Emerging roles of vascular brassinosteroid receptors of the BRI1-like family.

Brassinosteroids (BRs) are essential hormones for plant growth and development that are perceived at the plasma membrane by a group of Leucine-Rich Repeat Receptor-Like Kinases (LRR-RLKs) of the BRASSINOSTEROID INSENSITIVE 1 (BRI1) family. The BRI1 receptor was first discovered by genetic screenings based on the dwarfism of BR-deficient plants. There are three BRI1 homologs, named BRI1-like 1, 2 and 3 (BRLs), yet only BRL1 and BRL3 behave as functional BR receptors. Whereas the BRI1 pathway operates in the majority of cells to promote growth, BRL receptor signaling operates under specific spat

Interaction between BZR1 and PIF4 integrates brassinosteroid and environmental responses.

Plant growth is coordinately regulated by environmental and hormonal signals. Brassinosteroid (BR) plays essential roles in growth regulation by light and temperature, but the interactions between BR and these environmental signals remain poorly understood at the molecular level. Here, we show that direct interaction between the dark- and heat-activated transcription factor phytochrome-interacting factor 4 (PIF4) and the BR-activated transcription factor BZR1 integrates the hormonal and environmental signals. BZR1 and PIF4 interact with each other in vitro and in vivo, bind to nearly 2,000 com

Calcium and ZmCCaMK are involved in brassinosteroid-induced antioxidant defense in maize leaves.

Brassinosteroids (BRs) have been shown to enhance stress tolerance by inducing antioxidant defense systems. However, the mechanisms of BR-induced antioxidant defense in plants remain to be determined. In this study, the role of calcium (Ca(2+)) and maize calcium/calmodulin-dependent protein kinase (CCaMK), ZmCCaMK, in BR-induced antioxidant defense, and the relationship between ZmCCaMK and Ca(2+) in BR signaling were investigated. BR treatment led to a significant increase in cytosolic Ca(2+) concentration in protoplasts from maize mesophyll, and Ca(2+) was shown to be required for BR-induced

Effects of Lactone- and Ketone-Brassinosteroids of the 28-Homobrassinolide Series on Barley Plants under Water Deficit.

The aim of this work was to study the ability of 28-homobrassinolide (HBL) and 28-homocastasterone (HCS) to increase the resistance of barley (Hordeum vulgare L.) plants to drought and to alter their endogenous brassinosteroid status. Germinated barley seeds were treated with 0.1 nM HBL or HCS solutions for two hours. A water deficit was created by stopping the watering of 7-day-old plants for the next two weeks. Plants responded to drought through growth inhibition, impaired water status, increased lipid peroxidation, differential effects on antioxidant enzymes, intense proline accumulation,

Brassinosteroids rescue the deficiency of CYP90, a cytochrome P450, controlling cell elongation and de-etiolation in Arabidopsis.

The cpd mutation localized by T-DNA tagging on Arabidopsis chromosome 5-14.3 inhibits cell elongation controlled by the ecdysone-like brassinosteroid hormone brassinolide. The cpd mutant displays de-etiolation and derepression of light-induced genes in the dark, as well as dwarfism, male sterility, and activation of stress-regulated genes in the light. The CPD gene encodes a cytochrome P450 (CYP90) sharing homologous domains with steroid hydroxylases. The phenotype of the cpd mutant is restored to wild type both by feeding with C23-hydroxylated brassinolide precursors and by ectopic overexpres

LF1 mediates brassinosteroid signaling to regulate leaf angle development in rice.

Leaf angle is a key determinant of plant architecture and grain yield in rice, yet the complex genetic regulatory network remains unclear. Here, we reveal that the HD-ZIP III transcription factor LF1 mediates brassinosteroid (BR) signaling to modulate leaf angle. A miRNA165/166-resistant, gain-of-function lf1 mutant displays an enlarged leaf angle driven by elongated adaxial parenchymal cells and diminished abaxial sclerenchyma. At the transcriptional level, the BR-activated transcription factor OsBZR1 directly induces LF1, which in turn upregulates OsOFP8, forming an OsBZR1-LF1-OsOFP8 hierarc

StCYP90C1 modulates plant architecture via regulating brassinosteroid biosynthesis in potato.

Brassinosteroid (BR) exert a crucial regulatory role in shaping plant architecture. However, the BR biosynthesis and underlying genetic variations governing potato plant architecture remain poorly understood. Here, we identified a single recessive locus, ap, which controls the abnormal architectural plant (AP) with dark-green curled leaves, shortened internodes, and delayed flowering, using a BC4S1 population. The functional gene, StCYP90C1, encoding 3-epi-6-deoxocathasterone 23-monooxygenase, was identified at this locus. StCYP90C1 expression was significantly lower in the leaves of APs than

PmLBD3 links auxin and brassinosteroid signalling pathways on dwarfism in Prunus mume.

BACKGROUND: Grafting with dwarf rootstock is an efficient method to control plant height in fruit production. However, the molecular mechanism remains unclear. Our previous study showed that plants with Prunus mume (mume) rootstock exhibited a considerable reduction in plant height, internode length, and number of nodes compared with Prunus persica (peach) rootstock. The present study aimed to investigate the mechanism behind the regulation of plant height by mume rootstocks through transcriptomic and metabolomic analyses with two grafting combinations, 'Longyan/Mume' and 'Longyan/Peach'. Insights into Metabolic Reactions of Semi-Dwarf, Barley Brassinosteroid Mutants to Drought.

The roles of endogenous brassinosteroids (BRs) in the modulation of reaction to drought and genetic regulation of this process are still obscure. In this study, a multidirectional analysis was performed on semi-dwarf barley (Hordeum vulgare) Near-Isogenic Lines (NILs) and the reference cultivar "Bowman" to get insights into various aspects of metabolic reaction to drought. The NILs are defective in BR biosynthesis or signaling and displayed an enhanced tolerance to drought. The BR metabolism perturbations affected the glucose and fructose accumulation under the control and stress conditions. T

Brassinosteroid-regulated transcription factors confer epigenetic changes that repress plant immunity.

When organisms encounter pathogens, they rapidly activate complex defense programs to ensure survival. While these immune responses are vital, they often also incur trade-offs, such as reduced growth and development and must therefore be tightly controlled. In this study, we reveal that the steroid hormones brassinosteroids (BRs) contribute to this control in Arabidopsis thaliana by repressing immunity-related genes. We provide evidence that the BR-regulated basic helix-loop-helix (bHLH) transcription factor CESTA (CES), along with its homologs BR ENHANCED EXPRESSION (BEE)1-3, mediate DNA meth

Brassinosteroid signaling represses ZINC FINGER PROTEIN11 to regulate ovule development in Arabidopsis.

Brassinosteroid (BR) signaling and the C-class MADS-box gene AGAMOUS (AG) play important roles in ovule development in Arabidopsis (Arabidopsis thaliana). However, how BR signaling integrates with AG functions to control the female reproductive process remains elusive. Here, we showed that the regulatory role of BR signaling in proper ovule development is mediated by the transcriptional repressor gene ZINC FINGER PROTEIN 11 (ZFP11), which is a direct target of AG. ZFP11 expression initiates from the placenta upon AG induction and becomes prominent in the funiculus of ovule primordia. Plants ha

An interaction between BZR1 and DELLAs mediates direct signaling crosstalk between brassinosteroids and gibberellins in Arabidopsis.

Plant growth and development are coordinated by several groups of small-molecule hormones, including brassinosteroids (BRs) and gibberellins (GAs). Physiological and molecular studies have suggested the existence of crosstalk between BR and GA signaling. We report that BZR1, a key transcription factor activated by BR signaling, interacts in vitro and in vivo with REPRESSOR OF ga1-3 (RGA), a member of the DELLA family of transcriptional regulators that inhibits the GA signaling pathway in Arabidopsis thaliana. Genetic analyses of plants with mutations in the genes encoding RGA and BZR1 revealed

Induced variation in BRASSINOSTEROID INSENSITIVE 1 (BRI1) confers a compact wheat architecture.

BACKGROUND: The brassinosteroid (BR) plant hormones regulate numerous developmental processes, including those determining stem height, leaf angle, and grain size that have agronomic relevance in cereals. Indeed, barley (Hordeum vulgare) varieties containing uzu alleles that impair BR perception through mutations in the BR receptor BRASSINOSTEROID INSENSITIVE 1 (BRI1) exhibit a semi-dwarf growth habit and more upright leaves suitable for high-density planting. We used forward and reverse genetic approaches to develop novel BRI1 alleles in wheat (Triticum aestivum L.) and investigated their pot

Take a deep BReath: Manipulating brassinosteroid homeostasis helps cereals adapt to environmental stress.

Global climate change leads to the increased occurrence of environmental stress (including drought and heat stress) during the vegetative and reproductive stages of cereal crop development. Thus, more attention should be given to developing new cereal cultivars with improved tolerance to environmental stress. However, during the development of new stress-tolerant cereal cultivars, the balance between improved stress responses (which occur at the expense of growth) and plant yield needs to be maintained. Thus, the urgent need for developing new cereal germplasm with improved stress tolerance co

Roles of Nitric Oxide and Brassinosteroid in Improving Fruit Quality during Postharvest: Potential Regulators?

Most postharvest fruits are highly perishable, which directly impairs fruit taste and causes an economic loss of fresh products. Thus, it is necessary to find effective techniques to alleviate this issue. Recently, nitric oxide (NO) and brassinosteroid (BR) have been developed as postharvest alternatives to improve fruit quality. This work mainly reviews the recent processes of NO and BR in improving fruit quality during postharvest. Exogenous NO or BR treatments delayed fruit senescence, enhanced disease resistance, and alleviated chilling injury in postharvest fruit, and potential physiologi

Nitric oxide and brassinosteroids enhance chromium stress tolerance in Glycine max L. (Merr.) by modulating antioxidative defense and glyoxalase systems.

Chromium (Cr) contamination of agricultural soils is a major threat to human and plant health worldwide and causes reductions in plant growth and crop yields. 24-epibrassinolide (EBL) and nitric oxide (NO) have been shown to ameliorate the reductions in growth caused by the stresses induced by heavy metals; however, the interactions between EBL and NO on the alleviation of Cr-induced phytotoxicity have been poorly studied. Hence, this study was undertaken to examine any beneficial effects of EBL (0.01 µM) and NO (100 µM), applied alone or in combination, on the mitigation of stress induced by

Water deficit aggravated the inhibition of photosynthetic performance of maize under mercury stress but is alleviated by brassinosteroids.

Mercury (Hg) significantly inhibits maize (Zea mays L.) production, which could be aggravated by water deficit (WD) due to climate change. However, there is no report on the maize in response to combined their stresses. This work was conducted for assessing the response and adaptive mechanism of maize to combined Hg and WD stress using two maize cultivars, Xianyu (XY) 335 and Yudan (YD) 132. The analysis was based on plant growth, physiological function, and transcriptomic data. Compared with the single Hg stress, Hg accumulation in whole plant and translocation factor (TF) under Hg+WD were in

Brassinosteroids mediate susceptibility to brown planthopper by integrating with the salicylic acid and jasmonic acid pathways in rice.

Improved knowledge of the interactions between plants and insects will facilitate better insect control in crops. Brassinosteroids (BRs) play a vital role in plant growth, developmental processes, and responses to pathogen infection, but the role of BRs in interactions between plants and insects remain largely unknown. In this study, we characterized a negative role of BRs in rice defense against brown planthopper (BPH, Nilaparvata lugens) and examined its underlying mechanisms. We found that BPH infestation suppressed the BR pathway while successively activating the salicylic acid (SA) and ja

A BRASSINOSTEROID INSENSISTIVE 1 receptor kinase ortholog is required for sex determination in Ceratopteris richardii.

Most ferns, unlike all seed plants, are homosporous and produce sexually undifferentiated spores. Sex ratio in many homosporous species is environmentally established by the secretion of antheridiogen from female/hermaphrodite gametophytes. Nearby undetermined gametophytes perceive antheridiogen, which induces male development. In the fern Ceratopteris richardii (Ceratopteris), hermaphroditic (her) mutants develop as hermaphrodites even in the presence of antheridiogen. Modern sequencing and genomic tools make the molecular identification of mutants in the 11-Gbp genome of this fern possible.

CRISPR/Cas9-mediated editing of GmDWF1 brassinosteroid biosynthetic gene induces dwarfism in soybean.

The study on the GmDWF1-deficient mutant dwf1 showed that GmDWF1 plays a crucial role in determining soybean plant height and yield by influencing the biosynthesis of brassinosteroids. Soybean has not adopted the Green Revolution, such as reduced height for increased planting density, which have proven beneficial for cereal crops. Our research identified the soybean genes GmDWF1a and GmDWF1b, homologous to Arabidopsis AtDWF1, and found that they are widely expressed, especially in leaves, and linked to the cellular transport system, predominantly within the endoplasmic reticulum and intracellu
