Bioengineered skin has great potential for use in regenerative medicine for treatment of severe wounds such as burns or chronic ulcers. Genetically modified skin substitutes have also been used as cell-based devices or "live bioreactors" to deliver therapeutics locally or systemically. Finally, these tissue constructs are used as realistic models of human skin for toxicological testing, to speed drug development and replace traditional animal-based tests in a variety of industries. Here we describe a method of generating bioengineered skin based on a natural scaffold, namely, decellularized hu

Özugur et al. recently pushed the boundaries of augmented physiology through artificial symbioses. They microinjected algal cultures into tadpole (Xenopus laevis) hearts. The resulting capillary-bound algae produced physiologically relevant levels of oxygen, which could rescue neuronal hypoxia. This work contributes to the growing field of artificial photosymbioses.

Regenerative Medicine is an emerging field that combines basic research and clinical observations in order to identify the elements required to replace damaged tissues and organs in vivo and to stimulate the body's intrinsic regenerative capacity. Great benefits are expected in this field as researchers take advantage of the potential regenerative properties of both embryonic and adult stem cells, and more recently, of induced pluripotent stem cells. Bioengineered skin emerged mainly in response to a critical need for early permanent coverage of extensive burns. Later this technology was also

Cardiovascular disease (CVD) affecting blood vessel function is a leading cause of death around the world. A common treatment option to replace the diseased blood vessels is vascular grafting using the patient's own blood vessels. However, patients with CVD are usually lacking vessels for grafting. Recent advances in tissue engineering are now providing alternatives to autologous vascular grafts in the form of tissue-engineered blood vessels (TEBVs). In this review, we will describe the use of different scaffolding systems, cell sources and conditioning approaches for creating fully functional

Over 4.2 million people worldwide suffer from vision impairment related to corneal diseases, with the shortage of donor corneas limiting surgical interventions. The development of 3D printing provides a promising solution for corneal tissue engineering, offering precise control over dimensionality, structural organization, and cell-matrix interactions. The choice of biomaterials and printing strategies critically determines the properties of 3D-printed corneal constructs. While prior reviews have summarized general bioinks and techniques, few have addressed cornea-specific benchmarks such as t

Unraveling the complex behavior of healthy and disease podocytes by analyzing the changes in their unique arrangement of foot processes, slit diaphragm, and the three-dimensional (3D) morphology is a long-standing goal in kidney-glomerular research. The complexities surrounding the podocytes' accessibility in animal models and growing evidence of differences between humans and animal systems have compelled researchers to look for alternate approaches to study podocyte behaviors. With the advent of bioengineered models, an increasingly powerful and diverse set of tools is available to develop n

OBJECTIVE: The use of exogenous materials as scaffolds in cartilage tissue engineering has limited the clinical application of resultant constructs due to the risk of postoperative complications. In an effort to minimize such complications, we aim to generate human, scaffold-free auricular cartilaginous constructs.
STUDY DESIGN: Laboratory study using pediatric auricular cartilage.
METHODS: Remnant, normal pediatric auricular cartilage samples that would have otherwise been discarded were collected and digested to free cells. Harvested cells were cultured and expanded in vitro for two passag

Kidney regeneration is likely to provide an inexhaustible source of tissues and organs for immunosuppression-free transplantation. It is currently garnering considerable attention and might replace kidney dialysis as the ultimate therapeutic strategy for renal failure. However, anatomical complications make kidney regeneration difficult. Here, we review recent advances in the field of kidney regeneration, including (i) the directed differentiation of induced pluripotent stem cells/embryonic stem cells into kidney cells; (ii) blastocyst decomplementation; (iii) use of a decellularized cadaveric

Embryo transfer (ET) is the final manual intervention in extracorporeal fertilization in which an embryo is transferred into the uterus by a transcervical catheter. The low rates of embryo implantation within the uterus are attributed, among other factors, to the ET technique, which depends on a multitude of anatomical, physiological, and mechanical aspects. We developed computational and experimental models to simulate ET to examine the contribution of mechanical features to the success of this procedure. The experimental model allowed laboratory simulations of the dispersion of the catheter

Extracellular vesicles (EVs) are nano-scale structures produced by cells that transport biological substances for intercellular communication. The tetraspanins CD9, CD81, and CD63 are crucial to EV biogenesis and function. This study uses CRISPR-Cas9 system to knock out (KO) CD9, CD63, and CD81 in HEK293T cells. The goal is to investigate the role of these tetraspanins in EV bioengineering with the hypothesis that repressing endogenous production may increase the availability of exogenously introduced tetraspanin-fusion constructs and increase engineered EV production. First, it is observed th

Nanotechnology is transforming the area of corneal tissue engineering by improving scaffold design and enabling sophisticated therapeutic strategies. Nanomaterials are being used to improve the corneal scaffolds' mechanical strength, permeability, and transparency, as well as to enable the therapeutic agents' targeted delivery by nanocarriers. These improvements deal with important problems in corneal repair, like inflammation, infections, and neovascularization. While corneal transplantation remains a standard treatment, the risk of rejection and availability of donor tissue are the main limi

Over the past several decades, ex vivo perfusion has emerged as a promising technology for the assessment, preservation, and recovery of donor organs. Many exciting pre-clinical findings have now been translated to clinical use, and successful transplantation following ex vivo perfusion has been achieved for heart, lung, and liver. While machine perfusion provides distinct advantages over traditional cold preservation, many challenges remain, including that of long-term (multi-day) ex vivo support. Here, we provide an overview of the current status of ex vivo machine perfusion in the pre-clini

OBJECTIVE: Bioengineering of viable, functional, and implantable human lung grafts on porcine matrix.
SUMMARY BACKGROUND DATA: Implantable bioartificial organ grafts could revolutionize transplant surgery. To date, several milestones toward that goal have been achieved in rodent models. To make bioengineered organ grafts clinically relevant, scaling to human cells and graft size are the next steps.
METHODS: We seeded porcine decellularized lung scaffolds with human airway epithelial progenitor cells derived from rejected donor lungs, and banked human umbilical vein endothelial cells. We subs

RATIONALE: More than 25 million individuals have heart failure worldwide, with ≈4000 patients currently awaiting heart transplantation in the United States. Donor organ shortage and allograft rejection remain major limitations with only ≈2500 hearts transplanted each year. As a theoretical alternative to allotransplantation, patient-derived bioartificial myocardium could provide functional support and ultimately impact the treatment of heart failure.
OBJECTIVE: The objective of this study is to translate previous work to human scale and clinically relevant cells for the bioengineering of func

The present review aims to illustrate the strategies that are being implemented to regenerate or bioengineer livers for clinical purposes. There are two general pathways to liver bioengineering and regeneration. The first consists of creating a supporting scaffold, either synthetically or by decellularization of human or animal organs, and seeding cells on the scaffold, where they will mature either in bioreactors or in vivo. This strategy seems to offer the quickest route to clinical translation, as demonstrated by the development of liver organoids from rodent livers which were repopulated w

About 27,000 deaths are registered annually in the United States due to liver disease. At this time, the only definitive treatment of hepatic failure is orthotopic transplantation. However, there is a critical shortage of organs with the total waiting list for all organs currently at 100,000 requests. The number is increasing by 5% every year. Given that only organs in pristine condition are transplantable and that the hidden demand for organs as an anti-aging solution will be many times the current figures, orthotopic transplantation will always remain a limited pool. The increasing donor org
