, we report a dual-sided microwell array integrated with a track-etched polyester (PETE) nanopore membrane, enabling fed-batch bacterial culture with controlled mass transport and targeted extraction from individual wells. The device incorporates a stepped through-hole well structure that fac

BACKGROUND: Nanobodies (NB) are compact single-domain antibody fragments with substantial diagnostic and therapeutic potential, but scalable microbial production under controlled bioreactor conditions remains insufficiently characterized. In this study a high-cell-density cultivation (HCDC) fed-batch process for an anti-human CD45 NB in Escherichia coli was established and evaluated how process parameters, host background, and subcellular targeting (cytoplasmic vs. periplasmic) affect yield and product quality including correct disulfide bond formation.
RESULTS: Using a two-stage design-of-ex

Optimizing fed-batch fermentation strategies is key to maximizing bioprocess efficiency. While mathematical modeling can aid process design, its complexity often limits accessibility for experimental scientists. We present FedBatchDesigner, a user-friendly web tool for optimizing fed-batch processes with a growth-arrested production stage. With minimal input requirements, FedBatchDesigner enables rapid exploration of a process's titer, rate, and yield (TRY) landscape for constant, linear, and exponential feeding strategies. Interactive visualizations allow users to assess trade-offs between pr
