PREMISE OF THE STUDY: We developed microsatellite markers to investigate genetic diversity within and among populations of Capsella rubella and Capsella bursa-pastoris and between these two species.
METHODS AND RESULTS: Fourteen polymorphic microsatellite loci were identified in the two species and one more polymorphic microsatellite locus only in C. rubella. Samples from different European localities were genotyped. Up to six alleles per locus were observed in C. rubella, and up to 22 alleles per locus in C. bursa-pastoris. Observed heterozygosities were low, indicating high selfing rates, e

Evolution of chromosome complements can be resolved by genome sequencing, comparative genetic mapping, and comparative chromosome painting. Previously, comparison of genetic maps and gene-based phylogenies suggested that the karyotypes of Arabidopsis thaliana (n = 5) and of related species with six or seven chromosome pairs were derived from an ancestral karyotype with eight chromosome pairs. To test this hypothesis, we applied multicolor chromosome painting using contiguous bacterial artificial chromosome pools of A. thaliana arranged according to the genetic maps of Arabidopsis lyrata and Ca

Brassicaceae-based bio-disinfestation is a promising non-chemical option to manage lettuce Fusarium wilt caused by Fusarium oxysporum f. sp. lactucae (FOLac), but evidence integrating pathogen suppression, residue chemistry, and soil microbiome response remains limited. We conducted a greenhouse mesocosm assay with naturally infested soil to compare biofumigation (uncovered soil) and biosolarization (plastic-covered soil for 40 days) using five Brassicaceae residues (Brassica carinata, B. juncea, B. napus, Sinapis alba and Raphanus sativus) applied at two field-equivalent doses. Biosolarizatio

The Brassica database (BRAD) was built initially to assist users apply Brassica rapa and Arabidopsis thaliana genomic data efficiently to their research. However, many Brassicaceae genomes have been sequenced and released after its construction. These genomes are rich resources for comparative genomics, gene annotation and functional evolutionary studies of Brassica crops. Therefore, we have updated BRAD to version 2.0 (V2.0). In BRAD V2.0, 11 more Brassicaceae genomes have been integrated into the database, namely those of Arabidopsis lyrata, Aethionema arabicum, Brassica oleracea, Brassica n

Proteins containing domain of unknown function (DUF) are prevalent in eukaryotic genome. The DUF1216 proteins possess a conserved DUF1216 domain resembling to the mediator protein of Arabidopsis RNA polymerase II transcriptional subunit-like protein. The DUF1216 family are specifically existed in Brassicaceae, however, no comprehensive evolutionary analysis of DUF1216 genes have been performed. We performed a first comprehensive genome-wide analysis of DUF1216 proteins in Brassicaceae. Totally 284 DUF1216 genes were identified in 27 Brassicaceae species and classified into four subfamilies on

DNA methylation is an ancient molecular modification found in most eukaryotes. In plants, DNA methylation is not only critical for transcriptionally silencing transposons, but can also affect phenotype by altering expression of protein coding genes. The extent of its contribution to phenotypic diversity over evolutionary time is, however, unclear, because of limited stability of epialleles that are not linked to DNA mutations. To dissect the relative contribution of DNA methylation to transposon surveillance and host gene regulation, we leveraged information from three species in the Brassicac

Selecting insect-resistant plant varieties is a key component of integrated management programs of oligophagous pests such as diamondback moth, Plutella xylostella (L.) (Lepidoptera: Plutellidae), but rigorous research on important life history parameters of P. xylostella in relation to host plant resistance is rare. We evaluated six conventional brassicaceous species, namely, Brassica napus L. 'Q2', B. rapa L., B. juncea (L.) Czern., B. carinata L., B. oleracea L., and Sinapis alba L., and two herbicide-tolerant cultivars, namely, B. napus 'Liberty' and B. napus 'Conquest' for their resistanc

Different extracts of the aerial parts of Biscutella raphanifolia (Brassicaceae), which has not been the subject of any study, were screened for the phytochemical content, anti-microbial, antioxidant and anti-cholinesterase activities. We used four methods to identify the antioxidant activity namely, ABTS(•+), DPPH• scavenging, CUPRAC and ferrous-ions chelating methods. Since there is a relationship between antioxidants and cholinesterase enzyme inhibitors, we used two methods to determine the in vitro anti-cholinesterase activity by the use of the basic enzymes that occur in causing Alzheimer

The genus Iberis L. is mainly distributed in the Mediterranean basin, with Europe representing the primary center of diversity, while a limited number of species occur in Northwest Africa and Southwest to Central Asia. The genus is represented by ten taxa in Türkiye, including four endemic taxa: Iberis carica (Bornm.) Prain, Iberis halophila Vural & H. Duman, Iberis gypsicola Yıld. and Iberis saxatilis subsp. Magnesiana Oskay. Previous studies emphasized morphology and phylogeny, whereas comprehensive metabolomic and phytochemical analyses of the genus remain limited. In this study, plant mate

In Brassicaceae crops such as cabbage and turnip, which are harvested for their vegetative organs, floral induction can reduce the eating quality of these organs. Therefore, developing late-flowering varieties that are insensitive to vernalization is a key breeding objective. However, a major challenge in breeding these cultivars lies in the contradiction between the late-flowering trait and the need to induce flowering for crossing. To address this, we developed a flowering induction technique using grafting, in which gibberellin-induced elongated internodes are used as scions and grafted ont

The availability of a large number of whole-genome sequences allows comparative genomic analysis to reveal and understand evolution of regulatory regions and elements. The role played by events such as whole-genome and segmental duplications followed by genome fractionation in shaping genomic landscape and in expansion of gene families is crucial toward developing insights into evolutionary trends and consequences such as sequence and functional diversification. Members of Brassicaceae are known to have experienced several rounds of whole-genome duplication (WGD) that have been termed as paleo

Generation of competent offspring is vital for the prosperity of flowering plants. The pistil not only functions as a conduit for pollen tubes to grow to the ovary but also provides a selective venue for facilitating the growth of compatible pollen tubes and discouraging invaders and incompatible pollen. This review integrates recent advances in pollen-pistil interactions on dry stigmas of the Brassicaceae in the domains of self-incompatibility (SI) and cross-compatibility. We first outline the initial recognition mechanisms that distinguish self from nonself pollen and then highlight how key

BACKGROUND: Plants produce diverse bouquets of specialized metabolites (SMs), yet only a fraction of the vast phytochemical space hasbeen explored to date.Comparative analysis of SM profiles can reveal hotspots of biochemical novelty, while systematic profilingacross taxonomic levels does presently not cover large plant families.
MATERIALS AND METHODS: To study core and accessory SM profiles in the Brassicaceae plant family, we fingerprinted 14 species by Liquid-ChromatographyMass-Spectrometry (LC-MS/MS). We develop standardized experimental and computational workflows integrating in silicoan

INTRODUCTION: Helicobacter pylori (Hp) infection is highly prevalent globally and poses a significant public health challenge due to its link with chronic gastritis, peptic ulcers, and gastric malignancies. Hp's persistence within the gastric environment, particularly in case of infection with virulent strains, triggers chronic inflammatory responses and mucosal damage. Antibiotic therapy is the primary approach for Hp eradication, but antibiotic resistance and adverse effects hinder treatment efficacy. Emerging evidence suggests that Brassicaceae-derived metabolites could serve as adjunctive

Sesterterpenoids are a rare terpene class harboring untapped chemodiversity and bioactivities. Their structural diversity originates primarily from the scaffold-generating sesterterpene synthases (STSs). In fungi, all six known STSs are bifunctional, containing C-terminal trans-prenyltransferase (PT) and N-terminal terpene synthase (TPS) domains. In plants, two colocalized PT and TPS gene pairs from Arabidopsis thaliana were recently reported to synthesize sesterterpenes. However, the landscape of PT and TPS genes in plant genomes is unclear. Here, using a customized algorithm for systematical

Pervasive hybridization and whole-genome duplications (WGDs) influenced genome evolution in several eukaryotic lineages. Although frequent and recurrent hybridizations may result in reticulate phylogenies, the evolutionary events underlying these reticulations, including detailed structure of the ancestral diploid and polyploid genomes, were only rarely reconstructed. Here, we elucidate the complex genomic history of a monophyletic clade from the mustard family (Brassicaceae), showing contentious relationships to the early-diverging clades of this model plant family. Genome evolution in the cr

OBJECTIVES: The intracellular parasitic protozoan, Leishmania spp., causes several forms of diseases in humans. Cytotoxicity and emergence of new strains resistance to the current anti-leishmanial drugs have encouraged researchers to focus on new resources. Glucosinolates (GSL) are found mainly in the Brassicaceae family with potential cytotoxic and anti-parasitic properties. The present study reports in vitro antileishmanial activity of the GSL fraction from Alyssum linifolium seeds against Leishmania major.
MATERIALS AND METHODS: The GSL fraction was prepared by ion-exchange and reversed-ph

Brassicaceae is a major model system for studies of plant evolution, systematics, comparative genomics, and speciation; however, mitochondrial genomic resources remain limited compared to plastid genomic resources, constraining investigations of mitochondrial genome evolution. Here, we report the mitogenome of the Korean endemic species Arabis takesimana, representing only the third available mitogenome for the genus Arabis. The mitogenome is 291,633 bp in length with a GC content of 45.3%. It was assembled with an average sequencing depth of 298.9×. It contains 58 annotated genes, including 3

In plants, de novo DNA methylation is guided by 24-nt short interfering (si)RNAs in a process called RNA-directed DNA methylation (RdDM). Primarily targeted at transposons, RdDM causes transcriptional silencing and can indirectly influence expression of neighboring genes. During reproduction, a small number of siRNA loci are dramatically upregulated in the maternally derived seed coat, suggesting that RdDM might have a special function during reproduction. However, the developmental consequence of RdDM has been difficult to dissect because disruption of RdDM does not result in overt phenotypes

Capsella is a model plant genus of the Brassicaceae closely related to Arabidopsis. To disentangle its biogeographical history and intrageneric phylogenetic relationships, 282 individuals of all five currently recognized Capsella species were genotyped using a restriction digest-based next-generation sequencing method. Our analysis retrieved two main lineages within Capsella that split c. one million years ago, with western C. grandiflora and C. rubella forming a sister lineage to the eastern lineage consisting of C. orientalis. The split was attributed to continuous latitudinal displacements
