Restricted Arp3 expression in the testis prevents blood-testis barrier disruption during junction restructuring at spermatogenesis.

In epithelia, a primary damage of tight junctions (TJ) always leads to a secondary disruption of adherens junction (AJ), and vice versa. This response, if occurring in the testis, would disrupt spermatogenesis because the blood-testis barrier (BTB) must remain intact during the transit of spermatids in the seminiferous epithelium, which is associated with extensive apical ectoplasmic specialization (apical ES, a testis-specific AJ type) restructuring. As such, apical ES restructuring accompanied with the transit of developing spermatids during spermiogenesis must be segregated from the BTB to

The blood-testis and blood-epididymis barriers are more than just their tight junctions.

The terms blood-testis barrier (BTB) or blood-epididymis barrier (BEB), are often described as Sertoli cell-Sertoli cell tight junctions (TJs) or TJs between the epithelial cells in the epididymis, respectively. However, in reality, the BTB and BEB are much more complex than just the TJ. The focus of this minireview is to remind readers that the complete BTB and BEB are comprised of three components: anatomical, physiological, and immunological. The TJs form the anatomical (physical) barrier that restricts passage of molecules and cells from entering or exiting the lumen. The physiological bar

N-wasp is required for structural integrity of the blood-testis barrier.

During spermatogenesis, the blood-testis barrier (BTB) segregates the adluminal (apical) and basal compartments in the seminiferous epithelium, thereby creating a privileged adluminal environment that allows post-meiotic spermatid development to proceed without interference of the host immune system. A key feature of the BTB is its continuous remodeling within the Sertoli cells, the major somatic component of the seminiferous epithelium. This remodeling is necessary to allow the transport of germ cells towards the seminiferous tubule interior, while maintaining intact barrier properties. Here

The blood-testis barrier in men with varicocele: a lanthanum tracer study.

Testicular biopsy specimens were obtained from 28 otherwise healthy males (22 to 37 years old) with diagnosed varicocele. Tissue was processed for ultrastructural and intercellular tracer studies using lanthanum nitrate. In all cases Sertoli-Sertoli junctional complexes appeared structurally intact and functionally normal on the basis of their ability to occlude lanthanum from the adluminal testicular compartment. Our results indicate that, even in the presence of severe dissolution of the adluminal testicular compartment as evidenced in varicocele, both the basal compartment and the blood-tes

Mechanisms of Polystyrene Microplastics in Multipathway Disruption of the Blood-Testis Barrier.

BACKGROUND: Polystyrene microplastics (PS-MPs) represent an emerging environmental pollutant, yet their potential threat to male reproductive health, specifically the integrity of the blood-testicular barrier (BTB), remains incompletely understood. OBJECTIVE: This study aimed to investigate the toxicological effects and underlying mechanisms of PS-MPs exposure on testicular structure and BTB function in a mouse model. METHODS: Four-week-old male ICR mice were exposed to PS-MPs at concentrations of 0, 500 µg/L, 5 mg/L, and 50 mg/L via drinking water for up to 5 weeks. Animals (n = 10 per grou

Biomimetic Nanotechnology Overcoming the Blood-Testis Barrier for Testicular Protection in Chemotherapy.

Epidural electrical stimulation (EES) has emerged as a promising treatment for spinal cord injury (SCI). However, the therapeutic potential of EES in functional recovery following incomplete SCI remains limited, with few studies of a large sample size. This study included 11 patients who received EES combined with physical therapy (PT) and 10 who received only PT. Follow-ups were conducted pre-surgery, post-surgery, and at 19 to 25 months postoperatively. After the surgery, patients in the EES + PT group showed significant improvements in sensory function (P < 0.001) and muscle spasticity (P <

HDAC3 promotes Sertoli cell maturation and maintains the blood-testis barrier dynamics.

Germ cell development depends on the capacity of somatic Sertoli cells to undergo differentiation into a mature state and establish a germ cell-specific blood-testis barrier (BTB). The BTB structure confers an immunological barrier for meiotic and postmeiotic germ cells, and its dynamic permeability facilitates a transient movement of preleptotene spermatocytes through BTB to enter meiosis. However, the regulatory factors involved in Sertoli cell maturation and how BTB dynamics coordinate germ cell development remain unclear. Here, we found a histone deacetylase HDAC3 abundantly expresses in S

Fluorinated hyperbranched polymers mediate spermatogonia-directed transport across the blood-testis barrier.

Delivering therapeutics into seminiferous tubules remains a formidable challenge, limiting the treatment of reproductive and testis-related diseases, because the blood-testis barrier (BTB) restricts access to the luminal compartment. Here we report well-defined unimolecular fluorinated hyperbranched (Fhb) nanoparticles that can reach seminiferous tubules after systemic administration. Fhb polymers with tunable fluorine contents were synthesized via a B3 core-mediated chain-growth copper-catalyzed azide-alkyne cycloaddition polymerization, followed by terminal-group modification. Assessment of microplastic toxicity on blood-testis barrier using 3D cell spheroids.

Industrial production generates large quantities of microplastics (MPs), which can significantly impact ecosystems and pose potential risks to human health, both directly and indirectly, and the mechanisms and potential toxicity of microplastics affecting the reproductive system remain unclear. In this study, we investigated the cytotoxicity of microplastics on the blood-testis barrier using 3D Sertoli cell spheroids (TM4S) exposed to artificially manufactured primary polystyrene microplastics (MPS). To investigate these effects, we explored the invasiveness of MPS in relation to microplastic

Melatonin Regulates Clusterin to Reconstruct the Blood-Testis Barrier Disrupted by Atrazine.

Atrazine (ATZ), a commonly used triazine herbicide, is known to impair male reproductive function. Melatonin (MLT) is an endogenous hormone synthesized by the pineal gland. Its lipophilic nature enables it to freely penetrate the blood-testis barrier (BTB) and exert direct effects. However, it remains unclear whether MLT can prevent ATZ-induced impairment of BTB function. Pubertal male mice were randomly assigned to four groups and treated via gavage for 21 days: control group (Con), 5 mg/kg melatonin group (MLT), 170 mg/kg atrazine group (ATZ), and cotreatment group (ATZ+MLT). Furthermore, BT

HIV1-Nef perturbs the integrity of blood testis barrier in rat model.

The blood-testis barrier is a specialized feature within the mammalian testis, located in close proximity to the basement membrane of seminiferous tubules. This barrier serves to divide the seminiferous epithelium into distinct basal and adluminal (apical) compartments. The selectivity of the BTB to foreign particles makes it a safe haven for the virus, and the high affinity of HIV for testis might lead to the vertical transmission of the virus. In the present study, recombinant HIV1-Nef (rNef) protein was injected intravenously to examine the effect of rNef on BTB. SD male rats received 250 µ

LncRNA SNHG5/IGF2BP1/Occludin axis regulates Nd2O3 induced blood-testis barrier disruption.

Neodymium oxide (Nd2O3) is a rare earth element that can lead to various type of tissue and organ damage with prolonged exposure. The long noncoding RNA small nucleolar ribonucleic acid host gene 5 (lncRNA SNHG5) plays a role in disease progressiong. However, its connection with Nd2O3 induced reproductive harm in males has not been thoroughly investigated. Our research discovered that exposure to Nd2O3 increases the expression of SNHG5 in the testes of mice, which in turn binds directly to and reduces in the protein levels of insulin like growth factor 2 mRNA-binding protein 1 (IGF2BP1) both i

Granulocyte macrophage-colony stimulating factor crosses the blood-testis barrier in mice.

Granulocyte macrophage-colony stimulating factor (GM-CSF) has hormone-like effects on female reproductive systems. Recent evidence has suggested that GM-CSF also might be important to male testicular function. It is possible, however, that most sources of GM-CSF might not be able to reach the testis, since testes are shielded from contact with the general circulation by vascular and Sertoli cell barriers, which together comprise the blood-testis barrier (BTB). We used a sensitive in vivo method to determine whether blood-borne GM-CSF crossed the BTB in mice. 125I-GM-CSF was found to cross the

Testis and blood-testis barrier in Covid-19 infestation: role of angiotensin-converting enzyme 2 in male infertility.

Severe acute respiratory syndrome coronavirus 2 (SARS- CoV-2) that causes COVID-19 infections penetrates body cells by binding to angiotensin-converting enzyme-2 (ACE2) receptors. Evidence shows that SARS-CoV-2 can also affect the urogenital tract. Hence, it should be given serious attention when treating COVID-19-infected male patients of reproductive age group. Other viruses like HIV, mumps, papilloma and Epstein-Barr can induce viral orchitis, germ cell apoptosis, inflammation and germ cell destruction with attending infertility and tumors. The blood-testis barrier (BTB) and blood-epididymi

The blood-testis barrier disruption is a prerequisite for toxicant-induced peritubular macrophage increases in the testis of peripubertal rats.

Peritubular macrophages (PTMφ) are predominantly localized near spermatogonial stem cells in the testis. We previously revealed that exposure of peripubertal male Fischer rats to mono-(2-ethylhexyl) phthalate (MEHP) leads to increased PTMφs in the testis. The mechanisms that trigger increases in PTMφs in the testis are poorly understood. However, MEHP exposure is known to both induce spermatocyte apoptosis and to perturb the blood-testis barrier (BTB). This study aims to elucidate the association between the disruption of BTB and the increases of PTMφs in the testis by comparing the effects ob

GATA4 Regulates Blood-Testis Barrier Function and Lactate Metabolism in Mouse Sertoli Cells.

Conditional deletion of Gata4 in Sertoli cells (SCs) of adult mice has been shown to increase permeability of the blood-testis barrier (BTB) and disrupt spermatogenesis. To gain insight into the molecular underpinnings of these phenotypic abnormalities, we assessed the impact of Gata4 gene silencing in cell culture models. Microarray hybridization identified genes dysregulated by siRNA-mediated inhibition of Gata4 in TM4 cells, an immortalized mouse SC line. Differentially expressed genes were validated by quantitative RT-PCR analysis of primary cultures of Gata4(flox/flox) mouse SCs that had

Spermatogenesis and blood-testis barrier in rats after long-term Vitamin A deprivation.

It has been established that experimental avitaminosis A in rats results in a 'Sertoli cell-only situation' after about 10 weeks, and that replacing the vitamin immediately reinitiates spermatogenesis. The present study deals with testicular recovery after prolonged deprivation (up to 19 weeks). The Sertoli cell-only situation reached under this condition was thought to be refractory to Vitamin A replacement. However, spermatogenesis did reinitiate about 11 weeks after vitamin restoration, although in an atypical manner. The blood-testis barrier, normally assembled when spermatocytes reaches t

Mono-(2-ethylhexyl) phthalate reversibly disrupts the blood-testis barrier in pubertal rats.

The blood-testis barrier (BTB) is constituted by tight junctions between adjacent Sertoli cells (SCs) that create a specialized adluminal microenvironment to foster the development of spermatocytes and spermatids. The BTB is a well-studied target of numerous environmental toxicants, including di-(2-ethylhexyl) phthalate (DEHP), a compound widely used in various consumer products. Mono-(2-ethylhexyl) phthalate (MEHP) is the active toxic metabolite of DEHP that has long been recognized in postnatal rodents to disrupt SC function. This study evaluates the impact of MEHP on the integrity of the BT

Functional characteristics of 3'-azido-3'-deoxythymidine transport at the blood-testis barrier.

3'-Azido-3'-deoxythymidine (AZT), an antiretroviral drug, is often adopted in the therapy for human immunodeficiency virus (HIV) infection, and the characteristics of AZT transport at the blood-testis barrier (BTB) were investigated in this study. In the integration plot analysis that evaluates the transport activity in vivo, the apparent influx clearance of [3H]AZT was significantly greater than that of [14C]D-mannitol, a non-permeable paracellular transport marker. In the uptake study in vitro with TM4 cells derived from mouse Sertoli cells, [3H]AZT uptake exhibited a time- and concentration
