Evaluation of novel disposable bioreactors on pandemic influenza virus production.

Since 1997, the highly pathogenic influenza H5N1 virus has spread from Hong Kong. According to the WHO bulletin report, the H5N1 virus is a zoonotic disease threat that has infected more than 850 humans, causing over 450 deaths. In addition, an outbreak of another new and highly pathogenic influenza virus (H7N9) occurred in 2013 in China. These highly pathogenic influenza viruses could potentially cause a worldwide pandemic. it is crucial to develop a rapid production platform to meet this surge demand against any possible influenza pandemic. A potential solution for this problem is the use of

Different bioreactors for treating secondary effluent from recycled paper mill.

Secondary effluent from paper mill was characterized by poor biodegradability and containing recalcitrant compounds. In this study, four bioreactors, including a sequencing batch biofilm reactor (SBBR), a stirred-tank reactor (STR) and two submerged aeration reactors (SAR) were used to treat secondary effluent from a recycled paper mill respectively. The results indicated that chemical oxygen demand (COD) was increased by SAR2 treatment and COD removal efficiency for SBBR, SAR1 and STR was 39.7%, 15.7% and 30.9% respectively. It is suggested that recalcitrant compounds were removed by SBBR, SA

Optimizing the culture of Plasmodium falciparum in hollow fiber bioreactors.

The hollow fiber bioreactor (HFBR) is a cell culturing system allowing continuous perfusion of medium. It was designed to grow microorganisms in a dynamically altering medium mimicking change in the in vivo intravascular and extravascular compartments. The cell compartment (extra capillary space) and medium compartment (intra capillary space) are connected through pores of semipermeable fiber membranes. These membranes allow exchange of gas and nutrients. We have adapted this system for the ex vivo culture of Plasmodiumfalciparum at high parasite densities. A Thai P. falciparum isolate (TM036)

Optimization of batch and fed-batch bioreactors using simulated annealing.

In order for biobased industrial products to compete economically with petroleum-derived products, significant reduction in their processing cost is necessary. Since most bioprocesses are operated in batch or fed-batch mode, their optimization involves theoretical and computational challenges. Simulated annealing (SA), a stochastic optimization algorithm, is used in this study to solve a number of challenging optimization problems related to the design and operation of bioreactors. Two well-known case studies are considered in which the robustness and efficiency of the SA algorithm is demonstr

Ultrasound Imaging and Machine Learning for Nondestructive Sensing in Bioreactors.

The advancement of cell therapy and cellular agriculture underscores the need for noninvasive, cost-effective methods for continuous monitoring of large-scale cell production. Bioreactors, designed to mimic physiological conditions to facilitate cell growth, require reliable quality control measures. This study investigates the potential of ultrasound technology to characterize cellular growth and decellularization in spinach scaffolds. Ultrasound images were acquired of fresh spinach leaves, decellularized leaves, and leaves with cells seeded in defined cloning well regions on decellularized

Riverine mycobiome dynamics: From South African tributaries to laboratory bioreactors.

Riverine fungi have the capacity for both pathogenicity, pertinent for countries with elevated immunosuppressed individuals, and bioremediation potential. The purpose was (i) to screen for the presence of clinically relevant riverine fungi and associations with anthropogenic influence, and (ii) the acclimatisation of environmental communities toward potential bioremediation application. Communities were harvested from polluted rivers in Stellenbosch, South Africa, and mycobiomes characterised by high-throughput amplicon sequencing. The remainder of the biomass was inoculated into continuous bi

Microbial communities in full-scale woodchip bioreactors treating aquaculture effluents.

Woodchip bioreactors are being successfully applied to remove nitrate from commercial land-based recirculating aquaculture system (RAS) effluents. In order to understand and optimize the overall function of these bioreactors, knowledge on the microbial communities, especially on the microbes with potential for production or mitigation of harmful substances (e.g. hydrogen sulfide; H2S) is needed. In this study, we quantified and characterized bacterial and fungal communities, including potential H2S producers and consumers, using qPCR and high throughput sequencing of 16S rRNA gene. We took wat

Evaluation of Mechanistic Models for Nitrate Removal in Woodchip Bioreactors.

Woodchip bioreactors (WBRs) are increasingly being applied to remove nitrate from runoff. In this study, replicate columns with aged woodchips were subjected to a range of measured flow rates and influent nitrate concentrations with an artificial stormwater matrix. Dissolved oxygen (DO), nitrate, and dissolved organic carbon (DOC) were measured along the length of the columns. A multispecies reactive transport model with Michaelis-Menten kinetics was developed to explain the concentration profiles of DO, nitrate, and DOC. Four additional models were developed based on simplifying assumptions,

Hollow Fiber Bioreactors for In Vivo-like Mammalian Tissue Culture.

Tissue culture has been used for over 100 years to study cells and responses ex vivo. The convention of this technique is the growth of anchorage dependent cells on the 2-dimensional surface of tissue culture plastic. More recently, there is a growing body of data demonstrating more in vivo-like behaviors of cells grown in 3-dimensional culture systems. This manuscript describes in detail the set-up and operation of a hollow fiber bioreactor system for the in vivo-like culture of mammalian cells. The hollow fiber bioreactor system delivers media to the cells in a manner akin to the delivery of

Sustainable production of bioactive compounds from sponges: primmorphs as bioreactors.

Sponges [phylum Porifera] are a rich source for the isolation of biologically active and pharmacologically valuable compounds with a high potential to become effective drugs for therapeutic use. However, until now, only one compound has been introduced into clinics because of the limited amounts of starting material available for extraction. To overcome this serious problem in line with the rules for a sustainable use of marine resources, the following routes can be pursued; first, chemical synthesis, second, cultivation of sponges in the sea (mariculture), third, growth of sponge specimens in

Reactor engineering for the enzymatic synthesis of 5-hydroxymethylfurfural stearate in a batch bioreactor and a packed bed flow bioreactor.

The enzymatic esterification of 5-hydroxymethylfurfural (HMF) with long-chain fatty acids offers a sustainable route for producing biolubricants and other high-value chemicals. This work evaluates the synthesis of 5-hidroxymethylfurfural stearate catalyzed by immobilized lipases in both batch and continuous packed-bed bioreactors, combining molecular dynamics (MD) simulations with experimental validation to identify suitable green solvents. Four solvents were tested: 2-methyl-3-buten-2-ol (2-MB), tert-butanol (TB), 2-methyltetrahydrofuran (2-MeTHF), and cyclopentyl methyl ether (CPME). MD simu

Yellow Fever Virus Production in Bioreactor.

On a commercial scale, the vaccine against yellow fever is based on replication of the attenuated virus in embryonated eggs [2]. A very promising alternative platform to the production of the virus in eggs, which has spread as a worldwide trend, is production using mammalian cell lines. The advantage of using established animal cell lines in vaccine production lies in their greater reproducibility and convenience when compared to primary cells and animal models, as well as their great potential for large-scale production. Such cell lines can be cultivated in bioreactors, under monitored and co

Bioreactor maintained living skin matrix.

Numerous reconstructive procedures result in wounds that require skin grafting. Often, the amount of tissue available from donor sites is limited. In vivo tissue expanders have been used clinically to generate larger sections of skin, and other methods exist to cover large wounds, but all have significant limitations. We investigated whether these difficulties could be overcome by increasing the surface area of skin in vitro while maintaining tissue viability. Human foreskin was incrementally expanded in a computer-controlled bioreactor system over 6 days to increase its surface dimensions und

In vivo engineering of organs: the bone bioreactor.

Treatment of large defects requires the harvest of fresh living bone from the iliac crest. Harvest of this limited supply of bone is accompanied by extreme pain and morbidity. This has prompted the exploration of other alternatives to generate new bone using traditional principles of tissue engineering, wherein harvested cells are combined with porous scaffolds and stimulated with exogenous mitogens and morphogens in vitro and/or in vivo. We now show that large volumes of bone can be engineered in a predictable manner, without the need for cell transplantation and growth factor administration.

Strategies for improving the functionality of an affinity bioreactor.

Heparin employed in extracorporeal blood circulation (ECBC) procedures (e.g. open heart operations) often leads to a high incidence of bleeding complications. Protamine employed in heparin neutralization, on the other hand, can cause severe adverse reactions. We previously developed an approach that could prevent both heparin- and protamine-induced toxic side effects concomitantly. This approach consisted of placing a hollow fiber-based bioreactor device containing immobilized protamine (termed a "protamine bioreactor") at the distal end of the ECBC procedure. This protamine bioreactor would r

Removal of tetrachloroethylene in an anaerobic column bioreactor.

Removal of tetrachloroethylene (perchloroethylene; C2Cl4) by microbial consortia from two sites with different C2Cl4 exposure histories was examined in a bench-scale anaerobic column bioreactor. It was hypothesized that optimal removal would be observed in the reactor packed with sediments having an extensive exposure history. Microbial consortia were enriched from hyporheic-zone (HZ) sediments from the Portneuf aquifer near Pocatello, Idaho, and from industrial-zone (IZ) sediments from a highly contaminated aquifer in Portland, Oregon. Lactate and acetate were the electron donors during exper

Structural Design of Ceramic Membranes to Mitigate Fouling in Membrane Bioreactors.

Despite the robust mechanical and chemical stability that make hollow flat-sheet ceramic membranes highly attractive for membrane bioreactors (MBRs), the fundamental relationship between their structural design, specifically pore size and structural symmetry, and biological fouling behavior remains elusive. To decouple the effects of membrane architecture on fouling mechanisms, a series of symmetric and asymmetric hollow flat-sheet alumina membranes were systematically engineered. Symmetric architectures with tunable pore sizes were fabricated by controlling aggregate particle sizes, whereas a

Protocol for controlled gas-phase oxygen In-leakage in anaerobic bioreactors.

Trace oxygen may enter the gas supply line of an anaerobic bioreactor and affect reactor operation. In this study, we describe a method to expose a defined oxygen concentration for gas-fed anaerobic bioreactors. H2, CO2 and premixed CO2/O2 gas were supplied using mass flow controllers, and the reactor inlet O2 concentration was adjusted from 0 to 10,000 ppm. Both stepwise and constant exposure tests were carried out. The reactor outlet O2 concentration was measured by gas chromatography to confirm the actual exposure condition in bioreactors. Because oxygen exposure is defined at the reactor i

Engineered Living Material Bioreactors with Tunable Mechanical Properties using Vat Photopolymerization.

3D-printed engineered living materials (ELM) are promising bioproduction platforms for agriculture, biotechnology, sustainable energy, and green technology applications. However, the design of these platforms faces several challenges, such as the processability of these materials into complex form factors and control over their mechanical properties. Herein, ELM are presented as 3D-printed bioreactors with arbitrary shape geometries and tunable mechanical properties (moduli and toughness). Poly(ethylene glycol) diacrylate (PEGDA) is used as the precursor to create polymer networks that encapsu
