Comparative bursal cytokine gene expression and apoptosis in vaccinated chickens following virulent infectious bursal disease virus challenge.

The bursal cytokine gene expression and apoptosis were compared in vaccinated chickens with either live or immune-complex infectious bursal disease virus (IBDV) vaccines with or without virulent IBDV challenge. The cytokine gene expressions were evaluated at 5 and 12 day-post-challenge (DPC). The apoptotic marker Caspase-3 was determined by IHC on collected bursae, thymus, spleen, and kidneys at 12 DPC. A significantly decreased bursal cytokine levels were observed in the all-vaccinated birds except for IL-6 in the classic IBD vaccines at 5DPC. A significant upregulation of the IL-2 was observ

Immunoreactivity and morphological changes of bursal follicles in chickens infected with vaccine or wild-type strains of the infectious bursal disease virus.

Infectious bursal disease (IBD) is characterized by immunosuppression due to the depletion of lymphocytes in the atrophied bursa of Fabricius (BF). We have sometimes encountered contradictory findings: chickens infected with the vaccine IBD virus (IBDV) strain have sometimes exhibited a highly atrophied BF, but not immunosuppression. In this study, chickens administered vaccine or wild-type strains of IBDV were later vaccinated with the B1 strain of the Newcastle disease virus (NDV). Bursal changes were examined histologically with a focus on the bursal follicle. The immunoreactivity to NDV wa

Phylogenetic diversity of avian nephritis virus in Hungarian chicken flocks.

Avian nephritis virus (ANV) infection was detected in 4-day-old to 22-day-old chickens collected on Hungarian farms between 2002 and 2005. The animals suffered from diarrhoea, growth retardation, runting-stunting syndrome, and 2 to 6% mortality was reported. Tubulonephrosis, interstitial nephritis and uricosis (gout) was diagnosed. The presence of ANV RNA was detected in chicken carcasses using reverse transcriptase-polymerase chain reaction. The virus was demonstrated in 69% of the investigated farms. The nucleotide sequence of the amplification products (corresponding to part of the genome t

Mapping Molecular Determinants of Antigenicity and Pathogenicity of Infectious Bursal Disease Virus (IBDV): A Scoping Review.

Infectious bursal disease virus (IBDV) is an immunosuppressive pathogen posing a major threat to poultry health worldwide. Its marked phenotypic variability is driven by the rapid evolution of its double-stranded RNA genome, primarily achieved through mutation and reassortment. Although extensive evidence has been generated on molecular determinants of antigenicity and pathogenicity, interpretation is often hindered by heterogeneity and lack of systematicity. This scoping review synthesizes over 35 years of research on amino acid positions influencing IBDV phenotype. A total of 62 studies repo

Passive surveillance of infectious bursal disease virus circulating in Egyptian chicken flocks in 2025.

Infectious bursal disease virus (IBDV) causes severe immunosuppression diseases in poultry, resulting in substantial economic losses for the global poultry industry. This study aimed to characterize circulating IBDV strains in Egyptian chicken flocks and assess their potential impact on poultry health and production. We conducted a passive surveillance study by collecting bursal samples from a total of 30 flocks, including commercial broilers, layers, and Baladi chickens, across nine Egyptian governorates in 2025. These flocks exhibited clinical signs of depression, along with kidney and bursa

Characterization of the activity of 2'-C- methylcytidine against infectious pancreatic necrosis virus replication.

Infectious pancreatic necrosis virus (IPNV) is the pathogen of infectious pancreatic necrosis (IPN), which can cause high mortality in salmonids, harm the healthy development of salmon-trout aquaculture, and lead to huge economic losses. However, in China, there is currently neither a commercially available vaccine to prevent IPNV infection nor antiviral drugs to treat IPNV infection. The genome of IPNV consists of two segments of dsRNA named A and B. Segment B encodes the RNA-dependent RNA-polymerase (RdRp) VP1 which is essential for viral RNA replication and is therefore considered an import

Reverse genetics approaches for live-attenuated vaccine development of infectious bursal disease virus.

Infectious bursal disease (IBD), which is caused by infectious bursal disease virus (IBDV) infection, leads to severe immunosuppression in young chickens and results in significant economic losses in the poultry industry. To date, vaccination with live-attenuated vaccine (LAV) is a convenient method to provide effective protection against IBDV infection. Classical attenuated viruses are usually obtained by either passaging virus in cultured cells or natural isolation. However, these empiric attenuation methods, which are time-consuming and not guaranteed, are not reliable for emergent antigeni

Exacerbated Apoptosis of Cells Infected with Infectious Bursal Disease Virus upon Exposure to Interferon Alpha.

Infectious bursal disease virus (IBDV) belongs to the Birnaviridae family and is the etiological agent of a highly contagious and immunosuppressive disease (IBD) that affects domestic chickens (Gallus gallus). IBD or Gumboro disease leads to high rates of morbidity and mortality of infected animals and is responsible for major economic losses to the poultry industry worldwide. IBD is characterized by a massive loss of IgM-bearing B lymphocytes and the destruction of the bursa of Fabricius. The molecular bases of IBDV pathogenicity are still poorly understood; nonetheless, an exacerbated cytoki

Phylogenetic and pathogenic analyses of circulating infectious bursal disease virus strains in China.

Infectious bursal disease (IBD) remains one of the most important immunosuppressive diseases in poultry and has been endemic in China for over four decades. In recent years, outbreaks characterized by markedly reduced mortality have emerged. To investigate the current prevalence and pathogenicity of infectious bursal disease virus (IBDV), strains were isolated from vaccinated poultry farms in central and eastern China, and representative isolates of distinct genotypes were evaluated for pathogenicity. Phylogenetic analysis of segment A (HVR) and segment B (B-marker) classified segment A into e

Evaluation of five circulating strains of variant infectious bursal disease virus (varIBDV) for their immunogenicity as broiler breeder vaccines and protective efficacy in neonatal broiler chicks.

The majority of infectious bursal disease virus (IBDV) strains circulating in the broiler chicken industry in Canada are variant strains (varIBDV). Despite high levels of maternally derived antibodies (MtAb), the circulating varIBDVs can establish infection and cause severe immunosuppression in broiler chicks. The objective of this study was to evaluate circulating varIBDVs as broiler breeder vaccine candidates and investigate their protective efficacy against varIBDV challenge in their progeny chicks. Six groups of breeders (20 females/group) were vaccinated with varIBDV strains, SK09, SK10,

A multimodal evaluation of immunogenicity and field performance of infectious bursal disease vaccines in Ethiopian broilers.

Infectious bursal disease (IBD) continues to undermine poultry health and productivity in Ethiopia. Despite the availability of commercial vaccines, there are raising concerns about vaccine performance, particularly in broiler production systems. This study employed a multi-modal evaluation to compare the immunogenicity and field performance of commercially available IBD vaccines in Ethiopia. The research integrated a controlled in vivo trial assessing vaccine-induced antibody responses and a field-level serological survey of broiler flocks under typical management conditions. Additionally, a

Genotype classification and pathogenicity of infectious bursal disease virus circulating in vaccinated broiler chicken farms.

This study investigated the genotype classification and pathogenicity of infectious bursal disease virus (IBDV) circulating in vaccinated broiler chicken farms in Egypt. A total of 150 samples were collected from 30 vaccinated commercial broiler chicken farms and pooled into 30 working samples. IBDV was tested using reverse transcriptase polymerase chain reaction (RT-PCR) amplification of the hypervariable region of the viral protein 2 (hvVP2) and the VP1 gene 5' extremity. Both RT-PCR fragments were sequenced from six samples, and then the obtained nucleotide sequences were analyzed. The IBDV

Effect of Intermediate Plus Vaccine and vvIBDV on Bursa Secretory Cells and Their Glycoprotein Production.

There are two types of secretory cells in the chicken bursa of Fabricius (BF): (a) interfollicular epithelial cells (IFE), and (b) bursal secretory dendritic cells (BSDC) in the medulla of bursal follicles. Both cells produce secretory granules, and the cells are highly susceptible to IBDV vaccination and infection. Before and during embryonic follicular bud formation, an electron-dense, scarlet-acid fuchsin positive substance emerges in the bursal lumen, the role of which is unknown. In IFE cells, IBDV infection may induce rapid granular discharge, and in several cells, peculiar granule forma

Safety and Efficacy Profile of Live, Intermediate Plus Vaccine Against Infectious Bursal Disease Based on Strain G6.

Infectious bursal disease (IBD) is an acute, highly contagious, immunosuppressive disease of young chickens that causes considerable economic loss in the poultry industry worldwide. Vaccination with live attenuated vaccines is still the most important method used for the control and prevention of IBD in chickens. Here we present the results of in vitro characterization, as well as efficacy and safety testing of a live, intermediate plus vaccine against IBD based on strain G6. Strain characterization confirmed that G6 strain is an intermediate plus strain, showing a high degree of homology with

Gene expression comparison of resistant and susceptible Atlantic salmon fry challenged with Infectious Pancreatic Necrosis virus reveals a marked contrast in immune response.

BACKGROUND: Infectious Pancreatic Necrosis (IPN) is a highly contagious birnavirus disease of farmed salmonid fish, which often causes high levels of morbidity and mortality. A large host genetic component to resistance has been previously described for Atlantic salmon (Salmo salar L.), which mediates high mortality rates in some families and zero mortality in others. However, the molecular and immunological basis for this resistance is not yet fully known. This manuscript describes a global comparison of the gene expression profiles of resistant and susceptible Atlantic salmon fry following c
