Synergy between Cu-NPs and fungicides against Botrytis cinerea.

Combating drug-resistance is a daunting task, especially due to the shortage of available drug alternatives with multisite modes of action. In this study, the potential of copper nanoparticles (Cu-NPs) to suppress 15 Botrytis cinerea isolates, which are sensitive or resistant to fungicides, alone or in combination with conventional fungicides, was tested in vitro and in vivo. Sensitivity screening in vitro revealed two fungicide resistance phenotypes, resulting from target site mutations. DNA sequencing revealed three B. cinerea isolates highly resistant to benzimidazoles (BEN-R), thiophanare

Evidence for viable but nonculturable yeasts in botrytis-affected wine.

AIMS: In Botrytis-affected wine, high concentrations of SO2 are added to stop the alcoholic fermentation and to stabilize the wine. During maturing in barrels or bottle-ageing, an unwanted refermentation can sometimes occur. However, results of the usual plate count of wine samples at the beginning of maturing suggest wine microbial stability. The aim of this study was to investigate the mode of yeasts survival after the addition of SO2 and to identify surviving yeasts. METHODS AND RESULTS: Using direct epifluorescence technique, we observed the behaviour of cells after SO2 addition and compa

Botrytis virus X ORF2 suppresses RNA silencing in a trigger-restricted manner and is associated with altered Dicer-like gene expression in Botrytis cinerea.

RNA interference (RNAi) is a central antiviral defence mechanism in fungi, yet relatively few mycoviral suppressors of RNA silencing (VSRs) have been functionally characterized, particularly in phytopathogenic hosts. Botrytis virus X (BVX), a positive-sense RNA virus in the family Alphaflexiviridae, infects Botrytis cinerea and encodes five predicted ORFs, most of which have unknown functions. Here, we screened BVX ORFs 2-5 for RNA silencing suppressor activity using complementary GFP-based assays in Nicotiana benthamiana and examined the leading candidate in the fungal host B. cinerea. BVX OR

Biocontrol Efficacy of Pseudomonas Consortia Against Botrytis Blight in Petunias.

Botrytis cinerea, a fungal pathogen causing Botrytis blight, significantly impacts greenhouse crop management owing to its broad host range and infection capabilities at various growth stages. Traditional control methods, primarily reliant on fungicides, are challenged by environmental concerns and the rise of fungicide-resistant strains. This study investigates the use of beneficial Pseudomonas bacteria as a sustainable alternative. We hypothesized that specific Pseudomonas consortia could provide more effective biocontrol of B. cinerea than individual strains. Our research investigated five

Mechanisms of Action of Microbial Biocontrol Agents against Botrytis cinerea.

Botrytis cinerea is a phytopathogenic fungus responsible for economic losses from USD 10 to 100 billion worldwide. It affects more than 1400 plant species, thus becoming one of the main threats to the agriculture systems. The application of fungicides has for years been an efficient way to control this disease. However, fungicides have negative environmental consequences that have changed popular opinion and clarified the need for more sustainable solutions. Biopesticides are products formulated based on microorganisms (bacteria or fungi) with antifungal activity through various mechanisms. Th

Deep sequencing of mycovirus-derived small RNAs from Botrytis species.

RNA silencing is an ancient regulatory mechanism operating in all eukaryotic cells. In fungi, it was first discovered in Neurospora crassa, although its potential as a defence mechanism against mycoviruses was first reported in Cryphonectria parasitica and, later, in several fungal species. There is little evidence of the antiviral potential of RNA silencing in the phytopathogenic species of the fungal genus Botrytis. Moreover, little is known about the RNA silencing components in these fungi, although the analysis of public genome databases identified two Dicer-like genes in B. cinerea, as in

Protocol for the Extraction of High-Quality DNA from Botrytis cinerea.

Using Botrytis cinerea (ATCC 58025) as the material, genomic DNA was extracted using the SDS-CTAB method after liquid nitrogen freezing and extracted multiple times with organic reagents. Then the precipitate was washed with ethanol and dissolved in TE buffer. The cultured genomic DNA is high-yield, easy to extract, and has good purity, making it suitable for extracting from multiple samples at once. After experiments, the genomic DNA obtained by this method can be used for most basic molecular biology experiments such as PCR and DNA enzyme digestion.

The Defense Mechanism of PpCAD4 in Physcomitrium patens Against Botrytis cinerea.

The existence of lignin in Physcomitrium patens has been controversial. However, cinnamyl alcohol dehydrogenase (CAD), the key enzyme in monolignol biosynthesis, has been identified with four gene members in P. patens. Despite the roles of PpCAD1 in moss architecture being proven in a previous study, the functions and molecular mechanisms of PpCAD4 remain largely unexplored in early terrestrial plants. This study aims to unravel this mystery via a comprehensive analysis of the transcriptome and metabolome of PpCAD4-overexpression (OE) lines compared with wild type (WT) under Botrytis cinerea t

SmMYC2 positively regulates the resistance to Botrytis cinerea in Salvia miltiorrhiza.

This study unravels the function of SmMYC2 regulating resistance to B. cinerea in S. miltiorrhiza. Salvia miltiorrhiza is a traditional medicinal herb and commonly used to treat cardiovascular and cerebrovascular diseases. Gray mold caused by Botrytis cinerea poses a significant threat to production of many medicinal plants. The key transcription factor SmMYC2 has been found to contribute to promoting the biosynthesis of tanshinones or phenolic acids, and also mediated salt resistance in S. miltiorrhiza. However, limited information underlying its role of resistance to B. cinerea is available.

Hormetic Effects of Mixtures of Carbendazim and Iprodione on the Virulence of Botrytis cinerea.

Hormetic effects of fungicides on mycelial growth and virulence of plant pathogenic fungi have been reported, but the effects of fungicide mixtures on virulence hormesis of plant pathogens remain to be investigated. In this study, hormetic effects of mixtures of carbendazim and iprodione on the virulence of two carbendazim-resistant isolates of Botrytis cinerea were determined. Spraying carbendazim alone at 3 to 800 μg/ml exhibited hormetic effects on virulence to cucumber leaves, and carbendazim at 10 μg/ml had the maximum stimulation of 16.7% for isolate HBtom451. Spraying iprodione alone at

Characterization of iprodione resistance in Botrytis cinerea from strawberry and blackberry.

Gray mold, caused by the fungal pathogen Botrytis cinerea, is one of the most destructive diseases of strawberry. Control of the disease in commercial fields is largely dependent on the application of fungicides, including the dicarboximide iprodione. Single-spore isolates were collected from strawberry fields in Florida, North Carolina, and South Carolina and subjected to an assay using conidial germination that distinguished sensitive (S) isolates from isolates with various levels of resistance to iprodione. Of the 245 isolates, 1 was highly resistant (HR), 5 were moderately resistant (MR),

Assays of Antifungal Activity of an Antimicrobial Peptide Against Botrytis cinerea.

Antimicrobial peptides (AMPs) represent the first line of host immune defense against a broad spectrum of microorganisms. Whether naturally occurring or synthetically designed, these small peptides can potently inhibit microbial growth and remain stable under diverse conditions. Most AMPs act by directly disrupting fungal or bacterial membranes. In this study, we identified a glycine-rich peptide with strong antifungal activity against Botrytis cinerea. We further investigated its mode of action (MOA) and demonstrated its preventive antifungal activity in plants. Our work focuses primarily on

Genome-wide transcriptomic analysis of the response of Botrytis cinerea to wuyiencin.

Grey mould is caused by the ascomycetes Botrytis cinerea in a range of crop hosts. As a biological control agent, the nucleoside antibiotic wuyiencin has been industrially produced and widely used as an effective fungicide. To elucidate the effects of wuyiencin on the transcriptional regulation in B. cinerea, we, for the first time, report a genome-wide transcriptomic analysis of B. cinerea treated with wuyiencin. 2067 genes were differentially expressed, of them, 886 and 1181 genes were significantly upregulated and downregulated, respectively. Functional categorization indicated that transcr

Bacillus species compatibility enhances VOC-mediated systemic resistance against Botrytis cinerea.

Botrytis cinerea, the causal agent of gray mold, poses a significant threat to global tomato production. This study explores the role of volatile organic compounds (VOCs) from three Bacillus strains in eliciting systemic resistance in tomato plants against gray mold. We tested VOCs from individual strains (LNXM12, GBAC46, LLTC93) as well as from both compatible and incompatible combinations of these bacteria. The result showed that compatible strains exhibited mutual growth, while incompatible strains displayed growth antagonism. Our findings further revealed that VOCs from both compatible and

Enhanced Arabidopsis disease resistance against Botrytis cinerea induced by sulfur dioxide.

Sulfur dioxide (SO2) is a common air pollutant that has complex impacts on plants. The effect of prior exposure to 30mgm-3 SO2 on defence against Botrytis cinerea (B. cinerea) in Arabidopsis thaliana and the possible mechanisms of action were investigated. The results indicated that pre-exposure to 30mgm-3 SO2 resulted in significantly enhanced resistance to B. cinerea infection. SO2 pre-treatment significantly enhanced the activities of defence-related enzymes including phenylalanine ammonia-lyase (PAL), polyphenol oxidase (PPO), β-1,3-glucanase (BGL) and chitinase (CHI). Comparative Transcriptomic Analysis of Potato and Tomato Responses to Botrytis cinerea Infection.

Botrytis cinerea causes gray mold disease in numerous plant species, including economically important solanaceous crops. To investigate the molecular interaction between B. cinerea and Solanum tuberosum, we performed dual RNA-seq analyses of infected potato leaves at 24 and 72 h post inoculation (hpi). A total of 5932 potato genes were differentially expressed during infection, including genes associated with pathogen perception, hormone signaling, transcriptional regulation, and defense responses to biotic stress. In parallel, 2162 fungal genes showed differential expression, including genes

The effects and potential mechanisms of statins on growth, development and pathogenicity in Botrytis cinerea.

Statins that target HMG-CoA reductase (HMGR) in the mevalonate pathway have been used to inhibit cholesterol biosynthesis. Therefore, they have the potential as inhibitors targeting the sterol biosynthesis pathway of plant pathogenic fungi. Currently, their activity against Botrytis cinerea is unknown. In this study, we determined the effects of four statins on both development and pathogenicity of B. cinerea. The interaction modes between statins and HMGR in B. cinerea were characterized through qRT-PCR and molecular docking analysis. Fluvastatin, atorvastatin, simvastatin and lovastatin stro

Effects of SHAM on the Sensitivity of Sclerotinia sclerotiorum and Botrytis cinerea to QoI Fungicides.

It is a common practice to add salicylhydroxamic acid (SHAM) into artificial medium in the in vitro sensitivity assay of fungal phytopathogens to the quinone outside inhibitor (QoI) fungicides. The rationale for adding SHAM is to inhibit fungal alternative oxidase, which is presumed to be inhibited by secondary metabolites of plants. Therefore, the ideal characteristics of SHAM should be almost nontoxic to phytopathogens and have no significant effect on control efficacy of fungicides. However, this study showed that the average effective concentration for 50% inhibition (EC50) of mycelial gro
