TGF-β/SMAD Pathway Mediates Cadmium Poisoning-Induced Chicken Liver Fibrosis and Epithelial-Mesenchymal Transition.

Cadmium(Cd) is a toxic heavy metal widely present in the environment, capable of accumulating in the liver and causing liver damage. In this study, the mechanism of cadmium-induced liver fibrosis in chickens was investigated from the perspective of hepatocyte epithelial-mesenchymal transition (EMT) based on the establishment of a model of chicken cadmium toxicity and a model of cadmium-stained cells in a chicken hepatocellular carcinoma cell line (LMH). The 7-day-old chickens were randomly divided into the regular group (C group) and cadmium poisoning group (Cd group), and the entire test cycl

Experimental reproduction of itai-itai disease, a chronic cadmium poisoning of humans, in rats and monkeys.

To establish a useful animal model of Itai-Itai disease (IID) of humans, we conducted the following experiments. Experiment 1: Toxic effects of Cd were compared between ovariectomized (OX) and non-OX rats after daily, intravenous injection of cadmium (Cd) chloride for 14 days. In this experiment, we demonstrated that OX rats were more susceptible to Cd-induced nephrotoxicity and hepatotoxicity than non-OX rats. Experiment 2: OX rats were injected with Cd at doses of 1.0 and 2.0 mg/kg, 5 days a week, for 13 weeks. The bone Cd content was gradually increased for 13 weeks in a dose-dependent mann

Detoxification effect of sodium thiosulfate on cadmium poisoning in Litopenaeus vannamei and the mechanisms of intestinal flora regulation.

Cadmium (Cd) is currently one of the heavy metals with the highest environmental toxicity impact. Sodium thiosulfate (Na2S2O3) is a commonly used heavy metal detoxification drug in clinical practice, however, it has not been used for Cd detoxification of Litopenaeus vannamei. The present study used exposure of L. vannamei to 150 μg/L of Cd while mitigating in the addition of 75 μg/L of Na2S2O3 for 28 days. The goal was to study the detoxifying effect of Na2S2O3 on L. vannamei poisoning and its role in intestinal flora. The results showed that the growth of Cd group was inhibited, and the growt

Extracellular vesicles from human bone marrow mesenchymal stem cells repair organ damage caused by cadmium poisoning in a medaka model.

Treatment modalities for kidney disease caused by long-term exposure to heavy metals, such as cadmium (Cd), are limited. Often, chronic, long-term environmental exposure to heavy metal is not recognized in the early stages; therefore, chelation therapy is not an effective option. Extracellular vesicles (EVs) derived from stem cells have been demonstrated to reduce disease pathology in both acute and chronic kidney disease models. To test the ability of EVs derived from human bone marrow mesenchymal stem cells (hBM-MSCs) to treat Cd damage, we generated a Cd-exposed medaka model. This model dev

Effect of Tea Polyphenols on Nuclear Factor Erythroid 2-related Factor 2 (NRF2) and Kelch-like ECH-associated Protein 1 (KEAP1) Gene Expression in Mice with Acute Cadmium Poisoning.

BACKGROUND: Cadmium poisoning is mainly caused by inhalation of cadmium dust or cadmium compound dust, which greatly harms people's lives. Tea polyphenols extracted from green tea have wide biological properties, including anti-cardiovascular disease, anti-tumor, anti-inflammatory, and immune regulation. The transcription factor nuclear factor erythroid 2-related factor 2 (NRF2) and Kelch-like ECH-associated protein 1 (KEAP1) are involved in the regulation of cadmium-induced oxidative damage. However, whether tea polyphenols relieve acute cadmium poisoning via regulating NRF2 and KEAP1 gene ex

[Recent research progress on natural medicines in treatment of cadmium toxicity].

Cadmium contamination of environment is a subject of serious international concern. Bioaccumulation of cadmium occurs primarily through ingestion of contaminated water and food. Cadmium poisoning came into prominence with the "itai-itai" disease event in Japan in the 1950s. It could also cause damages to liver, kidney, lung and other organs. Thus, the treatment of cadmium poisoning has become a research hotspot. Researchers are trying their best to explore prophylactic and therapeutic medicines for prevention and treatment of cadmium-induced poisoning. So far, chelation therapy, the convention

The importance of cadmium as a factor in calcified upper urinary tract stone disease--a prospective 7-year study.

The non-essential trace metal cadmium may cause major pathological effects on the kidneys. A group of coppersmiths who have been chronically exposed to cadmium poisoning was systematically studied over a 6-year period. The stone prevalence in the group is now just under 40% as compared with 3.5% in a general population. The blood cadmium levels have remained persistently elevated despite removal of the cadmium source. The measurement of cadmium as a means of monitoring chronic poisoning has been undertaken in terms of the prospective study in the coppersmiths and also in its relationship to ot

Chelation of cadmium with BAL and DTPA in rats.

Cadmium is unique among non-essential metals in several of its toxicological effects because of its long biological half life, slow excretion and delayed action on the kidneys. Although cadmium poisoning in humans is uncommon, there are reports of chronic cadmium poisoning in workmen and also in others in certain polluted areas. At present there are no suitable methods either to measure the body burden of cadmium or to treat cadmium poisoning. The therapeutic effects of various chelating agents including 2,3-dimercaptopropanol (BAL) and its soluble glycosides have been studied without much suc

Health effects of cadmium with special reference to studies in Japan.

This paper gives an overview of the health effects of cadmium with special emphasis on the natural history of cadmium poisoning. The following questions are discussed: (1) Do the renal changes caused by cadmium result in both glomerular and tubular dysfunction? (2) What is the fate of these renal changes? (3) What is the relationship between renal changes and osteomalacia? (4) Is there a dose-response relationship between cadmium exposure and renal dysfunction? (5) What is the solution to the enigma of cadmium and respiratory cancer? Although innumerable papers have been published, the above q

The Recent Research Progress On Cadmium-Induced Kidney Injury.

Kidney diseases are a group of disorders that cause kidney damage, characterized by renal dysfunction, pathological changes in renal tissue, and abnormal components in blood or urine. A plethora of factors have been identified as causes of renal diseases, including cadmium exposure. Cadmium is an environmental pollutant, and excessive intake of cadmium in the human body can result in cadmium poisoning. Accumulating evidence has demonstrated that cadmium exposure is a contributing factor to the development of kidney diseases. The associated symptoms include damage to the renal tubules and glome

Cadmium toxicity and treatment: An update.

Cadmium poisoning has been reported from many parts of the world. It is one of the global health problems that affect many organs and in some cases it can cause deaths annually. Long-term exposure to cadmium through air, water, soil, and food leads to cancer and organ system toxicity such as skeletal, urinary, reproductive, cardiovascular, central and peripheral nervous, and respiratory systems. Cadmium levels can be measured in the blood, urine, hair, nail and saliva samples. Patients with cadmium toxicity need gastrointestinal tract irrigation, supportive care, and chemical decontamination t

[Behavior of selected indicators of lipid metabolism in kidney and liver of rats continuously exposed to effects of cadmium].

In this research, we attempted to demonstrate whether and to what extent chronic cadmium poisoning influences lipid metabolism in the kidney and liver of rats. The presence of small quantities of cadmium was discovered in the kidneys and livers of normal animals. In the tested organs of cadmium-poisoned rats, a significant increase of the concentration of this element was observed, depending on the dose of the compound administered. Regarding lipid components, a reduced concentration of phospholipids and an increased level of free fatty acids were found in both kidney and liver. Such changes a
