Cadmium in fertilisers used for cacao production: a case study in Colombia.

The cacao crop is important for Colombia due to its high economic impact. One concern is the presence of cadmium (Cd) in beans, which can enter the soil from various sources, including fertilisers. However, to date, no studies have been carried out to identify Cd in these inputs. Therefore, this study seeks to determine Cd concentrations in (i) organic matter, (ii) chemical fertilisers and (iii) lime, together with the available fraction accessible to the cacao tree. A total of 133 fertilisers used in eight departments of Colombia were assessed. For 10 of these fertilisers a sequential extract

Biosynthesis, accumulation and degradation of theobromine in developing Theobroma cacao fruits.

We have studied the purine alkaloid content and purine metabolism in Theobroma cacao fruits at differing growth stages: Stage A (young small fruit, fresh weight, ca. 2 g); stage B (medium size fruit, fresh weight, ca. 100 g) and stage C (large size, fresh weight, ca. 500 g). The major purine alkaloid in stage A fruits (mainly pericarp) was theobromine (0.7 micromol g(-1) fresh weight), followed by caffeine (0.09 micromol g(-1) fresh weight). The theobromine content of the pericarp decreased sharply with tissue age, and the caffeine content decreased gradually. A large amount of theobromine (22

Cocoa Honey (Theobroma cacao L.): A Promising Byproduct for Industrial Applications.

Cocoa honey is a sweet, acidic liquid obtained from the pulp of cocoa fruits (Theobroma cacao L.) during postharvest processing, distinct from bee honey and derived from cocoa pulp, being specifically defined as the liquid obtained by natural drainage of the pulp. Rich in soluble solids, total sugars, and essential minerals like magnesium and zinc, while low in sodium, its characteristic fruity, sweet, and floral aroma is attributed to high levels of esters. Despite its technological potential, the industrial exploitation of cocoa honey is hindered by significant research gaps, including a lac

Association between bacterial community and cadmium distribution across Colombian cacao crops.

UNLABELLED: Assessing the bacterial community composition across cacao crops is important to understand its potential role as a modulator of cadmium (Cd) translocation to plant tissues under field conditions; Cd mobility between soil and plants is a complex and multifactorial problem that cannot be captured only by experimentation. Although microbes have been shown to metabolize and drive the speciation of Cd under controlled conditions, regardless of the link between soil bacterial community (SBC) dynamics and Cd mobilization in the rhizosphere, only a few studies have addressed the relations

Potential anti-alopecia constituents from Theobroma cacao: An in silico study.

Tinea capitis is local alopecia caused by a dermatophyte infection of the scalp. Trichophyton rubrum produces the squalene epoxidase enzyme, which has a crucial role in prolonged dermatophyte infection, as well as in synthesizing fatty acids in this dermatophyte group. This study analyzes Trichophyton cacao compounds as anti-alopecia by inhibiting the squalene epoxidase enzyme formation, in silico. The structure of T. cacao compounds was prepared using the MolView Web application. The compound docked to squalene epoxidase using AutoDock Vina in PyRx 0.8, followed by PyMOL for visualization, an

The Impact of Controlled Fermentation Temperature on Chemical Composition and Sensory Properties of Cacao.

The content of phenolic compounds can affect the quality of cacao beans (Theobroma cacao L). The variation in the concentration of these compounds is influenced by factors such as cacao variety, fermentation conditions, and temperature, which play a crucial role in the method of bean drying. In this study, the analytical method of ultra-performance liquid chromatography (UPLC-DAD-RI) was developed to identify, quantify, and examine variations in the concentrations of catechins (catechin, epicatechin, and epigallocatechin) and methylxanthines (theobromine and caffeine) by subjecting the beans t

Integrative multi-omics approach identified emerging viral threat in Theobroma cacao plants.

BACKGROUND: Theobroma cacao seeds are the source of chocolate and other valuable products. Pathogens can damage cocoa beans, reducing yield and quality. This study examined viruses linked to chlorosis and leaf deformation in cocoa plants in Bahia, Brazil, using an integrative approach. RESULTS: Macroscopic and microscopic analysis of symptomatic T. cacao leaves revealed chlorosis and histological alterations consistent with viral infection, including cell and nuclear hypertrophy. Deep sequencing of small RNA (sRNA) from these tissues identified virus-derived sRNA (vsiRNAs) profile, which sugg

Volatile organic compounds from Pichia kluyveri inhibit the cacao pathogen Moniliophthora roreri.

Moniliophthora roreri is a major fungal pathogen that significantly reduces cacao yields in Colombia. Currently, farmers rely solely on fungicides for its control, and environmentally friendly alternatives remain limited. The objective of this study was to assess the antagonistic potential of yeasts isolated from diverse niches in Colombia against M. roreri as a first step toward developing sustainable biocontrol strategies. Initially, the pathogen was identified at the molecular level. Subsequently, seven yeast strains belonging to the genera Wickerhamomyces, Naganishia, Pichia, and Metschnik

Exploring variations in quality parameters of Theobroma cacao L.beans from Eastern Amazonia.

The cocoa tree (Theobroma cacao L.) is a native crop of the tropical forests of Central America and the Amazon. It plays an important role in the economy of producing regions; however, the infrastructure remains incipient, leading to inadequate processing practices and affecting the quality of the cocoa beans. Therefore, this research aimed to evaluate the quality of 55 samples of Amazonian cocoa beans produced in five regions of state of Pará (Lower Tocantins River, Northeast, West, Southeast, and Trans-Amazon) through physical and physicochemical evaluations (proteins, lipids, moisture, ash,

Preparative separation of cacao bean procyanidins by high-speed counter-current chromatography.

In this work, an efficient method for preparative separation of procyanidins from raw cacao bean extract by high-speed counter-current chromatography (HSCCC) was developed. Under the optimized solvent system of n-hexane-ethyl acetate-water (1:50:50, v/v/v) with a combination of head-tail and tail-head elution modes, various procyanidins fractions with different polymerization degrees were successfully separated. UPLC, QTOF-MS and 1H NMR analysis verified that these fractions contained monomer up to pentamer respectively. Dimeric procyanidin B2 (purity>86%) could be isolated by HSCCC in a singl

Chemical composition and volatile profile of cocoa liquors from Colombian Criollo cacao.

Theobroma cacao is commonly classified as Criollo, Forastero, or Trinitario. In recent years, Criollo cacao has been revalorized due to its low volume production and the increase in the premium chocolate market. Besides, the origin and post-harvest processes affect the chemical and volatile composition of the cocoa beans that are transformed into liquor as raw material for chocolate production. Further, there is no published information regarding the chemical composition and volatile profile of Colombian Criollo cocoa liquor. Based on this, the present study aimed to characterize and different

Longitudinal genomic prediction of cacao disease resilience identifies robust witches' broom disease targets.

INTRODUCTION: Genomic studies in cacao (Theobroma cacao L.) have largely emphasized single-locus discovery, but less is known about whether repeated-harvest disease trajectories can be predicted from genome-wide markers to support long-term breeding objectives. METHODS: We analyzed repeated-harvest phenotypes from 102 cacao accessions to derive longitudinal targets describing baseline performance and temporal change for healthy pod rate, frosty pod rot (FPR), and witches' broom disease (WBD). Genomic prediction was evaluated using random and relatedness-blocked cross-validation with single nu

Pentameric procyanidin from Theobroma cacao selectively inhibits growth of human breast cancer cells.

A naturally occurring, cocoa-derived pentameric procyanidin (pentamer) was previously shown to cause G0/G1 cell cycle arrest in human breast cancer cells by an unknown molecular mechanism. Here, we show that pentamer selectively inhibits the proliferation of human breast cancer cells (MDA MB-231, MDA MB-436, MDA MB-468, SKBR-3, and MCF-7) and benzo(a)pyrene-immortalized 184A1N4 and 184B5 cells. In contrast, normal human mammary epithelial cells in primary culture and spontaneously immortalized MCF-10A cells were significantly resistant. We evaluated whether this differential response to pentam

Analysis of catechins in Theobroma cacao beans by cyclodextrin-modified micellar electrokinetic chromatography.

A micellar electrokinetic chromatography (MEKC) method was developed for the quantitation of polyphenols (+)-catechin and (-)-epicatechin (catechin monomers) and the methylxanthine theobromine in Theobroma cacao beans. Owing to the poor stability of catechin monomers in alkaline conditions, a 50 mM Britton-Robinson buffer at a pH 2.50 was preferred as the background electrolyte. Under these conditions, the addition of hydroxypropyl-beta-cyclodextrin (HP-beta-CD) at a concentration of 12 mM to the SDS micellar solution (90 mM), resulted in a cyclodextrin-modified micellar electrokinetic chromat

Inhibitory effects of aqueous extract of cacao bean husk on collagenase of Bacteroides gingivalis.

Natural materials containing tannin were examined to determine whether they possessed any inhibitory activity against collagenase obtained from the culture supernatant of Bacteroides gingivalis. The collagenolytic activity was determined with Collagenokit CLN-100 (Collagen Technological Co., Tokyo) using FITC-conjugated collagen type 1. Among the test samples, only an aqueous extract of cacao (Theobroma cacao) bean husk strongly inhibited the bacterial collagenase. This inhibitory potency was at the same level as that of tetracycline-HCl.

Routine soil tests support screening of high aluminium saturation in acidic cacao soils.

High aluminium saturation is a frequent but unevenly monitored constraint in acidic tropical soils, where routine soil testing is often more accessible than complete exchangeable-acidity determinations. This study developed and internally validated a screening model for high or very high aluminium saturation in cacao soil samples from the central Peruvian Amazon. Of 1842 anonymized monitoring records, 1539 acidic samples (pH < 5.5) formed the analytical cohort; the outcome was laboratory-reported aluminium saturation ≥ 20%. Six logistic-regression models compared routine soil tests with gridde

Combining ability, heritability and genotypic relations of different physiological traits in cacao hybrids.

Selecting parents and evaluating progenies is a very important step in breeding programs and involves approaches such as understanding the initial stages of growth and characterizing the variability among genotypes for different parameters, such as physiological, growth, biomass partitioning and nutrient translocation to the aerial part. In these cases, facilitating tools can be used to understand the involved gene dynamics, such as diallel crosses and genetic and phenotypic correlations. Our main hypothesis is that the contrasting phenotypes of these parental genotypes of cocoa used are due t

Enhanced somatic embryogenesis in Theobroma cacao using the homologous BABY BOOM transcription factor.

BACKGROUND: Theobroma cacao, the chocolate tree, is an important economic crop in East Africa, South East Asia, and South and Central America. Propagation of elite varieties has been achieved through somatic embryogenesis (SE) but low efficiencies and genotype dependence still presents a significant limitation for its propagation at commercial scales. Manipulation of transcription factors has been used to enhance the formation of SEs in several other plant species. This work describes the use of the transcription factor Baby Boom (BBM) to promote the transition of somatic cacao cells from the

Changes in bioactive compounds during fermentation of cocoa (Theobroma cacao) harvested in Amazonas-Peru.

Cocoa (Theobroma cacao) is the main raw material for the production of chocolate; it is considered the food of the gods, as it possesses a diversity of bioactive compounds beneficial to human health. The abundance of bioactive compounds, among others, is conditioned by the post-harvest processing of cocoa beans, and fermentation is a major step in this regard. Consequently, this research evaluated the changes in phenolic compounds and methylxanthines occurred in the fermentation of Criollo and CCN-51 cocoa beans, varieties of great commercial interest for the cocoa-growing areas of Peru.
