Antimicrobial Resistance in Members of the Bacterial Bovine Respiratory Disease Complex Isolated from Lung Tissue of Cattle Mortalities Managed with or without the Use of Antimicrobials.

Over a two-year period, Mannheimia haemolytica (MH; n = 113), Pasteurella multocida (PM; n = 47), Histophilus somni (HS; n = 41) and Mycoplasma bovis (MB; n = 227) were isolated from bovine lung tissue at necropsy from cattle raised conventionally (CON, n = 29 feedlots) or without antimicrobials [natural (NAT), n = 2 feedlots]. Excluding MB, isolates were assayed by PCR to detect the presence of 13 antimicrobial resistance (AMR) genes and five core genes associated with integrative and conjugative elements (ICEs). Antimicrobial susceptibility phenotypes and minimum inhibitory concentrations (M

Development of a multiplex real-time PCR assay using two thermocycling platforms for detection of major bacterial pathogens associated with bovine respiratory disease complex from clinical samples.

Bovine respiratory disease complex (BRDC) is one of the most significant diseases of cattle. Bacterial pathogens involved in BRDC include Mannheimia haemolytica, Mycoplasma bovis, Histophilus somni, and Pasteurella multocida. We developed and evaluated a multiplexed real-time hydrolysis probe (rtPCR) assay using block-based Peltier and rotary-based thermocycling on lung tissue, nasal swabs, and deep nasopharyngeal swabs. The rtPCR results were compared to culture or a gel-based M. bovis PCR using statistical analysis to determine optimum quantification cycle (Cq) cutoffs to maximize agreement.…

The calf holobiont under challenge: longitudinal microbiota-pathogen dynamics and respiratory health.

The bovine respiratory disease complex (BRDC) is a major health and welfare challenge driven by multifactorial interactions among pathogens, the host, the microbiota, and environmental pressures, reflecting the holobiont nature of the calf. Many implicated bacterial pathogens are also respiratory tract commensals, complicating diagnosis, risk assessment, and prevention. Although the potential association of the respiratory and gut microbiotas in BRDC is only beginning to be recognized, their integrated effects have not yet been investigated. Here, we dynamically followed the microbiota, pathog…

Evaluation of reticulorumen temperature boluses for the diagnosis of subclinical cases of bovine respiratory disease in feedlot cattle.

Bovine respiratory disease (BRD) is the most important and costly health issue of the feedlot industry worldwide. Remote monitoring of reticulorumen temperature has been suggested as a potential tool to improve the diagnostic accuracy of BRD. The present study aimed to evaluate 1) the difference and degree of reticulorumen hyperthermia episodes between healthy and subclinical BRD feedlot steers, and 2) determine the correlation between reticulorumen hyperthermia and lung pathology, performance, and carcass traits. Mixed-breed feedlot steers (n = 148) with a mean arrival weight of 321 ± 3.34 kg

Viral Infections in Stocker Cattle and Their Implications for Bovine Respiratory Disease.

Bovine respiratory disease complex (BRDC) remains a leading cause of morbidity and economic loss in stocker cattle, particularly during the transition from weaning to feedlot placement. Virus-induced changes promote microbiome imbalance and facilitate secondary bacterial infections, leading to bronchopneumonia. The high transmissibility of these viruses, with basic reproduction numbers ranging from approximately 3 to 36, contributes to rapid spread under feedlot conditions. This article reviews the epidemiology, pathogenesis, transmission, diagnosis, and control of major viral pathogens associ…

Co-infection of epithelial cells established from the upper and lower bovine respiratory tract with bovine respiratory syncytial virus and bacteria.

Bovine respiratory disease complex is a major disease affecting the global cattle industry. Multiple infections by viruses and bacteria increase disease severity. Previously, we reported that bovine respiratory syncytial virus (BRSV) infection increases adherence of Pasteurella multocida to human respiratory and bovine kidney epithelial cells. To examine the interaction between the virus and bacteria in bovine respiratory cells, we generated respiratory epithelial cell lines from bovine trachea (bTEC), bronchus (bBEC), and lung (bLEC). Although all established cell lines were infected by BRSV …

Bacterial Respiratory Pathogens of Beef Stocker Cattle.

Bovine respiratory disese (BRD) is the leading cause of morbidity and mortlaity in stocker cattle, a high-risk sector of the beef cattle industry, characterized by stress, commingling, and limited or unknown prior health management. Key bacterial pathogens, including Mannheimia haemolytica, Pateurella multocida, Histophilus somni, and Mycoplasmopsis bovis, act as opportunists when host defenses are compromised. These microbes employ diverse virulence mechanisms, including toxins, adhesins, surface proteins, and immune evasion strategies to colonize the lower respiratory tract and cause disease

Behavior assessment and applications for BRD diagnosis: preweaned dairy calves.

Bovine respiratory disease (BRD) is an important disease in dairy calves due to its long-lasting effects. Early identification results in better outcomes for the animal, but producers struggle to identify all calves with BRD. Sickness behavior, or the behavioral changes that accompany illness, has been investigated for its usefulness as a disease detection tool. Behavioral changes associated with BRD include decreased milk intake and drinking speed, depressed attitude, and less likelihood of approaching a novel object or stationary human. Behavioral measurements are useful, as they can be coll

Infection of calves with in-vivo passaged bovine parainfluenza-3 virus, alone or in combination with bovine respiratory syncytial virus and bovine coronavirus.

Bovine respiratory disease complex is etiologically complex and usually involves co-infection by several agents, including bovine parainfluenza virus-3 (BPIV-3), bovine respiratory syncytial virus (BRSV), and bovine coronavirus (BCoV). Traditionally, vaccines have been tested in seronegative calves infected with a single in vitro-passaged agent, often with little disease, resulting in unvaccinated subjects. To overcome the potential problem of attenuation coincident with in vitro culture of the viruses, cocktails of field isolates of BPIV-3s and BCoVs were passaged in the lungs of neonatal col…

Hematological and immunological responses to naturally occurring bovine respiratory disease in newly received beef calves in a commercial stocker farm.

The objective was to determine temporal changes in hematological and immune parameters in response to naturally occurring bovine respiratory disease (BRD) in commercially managed stocker calves. Forty newly weaned beef steers purchased from auction markets were housed at a commercial stocker operation in Crossville, TN. Blood samples, rectal temperature, and thoracic ultrasonography (TUS; 1: normal to 3: severe) were collected on days 0, 7, 14, and 21. Castration status (FC: freshly castrated; PC: previously castrated) was determined on arrival based on presence of a fresh castration site at t

Growth performance and hematological changes of weaned beef calves diagnosed with respiratory disease using respiratory scoring and thoracic ultrasonography.

This study investigated 1) the effect of clinical bovine respiratory disease (BRD) and associated lung consolidations on growth performance and hematological profiles of recently weaned beef calves and 2) the relationship between clinical respiratory signs and lung consolidation detected by thoracic ultrasonography (TUS). One hundred and fifty-three weaned beef calves (209 days old [SD: 35.8] and 306 kg [SD: 26.3], at arrival) purchased and transported from auction markets were accommodated indoors in concrete slatted floor pens. Calves were weighed weekly from arrival until day 28 and on day

Behavior assessment and applications for BRD diagnosis: beef.

Assessment of behavior is a longstanding strategy to assist the diagnosis of clinical bovine respiratory disease (BRD) in beef cattle. Cattle with systemic inflammation caused by infectious pathogen(s) display predictable behavioral adaptations compared to healthy cohorts. Behavioral alterations in BRD-affected cattle include lethargy, social isolation, and anorexia. However, behavior assessment to support BRD case definition in the production setting is challenging because: (1) other bovine diseases cause behavior alterations similar to BRD; (2) cattle have inherent prey instinct to disguise

The role of the veterinary diagnostic lab in the management of BRD.

Veterinary diagnostic labs (VDLs) are important service agencies providing essential diagnostic testing for a wide variety of domestic animal species as well as wildlife. They serve key roles in disease monitoring and diagnosis as well as surveillance for diseases of consequence. Of the many roles VDLs serve, one is being a member of the larger team of professionals dealing with the management of the bovine respiratory disease (BRD) complex. VDLs provide a number of services related to the management of BRD. These include disease outbreak investigation, abnormal morbidity characterization, rou

Bovine respiratory coronavirus enhances bacterial adherence by upregulating expression of cellular receptors on bovine respiratory epithelial cells.

Bovine coronavirus (BCoV) is one of the agents causing bovine respiratory disease complex (BRDC), with single infection tending to be mild to moderate; the probability of developing pneumonia in BRDC may be affected by viral and bacterial combinations. Previously, we reported that bovine respiratory syncytial virus (BRSV) infection enhances adherence of Pasteurella multocida (PM) to cells derived from the bovine lower respiratory tract but that BRSV infection in cells derived from the upper respiratory tract reduces PM adherence. In this study, we sought to clarify whether the modulation of ba…

Transcriptomic analysis of naturally occurring bovine respiratory disease in Australian feedlot cattle identifies bovine viral diarrhoea virus-induced immunosuppression and Mollicutes as the dominant drivers of lung inflammation.

Bovine respiratory disease (BRD) is the most economically significant syndrome of feedlot cattle worldwide, yet the relative contributions of individual BRD pathogens to host transcriptional responses remain poorly understood. We performed pathogen-stratified transcriptomic analysis of lung tissue obtained from naturally occurring BRD cases (n = 22) and non-BRD controls (n = 9) in feedlot cattle. BRD-affected tissue showed significant immune activation with 3148 differentially expressed genes (DEGs; 18.7% of the 16,873 genes tested), of which 70.4% were upregulated. Pathogen-stratified analysi
