Interferon-β gene transfer induces a strong cytotoxic bystander effect on melanoma cells.

A local gene therapy scheme for the delivery of type I interferons could be an alternative for the treatment of melanoma. We evaluated the cytotoxic effects of interferon-β (IFNβ) gene lipofection on tumor cell lines derived from three human cutaneous and four canine mucosal melanomas. The cytotoxicity of human IFNβ gene lipofection resulted higher or equivalent to that of the corresponding addition of the recombinant protein (rhIFNβ) to human cells. IFNβ gene lipofection was not cytotoxic for only one canine melanoma cell line. When cultured as monolayers, three human and three canine IFNβ-li

Short-term and bystander effects of radiation on murine submandibular glands.

Many patients treated for head and neck cancers experience salivary gland hypofunction due to radiation damage. Understanding the mechanisms of cellular damage induced by radiation treatment is important in order to design methods of radioprotection. In addition, it is crucial to recognize the indirect effects of irradiation and the systemic responses that may alter saliva secretion. In this study, radiation was delivered to murine submandibular glands (SMGs) bilaterally, using a 137Cs gamma ray irradiator, or unilaterally, using a small-animal radiation research platform (SARRP). Analysis at

Genomic instability and bystander effects: a paradigm shift in radiation biology?

A basic paradigm in radiobiology is that, following exposure to ionizing radiation, the deposition of energy in the cell nucleus and the resulting damage to DNA, the principal target, are responsible for the radiation's deleterious biological effects. Findings in two rapidly expanding fields of research--radiation-induced genomic instability and bystander effects--have caused us to reevaluate these central tenets. In this article, the potential influence of induced genomic instability and bystander effects on cellular injury after exposure to low-level radiation will be reviewed.

Radiation-induced bystander effects in cultured human stem cells.

BACKGROUND: The radiation-induced "bystander effect" (RIBE) was shown to occur in a number of experimental systems both in vitro and in vivo as a result of exposure to ionizing radiation (IR). RIBE manifests itself by intercellular communication from irradiated cells to non-irradiated cells which may cause DNA damage and eventual death in these bystander cells. It is known that human stem cells (hSC) are ultimately involved in numerous crucial biological processes such as embryologic development; maintenance of normal homeostasis; aging; and aging-related pathologies such as cancerogenesis and

Bystander effects of ionizing radiation can be modulated by signaling amines.

Actual risk and risk management of exposure to ionizing radiation are among the most controversial areas in environmental health protection. Recent developments in radiobiology especially characterization of bystander effects have called into question established dogmas and are thought to cast doubt on the scientific basis of the risk assessment framework, leading to uncertainty for regulators and concern among affected populations. In this paper we test the hypothesis that small signaling molecules widely used throughout the animal kingdom for signaling stress or environmental change, such as

Bystander effect and abscopal effect in recurrent thymic carcinoma treated with carbon-ion radiation therapy: A case report.

BACKGROUND: Although the bystander effect and abscopal effect are familiar in medicine, they are relatively rare in clinical practice. Herein, we report the case of a patient who demonstrated an obvious bystander effect and abscopal effect response following carbon-ion irradiation for recurrent thymic carcinoma. CASE SUMMARY: A 44-year-old female presented with shortness of breath. Eleven years prior, she was diagnosed with athymic tumor located in the anterosuperior mediastinum. She underwent extensive tumor resection, and the postoperative pathologic diagnosis was thymic carcinoma. She was

The Effect of Moral Judgment on Bystander Cooperation Behavior: The Role of Personal Force.

BACKGROUND: While extensive research has examined the antecedents of utilitarian moral judgment, its subsequent social consequences remain less explored. Drawing on the moral reciprocal partner selection model and the moral intuition modular myopia hypothesis, this study investigates the impact of utilitarian moral judgment on bystander cooperation behavior and the moderating role of personal force. OBJECTIVES: This research aims to determine whether utilitarian moral judgments, compared to non-utilitarian ones, decrease bystander cooperation (Hypothesis 1), and whether this effect is more pr

Exosome-mediated modulation of radioresistance: The radiation-induced bystander effect in prostate cancer cells.

Exosomes are involved in intracellular communication and mediate the radiation-induced bystander effect (RIBE). We assessed the ability of exosomes to modify the radiation response of PC3 and DU145 prostate cancer cells exposed to X-rays. Irradiated cells were analyzed using clonogenic survival and apoptosis assays, while exosome-stimulated cells were evaluated for γH2AX immunostaining, immunoblotting, and apoptosis. Exosomes were isolated via size exclusion chromatography (SEC), characterized by nanoparticle tracking analysis (NTA) and immunoblotting, and ranged from 130 to 137 nm, containing

"Bystander effect" induced by photodynamically or heat-injured ovarian carcinoma cells (OVP10) in vitro.

BACKGROUND: The propagation of injury ("bystander effect") from directly damaged cells to other cells has been observed in cancer therapies. Some experiments suggested that this phenomenon was also detected in photodynamic therapy (PDT). The present study was undertaken to evaluate the bystander response in cells co-cultured with PDT- or heat-injured cells. MATERIAL/METHODS: Human ovary cancer cells (OVP10) were co-cultivated with PDT- and heat-damaged cells under various conditions. Fluorescence and light microscopy, shear test, clonogenic assays, and RT-PCR were used to estimate the vital f

Effect of Kinases in Extracellular Vesicles from HIV-1-Infected Cells on Bystander Cells.

As of 2023, there were 39.9 million people living with Human Immunodeficiency Virus type 1 (HIV-1). Although great strides have been made in treatment options for HIV-1, and our understanding of the HIV-1 life cycle has vastly improved since the start of this global health crisis, a functional cure remains elusive. One of the main barriers to a cure is latency, which allows the virus to persist despite combined antiretroviral therapy (cART). Recently, we have found that exosomes, which are small, membrane-enclosed particles released by virtually all cell types and known to mediate intercellula

Inter-species bystander effect: Eisenia fetida and Enchytraeus albidus exposed to uranium and cadmium.

The bystander effect is commonly defined as the observation of effects in nonirradiated cells and tissues when the later are in contact with irradiated cells/ tissues. More recently the occurrence of bystander effect between organisms of the same species has been also demonstrated. Nevertheless, there is limited knowledge about this effect between soil dwelling organisms from different taxonomic groups, as well as in response to stressors other than ionizing radiation. Moreover, data reporting this phenomenon for soil invertebrates are scarce. The results herein presented contribute for the un

An acute negative bystander effect of γ-irradiated recipients on transplanted hematopoietic stem cells.

Ultimate success of hematopoietic stem cell transplantation (HSCT) depends not only on donor HSCs themselves but also on the host environment. Total body irradiation is a component in various host conditioning regimens for HSCT. It is known that ionizing radiation exerts "bystander effects" on nontargeted cells and that HSCs transplanted into irradiated recipients undergo proliferative exhaustion. However, whether irradiated recipients pose a proliferation-independent bystander effect on transplanted HSCs is unclear. In this study, we found that irradiated mouse recipients significantly impair

Can bystander effects of metals in Daphnia magna be mediated by DNA methylation?

The bystander phenomenon can be described as the detection of effects in non-exposed cells/individuals when these come in contact with exposed ones. Bystander literature has focused mostly on the detection of effects rather than understanding the underlying mechanisms, thus these remain largely unknown from a molecular perspective. Epigenetic mechanisms play a major role in biological responses to environmental stressors, with limited evidence suggesting their involvement in the mechanisms behind bystander effects as well. To gain further insight on this specific topic, we performed bystander

Radiation-induced delayed bystander-type effects mediated by hemopoietic cells.

Genetic lesions and cell death associated with exposure to ionizing radiation have generally been attributed to DNA damage arising as a consequence of deposition of energy in the cell nucleus. However, reports of radiation-induced bystander effects, in which DNA damage is produced in nonirradiated cells as a consequence of communication with irradiated cells, indicate additional mechanisms. At present, most information has been obtained using in vitro systems, and the in vivo significance of bystander factors is not clear. In this study we show that signals generated in vivo in the bone marrow

Propolis reduced the radiation-induced bystander effect in rat liver exposed to total head irradiation.

In the realm of radiation oncology, the observation of the radiation-induced bystander effect (RIBE) has raised concerns about off-target tissue effects caused by radiation. The concept of RIBE encompasses the response of non-irradiated tissue to radiation exposure. This study investigated the impact of cranial irradiation on oxidative/nitrosative stress and inflammatory responses in liver tissue. Thirty-two male Sprague-Dawley rats were divided into four groups: sham, irradiation (IR), propolis plus IR, and propolis control. The IR group received an initial 1 ml oral dose of saline via an oro

Exosomal hnRNPA1-delivered miR-27a-3p inhibits the radiation-induced bystander effect in cervical cancer.

PURPOSE: The radiation-induced bystander effect (RIBE) refers to biological responses in non-irradiated cells exposed to signals from irradiated cells and may contribute to tumor control during radiotherapy. Periodic dose heterogeneity caused by the thread effect of helical tomotherapy (TOMO) may favor RIBE, but the underlying intercellular mechanisms remain unclear. This study investigated exosome-mediated bystander responses and their molecular and clinical relevance in cervical cancer. METHODS: Thread effect and uniform TOMO irradiation plans were established, and their effects on cervical

In silico decrypting of the bystander effect in antibody-drug conjugates for breast cancer therapy.

Antibody-Drug Conjugates (ADCs) are a promising cancer treatment that deliver toxic drugs directly to cancer cells, reducing harm to healthy tissue. A key feature of newer ADCs is the "bystander effect," in which nearby cancer cells are also affected by passive diffusion. However, the mechanisms underlying this effect remain unclear. Using computer simulations, this study investigates how the drug's ionization state and the linker connecting it to the antibody influence its ability to cross cell membranes. The results show that the ionization state of the drug impacts its membrane permeability

Distant Bystander Effect of REIC/DKK3 Gene Therapy Through Immune System Stimulation in Thoracic Malignancies.

BACKGROUND: Reduced expression in immortalized cell (REIC)/Dickkoph-3 (DKK3) is a tumor-suppressor gene, and its overexpression by adenovirus vector (Ad-REIC) exhibits a remarkable therapeutic effect on various human cancer types through a mechanism triggered by endoplasmic reticulum stress. MATERIALS AND METHODS: We examined the direct anti-tumor effect of Ad-REIC gene therapy on lung cancer and malignant mesothelioma cell lines in vitro, and the distant bystander effect using immunocompetent mouse allograft models with bilateral flank tumors. Tunneling nanotubes: an alternate route for propagation of the bystander effect following oncolytic viral infection.

Tunneling nanotubes (TNTs) are ultrafine, filamentous actin-based cytoplasmic extensions which form spontaneously to connect cells at short and long-range distances. We have previously described long-range intercellular communication via TNTs connecting mesothelioma cells in vitro and demonstrated TNTs in intact tumors from patients with mesothelioma. Here, we investigate the ability of TNTs to mediate a viral thymidine kinase based bystander effect after oncolytic viral infection and administration of the nucleoside analog ganciclovir. Using confocal microscopy we assessed the ability of TNTs
