Cadherins regulate nuclear topography and function of developing ocular motor circuitry.

In the vertebrate central nervous system, groups of functionally related neurons, including cranial motor neurons of the brainstem, are frequently organised as nuclei. The molecular mechanisms governing the emergence of nuclear topography and circuit function are poorly understood. Here we investigate the role of cadherin-mediated adhesion in the development of zebrafish ocular motor (sub)nuclei. We find that developing ocular motor (sub)nuclei differentially express classical cadherins. Perturbing cadherin function in these neurons results in distinct defects in neuronal positioning, includin

Cadherins in vascular smooth muscle cell (patho)biology: Quid nos scimus?

Vascular smooth muscle cells (SMCs) phenotypes span a reversible continuum from quiescent/contractile (differentiated) to proliferative/synthetic (dedifferentiated) enabling them to perform a diversity of functions that are context-dependent and important for vascular tone-diameter homeostasis, vasculogenesis, angiogenesis or vessel reparation after injury. Dysregulated phenotype modulation and failure to maintain/regain the mature differentiated and contractile phenotypic state is pivotal in the development of vascular diseases such as atherosclerosis and restenosis after angioplasty and coro

Usher syndrome type 1-associated cadherins shape the photoreceptor outer segment.

Usher syndrome type 1 (USH1) causes combined hearing and sight defects, but how mutations in USH1 genes lead to retinal dystrophy in patients remains elusive. The USH1 protein complex is associated with calyceal processes, which are microvilli of unknown function surrounding the base of the photoreceptor outer segment. We show that in Xenopus tropicalis, these processes are connected to the outer-segment membrane by links composed of protocadherin-15 (USH1F protein). Protocadherin-15 deficiency, obtained by a knockdown approach, leads to impaired photoreceptor function and abnormally shaped ph

Interplay between cadherins and α2β1 integrin differentially regulates melanoma cell invasion.

BACKGROUND: Malignant transformation of melanocytes frequently coincides with an alteration in the expression of cell-cell adhesion molecules (cadherins) and cell-extracellular matrix proteins (integrins). How these two adhesion systems interplay to impact on cell invasion remains to be described in melanoma. METHODS: Cell adhesion networks were localised by immunofluorescence in human primary cutaneous melanoma, metastatic melanoma in the lymph nodes, and melanoma cell lines. The role of these cell adhesion networks was assessed both in vivo, by analysing their impact on tumour growth in mic

Type II cadherins guide assembly of a direction-selective retinal circuit.

Complex retinal circuits process visual information and deliver it to the brain. Few molecular determinants of synaptic specificity in this system are known. Using genetic and optogenetic methods, we identified two types of bipolar interneurons that convey visual input from photoreceptors to a circuit that computes the direction in which objects are moving. We then sought recognition molecules that promote selective connections of these cells with previously characterized components of the circuit. We found that the type II cadherins, cdh8 and cdh9, are each expressed selectively by one of the

Cadherins: an integral role in inflammatory bowel disease and mucosal restitution.

The intestinal epithelium is characterized by rapid cellular turnover with continuous proliferation of multipotential stem cells within Lieberkuhn's crypts, cellular migration along the crypt-villus axis, cellular differentiation, polarization, apical apoptosis, and luminal shedding. These processes are critical for the development and maintenance of normal intestinal epithelial architecture and function and involve complex cell-cell and cell-substratum interactions, which are mediated by epithelial (E)-cadherin and the integrins, respectively. This review outlines the role of E-cadherin and i

Cadherins and cadherin-associated molecules in the developing and maturing rat testis.

The calcium-dependent class of cell adhesion molecules known as cadherins mediate homotypic cell interactions in most epithelia. We have now investigated the expression and distribution of cadherins and cadherin-associated molecules in the developing and maturing rat testis. E-Cadherin was not detected in the seminiferous tubule at any time in development or in the adult. In contrast, Leydig cells expressed E-cadherin between day 15 of gestation and postnatal day 3. alpha- and beta-catenins were expressed throughout the developing testis, but were particularly prominent in Leydig cells. In the

Recent progress in T-cadherin (CDH13, H-cadherin) research.

T-cadherin is a unique cadherin cell adhesion molecule that is anchored to the cell surface membrane through a glycosyl phosphatidyl inositol (GPI) moiety. The cytoplasmic domain, which T-cadherin lacks, is believed to be critical for homophilic binding through interaction with submembrane cytoskeletal proteins. Does this mean that T-cadherin is an unimportant molecule? However, the T-cadherin amino acid motif has been well conserved through evolution in vertebrates, suggesting that T-cadherin may have biological significance in higher animals. Consistent with this hypothesis, recent studies h

Auditory cortex interneuron development requires cadherins operating hair-cell mechanoelectrical transduction.

Many genetic forms of congenital deafness affect the sound reception antenna of cochlear sensory cells, the hair bundle. The resulting sensory deprivation jeopardizes auditory cortex (AC) maturation. Early prosthetic intervention should revive this process. Nevertheless, this view assumes that no intrinsic AC deficits coexist with the cochlear ones, a possibility as yet unexplored. We show here that many GABAergic interneurons, from their generation in the medial ganglionic eminence up to their settlement in the AC, express two cadherin-related (cdhr) proteins, cdhr23 and cdhr15, that form the

Cadherins and the formation of neural circuitry in the vertebrate CNS.

Cadherins are a family of calcium-dependent morphoregulatory molecules mediating cell-cell adhesion. More than a dozen cadherin subtypes are known to be expressed in the developing and mature CNS. Each of these subtypes shows a restricted and distinct expression pattern that differs from that of the other cadherins. During the formation of fiber tracts and neural circuits, each cadherin is expressed by a subset of neurite fascicles. The differential expression of cadherins provides a molecular code for the high degree of specificity and selectivity in neural circuit formation. This code may be

Dlg5 and Cadherins Cooperate to Ensure Peripheral Glia and Septate Junction Integrity.

The formation of the glial sheath is essential in nervous system development to insulate and protect peripheral nerves and axons, yet the regulation of adhesion junctions in nonmyelinating glia has not been clearly established. Many components of adhering junctions contain PDZ domains or are recruited by PDZ binding motifs. To identify PDZ domain proteins with roles in peripheral glia, we carried out an RNAi screen using Drosophila melanogaster to knockdown each of the 66 predicted PDZ proteins in larvae of either sex. We identified six PDZ genes with potential roles in glial morphology, and w

Mining the O-mannose glycoproteome reveals cadherins as major O-mannosylated glycoproteins.

The metazoan O-mannose (O-Man) glycoproteome is largely unknown. It has been shown that up to 30% of brain O-glycans are of the O-Man type, but essentially only alpha-dystroglycan (α-DG) of the dystrophin-glycoprotein complex is well characterized as an O-Man glycoprotein. Defects in O-Man glycosylation underlie congenital muscular dystrophies and considerable efforts have been devoted to explore this O-glycoproteome without much success. Here, we used our SimpleCell strategy using nuclease-mediated gene editing of a human cell line (MDA-MB-231) to reduce the structural heterogeneity of O-Man

Changing distribution of cadherins during gestation in the uterine epithelium of lizards.

The uterine epithelium provides the interface between an embryo and its mother during pregnancy. Calcium-dependent cadherins are adherens junction proteins that undergo major shifts in the uterine epithelium to facilitate the communication between maternal cells and the embryonic milieu during implantation in mammals. They are, therefore, important in trophoblast invasion and the maintenance of pregnancy. We investigated spatiotemporal changes of cadherins throughout pregnancy in the uterine epithelium of two viviparous skinks and one oviparous population, which all exhibit a noninvasive (epit

Cadherins in cerebellar development: translation of embryonic patterning into mature functional compartmentalization.

Cadherins are cell adhesion molecules with multiple morphogenic functions in brain development, for example, in neuroblast migration and aggregation, axon navigation, neural circuit formation, and synaptogenesis. More than 100 members of the cadherin superfamily are expressed in the developing and mature brain. Most of the cadherins investigated, in particular classic cadherins and δ-protocadherins, are expressed in the cerebellum. For several cadherin subtypes, expression begins at early embryonic stages and persists until mature stages of cerebellar development. At intermediate stages, disti

Cadherins as regulators for the emergence of neural nets from embryonic divisions.

Cadherins are a large family of cell adhesion molecules that are expressed in a spatially restricted fashion during vertebrate CNS development. Each cadherin shows a characteristic expression pattern that differs from that of other cadherins. Early in development, the cadherin expression domains relate to the neuromeric organization of the vertebrate CNS. Later, as functional structures (brain nuclei, cortical regions, fiber tracts and synapses) emerge, the expression patterns of each cadherin become restricted to subsets of these structures that form parts of specific neural nets. Cadherins t

Modulation of E-Cadherin and N-Cadherin by ovarian steroids and embryonic stimuli.

Endometrium is a dynamic tissue that undergoes extensive remodelling to attain a receptive state which is further modulated in presence of an embryo for successful initiation of pregnancy. Cadherins are the proteins of the junctional complex of which E-cadherin (E-Cad) is crucial for maintaining epithelial cell state and integrity of the epithelial barrier; gain of N-cadherin (N-Cad) in epithelial cells leads to epithelial to mesenchymal transition (EMT). In the present study, we investigated the expression of E-Cad and N-Cad in the mouse endometrial luminal epithelium and its modulation by es

BILL-cadherin/cadherin-17 contributes to the survival of memory B cells.

Memory B cells (MBCs) and long-lived plasma cells (LLPCs) are responsible for immunological "memory", which can last for many years. The long-term survival niche for LLPCs in the bone marrow is well characterized; however, the corresponding niche for MBCs is unclear. BILL-cadherin/cadherin-17 (CDH17) is the only member of the cadherin superfamily that is expressed on mouse B lymphocytes in a spatiotemporally regulated manner. Here, we show that half of all MBCs regain expression of CDH17 during the later stage of development. The maintenance of high affinity antigen-specific serum antibodies w

Reciprocal altered expression of T-cadherin and P-cadherin in psoriasis vulgaris.

BACKGROUND: The most characteristic change in psoriasis vulgaris is markedly increased, persistent keratinocyte proliferation. The underlying mechanism of excessive epidermal growth is controversial. We previously found and reported that T-cadherin was expressed in keratinocytes and confined to the basal layer of mouse and human skin. Invasive cutaneous squamous cell carcinoma showed a loss of T-cadherin expression. Another study showed that T-cadherin was a negative growth regulator of epidermal growth factor in T-cadherin transfectant neuroblastoma cells. OBJECTIVES: To obtain insight into
