Experimental population dynamics of Rhabdias bufonis (Nematoda) in toads (Bufo bufo): density-dependence in the primary infection.

Density-dependence in worm establishment, numbers, biomass and larval production were examined in primary infections of 0, 10, 40, 80 and 160 larvae of the lung nematode, Rhabdias bufonis in the common toad, Bufo bufo. The infection procedure established 4 non-overlapping levels of infection which persisted until 6 weeks post-infection (p.i.), after which there was an overall decline up to 12 weeks p.i. Worm numbers had no direct effect on adult worm survival but temporal changes in worm weight were density-dependent. Adult worm establishment in the lungs declined significantly as the numbers

N-methyl serotonin analogues from the Bufo bufo toad venom interact efficiently with the α7 nicotinic acetylcholine receptors.

Two low-molecular-weight compounds were isolated from the parotid gland secret of the toad Bufo bufo, which by absorption spectra and HPLC-MS/MS chromatography data correspond to di- and trimethyl derivatives of serotonin (5-hydorxytryptamine): bufotenine (confirmed by counter synthesis) and bufotenidine (5-HTQ). In experiments on competitive radioligand binding, these compounds showed a higher affinity and selectivity for neuronal α7 nicotinic acetylcholine receptors compared with the muscular cholinergic receptors. The most efficient compound in terms of binding value was bufotenine, the eff

Serotonin-degradative pathways in the toad (Bufo bufo japonicus) brain: clues to the pharmacological analysis of human psychiatric disorders.

Bufotenine (BUTN) is a hallucinogen with psychotropic effects. High levels of BUTN and its precursor, N-methylserotonin are shown for the first time to occur and to accumulate mainly in the brain during the degradation of serotonin in the central nervous system of the toad, Bufo bufo japonicus. These compounds are concentrated in the hindbrain, which includes the cerebellum and medulla oblongata. BUTN can also be detected in blood and urine specimens from the toad. In humans, autism is a subtype of schizophrenia that appears to be a functional disease of the brain. BUTN can be detected in urin

The antidromic activation of tectal neurons by electrical stimuli applied to the caudal medulla oblongata in the toad, Bufo bufo L.

In order to specify the tectal projection to the bulbar/spinal regions, the antidromic responses of the physiologically identified tectal neurons as well as the gross antidromic field responses in the optic tectum to electrical stimuli applied to the caudal medulla were examined in the paralyzed common toad, Bufo bufo. The antidromic field potential was recorded in the optic tectum in response to electrical stimuli applied to the ventral paramedian portion of the contralateral caudal medulla (where the crossed tecto-spinal pathway of Rubinson (1968) and Lázár (1969) runs), but generally not wh

From cells to tadpoles: mechanistic insights into ZnO-nanoparticle-induced oxidative stress and developmental toxicity in Bufo bufo.

Zinc oxide nanoparticles (ZnO-NPs) are widely used engineered nanomaterials increasingly present in freshwater ecosystems, yet their effects on amphibians remain poorly understood. Amphibians are ecologically important and highly sensitive to contaminants, making them valuable bioindicators. This study provides the first integrated assessment of ZnO-NP effects on the European toad (Bufo bufo, Gosner stages 26–27), combining biochemical, endocrine, and histological endpoints. Tadpoles were exposed for seven days to sub lethal ZnO-NP concentrations (0.1, 1.0, and 10 mg/L) under controlled labora

Individual variation in cardiotoxicity of parotoid secretion of the common toad, Bufo bufo, depends on body size - first results.

Anurans secrete a wide diversity of toxins from skin glands to defend themselves against predators and pathogens. Bufonids produce potent poison in parotoid macroglands located in the postorbital region. Parotoid secretion is a rich source of bioactive compounds with cardiotoxic, cytotoxic and hemolytic activity. Poison content and toxicity may vary between species, populations, and among conspecifics inhabiting the same area. In the present paper, we pre-analyzed the individual variation in cardiotoxicity of parotoid extract of common toads (Bufo bufo Linnaeus, 1758) and impact of body mass (

Nuclear maturation inhibitors in Bufo arenarum oocytes.

In Bufo arenarum, as in other amphibian species, nuclear maturation--characterized by germinal vesicle breakdown (GVBD) and meiosis resumption--is induced by progesterone. Seasonal variations in the sensitivity to progesterone were observed in Bufo arenarum oocytes. The gonadotropin FSH and drugs such as dbcAMP and xanthine did not affect nuclear maturation per se, although they acted as negative modulators of progesterone-induced nuclear maturation. Similar results to FSH in nuclear maturation were obtained with 17 beta-estradiol.

[Organization and differentiation of the segmentary plate in common toad embryo Bufo bufo L., following heat shock (author's transl)].

A heat shock greatly disturbs the shaping and the growth of embryos with 4 or 5 somites. One part of a segmentary plate with a size including four control somites stays inorganized, whereas posterior segmentation gets on normally. The differentiation of the unsegmented somitic block (fusing myoblasts, myogenesis) is the same as the one of the previous somites; the dermatome and the sclerotome issued from the unsegmented block are not cut up. Ultrastructural observations of treated segmentary plates show that the somitic block undergoes important changes. The non-segmented somitic mesoderm of t

Factors controlling the ovarian cycle in a temperate zone anuran, the toad Bufo bufo: food uptake, nutritional state, and gonadotropin.

The relations between nutritional state and vitellogenic growth of oocytes were studied in 87 sexually mature female toads Bufo bufo during the postbreeding season before initiation of the next annual ovarian cycle. Recruitment of small oocytes to vitellogenic growth was correlated with resumption of food uptake and the restoration of a positive energy balance in the toads that had become depleted of energy reserves during hibernation and breeding. Treatment with exogenous gonadotropin (hCG) immediately initiated vitellogenic growth, independently of the nutritional state. But the hCG-induced

K(+) transport in the mesonephric collecting duct system of the toad Bufo bufo: microelectrode recordings from isolated and perfused tubules.

We studied the mechanisms of K(+) transport in cells from isolated and perfused collecting tubules and ducts from the mesonephric kidney of the toad Bufo bufo. Cells were impaled with microelectrodes across the basal cell membrane. The basolateral membrane potential (V(bl)) depolarized upon change of bath [K(+)] from 3 to 20 mmol l(-1) demonstrating a large K(+) conductance in this membrane. In collecting tubules and collecting ducts a V(bl) of -66+/-2 mV and -74+/-4 mV depolarized by 30+/-2 mV and 36+/-3 mV, respectively (N=23; 15). The K(+) channel inhibitor Ba(2+) (1 mmol l(-1)) inhibited t

Mucor amphibiorum in the toad, Bufo marinus, in Australia.

Mucor amphibiorum, a fungus previously found in captive amphibians in Europe and the platypus in Australia, was observed in free-ranging toads, Bufo marinus, in Australia. In tissues the fungus occurred as sphaerules 4.9 to 36.4 microns in diameter; hyphae were not formed. Some spharules developed two to 11 daughter sphaerules internally and these were released into tissues by dissolution of the outer wall. Infected toads were found at 11 sites from nine locations in northern and eastern Australia. The overall prevalence of infection in 3,518 toads was 0.71%. Mucor amphibiorum was isolated fro

A new function for lactate in the toad Bufo marinus.

In the amphibian Bufo marinus, progressive hypoxia below a critical PO2 elicits a transient 50% increase in O2 consumption that coincides with the onset of lactate formation. The present study was designed to test the hypothesis that lactate causes the observed rise in metabolic rate. Arterial bolus infusions of pH-neutral sodium lactate solutions (4 mmol/kg body wt) in toads maintained under hypoxia actually elicit a similar increase in metabolic rate. The application of adrenergic antagonists (bretylium tosylate, phentolamine, propranolol, and reserpine) inhibits this response, suggesting th

Indication of spatially random occurrence of Chlamydia-like organisms in Bufo bufo tadpoles from ponds located in the Geneva metropolitan area.

Occurrence of bacteria belonging to the order Chlamydiales was investigated for the first time in common toad (Bufo bufo) tadpole populations collected from 41 ponds in the Geneva metropolitan area, Switzerland. A Chlamydiales-specific real-time PCR was used to detect and amplify the Chlamydiales 16S ribosomal RNA-encoding gene from the tails of 375 tadpoles. We found the studied amphibian populations to host Chlamydia-like organisms (CLOs) attributable to the genera Similichlamydia, Neochlamydia, Protochlamydia and Parachlamydia (all belonging to the family Parachlamydiaceae), Simkania (famil

Spontaneous chromomycosis in the marine toad (Bufo marinus).

Post-mortem examinations were performed on two marine toads, one animal showing neurological disorders and the other multifocal dermatitis. In one case, lesions consisted of a severe granulomatous encephalomyelitis and in the other of multiple granulomas in the nasal cavity, lungs, heart, bone marrow, ovaries and skin. Histologically, the lesions revealed varying amounts of dark brown fungal elements, predominantly sclerotic bodies indicative of a mycotic infection due to a pigmented fungus.

Leptospires in the marine toad (Bufo marinus) on Barbados.

Leptospires were isolated from the kidneys of four of 211 toads (Bufo marinus) caught on Barbados. Two of the isolates were identified as Leptospira interrogans serovar bim in the Autumnalis serogroup (the most common cause of leptospiral illness on Barbados), and two as possibly new serovars in the Australis serogroup. Sera from 198 of the toads were examined by the leptospire microscopic agglutination test. Forty-two (21%) were positive at titers of greater than or equal to 1:100, and 54 (27%) at greater than or equal to 1:50. The predominating serogroups were Australis (50%), Autumnalis (23

Motor innervation of the toad iris (Bufo marinus).

The sphincter pupillae muscle cells in the iris of Bufo marinus contract autonomously in response to bright light, causing a rapid constriction of the pupil. A strong sympathetic beta-adrenergic inhibition of the sphincter pupillae is apparent in this species. The inhibitory fibers can originate in the second, third, or fourth ventral spinal roots. No strong, consistent excitatory innervation of the toad iris was detected, even by transmural stimulation of the isolated iris. Pupilloconstriction occasionally resulted from stimulation of the 3rd or 5th cranial nerves, but the effect was small (1

Parallel metabotropic pathways in the heart of the toad, Bufo marinus.

This study examined the transduction pathways activated by epinephrine in the pacemaker region of the toad heart. Recordings of membrane potential, force, and intracellular Ca(2+) concentration ([Ca(2+)](i)) were made from arrested toad sinus venosus. Sympathetic nerve stimulation activated non-alpha-, non-beta-adrenoceptors to evoke a membrane depolarization and a transient increase in [Ca(2+)](i). In contrast, the beta-adrenoceptor agonist isoprenaline (10 microM) caused membrane hyperpolarization and decreased [Ca(2+)](i). The phosphodiesterase inhibitor 3-isobutyl-1-methylxanthine (0.5 mM)
