Investigations into the therapeutic effects of aerial and stem parts of Buxus papillosa C.K. Schneid.: In vitro chemical, biological and toxicological perspectives.

In this study, different solvent extracts (methanol, dichloromethane and n-hexane) from aerial and stem parts of Buxus papillosa C.K. Schneid (Buxaceae) were investigated for a panoply of bioassays. Biological profiles were established by determining antioxidant and enzyme inhibition profiles. Toxicity was tested using MTT cell viability assay on five different human cancer cell lines i.e, MCF-7, MDA-MB-231, CaSki, DU-145 and SW-480. For chemical fingerprinting, total bioactive contents and UHPLC-MS secondary metabolites profile were determined. Generally, both aerial and stem methanol extract

Danggui Buxue Decoction Alleviates Inflammation and Oxidative Stress in Mice with Escherichia coli-Induced Mastitis.

(1) Background: Bovine mastitis is a lactational disease caused by infection and milk stagnation in the mammary glands. Danggui buxue decoction (DBD), a traditional remedy for blood tonification, anti-inflammation, and antioxidation, has not been used previously to treat mastitis. (2) Methods: In this study, an Escherichia coli mastitis model was established by infecting lactating Kunming mice with clinically isolated bovine mastitis-derived E. coli. Based on this, the effects of DBD on inflammation and oxidative stress in mastitis model mice were evaluated by conducting routine blood tests, H

Danggui Buxue decoction activates AMPK/PGC-1α pathway to improve chronic heart failure.

ETHNOPHARMACOLOGICAL RELEVANCE: Danggui Buxue Decoction (DBD) is a famous traditional Chinese medicine (TCM) formula. It has been shown to have cardiovascular benefits. In past studies, DBD has successfully improved the cardiac function of patients with chronic heart disease. However, the mechanism of DBD on myocardial injury in chronic heart failure (CHF)models remains unclear. AIM: To investigate whether DBD can alleviate myocardial injury in CHF rats by improving abnormal myocardial energy metabolism. MATERIALS AND METHODS: To ensure the quality of DBD preparations, we employed high-perfo

Bone Protective Effects of Danggui Buxue Tang Alone and in Combination With Tamoxifen or Raloxifene in vivo and in vitro.

Danggui Buxue Tang (DBT), a traditional Chinese Medicine decoction containing Astragali Radix (AR) and Angelicae Sinensis Radix (ASR), is commonly prescribed for women in China as a remedy for menopausal symptoms. Previous study indicated that DBT stimulated cell growth and differentiation of human osteosarcoma MG-63 cells and exhibited estrogenic properties via estrogen receptors (ERs). The present study aimed to study the bone protective effects of DBT and its potential interactions with selective estrogen receptor modulators (SERMs, tamoxifen and raloxifene) in both in vivo and in vitro mod

Integrated metabolomics, transcriptomics and network pharmacology analysis to reveal the mechanisms of Danggui Buxue tang in treating Alzheimer's disease.

BACKGROUND: Alzheimer's disease (AD) is a prevalent, multifactorial, and multisystem chronic neurodegenerative disorder. Existing pharmacotherapeutic interventions for AD present significant limitations. Prior studies have indicated that Danggui Buxue Tang (DBT) reduces Aβ deposition and improves cognitive function. Nevertheless, the therapeutic mechanisms of DBT are not yet fully elucidated. PURPOSE: The objective of this study is to clarify the underlying mechanisms by which DBT alleviates AD. This will be accomplished through an integrated approach that encompasses network pharmacology, tr

Mechanism of modified danggui buxue decoction in glucocorticoid-induced osteoporosis: A discussion based on network pharmacology and molecular docking.

OBJECTIVE: Glucocorticoid-induced osteoporosis (GIOP) represents a major complication arising from the long-term use of glucocorticoids, which are widely prescribed for various inflammatory and autoimmune conditions. Despite its prevalence, the current therapeutic options for GIOP are limited in terms of efficacy, safety profiles, and patient compliance. The Modified Danggui Buxue Decoction (DGBXD), a traditional Chinese herbal formulation, has shown promise in preliminary studies for its potential osteoprotective effects. The present study aimed to explore the mechanistic underpinnings of DGB

A novel approach based on metabolomics coupled with network pharmacology to explain the effect mechanisms of Danggui Buxue Tang in anaemia.

Danggui Buxue Tang (DBT) is a famous Chinese medicinal decoction. Mechanism of DBT action is wide ranging and unclear. Exploring new ways of treatment with DBT is useful. Sprague-Dawley(SD) rats were randomly divided into 3 groups including control (NC, Saline), the DBT (at a dose of 8.10 g-1), and blood deficiency(BD) (Cyclophosphamide (APH)-andCyclophosphamide(CTX)-induced anaemia). A metabolomics approach using Liquid Chromatography-Quadrupole-Time-of-Flight/Mass Spectrometry (LC/Q-TOFMS) was developed to perform the plasma metabolic profiling analysis and differential metaboliteswerescreen

Analgesic Buxus alkaloids with Enhanced Selectivity for the Low-Voltage-Gated Calcium Channel Cav 3.2 over Cav 3.1 through a New Binding Mode.

Low-voltage-gated calcium channels (LVGCCs; Cav 3.1-3.3) represent promising drug targets for epilepsy, pain, and essential tremor. At present, modulators with heightened selectivity for a subtype of LVGCCs are still highly desired. In this study we explored three classes of Buxus alkaloids and identified 9(10/19)abeo-artanes Buxusemine H and Buxusemine L (BXSL) as an unprecedented type of Cav 3.2 inhibitors. Particularly, BXSL exhibited Cav 3.2 inhibition comparable to Z944, a non-subtype-selective LVGCCs inhibitor under clinical trial. While lacking specificity for Cav 3.3, BXSL showed a 30-

OBJECTIVE: To explore the mechanism of Danggui Buxue decoction (, DBD) for the treatment of gastric ulcer (GU), based on network pharmacology and in vivo experiments. METHODS: A network pharmacology strategy was used to predict the main components, candidate targets, and potential signaling pathways. Then, molecular docking was performed to further investigate the interactions and binding affinities between the main components and primary targets. Finally, a mouse model of ethanol-induced gastric ulcers was established to confirm the efficacy and potential therapeutic benefits of DBD, and can

Danggui Buxue decoction alleviates primary dysmenorrhea in rats by regulating the MEK1/2/ERK1/2/NF-κB pathway.

The study aimed to investigate the efficacy of Danggui Buxue Decoction (DBD) in treating primary dysmenorrhea (PD) and uncover its mechanisms, thereby validating its traditional use. Using ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF/MS), 38 compounds in DBD were identified. PD model rats were established via cold stimulation, estradiol benzoate, and oxytocin injections. DBD treatment improved behavioral parameters, reduced writhing response, and alleviated histopathological changes in uterus and ovary. Biochemical markers (PGE2, SOD and
