Gut microbiome and circulating bacterial DNA ("blood microbiome") in a mouse model of total parenteral nutrition: Evidence of two distinct separate microbiotic compartments.

BACKGROUND & AIMS: Total parenteral nutrition (TPN) causes gut atrophy, dysbiosis and leakage of the gut barrier. This study aimed to characterize the gut microbiome in response to different TPNs and tested the hypothesis whether increased gut permeability in TPN would lead to changes in the circulating bacterial DNA ("blood microbiome"). METHODS: Male C57BL/6J mice were randomly allocated to the following groups for seven days (1) chow-fed control (C) without jugular vein catheter (JVC, n=6) (2) chow-fed with JVC and infusion of saline (S) (n = 6) (3) Intralipid-based TPN (n-6:n-3 ratio 7:1)

Antibacterial Siderophores of Pandoraea Pathogens and Their Impact on the Diseased Lung Microbiota.

Antibiotic-resistant bacteria of the genus Pandoraea, frequently acquired from the environment, are an emerging cause of opportunistic respiratory infections, especially in cystic fibrosis (CF) patients. However, their specialized metabolites, including niche and virulence factors, remained unknown. Through genome mining of environmental and clinical isolates of diverse Pandoraea species, we identified a highly conserved biosynthesis gene cluster (pan) that codes for a nonribosomal peptide synthetase (NRPS) assembling a new siderophore. Using bioinformatics-guided metabolic profiling of wild t

Development of the Caecal Microbiota in Three Broiler Breeds.

The development of the caecal microbiota plays a role in the metabolism and immune competence of chickens. A detailed understanding of normal succession in the caecal microbiota can inform the use of probiotics and other interventions to optimize the caecal microbiota. The development of the microbiota in caecal mucus and lumen samples from three breeds of broiler chicken (Cobb 500, n = 36; Hubbard JA87, n = 38; and Ross 308, n = 36) was observed between 0 and 42 days post hatch. Chicks were housed in the same room of a climate-controlled, biosecure chicken housing unit. Between 0 and 14 days

Degradation of dichloromethane by an isolated strain Pandoraea pnomenusa and its performance in a biotrickling filter.

A strain Pandoraea pnomenusa LX-1 that uses dichloromethane (DCM) as sole carbon and energy source has been isolated and identified in our laboratory. The optimum aerobic biodegradation of DCM in batch culture was evaluated by response surface methodology. Maximum biodegradation (5.35 mg/(L·hr)) was achieved under cultivation at 32.8°C, pH 7.3, and 0.66% NaCl. The growth and biodegradation processes were well fitted by Haldane's kinetic model, yielding maximum specific growth and degradation rates of 0.133 hr(-1) and 0.856 hr(-1), respectively. The microorganism efficiently degraded a mixture

Characteristics of gallbladder microbiome in healthy dogs and cats, dogs with gallbladder mucocele, and cats with suspected cholangitis/cholangiohepatitis.

The aim of this study was to investigate and characterize the microbiome in bile samples obtained from dogs with gallbladder mucocele (6), cats with suspected cholangitis/cholangiohepatitis (4), as well as from healthy dogs (6) and cats (4). Our goal was to compare the microbiome patterns with clinical findings and bacterial culture results in diseases of the gallbladder and to identify a potential microbial biomarker of diseased groups. The microbial taxa composition revealed that Proteobacteria were the most dominant phylum in healthy and diseased individuals in all groups. Individuals from

Integrated genomic and experimental assessment of the biosafety of Paraburkholderia sacchari as a promising microbial chassis.

Paraburkholderia sacchari is an environmental member of the Burkholderiaceae with recognized biotechnological potential, yet its genomic diversity, biosafety profile, and evolutionary relationship to pathogenic Burkholderia species remain poorly understood. Here, we present a comprehensive genomic and phenotypic characterization of P. sacchari strain LMG 19,450T and compare it with three additional P. sacchari genomes and phylogenetically related Burkholderia species. A high-quality genome assembly confirmed its placement within the Paraburkholderia clade and species-level relatedness among P.

Outbreak of Pandoraea commovens Infections among Non-Cystic Fibrosis Intensive Care Patients, Germany, 2019-2021.

Pandoraea spp. are gram-negative, nonfermenting rods mainly known to infect patients with cystic fibrosis (CF). Outbreaks have been reported from several CF centers. We report a Pandoraea spp. outbreak comprising 24 non-CF patients at a large university hospital and a neighboring heart center in Germany during July 2019-December 2021. Common features in the patients were critical illness, invasive ventilation, antimicrobial pretreatment, and preceding surgery. Complicated and relapsing clinical courses were observed in cases with intraabdominal infections but not those with lower respiratory t

Plasmid-Host Networks Reveal Persistent Mobile Elements Across Wastewater Treatment.

Plasmids drive horizontal gene transfer and antimicrobial resistance spread in water resource recovery facilities, yet their in situ dynamics in complex microbial communities remain poorly characterized. We apply Hi-C metagenomics to resolve plasmid-host associations across influent, activated sludge, and effluent at three facilities. We identify 944 plasmid clusters with facility- and stage-specific distributions. Plasmid host range narrows from influent to effluent, indicating that treatment selectively constrains plasmid-host associations. Persistent plasmid clusters with detectable mobilit

Black soldier fly and uric acid-degrading bacteria contribute to bioconvert duck manure in an efficient manner.

Duck manure, a major pollutant of intensive poultry production, is characterized by high organic load and nutrient content. However, the high water content of duck manure virtually impedes effective manure management. Here, we report the suitability of duck manure as a rearing substrate for larvae of black soldier fly Hermetia illucens L. (Diptera: Stratiomyidae), achieving a corresponding reduction in organic liquid waste. First, duck manure efficiently sustains the development and growth of black soldier fly larvae and is optimized to rear black soldier fly larvae by appropriately adding cor

The melioidosis agent Burkholderia pseudomallei and related opportunistic pathogens detected in faecal matter of wildlife and livestock in northern Australia.

The Darwin region in northern Australia has experienced rapid population growth in recent years, and with it, an increased incidence of melioidosis. Previous studies in Darwin have associated the environmental presence of Burkholderia pseudomallei, the causative agent of melioidosis, with anthropogenic land usage and proximity to animals. In our study, we estimated the occurrence of B. pseudomallei and Burkholderia spp. relatives in faecal matter of wildlife, livestock and domestic animals in the Darwin region. A total of 357 faecal samples were collected and bacteria isolated through culture

Ottowia thiooxydans gen. nov., sp. nov., a novel facultatively anaerobic, N2O-producing bacterium isolated from activated sludge, and transfer of Aquaspirillum gracile to Hylemonella gracilis gen. nov., comb. nov.

Strain K11T was isolated from activated sludge of a municipal wastewater-treatment plant. Phylogenetic analysis of the 16S rRNA gene sequence revealed that it represents a distinct line of descent within the Comamonadaceae. The novel strain was a Gram-negative, catalase- and oxidase-positive, non-motile, straight to slightly curved rod. Polyhydroxyalkanoate granules were stored intracellularly as reserve material. Colonies on agar plates were small, regular and characterized by a water-insoluble yellow pigment. Unbranched fatty acids 16:1omega7c, 16:0 and 18:1omega7c dominated the cellular fat

Engineering a Paraburkholderia pallida Tannase for Improved Catalytic Efficiency, Thermal Stability, and Green Tea Clarification.

Tannases can reduce haze and astringency in tea beverages, but their application is often limited by poor thermal stability and catalytic efficiency. A tannase from Paraburkholderia pallida (Tanpp) was engineered using active-site mutagenesis and computational hotspot prediction. The double mutant F384T/N307A showed the best performance, with a 2.7-fold increase in catalytic efficiency and an 11-fold longer half-life at 50 °C than wild-type Tanpp. Structural modeling and molecular dynamics simulations suggested that N307A widened the substrate-entry region, whereas F384T compacted the catalyti

Trichloroethylene degradation by toluene-oxidizing bacteria grown on non-aromatic substrates.

The potential of trichloroethylene (TCE) to induce and non-aromatic growth substrates to support TCE degradation in five strains (Pseudomonas mendocina KR1, Ralstonia pickettii PKO1, Pseudomonas putida F1, Burkholderia cepacia G4, B. cepacia PR1) of toluene-oxidizing bacteria was examined. LB broth and acetate did not support TCE degradation in any of the wild-type strains. In contrast, fructose supported the highest specific levels of TCE oxidation observed in each of the strains tested, except B. cepacia G4. We discuss the potential mechanisms and implications of this observation. In particu

Constructed wetland-microbial fuel cells enhanced with iron carbon fillers for ciprofloxacin wastewater treatment and power generation.

In this study, the following three experimental devices were operated for 70 days for the treatment of ciprofloxacin pollutants in wastewater: constructed wetlands (CW), constructed wetland-microbial fuel cells (EG), and constructed wetland-microbial fuel cells with new iron-carbon fillers (TPFC). The water quality, power generation capacity, microbial community structure, and changes in the resistance gene qnrs were studied. The efficiency of removal of total phosphate in the TPFC (97.1% ± 2.5%) was significantly higher than that in the EG (51.6% ± 4.8%) and the CW (68.1% ± 2.9%). The efficie

Assessing the impact of lime on chromium migration in soil caused by basic chromium sulfate in tannery.

Chromium (Cr) pollution is the primary pollution problem of the soil in tannery. However, the effect of tanning chemicals on Cr migration in soil has not been clearly elucidated. Column leaching tests were designed in this study to reveal the transport and transformation of Cr from basic chromium sulfate (BCS) into soil and the effects of lime on Cr migration and transformation. The results showed that BCS was mainly leached out in the state of Cr(VI) after entering the soil, and the Cr concentration in leachate decreased with the increase of the bulking thickness of the BCS. Compared with the

Low-Diversity Microbiota in Apical Periodontitis and High Blood Pressure Are Signatures of the Severity of Apical Lesions in Humans.

(1) Background: In developed countries, the prevalence of apical periodontitis (AP) varies from 20% to 50% for reasons that could be associated with the apical periodontitis microbiota ecology. (2) Methods: We performed a clinical study in the Odontology department of Toulouse hospital in France, to sequence the 16S rRNA gene of AP microbiota and collect clinical parameters from 94 patients. Forty-four patients were characterized with a PAI (periapical index of AP severity) score lower or equal to 3, while the others had superior scores (n = 50). Endometrial microbiota is more diverse in people with endometriosis than symptomatic controls.

Endometriosis is a chronic, estrogen-dependent gynecological condition affecting approximately 10% of reproductive age women. The most widely accepted theory of its etiology includes retrograde menstruation. Recent reports suggest the uterus is not sterile. Thus, the refluxed menstrual effluent may carry bacteria, and contribute to inflammation, the establishment and growth of endometriotic lesions. Here, we compared and contrasted uterine bacteria (endometrial microbiota) in people with surgically confirmed presence (N = 12) or absence of endometriosis (N = 9) using next-generation 16S rRNA g

The modulatory effects of alfalfa polysaccharide on intestinal microbiota and systemic health of Salmonella serotype (ser.) Enteritidis-challenged broilers.

Salmonella serotype (ser.) Enteritidis infection in broilers is a main foodborne illness that substantially threatens food security. This study aimed to examine the effects of a novel polysaccharide isolated from alfalfa (APS) on the intestinal microbiome and systemic health of S. ser. Enteritidis-infected broilers. The results indicated that broilers receiving the APS-supplemented diet had the improved (P < 0.05) growth performance and gut health than those fed no APS-supplemented diet. Supplementation with APS enhanced (P < 0.05) the richness of gut beneficial microbes such as Bacteroidetes,
