Routine Use of Anaerobic Blood Culture Bottles for Specimens Collected from Adults and Children Enhances Microorganism Recovery and Improves Time to Positivity.

The utility of anaerobic blood culture bottles remains controversial, especially for specimens from children. Data are limited on the inclusion of an anaerobic bottle as part of a blood culture "set" when using contemporary blood culture instruments and media. Here, we evaluated the clinical utility of anaerobic blood culture bottles (FN Plus) and aerobic bottles (FA Plus) for the BacT/Alert Virtuo blood culture system (bioMérieux). A total of 158,710 bottles collected between November 2018 and October 2019 were evaluated. There were 6,652 positive anaerobic bottles, of which 384 (5.8%) contai

A protocol for testing the stability of biochemical analytes. Technical document.

Stability of a measurand in a specimen is a function of the property variation over time in specific storage conditions, which can be expressed as a stability equation, and is usually simplified to stability limits (SLs). Stability studies show differences or even inconsistent results due to the lack of standardized experimental designs and heterogeneity of the chosen specifications. Although guidelines for the validation of sample collection tubes have been published recently, the measurand stability evaluation is not addressed. This document provides an easy guideline for the development of

[Our Own Certification System for Education of Blood Sampling Techniques].

We have developed a certification system with objective scoring criteria to evaluate blood sampling techniques. Criteria included not only outcome evaluations, but process-oriented approaches and taxonomy in three fields. After introduction of this system, the time required for blood sampling was reduced and the attitudes of the medical technologists toward patients and blood sampling techniques improved. All members in our labora- tory department were involved in the establishment of this unique system. This system for medical tech- nologists was remarkably useful for education of blood sampl

[The procedures for sampling specimens in pediatrics].

Methods to obtain specimens from children are invasive, and the procedures require skill. Because children are smaller and weaker than adults, the procedures often become difficult. The equipment for the procedures must be chosen based on the physical constitution of the patient. In addition, pediatric patients often do not cooperate with procedures, and so restraint of the patient is extremely important. It is particularly important when obtaining specimens from small neonates and infants, after establishing the minimum quantity of specimens, to choose an appropriate procedure. Because the st

The struggle to detect circulating DNA.

In various diseases, such as cancer, autoimmune disease, sepsis or myocardial infarction, elevated levels of circulating DNA can be measured. However, its predictive value is under debate. Circulating DNA in plasma is protein-bound (nucleosomal) DNA. Quantification of circulating DNA can be performed by real-time quantitative PCR or immunological methods such as ELISA. The diagnostic value of both methods can be impaired by inappropriate handling of the samples. Assessment of circulating DNA in patients admitted to the intensive care unit offers a tool for predicting morbidity and mortality.

Single-site sampling strategy versus multisite sampling strategy in blood culture collection within the hospital setting: A systematic review.

BACKGROUND: Many global infectious disease and sepsis guidelines suggest a multisite sampling (MSS) strategy of blood cultures to diagnose bloodstream infections, however, single-site sampling (SSS) may be a viable alternative. This systematic review aims to identify the current state of literature comparing SSS to MSS. METHODS: MEDLINE, CINAHL, Pubmed, Web of Science, Scopus, and Google Scholar were searched from inception to August 2023. Studies comparing SSS and MSS in hospital settings among adult patients were included. Key outcomes being compared were bacteremia detection, contamination

Blood sampling technique in penguins (Sphenisciformes).

The blood vessels in the flippers of members of the Sphenisciformes were studied with the aim of locating a suitable anatomical site for the collection of blood from this order. The optimal site was found to be the brachial vein on the ventral surface of the flipper. Blood samples were taken using this method from five different species.

Low-volume collection devices for longitudinal biomarker monitoring in patient-centric sampling settings.

Advances in analytical techniques now enable biomarker detection from sample volumes as small as one microliter, reducing the need for large-volume specimen collection. This capability has supported the development of low-volume blood collection devices that allow frequent, longitudinal sampling outside traditional healthcare facilities, promoting decentralized and patient-centered sampling (PCS). The development of user-friendly, at-home sampling devices with reduced invasiveness and minimized sampling discomfort further enhances participant recruitment and retention in long-term longitudinal

Lack of fermentation in antemortem blood samples stored unstoppered in various locations.

A common defense challenge when antemortem blood ethanol results are presented at trial is the assertion that ethanol was formed in the blood tube after the blood draw through fermentation of the blood glucose by Candida albicans (C. Albicans). In contrast, decades of research into the stability of ethanol in antemortem blood collected for forensic purposes have consistently shown that any analytically significant change in ethanol concentration is a decrease and initially, ethanol-negative blood remains ethanol-negative with storage. For there to be any possibility of fermentation to occur by

Decreasing contamination rates in blood cultures: A plan-do-study-act approach.

Background The gold standard investigation to confirm a diagnosis of sepsis is blood culture. Contamination of blood culture with skin commensals contributes to increased health-care costs, unnecessary antibiotic treatment, and prolonged hospital stay. We did a study to identify the causes of contamination of blood culture samples and methods to decrease blood culture contamination rates. Methods This clinical audit study was a before-and-after, interventional study. Fish bone analysis was done to identify the possible issues responsible for blood culture contamination, following which interve

Causes, consequences and management of sample hemolysis in the clinical laboratory.

Preanalytical hemolysis of blood samples is a common problem in medical practice, especially in emergency departments. Several potential influences on sample hemolysis have been investigated, including sampling techniques, centrifugation and sample transport. In particular, the use of intravenous catheters and the vacuum sampling technique have often been demonstrated to provoke hemolysis. Other factors playing a role include the use of inappropriate puncture sites, complicated blood sampling, prolonged tourniquet application, underfilling of tubes and excessive shaking of specimens. Training

Importance of blood volume cultured in the detection of bacteremia.

The influence of the volume of blood cultured on the rate of detection of bacteremia was evaluated in a routine 12-tube blood culture system using 1693 samples from 1502 patients. Blood samples were drawn simultaneously into two transport tubes. The blood volume cultured was the only varying parameter. Generally, 17% more cultures with clinically significant microorganisms (both Enterobacteriaceae and gram-positive cocci) were found when blood from two instead of one tube was used (in most cases comparing 13-16 ml of blood with 6.5-8 ml). Of the most prevalent species, the maximum average extr

[Scribbling and doodling: social representations by children expressed in drawings, regarding the experience of blood sampling].

The scope of this article was to identify the social representations of children in a municipal public school in Rio de Janeiro, with respect to the blood sampling procedure and to discuss the social representations that emerge, in the light of the performance of the family health team in carrying out this procedure. Moscovisci's method/theory of analysis of social representations based on a qualitative approach was used. For data collection, the projective technique of the esthetic production scene and the student's speech was used. The results were grouped into four categories: 1) structural

Comparison of self-sampling blood collection for N-glycosylation analysis.

OBJECTIVE: Self-sampling of capillary blood provides easier sample collection, handling, and shipping compared to more invasive blood sampling via venepuncture. Recently, other means of capillary blood collection were introduced to the market, such as Neoteryx sticks and Noviplex cards. We tested the comparability of these two self-sampling methods, alongside dried blood spots (DBS), with plasma acquired from venepunctured blood in N-glycoprofiling of total proteins. We have also tested the intra-day repeatability of the three mentioned self-sampling methods. Capillary blood collection with Ne

Statistical comparisons of 0.08 g/100 ml fortified blood alcohol samples from 6-ml and 10-ml gray-top tubes.

This experiment supplements the study, "Statistical comparisons of blood alcohol samples from 6-ml and 10-ml grey-top tubes". The initial study analyzed fortified samples for blood alcohol concentration (BAC) using two sizes of gray-top tubes: a 10-ml tube containing a nominal 1% sodium fluoride (NaF), a preservative, and a 6-ml tube containing 0.25% NaF, using the variables of time, storage temperature, fill volume, and concentration. From that study, paired t-tests determined no difference in BAC from data sets of the two tube sizes, however, Analysis of Variance, or ANOVA, testing determine
